| 1 | Predictive value of multi-detector computed tomography for accurate diagnosis of serous cystadenoma:Radiologic-pathologic correlation显示文摘AIM:To identify multi-detector computed tomography(MDCT) features most predictive of serous cystadenomas(SCAs),correlating with histopathology,and to study the impact of cyst size and MDCT technique on reader performance.METHODS:The MDCT scans of 164 patients with surgically verified pancreatic cystic lesions were reviewed by two readers to study the predictive value of various morphological features for establishing a diagnosis of SCAs.Accuracy in lesion characterization and reader conf idence were correlated with lesion size(≤3cm or≥3cm) and scanning protocols(dedicated vs routine).RESULTS:28/164 cysts(mean size,39 mm;range,8-92mm) were diagnosed as SCA on pathology.The MDCT features predictive of diagnosis of SCA were microcystic appearance(22/28,78.6%),surface lobulations(25/28,89.3%) and central scar(9/28,32.4%).Stepwise logistic regression analysis showed that only microcystic appearance was signifi cant for CT diagnosis of SCA(P=0.0001).The sensitivity,specificity and PPV of central scar and of combined microcystic appearance and lobulations were 32.4%/100%/100% and 68%/100%/100%,respectively.The reader confidence was higher for lesions>3cm(P=0.02) and for MDCT scans performed using thin collimation(1.25-2.5mm) compared to routine 5 mm collimation exams(P>0.05).CONCLUSION:Central scar on MDCT is diagnostic of SCA but is seen in only one third of SCAs.Microcystic morphology is the most significant CT feature in diagnosis of SCA.A combination of microcystic appearance and surface lobulations offers accuracy comparable to central scar with higher sensitivity. | Anjuli A Shah Nisha I Sainani Avinash Kambadakone Ramesh Zarine K Shah Vikram Deshpande Peter F Hahn Dushyant V Sahani | 2009 | World Journal of Gastroenterology2009,15,22: | 11 |
| 2 | Arginine vasopressin as a target in the treatment of acute heart failure显示文摘Congestive heart failure(CHF) is one of the most common reasons for hospitalization in the United States. Despite multiple different beneficial medications for the treatment of chronic CHF, there are no therapies with a demonstrated mortality benefit in the treatment of acute decompensated heart failure. In fact, studies of inotropes used in this setting have demonstrated more harm than good. Arginine vasopressin has been shown to be up regulated in CHF. When bound to the V1 a and/or V2 receptors, vasopressin causes vasoconstriction, left ventricular remodeling and free water reabsorption. Recently, two drugs have been approved for use that antagonize these receptors. Studies thus far have indicated that these medications, while effective at aquaresis(free water removal), are safe and not associated with increased morbidity such as renal failure and arrhythmias. Both conivaptan and tolvaptan have been approved for the treatment of euvolemic and hypervolemic hyponatremia. Two test groups received D.carota seeds extract[DCSE) at doses of 200 mg/kg and 400 mg/kg. Standard group received silymarin(25 mg/kg) and toxic control received only thioacetamide. | 5 | Parenting preschoolers with autism: Socioeconomic influences on wellbeing and sense of competence显示文摘BACKGROUND Previous research suggests that parents raising a child with autism experience higher levels of psychological distress than parents of typically developing children and parents of children with other developmental disorders. Little is known, however, about the intersection between the effects of socioeconomic status(SES) on the wellbeing and sense of parental competency of parents of preschoolers with autism and how it relates to child symptom severity.AIM To examine the relationship between their child's symptom severity, SES, as measured by neighbourhood advantage and occupational status, on the psychological wellbeing and perceived parenting competence among parents of preschoolers with autism.METHODS Parents of 117 preschool-aged children with a diagnosis of autism spectrum disorder(ASD), 107 mothers and 54 fathers, completed questionnaires about their child's symptoms of ASD and functioning, their own perceptions of their wellbeing and parental competence on entry to an early intervention program in Sydney, Australia. Parents also provided demographic information pertaining to their occupation, level of education attained and address(postcode). All children were also assessed for their severity of symptoms using the Autism Diagnostic Observation Schedule. The Australian Socioeconomic Index of occupationalstatus as a measure of familial SES and the Index of Relative Socio-economic Advantage and Disadvantage as a measure of neighbourhood advantage were used to examine the impact of SES on parental sense of competence and wellbeing.RESULTS Compared to normative populations, both mothers and fathers in our sample reported significantly higher levels of parenting sense of efficacy but lower levels of interest in the parenting role. Mothers also displayed higher levels of satisfaction. Both