1Coxsackievirus B3 Infection Triggers Autophagy through 3 Pathways of Endoplasmic Reticulum Stress显示文摘Objective Autophagy is a highly conserved intracellular degradation pathway. Many picornaviruses induce autophagy to benefit viral replication, but an understanding of how autophagy occurs remains incomplete. In this study, we explored whether coxsackievirus B3(CVB3) infection induced autophagy through endoplasmic reticulum(ER) stress. Methods In CVB3-infected HeLa cells, the specific molecules of ER stress and autophagy were detected using Western blotting, reverse transcription polymerase chain reaction(RT-PCR), and confocal microscopy. Then PKR-like ER protein kinase(PERK) inhibitor, inositol-requiring protein-1(IRE1) inhibitor, or activating transcription factor-6(ATF6) inhibitor worked on CVB3-infected cells, their effect on autophagy was assessed by Western blotting for detecting microtubule-associated protein light chain 3(LC3). Results CVB3 infection induced ER stress, and ER stress sensors PERK/eIF2α, IRE1/XBP1, and ATF6 were activated. CVB3 infection increased the accumulation of green fluorescent protein(GFP)-LC3 punctuation and induced the conversion from LC3-Ⅰ to phosphatidylethanolamine-conjugated LC3-1(LC3-Ⅱ). CVB3 infection still decreased the expression of mammalian target of rapamycin(mTOR) and p-mTOR. Inhibition of PERK, IRE1, or ATF6 significantly decreased the ratio of LC3-Ⅱ to LC3-Ⅰ in CVB3-infected HeLa cells. Conclusion CVB3 infection induced autophagy through ER stress in HeL a cells, and PERK, IRE1, and ATF6 a pathways participated in the regulation of autophagy. 2Ginkgo biloba leaf extract improves the cognitive abilities of rats with D-galactose induced dementia显示文摘Standardized Ginkgo biloba leaf extract has been used in clinical trials for its beneficial effects on brain functions,particularly in dementia.Substantial experimental evidences indicated that Ginkgo biloba leaf extract (EGB) protected neuronal cells from a variety of insults.We investigated the effect of EGB on cognitive ability and protein kinase B (PKB) activity in hippocampal neuronal cells of dementia model rats.Rats received an intraperitoneal injection of D-galactose to induce dementia.Forty-eight Spraque-Dawley rats were randomly divided into six groups,including the control group,D-galactose group (Gal),low-dose EGB group (EGB-L),mid-dose EGB group (EGB-M),high-dose EGB group (EGB-H) and treatment group.The EGB-L,EGB-M and EGB-H groups were administered with EGB and D-galactose simultaneously.Y-maze,cresyl violet staining,TUNEL assays and immunohistochemistry staining were performed to detect learning and memory abilities,morphological changes in the hippocampus,neuronal apoptosis and the expressing level of phospho-PKB,respectively.Rats in the Gal group showed decreased abilities of learning and memory,and hippocampal pyramidal cell layer was damaged,while EGB administration improved learning and memory abilities.The Gal group exhibited many stained,condensed nuclei and micronuclei,either isolated or within the cytoplasm of cells (39.5±1.4).Apoptotic cells decreased in the groups of EGB-L (35.9±0.9),EGB-M (16.8±1.0) and EGB-H (10.1±0.8),and there were statistical significances compared with the Gal group.Immunoreactivity of phospho-PKB was localized diffusely throughout the cytosol of cells in all groups,while the immunoreactivity of the Gal group was weak.EGB significantly attenuated learning and memory impairment in a dose-dependent manner,while it could decrease the nmber of TUNEL-positive cells,and increase the activity of PKB.Our results demonstrated that EGB attenuated memory impairment and cell apoptosis in galactose-induced dementia model rats by activating PKB.Nuan Wang Xianming Chen Deqin Geng Hongli Huang Hao Zhou 2013The Journal of Biomedical Research2013,27,1:6
