| 1 | Safety of endovascular therapy for symptomatic intracranial artery stenosis:a national prospective registry显示文摘Introduction The safety outcomes of endovascular therapy for intracranial artery stenosis in a real-world stetting are largely unknown.The Clinical Registration Trial of Intracranial Stenting for Patients with Symptomatic Intracranial Artery Stenosis(CRTICAS)was a prospective,multicentre,real-world registry designed to assess these outcomes and the impact of centre experience.Methods 1140 severe,symptomatic intracranial arterial stenosis(ICAS)patients treated with endovascular therapy were included from 26 centres,further divided into three groups according to the annual centre volume of intracranial angioplasty and stent placement procedures over 2 years:(1)high volume for≥25 cases/year;(2)moderate volume for 10–25 cases/year and(3)low volume for<10 cases/year.Results The rate of 30-day stroke,transient ischaemic attack or death was 9.7%(111),with 5.4%,21.1%and 9.7%in high-volume,moderate-volume and low-volume centres,respectively(p<0.05).Multivariable logistic regression confirmed high-volume centres had a significantly lower primary endpoint compared with moderate-volume centres(OR=0.187,95%CI:0.056 to 0.627;p≤0.0001),while moderate-volume and low-volume centres showed no significant difference(p=0.8456).Conclusion Compared with the preceding randomised controlled trials,this real-world,prospective,multicentre registry shows a lower complication rate of endovascular treatment for symptomatic ICAS.Non-uniform utilisation in endovascular technology,institutional experience and patient selection in different volumes of centres may have an impact on overall safety of this treatment. | Yabing Wang Tao Wang Adam Andrew Dmytriw Kun Yang Liqun Jiao Huaizhang Shi Jie Lu Tianxiao Li Yujie Huang Zhenwei Zhao Wei Wu Jieqing Wan Qinjian Sun Bo Hong Yongli Li Liyong Zhang Jianfeng Chu Qiong Cheng Yiling Cai Pengfei Wang Qi Luo Hua Yang Baijing Dong Yang Zhang Jun Zhao Zuoquan Chen Wei Li Xiaoxin Bai Weiwen He Xueli Cai Maimai Ti Osama O Zaidat | 2022 | Stroke & Vascular Neurology2022,7,2: | 3 |
| 2 | A Convenient and Biosafe Replicon with Accessory Genes of SARS-CoV-2 and Its Potential Application in Antiviral Drug Discovery显示文摘SARS-CoV-2 causes the pandemic of COVID-19 and no effective drugs for this disease are available thus far.Due to the high infectivity and pathogenicity of this virus,all studies on the live virus are strictly confined in the biosafety level 3(BSL3)laboratory but this would hinder the basic research and antiviral drug development of SARS-CoV-2 because the BSL3 facility is not commonly available and the work in the containment is costly and laborious.In this study,we constructed a reverse genetics system of SARS-CoV-2 by assembling the viral cDNA in a bacterial artificial chromosome(BAC)vector with deletion of the spike(S)gene.Transfection of the cDNA into cells results in the production of an RNA replicon that keeps the capability of genome or subgenome replication but is deficient in virion assembly and infection due to the absence of S protein.Therefore,such a replicon system is not infectious and can be used in ordinary biological laboratories.We confirmed the efficient replication of the replicon by demonstrating the expression of the subgenomic RNAs which have similar profiles to the wild-type virus.By mutational analysis of nsp12 and nsp14,we showed that the RNA polymerase,exonuclease,and cap N7 methyltransferase play essential roles in genome replication and sgRNA production.We also created a SARS-CoV-2 replicon carrying a luciferase reporter gene and this system was validated by the inhibition assays with known anti-SARS-CoV-2 inhibitors.Thus,such a one-plasmid system is biosafe and convenient to use,which will benefit both fundamental research and development of antiviral drugs. | Yun-Yun Jin Hanwen Lin Liu Cao Wei-Chen Wu Yanxi Ji Liubing Du Yiling Jiang Yanchun Xie Kuijie Tong Fan Xing Fuxiang Zheng Mang Shi Ji-An Pan Xiaoxue Peng Deyin Guo | 2021 | Virologica Sinica2021,36,5: | 2 |

| 7 | Inhibition of glycogen synthase kinase 3βimproves cognitive function in aged mice by upregulating claudin presences in cerebral endothelial cells显示文摘Glycogen synthase kinase-3β(GSK-3β),a serine/threonine protein kinase,is widely distributed in mammalian brains.Since GSK-3βplays a vital role in the development of neurodegenerative disorders,the present study was designed to investigate the role of GSK-3βin the blood-brain barrier(BBB)permeability in aged mice.Morris water maze test was used to examine mouse cognitive function.BBB permeability was examined by the leakage of fluorescence signals of low-molecular weight dextran.GSK-3βinhibitor,4-benzyl-2-methyl-1,2,4-thiadiazolidine-3,5-dione(TDZD-8),was administrated in aged mice and in cultured mouse brain microvascular endothelial cells(bEnd.3).Compared with young mice,aged mice had increased leftover signals of dextran in the hippocampus and a lower score in the maze test,suggesting that aged mice have abnormal leakage of BBB and cognitive dysfunction.The protein expression of Toll-like