Application of atomic force microscopy to living samples from cells to fresh tissues显示文摘Atomic force microscopy is a novel method for imaging and characterization in biomedicine. However, high-resolution imaging of living samples from cells to tissues still remains a challenge. In this paper, two types of AFM working mode (contact mode and tapping mode) which are utilized for the imaging of living cells and tissues are discussed. A new magnetic tapping mode (MAC mode), which is more suitable for living samples, and a novel data collecting system named TREC, are also introduced.CHEN PeiPei DONG HongTao CHEN Long SUN QuanMei HAN Dong 2009Chinese Science Bulletin2009,54,14:8
Parathyroid hormone increases alveolar bone homoeostasis during orthodontic tooth movement in rats with periodontitis via crosstalk between STAT3 and β-catenin显示文摘Periodontitis patients are at risk of alveolar bone loss during orthodontic treatment. The aim of this study was to investigate whether intermittent parathyroid hormone(1–34)treatment(iPTH)could reduce alveolar bone loss during orthodontic tooth movement(OTM)in individuals with periodontitis and the underlying mechanism.A rat model of OTM in the context of periodontitis was established and alveolar bone loss was observed. The control,iPTH and iPTH+stattic groups received injections of vehicle,PTH and vehicle,or PTH and the signal transducer and activator of transcription 3(STAT3)inhibitor stattic, respectively.iPTH prevented alveolar bone loss by enhancing osteogenesis and suppressing bone resorption in the alveolar bone during OTM in rats with periodontitis. This effect of iPTH was along with STAT3 activation and reduced by a local injection of stattic.iPTH promoted osteoblastic differentiation and might further regulate the Wnt/β-catenin pathway in a STAT3-dependent manner. The findings of this study suggest that iPTH might reduce alveolar bone loss during OTM in rats with periodontitis through STAT3/β-catenin crosstalk.Cheng Zhang Tiancheng Li Chenchen Zhou Li Huang Yuyu Li Han Wang Peipei Duan Shujuan Zou Li Mei 2020International Journal of Oral Science2020,12,4:4
Analysis on Collapse Strength of Casing Wear显示文摘Carrying capacity of the casing will reduce after the casing is worn, which seriously affects the subsequent well drilling, well completion, oil extraction and well repair. A lot of researches on calculation of casing wear collapse strength have been done, but few of them focus on collapsing failure mechanism, and influencing factors and law of collapse strength. So, significant difference between estimated value and actual value of collapse strength comes into being. By theoretical analysis, numerical simulation and actual test, the collapsing failure mechanism of casing wear as well as the influencing factors and laws of collapse strength are investigated, and the investigation results show that collapse of crescent casing wear belongs to 'three hinged' instability. The severely-worn position on the casing is yielded into the plastic zone first then deformed greatly, which causes the plastic instability of the whole structure. The casing wear collapse strength presents changes of exponent, power function and linear trend with the residual casing wall thickness, wear radius and axial load, respectively. When the flexibility is less than 10°/30 m, the borehole bending has less impact on casing collapse strength. Thus, the computation model for the casing wear collapsing strength is established by introducing wear radius coefficient and casing equivalent yield strength, at the same time, the model is tested. The test results show that the relative error for the computation model is less than 5%. The research results provide a basis for design of the casing string strength and evaluation of down-hole safety.LIANG Erguo LI Zifeng HAN Yong LI Guoqin GUO Peipei 2013Chinese Journal of Mechanical Engineering2013,26,3:3
Lnc-RP5 Regulates the mi R-129-5p/Notch1/PFV Internal Promoter Axis to Promote the Expression of the Prototype Foamy Virus Transactivator Tas显示文摘Prototype foamy virus(PFV)is a unique retrovirus that infects animals and humans and does not cause clinical symptoms.Long noncoding RNAs(lncRNAs)are believed to exert multiple regulatory functions during viral infections.Previously,we utilized RNA sequencing(RNA-seq)to characterize and identify the lncRNA lnc-RP5-1086 D14.3.1-1:1(lnc-RP5),which is markedly decreased in PFV-infected cells. However, little is known about the function of lnc-RP5 during PFV infection. In this study,we identified lnc-RP5 as a regulator of the PFV transcriptional transactivator(Tas).Lnc-RP5 enhanced the activity of the PFV internal promoter(IP).The expression of PFV Tas was found to be promoted by lnc-RP5.Moreover,mi R-129-5 p was found to be involved in the lnc-RP5-mediated promotion of PFV IP activity, while the Notch1 protein suppressed the activity of PFV IP and the expression of Tas.Our results demonstrate that lnc-RP5 promotes the expression of PFV Tas through the miR-129-5 p/Notch1/PFV IP axis. This work provides evidence that host lnc RNAs can manipulate PFV replication by employing mi RNAs and proteins during an early viral infection.Shanshan Xu Liujun Chen Yinglian Tang Peipei Yuan Jun Yan Yingcheng Zheng Li Huang Zhi Li Yan Sun Song Han Jun Yin Qin Pan Biwen Peng Xiaohua He Wanhong Liu 2020Virologica Sinica2020,35,1:1

