1Clinicopathological significance of FHIT protein expression in gastric adenocarcinoma patients显示文摘AIM: To investigate the expression of fragile histidine triad (FHIT) protein, and the possible relationship between FHIT expression and clinicopathological indices in gastric carcinoma.METHODS: FHIT protein expression was examined in 76 cases of gastric carcinoma, 58 cases of intraepithelial neoplasia, and 76 cases of corresponding normal mucosae by immunohistochemical method to analyze its relationship to histological grade, clinical stage, metastatic status and prognosis.RESULTS: The FHIT protein expression was positive in 28/76 (36.8%) cases of adenocarcinoma tissue, 22/58(37.9%) cases of adjacent dysplastic tissue and 76/76(100%) cases of distal normal gastric mucosa. There was a significant difference in the expression of FHIT protein between cancer or adjacent intraepithelial neoplasia and normal gastric mucosa (P = 0.000). FHIT protein expression was found in 64.3% (18/28) of grades Ⅰ and Ⅱ cancers,and 20.8% (10/48) of grade Ⅲ cancers (P = 0.000), in 56.3% (18/32) of stages Ⅰ and Ⅱ cancers and 22.7%(10/44) of stages Ⅲ and Ⅳ cancers (P = 0.004), and in 63.6% (14/22) of cancers without metastasis but only 25.9% (14/54) of those with metastasis (P = 0.003). The significant difference in the expression of FHIT was found between histological grade, clinical stage and metastatic status of cancer. Follow-up data showed that there was a significant difference in median survival time between cancer patients with expression of FHIT (71 mo) and those without (33 mo, log rank = 20.78, P = 0.000).CONCLUSION: FHIT protein is an important tumor suppressor protein. 4Propofol differentially induces unconsciousness and respiratory depression through distinct interactions between GABAa receptor and GABAergic neuron in corresponding nuclei显示文摘Propofol is the most commonly used intravenous anesthetic worldwide.It can induce loss of consciousness prior to the occurrence of severe respiratory suppression,which is also a pharmacodynamic feature of all general anesthetics.However,the neural mechanisms underlying this natural phenomenon are controversial and highly related to patient safety.In the present study,we demonstrated that the pharmacodynamic effects of propofol(50 and 100 μM)on suppression of consciousness-related excitatory postsynaptic currents in the medial prefrontal cortex(mPFC)and centromedian nucleus of the thalamus(CMT)were lower than those in the kernel respiratory rhythmogenesis nucleus pre-Botzinger complex(PrBo).Furthermore,we unexpectedly found that the GABAa receptor β3 subunit is the key target for propofol's action and that it is mutually and exclusively expressed in GABAergic neurons.It is also more abundant in the mPFC and CMT,but mainly co-localized with GABAergic neurons in the PrBo.As a result,the differentiated expression pattern should mediate more neuron suppression through the activation of GABAergic neurons in the mPFC and CMT at low doses of propofol(50 μM).However,PrBo GABAergic neurons were only activated by propofol at a high dose(100 μM).These results highlight the detailed pharmacodynamic effects of propofol on consciousness-related and respiration-related nuclei and provide the distinct interaction mechanism between the β3 subunit and GABAergic neurons in mediating the suppression of consciousness compared to the inhibition of respiration.Junli Jiang Yingfu Jiao Po Gao Wen Yin Wei Zhou Yunchun Zhang Yanjun Liu Daxiang Wen Yuan Wang Liang Zhou Tian Yu Weifeng Yu 2021Acta Biochimica et Biophysica Sinica2021,53,8:11
5Advanced oxidation of bromide-containing drinking water: A balance between bromate and trihalomethane formation control显示文摘Addition of H2O2 has been employed to repress bromate formation during ozonation of bromide-containing source water.However,the addition of H2O2 will change the oxidation pathways of organic compounds due to the generation of abundant hydroxyl radicals,which could afect the removal efcacy of trihalomethane precursors via the combination of ozone and biological activated carbon(O3-BAC).In this study,we evaluated the efects of H2O2 addition on bromate formation and trihalomethane formation potential(THMFP)reduction during treatment of bromide-containing(97.6–129.1μg/L)source water by the O3-BAC process.At an ozone dose of 4.2 mg/L,an H2O2/O3(g/g)ratio of over 1.0 was required to maintain the bromate concentration below 10.0μg/L,while a much lower H2O2/O3ratio was sufcient for a lower ozone dose.An H2O2/O3(g/g)ratio below 0.3 should be avoided