| 3 | Sortilin promotes macrophage cholesterol accumulation and aortic atherosclerosis through lysosomal degradation of ATP-binding cassette transporter A1 protein显示文摘Sortilin is closely associated with hyperlipidemia and the risk of atherosclerosis(AS).The role of sortilin and the underlying mechanism in peripheral macrophage are not fully understood.In this study,we investigated the effect of macrophage sortilin on ATP-binding cassette transporter A1(ABCA1)expression,ABCA1-mediated cholesterol efflux,and aortic AS.Macrophage sortilin expression was upregulated by oxidized low-density lipoproteins(ox-LDLs)in both concentration-and time-dependent manners.Its expression reached the peak level when cells were incubated with 50μg/ml ox-LDL for 24 h.Overexpression of sortilin in macrophage reduced cholesterol efflux,leading to an increase in intracellular total cholesterol,free cholesterol,and cholesterol ester.Sortilin was found to bind with ABCA1 protein and suppress macrophage ABCA1 expression,resulting in a decrease in cholesterol efflux from macrophages.The inhibitory effect of sortilin in cholesterol efflux was partially reversed by treatment with chloroquine,a lysosomal inhibitor.On the contrary,the ABCA1 protein level and ABCA1-mediated cholesterol efflux is increased by sortilin short hairpin RNA transfection.The fecal and biliary cholesterol 3H-sterol from cholesterol-laden mouse peritoneal macrophage was reduced by sortilin overexpression through lentivirus vector(LV)-sortilin in low-density lipoprotein receptor knockout mice,which was prevented by co-treatment with chloroquine.Treatment with LV-sortilin reduced plasma high-density lipoprotein and increased plasma ox-LDL levels.Accordingly,aortic lipid deposition and plaque area were exacerbated,and ABCA1 expression was reduced in mice in response to infection with LV-sortilin alone.These effects of LV-sortilin were partially reversed by chloroquine.Sortilin enhances lysosomal degradation of ABCA1 protein and suppresses ABCA1-mediated cholesterol efflux from macrophages,leading to foam cell formation and AS development. | Yuncheng Lv Jing Yang Anbo Gao Sha Sun Xilong Zheng Xi Chen Wei Wan Chaoke Tang Wei Xie Suyun Li Dongming Guo Tianhong Peng Guojun Zhao Liyuan Zhong | 2019 | Acta Biochimica et Biophysica Sinica2019,51,5: | 4 |

| 5 | Behavior and mechanism of graphene oxide-tris(4-aminophenyl)amine composites in adsorption of rare earth elements显示文摘Three-dimensional(3D)graphene oxide-tris(4-aminophenyl)amine(GO-TAPAx:y)composites with different GO-to-TAPA mass ratios(x:y)were prepared by a facile one-step ultrasonic treatment.GOTAPAx:y composites were characterized by scanning electron microscopy(SEM),nitrogen(N2)adsorption-desorption isotherms,Fourier transform infrared spectroscopy(FTIR),X-ray photoelectron spectroscopy(XPS)and thermogravimetric analysis(TGA).Behavior and mechanism of GO-TAPAx:yco mposites for the adsorption of rare earth elements(REEs)were investigated.The adso rption capacities of GO-TAPA1;2 composite for Yb^3+,Er^3+,Nd^3+,La^3+and Y^3+are 30.88,26.52,20.60,11.24 and 10.52 mg/g,respectively.Effects of some important parameters including GO-to-TAPA mass ratios,contact time,reaction temperature and initial Yb^3+concentration on the adsorption were evaluated in detail.FTIR spectral and XPS analyses show that the metal complexations and ion exchanges between nitrogen/oxygen-containing species of the adsorbents and REEs are involved in the adsorption procedure besides the conventional adsorption based on specific surface area,indicating the introduction of specific molecules onto GO is beneficial to its adsorption ability.The convenient reusability and applicability of GO-TAPAx:y composites toward real samples also demonstrate their feasibility in the enrichment of REE ions. | Xiaoru Zhao Xinyu Jiang Dongming Peng Jie Teng Jingang Yu | 2021 | Journal of Rare Earths2021,39,1: | 3 |

| 19 | Time-Domain Full Waveform Inversion Using the Gradient Preconditioning Based on Transmitted Wave Energy显示文摘The gradient preconditioning approach based on seismic wave energy can effectively avoid the huge memory consumption of the gradient preconditioning algorithms based on the Hessian matrix. However, the accuracy of this approach is prone to be influ- enced by the energy of reflected waves. To tackle this problem, the paper proposes a new gradient preconditioning method based on the energy of transmitted waves. The approach scales the gradient through a precondition factor, which is calculated by the ‘ap- proximate transmission wavefield’ simulation based on the nonreflecting acoustic wave equation. The method requires no computing nor storing of the Hessian matrix and its inverse matrix. Furthermore, the proposed method can effectively eliminate the effects of geometric spreading and disproportionality in the gradient illumination. The results of model experiments show that the time-domain full waveform inversion (FWI) using the gradient preconditioning based on transmitted wave energy can achieve higher inversion accuracy for deep high-velocity bodies and their underlying strata in comparison with the one using the gradient preconditioning based on seismic wave energy.