Corrosion and irradiation behavior of Fe-based amorphous coating in lead-bismuth eutectic liquids显示文摘Lead-bismuth eutectics (LBE) have considerable potential as a candidate material for accelerator-driven sub-critical systems(ADS).However,LBE corrosion and irradiation damage are two urgent challenges remaining to be solved for impellers of primary pumps. In this study,we have explored the possibility of using Fe-based amorphous coatings to overcome LBE corrosion and concurrently to sustain irradiation damage.Specifically, the Fe_(54)Cr_(18)Mo_(2)Zr_(8)B_(18)amorphous coating was prepared by high-velocity oxygen-fuel (HVOF) spraying on 316L steel and exposed to saturated oxygen static LBE for 500 h at 400℃.The coating with high thermal stability (T_(g)=615℃ and T_(x)=660℃) effectively prevented the substrate steel from being corroded by LBE owing to its unique long-range disordered atomic packing. The coating also exhibited strong irradiation resistance when being subjected to 45 dpa (displacement per atom) Au ion irradiation at room temperature, with no sign of crystallization even at the maximum implantation depth of 300 nm.Consequently, the hardness of the coatings before and after irradiation increased slightly. The current findings shed new insights into understanding corrosion mechanism and irradiation behavior of amorphous solids in LBE and expand the application range of amorphous materials.LU ZhiChao PENG Xiang Yang TANG YuHai HAN ShuYin WU Yuan FU EnGang DING XiangBin HOU Shuo LU GuangYao CAO PeiPei ZHANG YiBo LIU XiongJun MA Dong WANG Hui JIANG SuiHe LU ZhaoPing 2022Science China(Technological Sciences)2022,65,2:1

Strong-field photoionization of intense laser fields by controlling optical singularities显示文摘The spatial features of a light field, such as in the form of the optical singularities, provide a new degree of freedom for the application of light fields in different areas of science and technology. However, although the exploration of structured light is growing rapidly, the investigation of strong-field photoionization using such light fields is noticeably lagging behind. Here, we present an experimental study that reveals the signatures of intense, structured light fields with controlled optical singularities in strong-field photoionization. The different types of optical singularities can be identified through photoionization observables,i.e., photoelectron momentum distributions(PMDs). By concurrently shifting the locations of the phase and polarization singularities, the focal electric field features can be designated, and subsequently, the photoionization appearances can be manipulated. In this process, the behaviors of the different intense optical singularities are clearly visualized by the PMDs. This work will advance both the strong-field science and singularity optics.Yiqi Fang Zhenning Guo Peipei Ge Xueyan Ma Meng Han Xiaoyang Yu Yongkai Deng Qihuang Gong Yunquan Liu 2021Science China(Physics,Mechanics & Astronomy)2021,64,7:1

Biomimetic selenium nanosystems for infectious wound healing显示文摘Bacteria-related wound infection and healing have been a major issue for patients and health-care systems for decades. The rise and evolution of effective treatment will result in significant benefits to human beings. In ad-dition to standard antibacterial drugs,a combination of nanoparticles(NPs)and biological membranes is widely applied as a novel antibacterial agent against infectious pathogens. In this paper, the red blood cell membrane-encapsulated selenium nanoparticles(R-SeNPs)were fabricated for infectious wound healing. The stability, the immune evading capability, and the internal circulation time of the R-SeNPs were notably enhanced compared with those of bare selenium nanoparticles(SeNPs).Moreover,in vivo studies demonstrated the outstanding per-formance of the R-SeNPs in infectious wound healing. The biomimetic selenium nanosystem demonstrated the benefits of the combination of nanotechnology and bionics design and will contribute to wound healing in the future.Mengkun Fang Han Zhang Yuze Wang Hui Zhang Dagan Zhang Peipei Xu 2023Engineered Regeneration2023,4,2:0
High-throughput screening of SARS-CoV-2 main and papain-like protease inhibitors显示文摘The global COVID-19 coronavirus pandemic has infected over 109 million people, leading to over 2 million deaths up to date and still lacking of effective drugs for patient treatment. Here,we screened about 1.8 million small molecules against the main protease(M^(pro))and papain like protease(PL^(pro)),two major proteases in severe acute respiratory syndrome-coronavirus 2 genome, and identified 1851M^(pro)inhibitors and 205 PL^(pro)inhibitors with low nmol/l activity of the best hits.Among these inhibitors, eight small molecules showed dual inhibition effects on both M^(pro)and PL^(pro),exhibiting potential as better candidates for COVID-19 treatment. The best inhibitors of each protease were tested in antiviral assay, with over 40%of M^(pro)inhibitors and over 20%of PL^(pro)inhibitors showing high potency in viral inhibition with low cytotoxicity. The X-ray crystal structure of SARS-CoV-2 M^(pro)in complex with its potent inhibitor 4a was determined at 1.8Åresolution.Together with docking assays, our results provide a comprehensive resource for future research on anti-SARS-CoV-2 drug development.Yi Zang Mingbo Su Qingxing Wang Xi Cheng Wenru Zhang Yao Zhao Tong Chen Yingyan Jiang Qiang Shen Juan Du Qiuxiang Tan Peipei Wang Lixin Gao Zhenming Jin Mengmeng Zhang Cong Li Ya Zhu Bo Feng Bixi Tang Han Xie Ming-Wei Wang Mingyue Zheng Xiaoyan Pan Haitao Yang Yechun Xu Beili Wu Leike Zhang Zihe Rao Xiuna Yang Hualiang Jiang Gengfu Xiao Qiang Zhao Jia Li 2023Protein & Cell2023,14,1:0
