18地氟烷和七氟烷预处理信号链中活性氧生成和线粒体解偶联的差异显示文摘背景麻醉药预处理的信号传导链已被广泛研究,但是许多问题仍然有待解决。本研究拟评价两种现代麻醉药地氟烷和七氰烷进行心肌细胞预处理过程中,活性氧(ROS)和线粒体解偶联的作用。方法酶解分离成年大鼠心室肌细胞,利用细胞存活实验评估地氟烷和七氟垸预处理保护心肌细胞免受氧化应激诱导的细胞死亡的效力。应用共聚焦显微镜实时监测ROS生成和黄素蛋白荧光。黄素蛋白荧光是黄素蛋白氧化和线粒体解偶联的标志。应用ROS清除剂Trolox通过细胞生存和共聚焦实验评估麻醉药导致ROS生成增加的作用。结果地氟烷或七氟烷参与的心肌细胞预处理明显减少氧化应激诱导的细胞死亡。这种效应与心肌细胞急性接触麻醉药过程中增加ROS生成和黄素蛋白氧化相一致。地氟烷诱导较七氟垸明显提高ROS生成和黄素蛋白氧化。ROS清除剂Trolox取消麻醉药的预处理效力,减弱黄素蛋白氧化。结论地氟烷和七氟烷预处理保护大鼠分离心肌细胞免受氧化应激诱导的细胞死亡。ROS清除剂废除两种麻醉药的预处理作用,减弱麻醉药诱导的线粒体解偶联,这表明ROS在麻醉药诱导预处理中起关键作用,提示ROS是线粒体解偶联的上游信号。地氟烷与七氟烷相比,刺激ROS生成和线粒体解偶联的作用更强。Filip Sedlic, MD Danijel Pravdic, MD Marko Ljubkovic, MD, PhD Jasna Marinovic, MD Anna Stadnicka, PhD Zeljko J.
