| 1 | Iron deprivation results in a rapid but not sustained increase of the expression of genes involved in iron metabolism and sulfate uptake in tomato(Solanum lycopersicum L.) seedlings显示文摘Characterization of the relationship between sulfur and iron in both Strategy I and Strategy II plants, has proven that low sulfur availability often limits plant capability to cope with iron shortage. Here it was investigated whether the adaptation to iron deficiency in tomato (Solanum lycopersicum L.) plants was associated with an increased root sulfate uptake and translocation capacity, and modified dynamics of total sulfur and thiols accumulation between roots and shoots. Most of the tomato sulfate transporter genes belonging to Groups 1,2, and 4 were significantly upregulated in iron-deficient roots,as it commonly occurs under S-deficient conditions. The upregulation of the two high affinity sulfate transporter genes,SlST1.1 and SlST1.2, by iron deprivation clearly suggests an increased root capability to take up sulfate. Furthermore, the upregulation of the two low affinity sulfate transporter genes SlST2.1 and SlST4.1 in iron-deficient roots, accompanied by a substantial accumulation of total sulfur and thiols in shoots of iron-starved plants, likely supports an increased root-to-shoot translocation of sulfate. | 2 | Next-generation sequencing:toward an increase in the diagnostic yield in patients with apparently idiopathic spermatogenic failure显示文摘A large proportion of patients with idiopathic spermatogenic failure(SPGF;oligozoospermia or nonobstructive azoospermia[NOA])do not receive a diagnosis despite an extensive diagnostic workup.Recent evidence has shown that the etiology remains undefined in up to 75%of these patients.A number of genes involved in germ-cell proliferation,spermatocyte meiotic divisions,and spermatid development have been called into play in the pathogenesis of idiopathic oligozoospermia or NOA.However,this evidence mainly comes from case reports.Therefore,this study was undertaken to identify the molecular causes of SPGF.To accomplish this,15 genes(USP9Y,NR5A1,KLHL10,ZMYND15,PLK4,TEX15,TEX11,MEIOB,SOHLH1,HSF2,SYCP3,TAF4B,NANOS1,SYCE1,and RHOXF2)involved in idiopathic SPGF were simultaneously analyzed in a cohort of 25 patients with idiopathic oligozoospermia or NOA,accurately selected after a thorough diagnostic workup.After next-generation sequencing(NGS)analysis,we identified the presence of rare variants in the NR5A1 and TEX11 genes with a pathogenic role in 3/25(12.0%)patients.Seventeen other different variants were identified,and among them,13 have never been reported before.Eleven out of 17 variants were likely pathogenic and deserve functional or segregation studies.The genes most frequently mutated were MEIOB,followed by USP9Y,KLHL10,NR5A1,and SOHLH1.No alterations were found in the SYCP3,TAF4B,NANOS1,SYCE1,or RH0XF2 genes.In conclusion,NGS technology,by screening a specific custom-made panel of genes,could help increase the diagnostic rate in patients with idiopathic oligozoospermia or NOA. | Rossella Cannarella Rosita A Condorelli Stefano Paolacci Federica Barbagallo Giulia Guerri Matteo Bertelli Sandro La Vignera Aldo E Calogero | 2021 | Asian Journal of Andrology2021,23,1: | 2 |
| 3 | On the cyclic behaviour of new pier-to-deck connections for short-medium span composite I-girder bridges显示文摘Short-medium span steeleconcrete composite I-girder bridges are becoming more and more popular, because of the reduced construction time and costs. The light weight renders them particularly suitable also is seismic areas, even though this advantage has not been yet adequately investigated. In fact, in case of seismic loading, significant tensile forces might be exhibited at the bottom flange of the steel girder, especially when monolithic connection between deck and pier is adopted, and the connection, conceived for sustaining prevailing hogging bending moments, could experience an excessive damage.With this aim, the cyclic behaviour of new pier-to-deck joints involving the use of concrete cross-beams(CCB) has been recently analysed within the European project(SEQBRI). This paper deals with the results of a wide experimental investigation of these new joints,performed with the aim of characterizing their hysteretic behaviour. Three different typologies were tested: one designed according to the German standard DIN-FB-104, generally utilized for gravity loads only, and other two types proposed for bridges located in low(VAR-1) and medium(VAR-2) seismicity. A series of displacement-imposed cyclic tests highlighted a good seismic behaviour of all the tested solutions. In particular, the DIN-FB-104 VAR C and VAR-2 displayed a similar global behaviour in terms of maximum force and displacement, but with a more pronounced crack development and buckling of steel girders of the first solution.
