1Single-cell transcriptomic analysis defines the interplay between tumor cells,viral infection,and the microenvironment in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an Epstein-Barr virus(EBV)-associated malignancy with a complex tumor ecosystem.How the interplay between tumor cells,EBV,and the microenvironment contributes to NPC progression and immune evasion remains unclear.Here we performed single-cell RNA sequencing on^104,000 cells from 19 EBV^+NPCs and 7 nonmalignant nasopharyngeal biopsies,simultaneously profiling the transcriptomes of malignant cells,EBV,stromal and immune cells.Overall,we identified global upregulation of interferon responses in the multicellular ecosystem of NPC.Notably,an epithelial–immune dual feature of malignant cells was discovered and associated with poor prognosis.Functional experiments revealed that tumor cells with this dual feature exhibited a higher capacity for tumorigenesis.Further characterization of the cellular components of the tumor microenvironment(TME)and their interactions with tumor cells revealed that the dual feature of tumor cells was positively correlated with the expression of co-inhibitory receptors on CD8^+tumor-infiltrating T cells.In addition,tumor cells with the dual feature were found to repress IFN-γproduction by T cells,demonstrating their capacity for immune suppression.Our results provide new insights into the multicellular ecosystem of NPC and offer important clinical implications.Shanzhao Jin Ruoyan Li Ming-Yuan Chen Chao Yu Lin-Quan Tang Yan-Min Liu Jiang-Ping Li Yi-Na Liu Yi-Ling Luo Yifan Zhao Yu Zhang Tian-Liang Xia Shang-Xin Liu Qi Liu Guan-Nan Wang Rui You Jing-Yun Peng Jiang Li Feng Han Jianwei Wang Qiu-Yan Chen Li Zhang Hai-Qiang Mai Benjamin E.Gewurz Bo Zhao Lawrence S.Young Qian Zhong Fan Bai Mu-Sheng Zeng 2020Cell Research2020,30,11:16

3Synthesis of layered double hydroxides/graphene oxide nanocomposite as a novel high-temperature CO_2 adsorbent显示文摘In this contribution, a novel high-temperature CO2 adsorbent consisting of Mg-Al layered double hydroxide(LDH) and graphene oxide(GO)nanosheets was prepared and evaluated. The nanocomposite-type adsorbent was synthesized based on the electrostatically driven self-assembly between positively charged Mg-Al LDH single sheet and negatively charged GO monolayer. The characteristics of this novel adsorbent were investigated using XRD, FE-SEM, HRTEM, FT-IR, BET and TGA. The results showed that both the CO2 adsorption capacity and the multicycle stability of LDH were increased with the addition of GO owing to the enhanced particle dispersion and stabilization. In particular, the absolute CO2 capture capacity of LDH was increased by more than twice by adding 6.54 wt% GO as support. GO appeared to be especially effective for supporting LDH sheets. Moreover, the CO2 capture capacity of the adsorbent could be further increased by doping with 15 wt%K2CO3. 4SPDEF suppresses head and neck squamous cell carcinoma progression by transcriptionally activating NR4A1显示文摘SAM pointed domain containing E26 transformation-specific transcription factor(SPDEF)plays dual roles in the initiation and development of human malignancies.However,the biological role of SPDEF in head and neck squamous cell carcinoma(HNSCC)remains unclear.In this study,the expression level of SPDEF and its correlation with the clinical parameters of patients with HNSCC were determined using TCGA-HNSC,GSE65858,and our own clinical cohorts.CCK8,colony formation,cell cycle analysis,and a xenograft tumor growth model were used to determine the molecular functions of SPDEF in HNSCC.ChIP-qPCR,dual luciferase reporter assay,and rescue experiments were conducted to explore the potential molecular mechanism of SPDEF in HNSCC.Compared with normal epithelial tissues,SPDEF was significantly downregulated in HNSCC tissues.Patients with HNSCC with low SPDEF mRNA levels exhibited poor clinical outcomes.Restoring SPDEF inhibited HNSCC cell viability and colony formation and induced G0/G1 cell cycle arrest,while silencing SPDEF promoted cell proliferation in vitro.The xenograft tumor