| 1 | TNF-α suppresses sweat gland differentiation of MSCs by reducing FTO-mediated m^6 A-demethylation of Nanog mRNA显示文摘An effect of inhibition of tumor necrosis factor-α(TNF-α)on differentiation of mesenchymal stromal cells(MSCs)has been demonstrated,but the exact mechanisms that govern MSCs differentiation remain to be further elucidated.Here,we show that TNF-αinhibits the differentiation of MSCs to sweat glands in a specific sweat gland-inducing environment,accompanied with reduced expression of Nanog,a core pluripotency factor.We elucidated that fat mass and obesity-associated protein(FTO)-mediated m^6 A demethylation is involved in the regulation of MSCs differentiation potential.Exposure of MSCs to TNF-αreduced expression of FTO,which demethylated Nanog m RNA.Reduced expression of FTO increased Nanog m RNA methylation,decreased Nanog m RNA and protein expression,and significantly inhibited MSCs capacity for differentiation to sweat gland cells.Our finding is the first to elucidate the functional importance of m^6 A modification in MSCs,providing new insights that the microenvironment can regulate the multipotency of MSCs at the post-transcriptional level.Moreover,to maintain differentiation capacity of MSCs by regulating m^6 A modification suggested a novel potential therapeutic target for stem cellmediated regenerative medicine. | Yihui Wang Rui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu | 2020 | Science China(Life Sciences)2020,63,1: | 9 |
| 2 | Advances and development trends in eco-friendly pavements显示文摘The major contemporary in road pavement engineering is related to the creation of green and sustainable infrastructures,e.g.,reduction of environmental impacts,increase in traffic safety,and transportation efficiency,etc.This review presents the recent trends in research and the technical solutions developed so far to address these challenges.After the analysis of research status in the past decades,a novel technology system of eco-friendly pavements is proposed considering two solutions,materials modification and structure improvement.The construction of an eco-friendly pavement can be achieved thanks to several different technologies ensuring permeable,noise-reducing,self-luminous,and exhaust-decomposing properties as well as apporting lower heat absorbing and enhanced anti-/de-icing characteristics.A systematic review of these technologies is presented pivoting on four main aspects:technical principle,material and structural composition,performance evaluation,and engineering application.The current trend in road engineering is combining the pavement infrastructure with various eco-friendly functions,e.g.,water permeability,noise reduction,low heat absorption,exhaust gas decomposition,and anti-/de-icing.Finally,the review lists the drawbacks of the existing technologies,including high cost,single function,etc.,and depicts the future developing direction and architecture of the next generation of eco-friendly pavements in which the road infrastructure should have more environmentally friendly functions than the existing technology. | Aimin Sha Zhuangzhuang Liu Wei Jiang Lin Qi Liqun Hu Wenxiu Jiao Diego Maria Barbieri | 2021 | Journal of Road Engineering2021,1,1: | 9 |

| 5 | Comparison of extended spectrum β-lactamasesproducing Escherichia coli with non-ESBLsproducing E.coli:drug-resistance and virulence显示文摘BACKGROUND:The virulent factors of Escherichia coli(E.coli) play an important role in the process of pathopoiesis.The study aimed to compare drug-resistant genes and virulence genes between extended spectrum β-lactamases(ESBLs)-producing E.coli and non-ESBLs-producing E.coli to provide a reference for physicians in management of hospital infection.METHODS:From October 2010 to August 2011,96 drug-resistant strains of E.coli isolated were collected from the specimens in Qingdao Municipal Hospital,Qingdao,China.These bacteria strains were divided into a ESBLs-producing group and a non-ESBLs-producing group.Drug sensitivity tests were performed using the Kirby-Bauer(K-B) method.Disinfectant gene,qacEA1-sull and 8 virulence genes(CNF2,hlyA,eaeA,VT1,est,bfpA,elt,and CNF1) were tested by polymerase chain reaction(PCR).RESULTS:Among the 96 E.coli isolates,the ESBLs-producing E.coli comprised 46(47.9%)strains and the non-ESBLs-producing E.coli consisted of 50(52.1%) strains.The detection rates of multiple drug-resistant strain,qacEA1-sull,CNF2,hlyA,eaeA,VT1,est,bfpA,elt,and CNF1 in 46ESBLs-producing E.coli isolates were 89.1%,76.1%,6.5%,69.6%,69.6%,89.1%,10.9%,26.1%,8.7%,and 19.6%,respectively.In the non-ESBLs-producing E.coli strains,the positive rates of multiple drug-resistant strain,qacEA1-sull,CNF2,hlyA,eaeA,VT1,est,bfpA,elt,and CNF1 were 62.0%,80.0%,16.0%,28.0%,64.0%,38.0%,6.0%,34.0%,10.0%,and 24.0%,respectively.The difference in the detection rates of multiple drug-resistant strain,hlyA and VT1 between the ESBLs-producing E.coli strains and the non-ESBLs-producing E.coli strains was statistically significant(P<0.05).CONCLUSION:The positive rate of multiple drug-resistant strains is higher in the ESBLsproducing strains than in the non-ESBLs-producing strains.The expression of some virulence genes hlyA and VT1 varies between the ESBLs-producing strains and the non-ESBLs-producing strains.Increased awareness of clinicians and enhanced testing by laboratories are required to reduce treatment failures and prevent the spread of multiple drug-resistant strains. | Sha Li Yan Qu Dan Hu Yong-xin Shi | 2012 | World Journal of Emergency Medicine2012,3,3: | 7 |

