| 1 | Propofol blocks apoptosis and Bcl-2 and Bax expression induced by hypoxia-reoxygenation in primary cultures of rat hippocampal astrocytes显示文摘BACKGROUND:Cerebral hippocampal astrocytes are more sensitive to ischemic injury than neurons.Hypoxic-ischemic brain injury induces profound astrocyte apoptosis,and propofol may protect against astrocyte apoptosis. OBJECTIVE:To verify the protective effects of propofol against astrocyte apoptosis and to investigate anti-apoptotic Bcl-2 and pro-apoptotic Bax expression in primary cultures of rat hippocampal astrocytes exposed to hypoxia-reoxygenation for different periods of time following propofol treatment. DESIGN,TIME,AND SETTING:In vitro neural immunocytochemistry was performed at the Central Laboratory of Yunyang Medical College between September 2007 and March 2008. MATERIALS:A total of 30 Wistar rats,aged 1-3 days,with equal numbers of males and females, were included for isolation and culture of hippocampal astrocytes. METHODS:Hippocampal astrocytes were purified and cultured for 3 weeks and treated with four culture conditions:50μL Hank's solution(normal control);0.2 mL/L Intralipid;50μL Hank's solution for 10 minutes followed by hypoxic incubation for 4 hours and normoxic incubation for 12,24,36,48, 60 or 72 hours;propofol(250μmol/L final) for 10 minutes followed by hypoxic incubation for 4 hours and normoxic incubation for 12,24,36,48,60 and 72 hours. MAIN OUTCOME MEASURES:(1) Morphologic changes in hippocampal astrocytes.(2) Levels of astrocyte apoptosis and Bcl-2 and Bax expression. RESULTS:Hypoxia and reoxygenation increased apoptosis over time,with Bcl-2 expression peaking at 24 hours and decreasing gradually(P<0.01);Bax expression peaked at 72 hours(P< 0.01);the ratio of Bcl-2/Bax was 1.4,0.8,and 0.6,respectively,at 24,48 and 72 hours. Non-apoptotic astrocytes showed significant proliferation and swelling.Propofol treatment decreased apoptosis after hypoxia-reoxygenation(P<0.01),as well as Bcl-2 and Bax expression (P<0.05,P<0.01),with Bcl-2/Bax ratios of 1.6-1.8.Propofol treatment also blocked astrocyte proliferation and swelling.No apoptotic cells or Bcl-2/Bax expression was detected in astrocytes cultured in Hank's or Intralipid solution. | 4 | Effect of propofol on glutamate-induced activation and related inflammatory cytokines of astrocytes from spinal cord dorsal horn显示文摘BACKGROUND:Astrocytes participate in central nervous system-mediated physiological or pathological processes,such as pain.Activated dorsal horn astrocytes from the spinal cord produce nerve active substances and proinflammatory cytokines,such as interleukin-1beta(IL-1β),IL-6,and tumor necrosis factor-α(TNF-α),which play important roles in pain transduction and regulation.OBJECTIVE:To investigate the effects of different doses of propofol on activation of cultured spinal cord dorsal horn astrocytes induced by glutamate,as well as changes in IL-1β,IL-6,and TNF-α,and IL-10(anti-inflammatory cytokine) expression in rats,and to explore the dose relationship of propofol.DESIGN,TIME AND SETTING:The cellular and molecular biology experiment was performed at the Central Laboratory of Yunyang Medical College between March 2006 and December 2007.MATERIALS:Forty healthy,Wistar rats,aged 2-3 days,were selected.Propofol was provided by Zeneca,UK;glutamate by Sigma,USA;EPICS XL flow cytometry by Beckman culture,USA;rabbit-anti-mouse glial fibrillary acidic protein(GFAP) antibody kit and inflammatory cytokine detection kit were provided by Zhongshan Biotechnology Company Ltd.,Beijing;multimedia color pathologic image analysis system was a product of Nikon,Japan.METHODS:Astrocytes were harvested from T11-L6 spinal cord dorsal horn of Wistar rats and incubated for 3 weeks.The cells were divided into seven groups,according to various treatment conditions:control group was cells cultured in Hank's buffered saline solution;intralipid group was cells cultured in intralipid(0.2 mL/L);glutamate group was cells cultured with 100 μmol/L glutamate;propofol group was cells cultured with 250 μmol/L propofol;three glutamate plus propofol groups were cultured in 100 μmol/L of glutamate,followed by 5,25,and 250 μmol/L of propofol 10 minutes later.MAIN OUTCOME MEASURES:GFAP-labeled astrocytes were analyzed using a multimedia pathology imaging analysis system to detect area density(AD) and average optical density(AOD) of positive cells.The supernatant concentrations of IL-1β,TNF-α,IL-6,and IL-10 were determined using radioimmune assays.RESULTS:Compared with the control group,cells in the glutamate plus low-dose propofol group were activated and hypertrophic,and AD and AOD were significantly increased(P < 0.01).Concentrations