mothers and fathers displayed higher levels of depression than normative populations with mothers also reporting greater levels of stress and anxiety. Child symptom severity was associated with maternal parenting competency with these relationships amplified among mothers with higher familial SES and who lived in areas of greater neighbourhood advantage.Increased adaptive functioning was associated with better maternal wellbeing,particularly among mothers who lived in areas of greater neighbourhood advantage. Contrastingly, paternal parenting competence was generally not influenced by child adaptive functioning or symptom severity, although for those in higher familial SES brackets, children's symptom severity and maladaptive symptoms were negatively related to paternal sense of parenting efficacy. | 6 | Sustained high level transgene expression in mammalian cells mediated by the optimized piggyBac transposon system显示文摘Sustained,high level transgene expression in mammalian cells is desired in many cases for studying gene functions.Traditionally,stable transgene expression has been accomplished by using retroviral or lentiviral vectors.However,such viral vector-mediated transgene expression is often at low levels and can be reduced over time due to low copy numbers and/or chromatin remodeling repression.The piggyBac transposon has emerged as a promising nonviral vector system for efficient gene transfer into mammalian cells.Despite its inherent advantages over lentiviral and retroviral systems,piggyBac system has not been widely used,at least in part due to their limited manipulation flexibilities.Here,we seek to optimize piggyBac-mediated transgene expression and generate a more efficient,user-friendly piggyBac system.By engineering a panel of versatile piggyBac vectors and constructing recombinant adenoviruses expressing piggyBac transposase(PBase),we demonstrate that adenovirusmediated PBase expression significantly enhances the integration efficiency and expression level of transgenes in mesenchymal stem cells and osteosarcoma cells,compared to that obtained from co-transfection of the CMV-PBase plasmid.We further determine the drug selection timeline to achieve optimal stable transgene expression.Moreover,we demonstrate that the transgene copy number of piggyBac-mediated integration is approximately 10 times higher than that mediated by retroviral vectors.Using the engineered tandem expression vector,we show that three transgenes can be simultaneously expressed in a single vector with high efficiency.Thus,these results strongly suggest that the optimized piggyBac system is a valuable tool for making stable cell lines with sustained,high transgene expression. | Xiang Chen Jing Cui Zhengjian Yan Hongmei Zhang Xian Chen Ning Wang Palak Shah Fang Deng Chen Zhao Nisha Geng Melissa Li Sahitya K.Denduluri Rex C.Haydon Hue H.Luu Russell R.Reid Tong-Chuan He | 2015 | Genes & Diseases2015,2,1: | 5 |

| 8 | Influence of feeding crimped kernel maize silage on the course of subclinical necrotic enteritis in a broiler disease model显示文摘This experiment was carried out with 375 male broilers(Ross 308) from days 1 to 28 to evaluate the influence of crimped kernel maize silage(CKMS) on the manifestation of subclinical necrotic enteritis,microbiota counts, organic acid production and relative weights of gastrointestinal segments. A necrotic enteritis disease model was applied. Birds were allocated into 3 different dietary treatments: a maizebased feed(MBF, control diet), and 2 diets supplemented with 15%(CKMS15) or 30%(CKMS30) of crimped ensiled kernel maize. The disease model involved a 10-time overdose of an attenuated live vaccine against coccidiosis given orally on day 17, followed by oral inoculation of Clostridium perfringens Type A(S48,10~8 to 10~9 bacteria/bird) twice daily on days 18,19, 20 and 21. Scoring of intestinal lesions was performed on days 22, 23, 25 and 28. Ileal and caecal digesta samples were collected for the quantification of selected bacterial groups and organic acids. The results showed that there was no effect of dietary treatments on small intestinal lesion scores(P> 0.05). Lesions scores peaked on days 23 and 25 and decreased again on day 28(P = 0.001). No effect of age on microbiota counts was observed, but feeding of CKMS30 reduced the number of coliforms in ileal contents(P = 0.01). Dietary treatments did not affect organic acid concentrations in ileum and caeca, but there was an effect of age; butyric acid was higher on days 22, 23 and 25 than on day 28(P = 0.04). Acetic acid and propionic acid concentrations in caeca were the highest on days 22 and 28 but the lowest on days 23 and 25. Relative gizzard and caeca weights were increased, and relative ileum weights were decreased when birds were fed CKMS30(P < 0.05). | 10 | Spectroscopic investigations on Dy^(3+) ions doped zinc lead alumino borate glasses for photonic device applications显示文摘This paper presents the structural, optical absorption, photoluminescence (PL) and decay spectral properties of Dy^(3+)ions doped zinc lead