growth model showed that tumors with SPDEF overexpression had slower growth rates,smaller volumes,and lower weights.SPDEF could directly bind to the promoter region of NR4A1 and promoted its transcription,inducing the suppression of AKT,MAPK,and NF-κB signaling pathways.Moreover,silencing NR4A1 blocked the suppressive effect of SPDEF in HNSCC cells.Here,we demonstrate that SPDEF acts as a tumor suppressor by transcriptionally activating NR4A1 in HNSCC.Our findings provide novel insights into the molecular mechanism of SPDEF in tumorigenesis and a novel potential therapeutic target for HNSCC.Yanting Wang Xianyue Ren Weiyu Li Ruoyan Cao Suyang Liu Laibo Jiang Bin Cheng Juan Xia 2021International Journal of Oral Science2021,13,4:3
5Phylogenetic Analysis of the Dengue Virus Strains Causing the 2019 Dengue Fever Outbreak in Hainan,China显示文摘Dengue virus is an arthropod-borne pathogen that is transmitted to humans primarily by Aedes spp.mosquitos,causing the acute infectious disease,dengue fever(DF).Until 2019,no dengue outbreak had been reported in Hainan Province for over 20 years.However,in early September of 2019,an increasing number of infected cases appeared and the DF outbreak lasted for over one month in Haikou City,Hainan Province.In our study,we collected 97 plasma samples from DF patients at three hospitals,as well as 1585 mosquito larvae samples from puddles in different areas of Haikou.There were 49(50.5%)plasma samples found to be strongly positive and 9(9.3%)plasma samples were weakly positive against the NS1 antigen.We discovered DENV both in the patient's plasma samples and mosquito larvae samples,and isolated the virus from C6/36 cells inoculated with the acute phase serum of patients.Phylogenetic analysis revealed that the new strains were the most closely related to the epidemic strain in the southern regions of China,belonging to lineageⅣ,genotypeⅠ,DENV-1.Compared to the seven closest strains from neighboring countries and provinces,a total of 18 amino acid mutations occurred in the coding sequences(CDS)of the new isolated strain,DENV1 HMU-HKU-2.Our data shows that dengue virus is re-emerged in Hainan,and pose new threats for public health.Thus regular molecular epidemiological surveillance is necessary for control and prevention of DENV transmission.Jiang Du Liyuan Zhang Xiaoyuan Hu Ruoyan Peng Gaoyu Wang Yi Huang Wenqi Wang Kunliang Wu Qiang Wang Haoxiang Su Fan Yang Yun Zhang Chuanning Tang Xiuji Cui Lina Niu Gang Lu Meifang Xiao Yongguo Du Feifei Yin 2021Virologica Sinica2021,36,4:2

10Metagenomic insights into responses of microbial population and key functional genes to fulvic acid during partial nitritation显示文摘The long-term impact of fulvic acid(FA)on partial nitritation(PN)systemwas initially examined in this study.The obtained results revealed that the FA lower than 50 mg/L had negligible effect on the nitrite accumulation rate(NAR nearly 100%)and ammonium removal rate(ARR 56.85%),while FA over 50 mg/L decreased ARR from 56.85%to 0.7%.Sludge characteristics analysis found that appropriate FA(<50 mg/L)exposure promoted the settling performance and granulation of PN sludge by removing Bacteroidetes and accumulating Chloroflexi.The analysis of metagenomics suggested that the presence of limited FA(0-50 mg/L)stimulated the generation of NADH,which favors the denitrification and nitrite reduction.The negative impact of FA on the PN system could be divided into two stages.Initially,limited FA(50-120 mg/L)was decomposed by Anaerolineae to stimulate the growth and propagation of heterotrophic bacteria(Thauera).Increasing heterotrophs competed with AOB(Nitrosomonas)for dissolved oxygen,causing AOB to be eliminated and ARR to declined.Subsequently,when FA dosage was over 120 mg/L,Anaerolineae were inhibited and heterotrophic bacteria reduced,resulting in the abundance of AOB recovered.Nevertheless,the ammonium transformation pathway was suppressed because genes amoABC and hao were obviously reduced,leading to the deterioration of reactor performance.Overall,these results provide theoretical guidance for the practical application of PN for the treatment of FA-containing sewage.Li Zhang Shuang Lan Quanhao Dou Shiwei Hao Yueping Wang Xiaoxuan Wang Ruoyan Zhang Yongzhen Peng Jiachun Yang 2023Journal of Environmental Sciences2023,,2:1