of IL-1 β,TNF-α,and IL-6 were also significantly increased(P < 0.01),while IL-10 levels remained unchanged(P > 0.05),but still higher than the control and glutamate groups(P > 0.05).Compared with the glutamate group,astrocyte activation was inhibited by moderate and high-dose propofol.In addition,with moderate and high-dose propofol,AD,AOD,IL-1β,TNF-α,and IL-6 concentrations were significantly decreased(P < 0.05-0.01),and IL-10 levels were increased(P < 0.01).CONCLUSION:Propofol can effectively inhibit glutamate-induced astrocyte activation in the spinal cord dorsal horn,significantly inhibit production of IL-1β,TNF-α,and IL-6,and increase IL-10 synthesis and release in a dose-dependent manner. | Chengming Qin Qing Li Juying Liu Tao Zhu Yong Xiang | 2008 | Neural Regeneration Research2008,3,9: | 0 |
| 5 | N-methyl-D-aspartate receptor effects on p38 mitogen activated protein kinase and its role in a rat model of diabetic neuropathic pain显示文摘BACKGROUND:Activated N-methyl-D-aspartate(NMDA) receptor is involved in the formation of chronic neuropathic pain,and its antagonist,ketamine,exhibits effective amelioration of diabetic neuropathic pain(DNP).However,the mechanisms of NMDA receptor participation in the formation and maintenance of DNP remain poorly understood. OBJECTIVE:To evaluate the role NMDA receptor plays in DNP and effects on p38 mitogen activated protein kinase(p38 MAPK) in a rat model of DNP. DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Human Embryonic Stem Cell Research Institute of Yunyang Medical College Affiliated Taihe Hospital between July 2005 and September 2007. MATERIALS:Streptozotocin was provided by Sigma,USA;p38 MAPK inhibitor(SB203580) was provided by Shanghai KangChen Biotech,China;NMDA receptor antagonist(MK-801) was purchased from Shanghai Yope Biotech,China. METHODS:A total of 128 healthy,Wistar rats of clean grade,aged 3 months and weighing 180- 220 g,were randomly assigned to 4 groups:control,DNP model,p38 MAPK,and NMDA receptor. Each group contained 32 rats.DNP was established in all groups except for the control group by intraperitoneal injection of streptozocin(65 mg/kg).Subsequently,1 mg/kg SB203580 and 1 mg/kg MK-801 were injected once each week via intraperitoneal injection in the p38 MAPK and NMDA receptor groups,respectively. MAIN OUTCOME MEASURES:At the end of 2,4,6,and 8 weeks following streptozotocin injection, mechanical withdrawal threshold was measured in 8 animals from each group following von Frey filament stimulation.The rats were anesthetized and nerve conduction velocity of the left sciatic nerve was measured.Subsequently,the right sciatic nerve,the lumbar segment of the spinal cord, and dorsal root ganglia were removed from the L_(3-6) segment for microscopic examination.p38 MAPK expression was determined using immunohistochemistry and Western blot analysis. Expression of NMDA receptor 1 mRNA in dorsal root ganglion and spinal cord neurons was detected using RT-PCR. RESULTS:Mechanical withdrawal threshold and nerve conduction velocity were significantly reduced,and p38 MAPK and NMDA receptor 1 mRNA expression in the spinal cord and dorsal root ganglia were significantly increased,in the model,p38 MAPK,and NMDA receptor groups compared with the control group at all time points(P<0.05).At 4-8 weeks following successful DNP model establishment,SB203580 and MK-801 increased mechanical withdrawal threshold,accelerated nerve conduction velocity,and attenuated p38 MAPK expression,compared with the model group. The NMDA receptor group exhibited downregulated mRNA expression of NMDA receptor 1 compared with the model and p38 MAPK groups(P<0.05). | 7 | Edge localized modes suppression via edge E × B velocity shear induced by RF sheath of ion cyclotron resonance heating in EAST显示文摘The control of large edge localized modes(ELMs) is a critical issue for the successful operation of future burning plasma devices,such as the international thermonuclear experimental reactor(ITER) and China fusion engineering test reactor(CFETR). In this paper, we present a new active and effective means of ELM suppression using ion cyclotron resonant heating(ICRH) on the experimental advanced superconducting tokamak(EAST). We obtained the key role of the external E × B velocity shear near the pedestal top and the scrape-off-layer(SOL) induced by the RF sheath potential of ICRH in ELM suppression. The experimental results showed a positive correlation between the RF sheath and the E × B shear rate in SOL. BOUT++ simulations indicate that increased E × B velocity shear rates in the pedestal and SOL regions promote ELM suppression;thereby, supporting the experimental observations on EAST.