3Effect of air-exposed biocathode on the performance of a Thauera-dominated membraneless single-chamber microbial fuel cell (SCMFC)显示文摘To investigate the effect of air-exposed biocathode(AEB) on the performance of singlechamber microbial fuel cell(SCMFC), wastewater quality, bioelectrochemical characteristics and the electrode biofilms were researched. It was demonstrated that exposing the biocathode to air was beneficial to nitrogen removal and current generation. In Test 1 of 95%AEB, removal rates of ammonia, total nitrogen(TN) and chemical oxygen demand(COD)reached 99.34% ± 0.11%, 99.34% ± 0.10% and 90.79% ± 0.12%, respectively. The nitrogen removal loading rates were 36.38 g N/m^3/day. Meanwhile, current density and power density obtained at 0.7 A/m3 and 104 m W/m^3 respectively. Further experiments on opencircuit(Test 2) and carbon source(Test 3) indicated that this high performance could be attributed to simultaneous biological nitrification/denitrification and aerobic denitrification, as well as bioelectrochemical denitrification. Results of community analysis demonstrated that both microbial community structures on the surface of the cathode and in the liquid of the chamber were different. The percentage of Thauera, identified as denitrifying bacteria, maintained at a high level of over 50% in water, but decreased gradually in the AEB. 4FPGA IMPLEMENTATION OF RSA PUBLIC-KEY CRYPTOGRAPHIC COPROCESSOR BASED ON SYSTOLIC LINEAR ARRAY ARCHITECTURE显示文摘In order to make the typical Montgomery’s algorithm suitable for implementation on FPGA, a modified version is proposed and then a high-performance systolic linear array architecture is designed for RSA cryptosystem on the basis of the optimized algorithm. The proposed systolic array architecture has dis- tinctive features, i.e. not only the computation speed is significantly fast but also the hardware overhead is drastically decreased. 5Synthesis and Adsorption Properties of Polystyrene-supported Chelating Resins Containing Heterocyclic Functional Groups显示文摘A series of new chelating resins with incorporating heterocyclic functional groups: pyridine, thiadizole, benzothizole into macroporous chloromethylated polystyrene were synthesized via hydrophilic spacer arm of polyethylene glycol containing sulfur. 7Synthesis and Adsorption Properties of Chelating Resins Containing Sulfoxide and Heterocyclic Functional Groups显示文摘Several of new chelating resins containing sulfoxide and heterocyclic functional groups (3-aminopyridine and 2-mercaptobenzothiazole) based on macroporous chloromethylated polystyrene were synthesized and characterized by elemental analysis and infrared spectra. Their adsorption capacities towards Zn2+, Cu2+, Pb2+, Hg2+ and Ag+ at pH 3.0 and 6.0 were investigated in detail. 8Restored intestinal integrity,nutrients transporters,energy metabolism,antioxidative capacity and decreased harmful microbiota were associated with IUGR piglet's catch-up growth before weanling显示文摘Background:Intrauterine growth restriction(IUGR)is a major inducer of higher morbidity and mortality in the pig industry and catch-up growth(CUG)before weanling could significantly restore this negative influence.But there was limited knowledge about the underlying mechanism of CUG occurrence.Methods:Eighty litters of newborn piglets were divided into normal birth weight(NBW)and IUGR groups according to birth weight.At 26 d,those piglets with IUGR but over average body weight of eighty litters of weaned piglets were considered as CUG,and the piglets with IUGR still below average body weight were considered as NCUG.This study was conducted to systemically compare the intestinal difference among NBW,CUG and NCUG weaned piglets considering the crucial role of the intestine for piglet growth.Results:The results indicated that the m RNA expression of nutrients(amino acids,glucose,and fatty acids)transporters,and mitochondrial electron transport chain(ETC)I were upregulated in CUG piglets'gut with improved morphology compared with those NCUG,as well as the ratio of P-AMPK/AMPK protein expression which is the indicator of energy metabolism.Meanwhile,CUG piglet's gut showed higher antioxidative capacity with increased SOD and GSHPx activity,decreased MDA levels,as well as higher m RNA expressions of Nrf2,Keap1,SOD,and GSH-Px.Furthermore,inflammatory parameters including TNF-α,IL-1β,IL-6,and IL-12 factors,and the activation of MAPK and NF-κB signaling pathways were significantly elevated in the NCUG intestine,while the protein expression of ZO-1,Occludin and Claudin-1 was reduced.The alpha diversity of fecal microbiota was higher in CUG piglets in contrast with NCUG piglets,and the increased beneficial bacteria and decreased pathogenic bacteria was also observed in CUG piglets.Conclusions:CUG piglet's intestine showed comprehensive restoration including higher nutrients transport,energy metabolism,antioxidant capacity,and intestinal physical barrier,while lower oxidative stress,inflammatory response,and pathogenic microbiota.Chang Cui Caichi Wu Jun Wang Ziwei Ma Xiaoyu Zheng Pengwei Zhu Nuan Wang Yuhua Zhu Wutai Guan Fang Chen 2023Journal of Animal Science and Biotechnology2023,14,1:1