since the bromate concentration will increase with increasing H2O2 dose below this ratio.However,the addition of H2O2 at an ozone dose of 3.2 mg/L and an H2O2/O3 ratio of 1.0 resulted in a 43% decrease in THMFP removal when compared with the O3-BAC process.The optimum H2O2/O3(g/g) ratio for balancing bromate and trihalomethane control was about 0.7–1.0.Fractionation of organic materials showed that the addition of H2O2 decreased the removal efcacy of the hydrophilic matter fraction of DOC by ozonation and increased the reactivity of the hydrophobic fractions during formation of trihalomethane,which may be the two main reasons responsible for the decrease in THMFP reduction efcacy.Overall,this study clearly demonstrated that it is necessary to balance bromate reduction and THMFP control when adopting an H2O2 addition strategy.Yongjing Wang Jianwei Yu Po Han Jing Sha Tao Li Wei An Juan Liu Min Yang 2013Journal of Environmental Sciences2013,25,11:9

9Efficacy and safety of the Qiguiyin formula in severe pneumonia: study protocol for a randomized, double-blind, placebo-controlled clinical trial显示文摘OBJECTIVE: To evaluate the clinical efficacy and safety of Qiguiyin(QGY) formula in patients with severe pneumonia in China compared with a placebo.METHODS: This is a multicenter double-blind, placebo-controlled, randomized clinical trial with two parallel arms. There will be 530 patients enrolled and randomized into either the experimental group(QGY formula) or the control group(placebo). Therapies for patients in the two groups above will be based on the conventional therapy. The primary outcome is 28-day mortality. Secondary outcomes include:(a) duration of hospital stay;(b) duration of time in the intensive care unit(ICU) stays;(c) duration of mechanical ventilation;(d) antibiotic DDD value(which means the doses of antibotics during the treatment period);(e) serum procalcitonin(PCT) level;(f) serum C-reactive protein(CRP)level;(g) Pneumonia severity index(PSI) score;(h)Sequential Organ Failure Assessment(SOFA) score;(i) sputum culture results;(j) blood routine examination results;(k) routine urine test results;(l) stool routine examination results;(m) electrocardiogram results;(n) alanine aminotransferase levels;(o) aspartate amino transferase levels;(p) total bilirubin;(q) creatinine levels;(r) urea nitrogen levels;and(s)adverse events.ETHICS AND DISSEMINATION: The protocol has been approved by the Research Ethics Committee of Beijing Hospital of Traditional Chinese Medicine,Affiliated with Capital Medical University(2018 BL-053-02). 10Overexpression of carbonic anhydrase Ⅱ and Ki-67 proteins in prognosis of gastrointestinal stromal tumors显示文摘AIM: To investigate the expression and prognostic value of carbonic anhydrase Ⅱ (CA Ⅱ) and Ki-67 in gastrointestinal stromal tumors (GISTs). METHODS: One hundred and thirteen GIST patients admitted to Chinese People's Liberation Army General Hospital from January 2004 to December 2010 were retrospectively followed up, and immunohistochemistry was used to detect CA Ⅱ, Ki-67 and CD117 expression in tumor samples. The survival rates of the patients were analyzed using the Kaplan-Meier method. Log-rank test, χ 2 test and Cox proportional hazards model were used to determine the relationships between CA Ⅱ, Ki-67 and CD117 expression and prognostic value in GISTs. RESULTS: The survival rates at 1, 3 and 5 years were 90.0%, 82.0% and 72.0% in all patients. However, in patients with positive CA Ⅱ or Ki-67, the survival rates were 92.0%, 83.0% and 77.0% or 83.0%, 66.6% and 53.0%, respectively. Compared with the negative groups, the survival rates in the positive groups were significantly lower (CA Ⅱ log-rankP = 0.000; Ki-67 logrank P = 0.004). Multivariate Cox analysis revealed that CA Ⅱ, CD117 and Ki-67 were considerable immune factors in prognosis of GIST patients (CA Ⅱ P = 0.043; CD117 P = 0.042; Ki-67 P = 0.007). Besides, tumor diameter, mitotic rate, tumor site, depth of invasion, complete resection, intraoperative rupture, and adjuvant therapy were important prognosis predictive factors. Our study indicated that CA Ⅱ had strong expression in GISTs and the prognosis of GISTs with high CA Ⅱ expression was better than that of GISTs with low or no expression, suggesting that CA Ⅱ is both a diagnostic and prognostic biomarker for GIST. CONCLUSION: CA Ⅱ and Ki-67 are significant prognostic factors for GISTs. 