| 8 | 6-O-angeloylplenolin exerts neuroprotection against lipopolysaccharide-induced neuroinflammation in vitro and in vivo显示文摘Neuroinflammation is one of the critical events in neurodegenerative diseases,whereas microglia play an important role in the pathogenesis of neuroinflammation.In this study,we investigated the effects of a natural sesquiterpene lactone,6-O-angeloylplenolin(6-OAP),isolated from the traditional Chinese medicine Centipeda minima(L.)A.Br.,on neuroinflammation and the underlying mechanisms.We showed that treatment with lipopolysaccharide(LPS)caused activation of BV2 and primary microglial cells and development of neuroinflammation in vitro,evidenced by increased production of inflammatory cytokines TNF-αand IL-1β,the phosphorylation and nuclear translocation of NF-κB,and the transcriptional upregulation of COX-2 and iNOS,leading to increased production of proinflammatory factors NO and PGE2.Moreover,LPS treatment induced oxidative stress through increasing the expression levels of NOX2 and NOX4.Pretreatment with 6-OAP(0.5−4μM)dose-dependently attenuated LPS-induced NF-κB activation and oxidative stress,thus suppressed neuroinflammation in the cells.In a mouse model of LPS-induced neuroinflammation,6-OAP(5−20 mg·kg^−1·d^−1,ip,for 7 days before LPS injection)dose-dependently inhibited the production of inflammatory cytokines,the activation of the NF-κB signaling pathway,and the expression of inflammatory enzymes in brain tissues.6-OAP pretreatment significantly ameliorated the activation of microglia and astrocytes in the brains.6-OAP at a high dose caused a much stronger antineuroinflammatory effect than dexamethansone(DEX).Furthermore,we demonstrated that 6-OAP pretreatment could inhibit LPS-induced neurite and synaptic loss in vitro and in vivo.In conclusion,our results demonstrate that 6-OAP exerts antineuroinflammatory effects and can be considered a novel drug candidate for the treatment of neuroinflammatory diseases. | Yi-le Zhou Yong-ming Yan Si-yi Li Dan-hua He Sha Xiong Su-fen Wei Wei Liu Ling Hu Qi Wang Hua-feng Pan Yong-xian Cheng Yong-qiang Liu | 2020 | Acta Pharmacologica Sinica2020,41,1: | 5 |
| 9 | Redirecting differentiation of mammary progenitor cells by 3D bioprinted sweat gland microenvironment显示文摘Background:Mammary progenitor cells(MPCs)maintain their reproductive potency through life,and their specific microenvironments exert a deterministic control over these cells.MPCs provides one kind of ideal tools for studying engineered microenvironmental influence because of its accessibility and continually undergoes postnatal developmental changes.The aim of our study is to explore the critical role of the engineered sweat gland(SG)microenvironment in reprogramming MPCs into functional SG cells.Methods:We have utilized a three-dimensional(3D)SG microenvironment composed of gelatin-alginate hydrogels and components from mouse SG extracellular matrix(SG-ECM)proteins to reroute the differentiation of MPCs to study the functions of this microenvironment.MPCs were encapsulated into the artificial SG microenvironment and were printed into a 3D cell-laden construct.The expression of specific markers at the protein and gene levels was detected after cultured 14 days.Results:Compared with the control group,immunofluorescence and gene expression assay demonstrated that MPCs encapsulated in the bioprinted 3D-SG microenvironment could significantly express the functional marker of mouse SG,sodium/potassium channel protein ATP1a1,and tend to express the specific marker of luminal epithelial cells,keratin-8.When the Shh pathway is inhibited,the expression of SG-associated proteins in MPCs under the same induction environment is significantly reduced.Conclusions:Our evidence proved the ability of differentiated mouse MPCs to regenerate SG cells by engineered SG microenvironment in vitro and Shh pathway was found to be correlated with the changes in the differentiation.These results provide insights into regeneration of damaged SG by MPCs and the role of the engineered microenvironment in reprogramming cell fate. | Rui Wang Yihui Wang Bin Yao Tian Hu Zhao Li Yufan Liu Xiaoli Cui Liuhanghang Cheng Wei Song Sha Huang Xiaobing Fu | 2019 | Burns & Trauma2019,7,1: | 5 |