11Angiogenesis function of astragaloside Ⅳ in rats with myocardial infarction via PKD1-HDAC5-VEGF pathway显示文摘OBJECTⅣE This study aimed to investigate the role and mechanism of Astragaloside Ⅳ(AS-Ⅳ) in rats with myocardial infarction.METHODS The myocardial infarction model was established by ligation of the left anterior descending artery.The rats were randomly divided into sham,DMSO,model group,AS-Ⅳ and CID755673 groups.The rats were sacrificed 4 weeks later,and segmental heart samples were used for hematoxylin and eosin staining and masson staining.The expression of PKD1,HDAC5 and VEGF were analyzed using immunohistochemistry,reverse transcription poly.merase chain reaction and western blot.RESULTS Compared with the sham operation and DMSO groups,morphology of myocardium in model group was disordered,accompanied with necrotic myocar.dial cells and obvious collagen tissues.After treatment with AS-Ⅳ,the morphology of myocardium was obviously improved,and the number of new blood vessels increased significantly.However,after treatment with CID755673,the myocardial tissue of rats became disordered again,the necrotic cells increased,and some vessels closed.The expression levels of PKD1,HDAC5 and VEGF mRNA and protein in myocardial tissue of model group were significantly lower than the other four groups(P<0.05),whereas these levels in the AS-Ⅳ group were significantly higher than those in the other four groups(P<0.01).Additionally,the CID755673 group had significantly higher levels of PKD1,HDAC5 and VEGF mRNA and protein than the sham group,DMSO group and model group(P<0.05).CONCLUSION AS-Ⅳ may partly promote the angiogenesis of myocardial tissue in rats with myocardial infarction via the PKD1-HDAC5-VEGF pathway.Lei YANG Nuan LIU Wei ZHAO Xing LI Qian WANG Li HAN Wen-jing QI Yan-ke WANG Bing-yu MAO 2018中国药理学与毒理学杂志2018,32,4:1

19Sedimentary and tectonic evolution of the Banquan pull-apart basin and implications for late Cenozoic dextral strike-slip movement of the Tanlu Fault Zone显示文摘The Banquan Basin is a pull-apart basin with the largest scale and the most prominent structure due to dextral slip of the Tanlu Fault Zone(TLFZ) in late Cenozoic. The depositional history of the basin records the start time and evolution of the right-lateral strike-slip movement of the TLFZ. This paper studies the sedimentary and tectonic evolution of the Banquan Basin by seismic reflection exploration, borehole detection and cosmogenic nuclide chronology. We analyze the coupling relationship between the pull-apart basin and the strike-slip fault and discuss the start time and tectonic significance of the right-lateral strikeslip of the TLFZ. Our study indicates that the Banquan Basin has undergone three evolutionary stages: weak rifting during the pre-pull-apart period, strong extension during the syn-pull-apart period and subsidence during the post-pull-apart period. This implies that the TLFZ, which controls the evolution of the basin, experienced an evolutionary process of weak activity,intensified activity and migration of activity toward the central basin. The sedimentary filling of the basin has strong response to the episodic pull-apart and extension of the basin. Lying upon the basement of the basin, a thin layer of Miocene mudstone slowly accumulated due to local rifting before the strong pull-apart event. Along with the dextral slipping and pull-apart process, the basin was filled with alluvial fan facies, fluvial facies and floodplain facies strata from bottom to top. The latest tectonic movement of the TLFZ in the North China Block in late Cenozoic was dominated by episodic dextral strike-slip motion, and this deformation pattern started at 4.01±1.27 Ma. The latest tectonic deformation in North China since late Cenozoic was governed by eastward extrusion and tectonic orogenesis of the eastern margin of the Qinghai-Xizang Plateau since late Miocene. The eastward thrusting of the Liupanshan fault zone and sinistral shearing of the Qinling fault zone led to the anticlockwise rotation and pushing of secondary blocks in North China, resulting in a planar bookshelf faulting and rotation pattern.