13A Natural Vaccine Candidate Strain Against Cholera显示文摘E1 Tor Vibrio cholerae (EVC) strains may be classifled into two kinds-epidemigenic (EEVC) strains and non-epidemigenic (NEEVC) strains-based on a phage-biotyping system. A large number of EEVC strains have been screened for toxigenic and putative colonization attributes. One such naturally occurring strain (designated IEM 101) has been found which is devoid of genes encoding cholera toxin (CT), accessory cholera enterotoxin (ACE), zonula occludens toxin (ZOT), but possesses RS1 sequences and toxin-coregulated pilus A gene (tcpA) although tcpA is poorly expressed. It expresses type B pili but does not posses type C pili. It is an E1 Tor Ogawa strain and does not cause fluid accumulation in rabbit ileal loop tests. 14Chromosome-scale genome assembly of sweet cherry(Prunus avium L.)cv.Tieton obtained using long-read and Hi-C sequencing显示文摘Sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.Jiawei Wang Weizhen Liu Dongzi Zhu Po Hong Shizhong Zhang Shijun Xiao Yue Tan Xin Chen Li Xu Xiaojuan Zong Lisi Zhang Hairong Wei Xiaohui Yuan Qingzhong Liu 2020Horticulture Research2020,7,1:4
15Therapeutic effect of autologous concentrated growth factor on lower-extremity chronic refractory wounds:A case report显示文摘BACKGROUND Management of chronic refractory wounds is one of the toughest clinical challenges for surgeons.Because of poor blood supply,less tissue coverage,and easy exposure,the lower leg is a common site for chronic refractory wounds.The current therapeutic regimens often lead to prolonged hospital stay and higher healthcare costs.Concentrated growth factor(CGF)is a novel blood extract that contains various growth factors,platelets,and fibrins to promote wound healing process.However,there has been little research reported on the treatment of lower extremity wounds with CGF.CASE SUMMARY A 37-year-old man,without any past medical history,presented an ulcerated chronic wound on his right lower leg.The skin defect exhibited clear boundaries,with a size of 2.0 cm×3.5 cm.The depth of wound was up to the layer of deep fascia.Staphylococcus aureus was detected by bacterial culture.The final diagnosis was right lower extremity ulcers with infection.Cefathiamidine,silver sulfadiazine,and mupirocin cream were applied to control the infection.CGF gel was prepared from the patient’s blood sample,and was used to cover the wound after thorough debridement.The skin wound was successfully healed after three times of CGF treatment.CONCLUSION CGF displays an excellent wound healing promoting effect in patients with lowerextremity chronic refractory wounds.Po Liu Yang Liu Chang-Neng Ke Wei-Shan Li Yue-Ming Liu Shi Xu 2021World Journal of Clinical Cases2021,9,18:3
16Activation of anterior thalamic reticular nucleus GABAergic neurons promotes arousal from propofol anesthesia in mice显示文摘Propofol is widely used for the induction and maintenance of anesthesia,which causes a rapid loss of consciousness.However,the mechanisms underlying the hypnosis effect of propofol are still not fully understood.The thalamic reticular nucleus(TRN)is crucial for regulating wakefulness,sleep rhythm generation,and sleep stability,while the role of TRN in the process of propofol-induced anesthesia is still unknown.Here,we investigated the function of the anterior TRN in propofol general anesthesia.Our results demonstrated that the neural activity of anterior TRN is suppressed during propofol anesthesia,whereas it is robustly activated from anesthesia by recording the calcium signals using fiber photometry technology.The results showed that the activation of anterior TRN neurons by chemogenetic and optogenetic methods shortens the emergency time without changing the induction time.Conversely,chemogenetic or optogenetic inhibition of the TRN neurons leads to a delay in the recovery time.Our study showed that anterior TRN is crucial for behavioral arousal without affecting the induction time of propofol anesthesia.Yanjun Liu Bing Chen Yirong Cai Yuan Han Ying Xia Nanqi Li Bingqian Fan Tianjie Yuan Junli Jiang Po Gao Weifeng Yu Yingfu Jiao Wenxian Li 2021Acta Biochimica et Biophysica Sinica2021,53,7:3