| 12 | A Teosinte-derived Allele of a MYB Transcription Repressor Confers Multiple Disease Resistance in Maize.显示文摘Natural alleles controlling multiple disease resistances (MDR) are valuable for crop breeding. However, only one MDR gene have been cloned in maize, and molecular mechanisms of MDR are not clear. By map-based cloning, we have cloned a teosinte-derived allele of a resistance gene, Mexicana lesion mimic 1 (ZmMM1), which has a lesion mimic phenotype and confers resistance to northern leaf blight (NLB), gray leaf spot (GLS) and southern corn rust (SCR). Strong MDR conferred by the teosinte allele is linked with the polymorphisms in the 3' untranslated region of the ZmMM1 gene that cause increased accumulation of ZmMM1 protein. ZmMM1 acts as a transcription repressor and negatively regulates transcription of specific target genes including ZmMM1-target gene 3 (ZmMT3), which functions as a negative regulator of plant immunity and associated cell death. | 13 | Highway constructions on the Qinghai-Tibet Plateau:Challenge,research and practice显示文摘Highway constructions on the Qinghai-Tibet Plateau(QTP)face great challenges induced by the unique local environmental,geological,and engineering conditions.The large area of permafrost,great temperature variability,strong UV rays,and complex geological conditions are the major factors that adversely influence the longterm performance of pavement systems.Since 1960s,Chinese engineers and researchers have started conducting research on the QTP to enhance the performance and durability of pavement systems.The present paper provide a comprehensive review of challenge,research and practice of highway constructions on the QTP including the special environmental and geological conditions,history of highway constructions on the QTP,major challenges and the state-of-the-art technology of subgrade constructions on permafrost,developments of the pavement structures and materials,performance prediction and maintenance methods of pavement surfaces,and applications of the research achievements on the first expressway on the QTP(i.e.,Gongyu Expressway).Based on the comprehensive literature review,it can be found that(1)frost heave and thaw weakening induced subgrade disease and longitudinal cracks on the pavement surface are complex coupled water-thermal-load problems.Engineering solutions are focusing on active cooling and thermal insulation methods,which can help to reduce temperature variations in the subgrade and thus improving its stability,(2)the harsh environmental and construction conditions may reduce the early strength and induce premature damage of cement-treated base materials.Some field validations showed that geocell-reinforced or asphalt-treated flexible base materials can provide better long-term performance,(3)the large temperature variability and strong UV rays can significantly accelerate aging of asphalt binders and greatly reduce the service life of asphalt mixtures.Various binder modification methods were developed for improving their viscoelasticity and enhance the low-temperature cracking resistance of pavement surface materials but are still lack of field validation data and comparisons of their life cycle costs.Therefore,it is recommended that a demonstration research project build test sections to examine a range of pavement structures and materials,and compare their long-term performance and life cycle costs,which can serve as important reference for future highway constructions on the QTP. | Aimin Sha Biao Ma Hainian Wang Liqun Hu Xuesong Mao Xilan Zhi Huaxin Chen Yu Liu Feng Ma Zhuangzhuang Liu Rui He Wei Si Xuhao Wang Cheng Li | 2022 | Journal of Road Engineering2022,2,1: | 4 |

To further investigate the mechanisms on the effect of E2 on NK cells, expressions of activation-associated markers (CD69, CD122) and inhibitory receptors (CD94, Ly49), and intracellular cytokine production were analyzed. At last, we performed the cDNA microarray to explore the possible involved genes. We found that E2 could suppress NK cell cytotoxicity and proliferative capacity in vitro. E2 reduced NK cell cytotoxicity and proliferative capacity, which may be through influencing the phenotypes and cytokine expression of NK cells, mainly involving CD94 and IFN-γ. | 20 | Frequency Loss and Recovery in Rolling Bearing Fault Detection显示文摘Rolling element bearings are key components of mechanical equipment. The bearing fault characteristics are a ected by the interaction in the vibration signals. The low harmonics of the bearing characteristic frequencies cannot be usually observed in the Fourier spectrum. The frequency loss in the bearing vibration signal is presented through two independent experiments in this paper. The existence of frequency loss phenomenon in the low frequencies, side band frequencies and resonant frequencies and revealed. It is demonstrated that the lost frequencies are actually suppressed by the internal action in the bearing fault signal rather than the external interference. The amplitude and distribution of the spectrum are changed due to the interaction of the bearing fault signal. The interaction mechanism of bearing fault signal is revealed through theoretical and practical analysis. Based on mathematical morphology, a new method is provided to recover the lost frequencies. The multi-resonant response signal of the defective bearing are decomposed into low frequency and high frequency response, and the lost frequencies are recovered by the combination morphological filter(CMF).