alumino borate (ZPAB) glasses to elucidate their possible usage in photonic devices such as w-LEDs and lasers. A broad hump shown by the XRD spectrum recorded for an un-doped ZPAB glass confirms its non-crystalline nature. The Judd-Ofelt (J-O) intensity parameter evaluated from the measured oscillator strengths of the absorption spectral features were used to estimate various radiative parameters and also to understand the nature of bonding between Dy^(3+)ions and oxygen ligands. Under 350 nm excitation, the as-prepared glasses are exhibiting two emission bands^4F_(9/2)→~6H_(15/2)(blue),and^4F_(9/2)→~6H_(13/2)(yellow) at 483 and 575 nm,respectively. From the PL spectra,the Y/B ratio values, CIE chromaticity color coordinates and color correlated temperature (CCT) were evaluated. The experimental lifetimes measured from the decay profiles are decreasing with increase in Dy^(3+)ions concentration in these glasses which may be attributed to the cross-relaxation and nonradiative multiphonon relaxation process. Decay profiles observed for higher concentration were well fitted to Inokuti-Hirayama (I-H) model to understand the energy transfer process and subsequent decrease in experimental lifetimes. | 13 | Performance,litter quality and gaseous odour emissions of broilers fed phytase supplemented diets显示文摘The effect of graded levels of phytase on performance, bone characteristics, excreta/litter quality and odorant emissions was examined using 720 Ross 308 male d-old broilers. A 2 x 4 factorial arrangement of treatments was employed with 6 replicates of 15 birds per pen. Factors were: diets-positive and negative control(PC, NC); phytase-0, 500,1,000.1,500 FTU/kg. The PC was formulated to meet the 2014 Ross 308 nutrient specifications, whereas the NC was formulated with lower Ca(-1.4 g/kg), available P(-1.5 g/kg), Na(-0.3 g/kg), dLys(-0.2 g/kg) and MEn(-0.28 MJ/kg) equivalent to nutrient matrix values for 500 FTU/kg phytase in the starter,grower and finisher periods(i.e.,downspec diet). On d 24, phytase decreased FCR by 1.6, 4.3 and 4.6 points at inclusion levels of 500.1,000 and 1,500 FTU/kg, respectively(P < 0.01) across all diets. Phytase by diet interactions on BW gain were observed on d 24 and 35(P < 0.01). The effect of phytase was much more pronounced in the NC diet as compared with the PC diet.On d 24, phytase increased BW gain by 37, 55 and 68 g in the PC and 127.233 and 173 g in the NC at 500,1,000 and 1,500 FTU/kg, respectively. Diet by phytase interactions were also observed for tibia ash, litter quality and water to feed intake ratio(P < 0.01) with higher phytase effect in NC as compared with PC.Neither diet nor phytase impacted excreta moisture content on d 18 or 21(P> 0.05). Solid phase microextraction gas chromatography-mass spectrometry(SPME-GC-MS) analysis of gaseous emissions on d 39 indicated no difference in the emission of alcohols, aldehydes, ketones, volatile fatty acids and phenols between treatments(P> 0.05). The results indicate that phytase has greater benefits when formulated using nutrient matrix values as compared with adding it over the top in an already nutrient sufficient diet. | 15 | 2,3-Diaryl-3H-imidazo[4,5-b]pyridine derivatives as potential anticancer and anti-inflammatory agents显示文摘In this study we examined the suitability of the 3H-imidazo[4,5-b]pyridine ring system in developing novel anticancer and anti-inflammatory agents incorporating a diaryl pharmacophore. Eight 2,3-diaryl-3H-imidazo[4,5-b]pyridine derivatives retrieved from our in-house database were evaluated for their cytotoxic activity against nine cancer cell lines. The results indicated that the compounds showed moderate cytotoxic activity against MCF-7, MDA-MB-468, K562 and SaOS2 cells, with K562 being the most sensitive among the four cancer cell lines. The eight 2,3-diaryl-3H-imidazo[4,5-b]pyridine derivatives were also evaluated for their COX-1 and COX-2 inhibitory activity in vitro. The results showed that compound 3f exhibited 2-fold selectivity with IC_(50) values of 9.2 and 21.8 mmol/L against COX-2 and COX-1, respectively. Since ancient time, different local ethnic groups have used various part of J.regia for a wide array of ailments including helminthiasis, diarrhea, sinusitis, stomach ache, arthritis, asthma, eczema, scrofula, skin disorders, diabetes mellitus, anorexia, thyroid dysfunction, cancer and infectious diseases. Biological activities of J.regia have been reported in several peer review journals and scientific attention is increasing. The present review attempts to provide comprehensive information on plant description, ethnobotanical use, toxicity, phytochemical profile, pharmacology, clinical studies and current research prospective of the J.regia. Currently, there is an immense interest on isolation/identification of active constituents from walnut and screening those active compounds for pharmacological activities. In addition, researchers are performing clinical trials as well as screening various solvent extracts or fractions of J.regia in several animal diseases models to identify