1Scutellarin protects oxygen/glucose-deprived astrocytes and reduces focal cerebral ischemic injury显示文摘Scutellarin, a bioactive flavone isolated from Scutellaria baicalensis, has anti-inflammatory, anti-neurotoxic, anti-apoptotic and anti-oxidative effects and has been used to treat cardiovascular and cerebrovascular diseases in China. However, the mechanisms by which scutellarin mediates neuroprotection in cerebral ischemia remain unclear. The interaction between scutellarin and nicotinamide adenine dinucleotide phosphate oxidase 2(NOX2) was assessed by molecular docking study, which showed that scutellarin selectively binds to NOX2 with high affinity. Cultures of primary astrocytes isolated from the cerebral cortex of neonatal Sprague-Dawley rats were pretreated with 2, 10 or 50 μM scutellarin for 30 minutes. The astrocytes were then subjected to oxygen/glucose deprivation by incubation for 2 hours in glucose-free Dulbecco's modified Eagle's medium in a 95% N2/5% CO_2 incubator, followed by simulated reperfusion for 22 hours. Cell viability was assessed by cell counting kit-8 assay. Expression levels of NOX2, connexin 43 and caspase-3 were assessed by western blot assay. Reactive oxygen species were measured spectrophotometrically. Pretreatment with 10 or 50 μM scutellarin substantially increased viability, reduced the expression of NOX2 and caspase-3, increased the expression of connexin 43, and diminished the levels of reactive oxygen species in astrocytes subjected to ischemia-reperfusion. We also assessed the effects of scutellarin in vivo in the rat transient middle cerebral artery occlusion model of cerebral ischemia-reperfusion injury. Rats were given intraperitoneal injection of 100 mg/kg scutellarin 2 hours before surgery. The Bederson scale was used to assess neurological deficit, and 2,3,5-triphenyltetrazolium chloride staining was used to measure infarct size. Western blot assay was used to assess expression of NOX2 and connexin 43 in brain tissue. Enzyme-linked immunosorbent assay was used to detect 8-hydroxydeoxyguanosine(8-OHd G), 4-hydroxy-2-nonenal(4-HNE) and 3-nitrotyrosin(3-NT) in brain tissue. Immunofluorescence double staining was used to determine the co-expression of caspase-3 and Neu N. Pretreatment with scutellarin improved the neurological function of rats with focal cerebral ischemia, reduced infarct size, diminished the expression of NOX2, reduced levels of 8-OHd G, 4-HNE and 3-NT, and reduced the number of cells co-expressing caspase-3 and Neu N in the injured brain tissue. Furthermore, we examined the effect of the NOX2 inhibitor apocynin. Apocynin substantially increased connexin 43 expression in vivo and in vitro. 2Epac2-deficiency leads to more severe retinal swelling, glial reactivity and oxidative stress in transient middle cerebral artery occlusion induced ischemic retinopathy显示文摘Ischemia occurs in diabetic retinopathy with neuronal loss, edema, glial cell reactivity and oxidative stress. Epacs, consisting of Epac1 and Epac2, are c AMP mediators playing important roles in maintenance of endothelial barrier and neuronal functions. To investigate the roles of Epacs in the pathogenesis of ischemic retinopathy, transient middle cerebral artery occlusion(t MCAO) was performed on Epac1-deficient(Epac1-/-) mice, Epac2-deficient(Epac2-/-) mice, and their wild type counterparts(Epac1+/+ and Epac2+/+). Two-hour occlusion and 22-hour reperfusion were conducted to induce ischemia/reperfusion injury to the retina. After t MCAO, the contralateral retinae displayed similar morphology between different genotypes. Neuronal loss, retinal edema and increase in immunoreactivity for aquaporin 4(AQP4), glial fibrillary acidic protein(GFAP), peroxiredoxin 6(Prx6) were observed in ipsilateral retinae. Epac2-/- ipsilateral retinae showed more neuronal loss in retinal ganglion cell layer, increased retinal thickness and stronger immunostaining of AQP4, GFAP, and Prx6 than those of Epac2+/+. However, Epac1-/- ipsilateral retinae displayed similar pathology as those in Epac1+/+ mice. 3Die design and process optimization of die cast V6 engine blocks显示文摘The use of aluminum, particularly for engine blocks, has grown considerably in the past ten years, and continues to rise in the automotive industry. In order to enhance the quality and engineering functionality of die cast engine blocks, die design and processes have to be optimized. In this study, a computer simulation software, MAGMAsoft, as an advanced tool for optimizing die design and casting process, was employed to virtually visualize cavity filling and patterns of a V6 engine block. The original die design and process was simulated first to establish a baseline. A reality check was used to verify the predicted results. Then, the die modification with a different runner system was made by using a CAD software, Unigraphics (UG). The simulation on combinations of the modified die design and revised process was performed to examine the effect of die modification and process change on flow filling of V6 engine blocks. The simulated prediction indicates that the enhancement of cavity filling due to the die and process modification minimizes the occurrence of defects during casting, and consequently improves the quality of blocks. 4High-speed dual-view band-limited illumination profilometry using temporally interlaced acquisition显示文摘We report dual-view band-limited illumination profilometry(BLIP)with temporally interlaced acquisition(TIA)for high-speed,three-dimensional(3D)imaging.Band-limited illumination based on a digital micromirror device enables sinusoidal fringe projection at up to 4.8 kHz.The fringe patterns are captured alternately by two high-speed cameras.A new algorithm,which robustly matches pixels in acquired images,recovers the object’s 3D shape.The resultant TIA–BLIP system enables 3D imaging over 1000 frames per second on a field of view(FOV)of up to 180 mm×130 mm(corresponding to 1180×860 pixels)in captured images.We demonstrated TIA–BLIP’s performance by imaging various static and fast-moving 3D objects.TIA–BLIP was applied to imaging glass vibration induced by sound and glass breakage by a hammer.Compared to existing methods in multiview phase-shifting fringe projection profilometry,TIA–BLIP eliminates information redundancy in data acquisition,which improves the 3D imaging speed and the FOV.We envision TIA–BLIP to be broadly implemented in diverse scientific studies and industrial applications.Cheng Jiang Patrick Kilcullen Yingming Lai Tsuneyuki Ozaki Jinyang Liang 2020Photonics Research2020,8,11:3

9Raman microscopy mapping for the purity assessment of chirality enriched carbon nanotube networks in thin- film transistors显示文摘With recent improvements in carbon nanotube separation methods, the accurate determination of residual metallic carbon nanotubes in a purified nanotube sample is important, particularly for those interested in using semiconducting single-walled carbon nanotubes (SWCNTs) in electronic device applications such as thin-film transistors (TFTs). This work demonstrates that Raman microscopy mapping is a powerful characterization tool for quantifying residual metallic carbon nanotubes present in highly enriched semiconducting nanotube networks. Raman mapping correlates well with absorption spectroscopy, yet it provides greater differentiation in purity. Electrical data from TFTs with channel lengths of 2.5 and 5μ m demonstrate the utility of the method. By comparing samples with nominal purities of 99.0% and 99.8%, a clear differentiation can be made when evaluating the current on/off ratio as a function of channel length, and thus the Raman mapping method provides a means to guide device fabrication by correlating SWCNT network density and purity with TFT channel scaling.Zhao Li Jianfu Ding Paul Finnie Jacques Lefebvre Fuyong Cheng ChristopherT.
