| 1 | Root microbiota shift in rice correlates with resident time in the field and developmental stage显示文摘Land plants in natural soil form intimate relationships with the diverse root bacterial microbiota. A growing body of evidence shows that these microbes are important for plant growth and health. Root microbiota composition has been widely studied in several model plants and crops; however, little is known about how root microbiota vary throughout the plant's life cycle under field conditions. We performed longitudinal dense sampling in field trials to track the time-series shift of the root microbiota from two representative rice cultivars in two separate locations in China. We found that the rice root microbiota varied dramatically during the vegetative stages and stabilized from the beginning of the reproductive stage, after which the root microbiota underwent relatively minor changes until rice ripening. Notably, both rice genotype and geographical location influenced the patterns of root microbiota shift that occurred during plant growth. The relative abundance of Deltaproteobacteria in roots significantly increased overtime throughout the entire life cycle of rice, while that of Betaproteobacteria, Firmicutes, and Gammaproteobacteria decreased. By a machine learning approach, we identified biomarker taxa and established a model to correlate root microbiota with rice resident time in the field(e.g., Nitrospira accumulated from 5 weeks/tillering in field-grown rice). | 5 | Optimization of food deprivation and sucrose preference test in SD rat model undergoing chronic unpredictable mild stress显示文摘Background:The chronic unpredictable mild stress(CUMS)model has long been considered the best model for exploring the pathophysiological mechanisms underlying depression.However,there are no widely recognised standards for strategies for modeling and for behavioral testing.The present study aimed to optimize the protocols for food deprivation and the sucrose preference test(SPT)for the CUMS model.Methods:We first evaluated the effects of different long periods of food deprivation on the body weight of Sprague Dawley(SD)rats by testing food deprivation for 24 hours(8:00-8:00^+),food deprivation for 12 hours during the daytime(8:00-20:00)and food deprivation for 12 hours at night(20:00-8:00^+).Next,we established a SD rat CUMS model with 15 different stimulations,and used body weight measurement,SPT,forced swim test(FST),open field test(OFT)and Morris water maze(MWM)test to verify the success of the modeling.In the SPT,consumption of sucrose and pure water within 1 and 12 hours was measured.Results:Twelve hours of food deprivation during the daytime(8:00-20:00)had no effect on body weight,while 12 hours of food deprivation at night(20:00-8:00^+)and 24 hours of food deprivation(8:00-8:00^+)significantly reduced the mean body weight of the SD rats.When SPT was used to verify the successful establishment of the CUMS rat model,sucrose consumption measured within 12 hours was less variable than that measured within 1 hour.Conclusions:Twelve hours of food deprivation in the daytime(8:00-20:00)may be considered a mild stimulus for the establishment of a CUMS rat model.Measuring sucrose consumption over 12 hours is recommended for SPT. | Li-Wen He Li Zeng Na Tian Yi Li Tong He Dong-Mei Tan Qian Zhang Yi Tan | 2020 | Animal Models and Experimental Medicine2020,3,1: | 7 |

| 7 | MicroRNA-125b regulates Th17/Treg cell differentiation and is associated with juvenile idiopathic arthritis显示文摘Background Juvenile idiopathic arthritis(JIA)is the most common rheumatic disease in childhood driven by aberrant pathways of T-cell activation.T helper 17(Th17)/regulatory T cell(Treg)imbalance plays critical roles in the pathogenesis of arthritis.MicroRNA-125b(miR-125b)was upregulated after the activation of the initial CD4^+T cells,and could regulate the differentiation of CD4^+T cells.However,the effects of miR-125b on Th17/Treg imbalance and differentiation of Th 17/Treg cells remain unknown.Methods In this study,we evaluated the expression of miR-125b in the peripheral blood mononuclear cells(PBMCs)of children with JIA,and the relationship of miR-125b with Th17/Treg imbalance.Then,we used lentivirus vector-mediated overexpression technology to investigate the regulatory function of miR-125b in CD4^+T cells or dendritic cell/CD4^+T co-culture system.Results Decreased miR-125b expression in PBMCs and CD4^+T cells of JIA patients was negatively correlated with the ratio of Th17/Treg cells.It also correlated negatively with retinoic acid receptor-related orphan receptorγt but positively with Forkhead box protein 3 at transcriptional levels.Furthermore,we found that miR-125b overexpression inhibited Th17 cell differentiation,whereas facilitated the differentiation of Treg cells.MiR-125b upregulation led to the decrease of Th 17-secreting cytokines but the increase of the Treg-secreting cytokines.Conclusions Our results demonstrate that miR-125b participated in regulating Thl7/Treg cell differentiation and imbalance in JIA patients.These findings provide novel insight into the critical role of miR-125b in the Th17/Treg imbalance of JIA,and raise the distinct possibility that miR-125b may prove to be a potential therapeutic target for JIA. | Zhi-Dan Fan Qian Cao Na Huang Le Ma Hui-Hui Ma Ya-Yuan Zhang Hai-Guo Yu Guo-Ping Zhou | 2020 | World Journal of Pediatrics2020,16,1: | 7 |