promising therapeutic benefits. | 17 | Lycopene modulates cellular proliferation, glycolysis and hepatic ultrastructure during hepatocellular carcinoma显示文摘AIM To investigate the effect of lycopene extracted from tomatoes(LycT) on ultrastructure, glycolytic enzymes, cell proliferation markers and hypoxia during N-Nitrosodiethylamine(NDEA)-induced hepatocarcinogenesis. METHODS Female BALB/c mice were randomly divided into four groups: The Control, NDEA(200 mg NDEA/kg b.w. given i.p.), LycT(5 mg/kg b.w. given orally on alternate days) and Lyc T + NDEA group. The m RNA and protein expression of various cell proliferation markers(PCNA, Cyclin D1, and p21) were assessed by reverse transcription-polymerase chain reaction and enzyme linked immunosorbent assay, respectively. The ultrastructure of hepatic tissue was analyzed using scanning and transmission electron microscopy. The enzymatic activity of glycolytic enzymes was estimated using standardized protocols, while glucose-6-phosphate dehydrogenase activity level was estimated using a kit obtained from Reckon Diagnostic P. Ltd.(India). RESULTS Uncontrolled proliferation in the liver of NDEA(P ≤ 0.001) mice was evident from the high expression of cell-proliferation associated genes(PCNA, Cyclin D1, and p21) when compared to control and Lyc T mice. In addition, enhanced activities of hexokinase, phosphoglucoisomerase, aldolase, glucose-6-phosphatedehydrogenase and hypoxia-inducible factor-1α were observed in NDEA mice as compared to control(P ≤ 0.001) and Lyc T(P ≤ 0.001) mice. The alterations in hepatic ultrastructure observed in the NDEA group correlated with the changes in the above parameters. Lyc T pre-treatment in NDEA-challenged mice ameliorated the investigated pathways disrupted by NDEA treatment. Moreover, hepatic electron micrographs from the Lyc T + NDEA group showed increased macrophages, apoptotic bodies and well-differentiated hepatocellular carcinoma(HCC) in comparison to undifferentiated HCC as observed in the NDEA treated group. | 18 | Indigenous plant medicines for health care:treatment of Diabetes mellitus and hyperlipidemia显示文摘Medicinal plants have played an important role in treating and preventing a variety of diseases throughout the world. Metabolic syndrome had become a global epidemic, defined as a cluster of three of five criteria: insulin resistance and glucose intolerance, abdominal obesity, hypertension, low high-density cholesterol, and hypertriglyceridemia. The current review focuses on Indian medicinal plant drugs and plants used in the treatment of diabetes and hyperlipidemia. Though there are various approaches to reduce the ill-effects of diabetes and hyperlipidemia and its secondary complications, plant-based drugs are preferred due to lesser side effects and low cost. The current review focuses on twenty-three medicinal plants used in the treatment of Diabetes mellitus and nine medicinal plants used in the treatment of hyperlipidemia. The wealth of knowledge on medicinal plants points to a great potential for research and the discovery of new drugs to fight diseases, including diabetes and hyperlipidemia. | Nisha H. | 19 | Basement tectonics and flexural subsidence along western continental margin of India显示文摘The Paleocene-recent post-rift subsidence history recorded in the Mumbai Offshore Basin off western continental margin of India is examined. Results obtained through 2-D flexural backstripping modelling of new seismic data reveal considerable thermo-tectonic subsidence over last ca. 56 Myr. Reverse postrift subsidence modelling with variable β stretching factor predicts residual topography of ca. 2000 m to the west of Shelf Margin Basin and fails to restore late Paleocene horizon and the underlying igneous basement to the sea level. This potentially implies that:(1) either the igneous basement formed during the late Cretaceous was emplaced under open marine environs; or(2) a laterally varying cumulative subsidence occurred within Mumbai Offshore Basin(MOB) during ca. 68 to ca. 56 Ma. Pre-depositional topographic variations at ca. 56 Ma across the basin could be attributed to the extensional processes such as varied lower crustal underplating along Western Continental Margin of India(WCMI). Investigations about basement tectonics after unroofing of sediments since late Paleocene from this region support a transitional and heavily stretched nature of crust with high to very high β factors. Computations of past sediment accumulation rates show that the basin sedimentation peaked during late Miocene concurrently with uplift of Himalayan-Tibetan Plateau and intensification of Indian monsoon system. Pugsley(Fumariaceae),known as 'Fumitory',is an annual herb found as a common weed all over the plains of India and Pakistan.The whole plant is widely used in traditional and folkloric systems of medicine.In traditional systems of medicine,the plant is reputed for its anthelmintic,diuretic,diaphoretic,laxative,cholagoguc,stomachic and sedative activities and is used to purify blood and in liver obstruction in ethnopharmacology.