17Hippocampus is more vulnerable to neural damages induced by repeated sevoflurane exposure in the second trimester than other brain areas显示文摘During the rapidly developing and sensitive period of the central nervous system (CNS), a harmful stimulus may have serious consequences. The effect of anesthetic exposure on the development of the offspring’s CNS during pregnancy is still unclear and has been widely concerned. In the present study, we compared the susceptibility of the hippocampus with those of other brain regions in offsprings when the mother mice were exposed to repeated sevoflurane. We found that other than affecting motor sensation, emotion, or social behavior of offspring mice, repeated sevoflurane exposure induced significant memory deficiency. Compared with other brain regions, the hippocampus, which is the key component of the brain serving for learning and memory, was more vulnerable to repeated sevoflurane exposure. We also found that repeated sevoflurane exposure to mother mice could inhibit the axon development of hippocampal neurons. We also predicted that N6-methyladenosine modification of mRNA might play an essential role in the vulnerability of the hippocampus to sevoflurane, while the underlying cellular mechanism needs to be explored in the future. 18Growth of cucumber seedlings in different varieties as affected by light environment显示文摘High-quality cucumber seedlings are a prerequisite for ensuring high yield of cucumbers.With the continuous increase of cucumber planting area in China,the demand for high-quality cucumber seedlings is also increasing.One of the important ways to improve the quality of cucumber seedlings is to improve the light environment by using artificial light source.In this study,three cucumber seedlings(cv.Jintong,cv.Yunv and cv.Xiazhiguang)were grown for 23 d under eight levels of daily light integral(DLI)at 4.3 mol/(m^(2)·d),8.6 mol/(m^(2)·d),10.1 mol/(m^(2)·d),13.0 mol/(m^(2)·d),5.8 mol/(m^(2)·d),11.5 mol/(m^(2)·d),14.4 mol/(m^(2)·d)and 17.3 mol/(m^(2)·d),respectively.The results showed that when DLI was 14.4 mol/(m^(2)·d),the seedling height,stem diameter,total leaf area and shoot dry/fresh weight of all three cucumber cultivars reached the maximum,while hypocotyl length decreased with the increase of light intensity.When DLI was 14.4 mol/(m^(2)·d),Jintong and Xiazhiguang had the highest health index,which were 49.29 and 28.56,respectively,while that of Yunv was 81.59(DLI=14.4 mol/(m^(2)·d)).With the increase of DLI,the photosynthetic capacity of cucumber increases gradually.The highest net photosynthetic rate was shown at DLI of 14.4 mol/(m^(2)·d),while the chlorophyll content of cucumber seedlings of all three cultivars were less affected by DLI.Jintong and Yunv had the highest chlorophyll content when DLI was 8.6 mol/(m^(2)·d)because they were adapted to low-light environment.In conclusion,the DLI of 14.4 mol/(m^(2)·d)for Jintong and Xiazhiguang,while that of 17.3 mol/(m^(2)·d)for Yunv are suggested for the light environment design of factory-cultivated seedling for cucumber.Fang Ji Siqi Wei Nan Liu Lijun Xu Po Yang 2020International Journal of Agricultural and Biological Engineering2020,13,5:2
19Fronts,baroclinic transport,and mesoscale variability of the Antarctic Circumpolar Current in the southeast Indian Ocean显示文摘Fronts,baroclinic transport,and mesoscale variability of the Antarctic Circumpolar Current(ACC) along 115°E are examined on the basis of CTD data from two hydrographic cruises occupied in 1995 as a part of the World Ocean Circulation Experiment(WOCE cruise I9S) and in 2004 as a part of CLIVAR/CO2 repeat hydrography program.The integrated baroclinic transport across I9S section is(97.2×106±2.2×106) m3/s relative to the deepest common level(DCL).The net transport at the north end of I9S,determined by the south Australian circulation system,is about 16.5×106m3/s westward.Relying on a consistent set of water mass criteria and transport maxima,the ACC baroclinic transport,(117×106 ±6.7×10 6)m3/s to the east,is carried along three fronts:the Subantarctic Front(SAF) at a mean latitude of 44°-49°S carries(50.6×10 6 ±13.4×106)m3/s;the Polar Front(PF),with the northern branch(PF-N) at 50.5°S and the southern branch(PFS) at 58°S,carries(51.3×106 ±8.7×106)m3/s;finally,the southern ACC front(SACCF) and the southern boundary of the ACC(SB) consist of three cores between 59°S and 65°S that combined carry(15.2×106 ±1.8×106)m3/s.Mesoscale eddy features are identifiable in the CTD sections and tracked in concurrent maps of altimetric sea level anomalies(SLA) between 44°-48°S and 53°-57°S.Because of the remarkable mesoscale eddy features within the SAF observed in both the tracks of the cruises,the eastward transport of the SAF occurs at two latitude bands separating by 1°.Both the CTD and the altimetric data suggest that the mesoscale variability is concentrated around the Antarctic Polar Frontal Zone(APFZ) and causes the ACC fronts to merge,diverge,and to fluctuate in intensity and position along their paths.DUAN Yongliang HOU Yijun LIU Hongwei HU Po 2012Acta Oceanologica Sinica2012,31,6:2