| 9 | Intranasal administration of tetrabromobisphenol A bis(2-hydroxyethyl ether) induces neurobehavioral changes in neonatal Sprague Dawley rats显示文摘Tetrabromobisphenol A(TBBPA) and its derivatives are now being highly concerned due to their emerging environmental occurrence and deleterious effects on non-target organisms.Considering the potential neurotoxicity of TBBPA derivatives which has been demonstrated in vitro, what could happen in vivo is worthy of being studied. Tetrabromobisphenol A bis(2-hydroxyethyl ether)(TBBPA-BHEE), a representative TBBPA derivative, was selected for a21-day exposure experiment on neonatal Sprague Dawley(SD) rats through intranasal administration. The neurobehavioral, histopathological changes, and differentially expressed genes based on RNA microarray were investigated to evaluate the neurological effects of this chemical. The results indicated that TBBPA-BHEE exposure significantly compromised the motor co-ordination performance and the locomotor activities(p < 0.05). The neurobehavioral phenotype could be attributed to the obvious histopathological changes in both cerebrum and cerebellum, such as neural cell swelling, microglial activation and proliferation. A total of 911 genes were up-regulated, whereas 433 genes were down-regulated. Gene set enrichment analysis showed multiple signaling pathways, including ubiquitin-mediated proteolysis and wingless-int(Wnt) signaling pathway etc. were involved due to TBBPA-BHEE exposure. The gene ontology enrichment analysis showed the basic cellular function and the neurological processes like synaptic transmission were influenced. | 12 | Serum microRNA-221 as a biomarker for diabetic retinopathy in patients associated with type 2 diabetes显示文摘AIM: To investigate the candidate microRNA(miRNA), miR-221 as a novel biomarker for diabetic retinopathy(DR) in patients associated with type 2 diabetes(T2D).METHODS: The subjects involved were divided into four groups: healthy control(HC), no diabetic retinopathy(NDR), non-proliferative diabetic retinopathy(NPDR) and proliferative diabetic retinopathy(PDR) group. Serum miR-221 was validated by real-time quantitative reversetranscription polymerase chain reaction(qRT-PCR). Also, serum angiotensin II(Ang II) and vascular endothelial growth factor(VEGF) were examined by enzyme-linked immunosorbent assay. In addition, receiver operating characteristic(ROC) curve was performed to explore the diagnostic accuracy of miR-221, Ang Ⅱ and VEGF for DR in patients with T2D. Spearman's rank correlation coefficient was executed to estimate the correlations of serum miR-221 with metabolic parameters and serum markers in patients with T2D.RESULTS: Primarily, serum miR-221, Ang Ⅱ and VEGF were increased significantly in T2D patients compared to HC participant respectively, and progressive up-regulated in NDR, NPDR and PDR groups(P<0.001). Additionally, miR-221 in serum was remarkably positively correlatedwith metabolic parameters such as glycated hemoglobin(r=0.310, P=0.002) and homeostasis model assessment for insulin resistance(r=0.413, P<0.001), as well as serum markers for instance Ang Ⅱ(r=0.667, P<0.001) and VEGF(r=0.499, P<0.001). Furthermore, serum miR-221(AUC, 0.894; 95%CI, 0.833-0.955; P<0.001), Ang Ⅱ(AUC, 0.888; 95%CI, 0.828-0.949; P<0.001) and VEGF(AUC, 0.785; 95%CI, 0.695-0.875; P<0.001) had evidently diagnostic efficiency in DR, and miR-221 is the most effective among them.CONCLUSION: Serum miR-221 as a potential biomarker could be related to not only occurrence but also progression for DR in patients with T2D. | 13 | The association between the genetic polymorphisms of LMP2/LMP7 and the outcomes of HCV infection among drug users显示文摘Objective:To investigate a possible association of LMP2/LMP7 genes with chronic hepatitis C virus(HCV) infection,and to assess whether LMP2/LMP7 genes could influence the outcomes of HCV infection among drug users.Methods:Genomic DNAs of 362 anti-HCV sero-positive drug users and 225 control drug users were extracted from the peripheral blood leukocytes.The sero-positive patients were divided into those who had persistent infection and those who had spontaneously cleared the infection.Polymorphisms of LMP genes were determined by PCR combined with restriction fragment length polymorphism(RFLP).Results:The distribution of LMP2 genotypes among the control,persistent infection and spontaneous clearance groups were not different.However,the LMP7 codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in control group(OR=1.75,95%CI=1.06~2.90;OR=3.16,95%CI=1.23-8.12;OR=1.94,95%CI=1.21-3.12,respectively).Similarly,the frequencies of the codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in the spontaneous clearance group(OR=1.64,95%CI=1.04-2.57;OR=2.40,95%CI=1.09-5.28;OR=1.76,95%CI=1.152.69,respectively).Stratified analysis indicated that combined genotype Gln/Lys+Lys/Lys of the LMP7 gene was related to an increasing susceptibility to HCV infection(OR=1.91,95%CI=1.02-3.55;OR=2.19,95%CI=1.243.89;OR=1.91,95%CI=1.05-3.48,OR=2.86,95%CI=1.41-5.78,respectively)and the risk of persistent HCV infection(OR=1.94,95%CI=1.12-3.34;OR=2.02,95%CI=1.21-3.38;OR=1.78,95%CI=1.12-2.85,OR=2.23,95%CI=1.09-4.58,respectively)among>30-year-old,males,the injection drug user(IDU)subjects and/or the shorter duration drug users(≤5 y).Conclusion:These results suggest that polymorphism of the LMP7 gene may have an influence on the outcomes of HCV infection,and is one of the factors accounting for the genetic susceptibility to HCV infection among drug users. | Qian Cui Yongxiang Zhang Jing Su Chao Shi Na Lei Keqin Ding Jun Li Rongbin Yu L u Wang Ning Wang | 2010 | The Journal of Biomedical Research2010,24,5: | 6 |

Real-time qPCR and immunohistochemistry were applied to detect dectin-1 expression in corneal epithelium of both groups. HCE cells were cultured with aspergillus fumigatus(AF) antigens in vitro. The expression of dectin-1 mRNA was measured by real-time qPCR at the stimulation of 0,4,8 and 24h separately. | 15 | An adjustment method for active reflector of large high-frequency antennas considering gain and boresight显示文摘The design of the Qitai 110 m Radio Telescope(QTT) with large aperture and very high working frequency(115 GHz) was investigated in Xinjiang, China. The results lead to a main reflector with high surface precision and high pointing precision. In this paper, the properties of active surface adjustment in a deformed parabolic reflector antenna are analyzed. To assure the performance of large reflector antennas such as gain and boresight, which can be obtained by utilizing an electromechanical coupling model, and satisfy them simultaneously, research on active surface adjustment applied to a new parabolic reflector as target surface has been done. Based on the initial position of actuators and the relationship between adjustment points and target points, a novel mathematical model and a program thatdirectly calculates the movements of actuators have been developed for guiding the active surface adjustment of large reflector antennas. | 16 | Lipoprotein(a) as a Marker for Predicting the Presence and Severity of Coronary Artery Disease in Untreated Chinese Patients Undergoing Coronary Angiography显示文摘Objective Low-density lipoprotein cholesterol(LDL-C) has been well known as the risk factor of coronary artery disease(CAD). However, the role of lipoprotein(a) [Lp(a)] in the development of CAD is of great interest but still controversial. Thus, we aim to explore the effect of Lp(a) on predicting the presence and severity of CAD in Chinese untreated patients, especially in combination with LDL-C. Methods We consecutively recruited 1,980 non-treated patients undergoing coronary angiography, among which 1,162 patients were diagnosed with CAD. Gensini score(GS) was used to assess the severity of CAD. Lp(a) was measured by immunoturbidimetric method. Results Patients with CAD had higher level of LDL-C and Lp(a) compared with non-CAD(P < 0.05). Multivariable logistic regression revealed that Lp(a) > 205 mg/L(highest tertile) predicted 1.437-fold risk for CAD(95% CI: 1.108-1.865, P = 0.006) and 1.480-fold risk for high GS(95% CI: 1.090-2.009, P = 0.012) respectively. Interestingly, concomitant elevated level of Lp(a) and LDL-C conferred the highest risk for both presence [OR = 1.845, 95% CI: 1.339-2.541, P < 0.001] and severity [OR = 1.736, 95% CI: 1.188-2.538, P = 0.004] of CAD. | 18 | Atractylodes lancea rhizome water extract reduces triptolide-induced toxicity and enhances anti-inflammatory effects显示文摘The present study was designed to explore the influence of water extracts of Atractylodes lancea rhizomes on the toxicity and anti-inflammatory effects of triptolide(TP). A water extract was prepared from A. lancea rhizomes and co-administered with TP in C57BL/6 mice. The toxicity was assayed by determining serum biochemical parameters and visceral indexes and by liver histopathological analysis. The hepatic CYP3A expression levels were detected using Western blotting and RT-PCR methods. The data showed that the water extract of A. lancea rhizomes reduced triptolide-induced toxicity, probably by inducing the hepatic expression of CYP3A. The anti-inflammatory effects of TP were evaluated in mice using a xylene-induced ear edema test. By comparing ear edema inhibition rates, we found that the water extract could also increase the anti-inflammatory effects of TP.