14A“T.E.S.T.”hydrogel bioadhesive assisted by corneal cross-linking for in situ sutureless corneal repair显示文摘Corneal transplantation is an effective clinical treatment for corneal diseases,which,however,is limited by donor corneas.It is of great clinical value to develop bioadhesive corneal patches with functions of“Transparency”and“Epithelium&Stroma generation”,as well as“Suturelessness”and“Toughness”.To simultaneously meet the“T.E.S.T.”requirements,a light-curable hydrogel is designed based on methacryloylated gelatin(GelMA),Pluronic F127 diacrylate(F127DA)&Aldehyded Pluronic F127(AF127)co-assembled bi-functional micelles and collagen type I(COL I),combined with clinically applied corneal cross-linking(CXL)technology for repairing damaged cornea.The patch formed after 5 min of ultraviolet irradiation possesses transparent,highly tough,and strongly bio-adhesive performance.Multiple cross-linking makes the patch withstand deformation near 600%and exhibit a burst pressure larger than 400 mmHg,significantly higher than normal intraocular pressure(10-21 mmHg).Besides,the slower degradation than GelMA-F127DA&AF127 hydrogel without COL I makes hydrogel patch stable on stromal beds in vivo,supporting the regrowth of corneal epithelium and stroma.The hydrogel patch can replace deep corneal stromal defects and well bio-integrate into the corneal tissue in rabbit models within 4 weeks,showing great potential in surgeries for keratoconus and other corneal diseases by combining with CXL.Meiyan Li Ruoyan Wei Chang Liu Haowei Fang Weiming Yang Yunzhe Wang Yiyong Xian Kunxi Zhang Yong He Xingtao Zhou 2023Bioactive Materials2023,,7:1
15Late Jurassic-Early Cretaceous tectonic switching in Liaodong Peninsula of the North China Craton and the implications for gold mineralisation显示文摘Constraining the processes of deformation during tectonic switching from compression to extension is difficult because of the scarcity or absence of associated sedimentary and magmatic rocks and weak metamorphism.The east margin of the eastern North China Craton experienced Early to Middle Jurassic compression and Early Cretaceous extension.However,the period of tectonic quiescence lasting~13 million years(between 153 and 140 Ma)during which this transition occurred is poorly understood.This paper reports the identification of small-scale N-S-trending thrust and sinistral strike-slip faults(TSS)and NWtrending thrust and dextral strike-slip faults(TDS)in the Tongyuanpu-Aiyang region,which is part of the northern Liaodong Peninsula.Calculation of the tectonic stress field using striations,fault planes and kinematics reveals a NW-SE-oriented maximum principal axis(σ1),and sub-horizontalσ2 andσ3.Assemblages of N-S-trending TSS and NW-trending TDS were arranged with right stepping and resulted in local uplift and erosion at the junctions between fault terminations,which partitioned the Early and Middle Jurassic basins into residual smaller volcanic-sedimentary basins.These basins were unconformably overlain by small Early Cretaceous volcanic-sedimentary basins,such as those at Tongyuanpu and Fangjiaweizi.Magmatic ages and the timing of basin formation constrain the initiation of tectonic switching to 156–153 Ma,and its termination to 140–139 Ma.We suggest that R-R’Riedel shears controlled the formation of the N-S-trending TSS and NW-trending TDS.The R-R’shears were produced by continued sinistral strike-slip and northward growth along the Bohai Bay segment of the Tan-Lu fault zone during the Late Jurassic to early Early Cretaceous,simultaneous with a gradual weakening in the tectonic stress field during a switch in the direction of subduction of the Palaeo-Pacific plate from NW-ward to NNW-ward.This tectonic switching might have promoted the dissolution of gold and migration of gold-bearing fluids.Dan-Ping YAN Ruoyan KONG Xiaoyu DONG Liang QIU Huilong LIU 2021Science China Earth Sciences2021,64,9:1