receptor 4(TLR4)was increased,whereas the protein expressions of junction proteins(claudinl and claudin5)were reduced in endothelial cells of BBB in aged mice.Phosphorylated level of serine 9,an inhibitory residue in GSK-3βprotein,was decreased.TDZD-8 treatment downregulated TLR4 protein expression,upregulated claudinl and claudin5 protein expressions,and significantly improved cognitive function in aged mice.In bEnd.3 cells,TDZD-8 treatment reduced TLR4 expression and increased claudin5 expression in cells stimulated with lipopolysaccharides.In conclusion,the inhibition of GSK-3βactivity downregulates aging-induced TLR4 expression and restores the BBB integrity,resulting in the improvement of cognitive function in aged mice. | Tao Liang Huihui Ju Yile Zhou Yajie Yang Yi Shi Hao Fang | 2020 | Acta Biochimica et Biophysica Sinica2020,52,4: | 0 |
| 8 | Multi-locus identification of Psilocybe cubensis by high-resolution melting(HRM)显示文摘Hallucinogenic mushroom is a kind of toxic strain containing psychoactive tryptamine substances such as psilocybin,psilocin and ibotenic acid,etc.The mushrooms containing hallucinogenic components are various,widely distributed and lack of standard to define,which made a great challenge to identification.Traditional identification methods,such as morphology and toxicology analysis,showed shortcomings in old or processed samples,while the DNA-based identification of hallucinogenic mushrooms would allow to identify these samples due to the stability of DNA.In this paper,four primer sets are designed to target Psilocybe cubensis DNA for increasing resolution of present identification method,and the target markers include largest subunit of RNA polymerase II(marked as PC-R1),psilocybin-related phosphotransferase gene(marked as PC-PT),glyceraldehyde 3-phosphate dehydrogenase(marked as PC-3)and translation EF1α(marked as PC-EF).Real-time PCR with high-resolution melting(HRM)assay were used for the differentiation of the fragments amplified by these primer sets,which were tested for specificity,reproducibility,sensitivity,mixture analysis and multiplex PCR.It was shown that the melting temperatures of PC-R1,PC-PT,PC-3 and PC-EF of P.cubensis were(87.93±0.12)℃,(82.21±0.14)℃,(79.72±0.12)℃ and(80.11±0.19)℃ in our kinds of independent experiments.Significant HRM characteristic can be shown with a low concentration of 62.5pg/µL DNA sample,and P.cubensis could be detected in mixtures with Homo sapiens or Cannabis sativa.In summary,the method of HRM analysis can quickly and specifically distinguish P.cubensis from other species,which could be utilized for forensic science,medical diagnosis and drug trafficking cases. | Xiaochun Zhang Huan Yu Ziwei Wang Qi Yang Ruocheng Xia Yiling Qu Ruiyang Tao Yan Shi Ping Xiang Suhua Zhang Chengtao Li | 2022 | Forensic Sciences Research2022,7,3: | 0 |
| 9 | Phytochemical activators of Nrf2:a review of therapeutic strategies in diabetes显示文摘Insulin resistance(IR)is fundamental to the development of type 2 diabetes(T2D),and altered mitochondrial function and abnormal lipid distribution are closely associated with IR or T2D.Excess oxidative stress-induced mitochondrial damage leads to an imbalance in redox homeostasis,which is considered the major contributor to the progression of diabetes.A key cellular defense mechanism,namely,the nuclear factor-E2 p45-related factor 2(Nrf2)-antioxidant response element(ARE)pathway,plays an essential protective role in combating excess oxidative stress.A series of phytochemicals are reported to improve IR and restore mitochondrial function against excess oxidative stress by activating the Nrf2-ARE signaling pathway to maintain cellular reactive oxygen species(ROS)homeostasis.The present review focuses on key knowledge gaps in the Nrf2-ARE system targeted by phytochemicals and its correlation to diabetes both in the in vitro and in vivo models and recent achievements in human clinical trials to evaluate its efficiency and safety.In addition,we provide an overview of recent research progress in nutrigenomics,precision nutrition and the interactions occurring in gut microbiota associated with the Nrf2-ARE signaling pathway and diabetes chemoprevention by phytochemicals and finally propose a future research strategy for regulating redox and microbiota balance via the Nrf2-ARE pathway.The present review aims to help us comprehensively understand the critical chemopreventive role of the Nrf2-ARE pathway targeted by phytochemicals in diabetes. | Si Qin Zhong Chen Yiling Wen Yuhang Yi Chenghao Lv Chaoxi Zeng Lanming Chen Meng Shi | 2023 | Acta Biochimica et Biophysica Sinica2023,55,1: | 0 |
| 10 | 中国阴阳文化背景下的躯体症状障碍1例并文献复习显示文摘躯体症状障碍(somatic symptom disorder,SSD)的特征是患者长期对各种躯体症状感到过度的担忧,感到十分痛苦,明显影响其社会功能及个人生活。SSD患者的临床表现与其文化背景存在联系;本例SSD患者在躯体检查时未见任何异常,但受到我国传统阴阳文化的影响其临床表现具有显著的中华文化色彩,以与中华传统阴阳文化联系紧密的性功能障碍和心境障碍为主。笔者采用汉密尔顿焦虑量表、汉密尔顿抑郁量表及国际勃起功能问卷表分别对患者的焦虑情绪、抑郁情绪及性功能进行评价,分值分别为32,33及9。经5周的文拉法辛和米氮平联合用药治疗,临床症状得到显著改善。我国SSD患者的临床表现有时具有明显的中华阴阳文化色彩,在临床工作中很容易被误诊,临床医生应对此提高警惕。此外,文拉法辛和米氮平联合用药对以慢性疼痛及性功能障碍为主要表现的SSD患者可能具有较好的疗效。 | CHENG Peng SHI Yiling XU Lizhi ZHOU Ying LIWeihui | 2020 | 中南大学学报(医学版)2020,45,9: | 0 |
