| 1 | Intranasal delivery of nanostructured lipid carriers,solid lipid nanoparticles and nanoemulsions:A current overview of in vivo studies显示文摘The management of the central nervous system(CNS)disorders is challenging,due to the need of drugs to cross the blood-brain barrier(BBB)and reach the brain.Among the various strategies that have been studied to circumvent this challenge,the use of the intranasal route to transport drugs from the nose directly to the brain has been showing promising results.In addition,the encapsulation of the drugs in lipid-based nanocarriers,such as solid lipid nanoparticles(SLNs),nanostructured lipid carriers(NLCs)or nanoemulsions(NEs),can improve nose-to-brain transport by increasing the bioavailability and site-specifc delivery.This review provides the state-of-the-art of in vivo studies with lipid-based nanocarriers(SLNs,NLCs and NEs)for nose-to-brain delivery.Based on the literature available from the past two years,we present an insight into the different mechanisms that drugs can follow to reach the brain after intranasal administration.The results of pharmacokinetic and pharmacodynamics studies are reported and a critical analysis of the differences between the anatomy of the nasal cavity of the different animal species used in in vivo studies is carried out.Although the exact mechanism of drug transport from the nose to the brain is not fully understood and its effectiveness in humans is unclear,it appears that the intranasal route together with the use of NLCs,SLNs or NEs is advantageous for targeting drugs to the brain.These systems have been shown to be more effective for nose-to-brain delivery than other routes or formulations with non-encapsulated drugs,so they are expected to be approved by regulatory authorities in the coming years. | Cláudia Pina Costa Joao Nuno Moreira JoséManuel Sousa Lobo Ana Catarina Silva | 2021 | Acta Pharmaceutica Sinica B2021,11,4: | 5 |
| 2 | Parathyroid ultrasonography and bone metabolic profile of patients on dialysis with hyperparathyroidism显示文摘AIM To evaluate the parathyroid ultrasonography and define parameters that can predict poor response to treatment in patients with secondary hyperparathyroidism due to renal failure.METHODS This cohort study evaluated 85 patients with chronic kidney disease stage V with parathyroid hormone levels above 800 pg/mL. All patients underwent ultrasonography of the parathyroids and the following parameters were analyzed: Demographic characteristics(etiology of chronic kidney disease, gender, age, dialysis vintage, vascular access, use of vitamin D), laboratory(calcium, phosphorus, parathyroid hormone, alkaline phosphatase, bone alkaline phosphatase), and the occurrence of bone changes, cardiovascular events and death. The χ~2 test were used to compare proportions or the Fisher exact test for small sample frequencies. Student t-test was used to detect differences between the two groups regarding continuous variables.RESULTS Fifty-three patients(66.4%) had parathyroid nodules with higher levels of parathyroid hormone, calcium and phosphorus. Sixteen patients underwent parathyroidectomy and had higher levels of phosphorus and calcium × phosphorus product(P = 0.03 and P = 0.006, respectively). They also had lower mortality(32% vs 68%, P = 0.01) and lower incidence of cardiovascular or cerebrovascular events(27% vs 73%, P = 0.02). Calcium × phosphorus product above 55 mg^2/dL^2 [RR 1.48(1.06, 2.08), P = 0.03], presence of vascular calcification [1.33(1.01, 1.76), P = 0.015] and previous occurrence of vascular events [RR 2.25(1.27, 3.98), P < 0.001] were risk factors for mortality in this population. | 11 | Can gadoxetic acid–enhanced magnetic resonance imaging be used to avoid liver biopsy in patients with nonalcoholic fatty liver disease?显示文摘BACKGROUND Nonalcoholic fatty liver disease(NAFLD)is a major cause of liver disease worldwide.The diagnosis of nonalcoholic steatohepatitis(NASH),the most severe form of NAFLD,is crucial and has prognostic and therapeutic implications.However,currently this diagnosis is based on liver biopsy and has several limitations.AIM To evaluate the performance of gadoxetic acid–enhanced magnetic resonance imaging(GA-MRI)in differentiating isolated steatosis from NASH in patients with NAFLD.METHODS In this prospective study,56 patients with NAFLD(18 with isolated steatosis and 38 with NASH)underwent GA-MRI.The contrast enhancement index(CEI)was calculated as the rate of increase of the liver-to-muscle signal intensity ratio from before and 20 min after intravenous GA administration.Between-group differences in mean CEI were examined using Student's t test.The area under the receiver operator characteristic curve and the diagnostic performance of gadoxetic acid–enhanced magnetic resonance imaging were evaluated.RESULTS The mean CEI for all subjects was 1.82±0.19.The mean CEI was significantly lower in patients with NASH than in those with isolated steatosis(P=0.008).Two CEI cut-off points were used:<1.66(94%specificity)to characterize NASH and>2.00(89%sensitivity)to characterize isolated steatosis.CEI values between 1.66 and 2.00 indicated liver biopsy,and the procedure could be avoided in 40%of patients with NAFLD.CONCLUSION GA-MRI is an effective noninvasive method that may be useful for the differentiation of NASH from isolated steatosis,and could help to avoid liver biopsy in patients with NAFLD. | Viviane Brandão Amorim Daniella Braz Parente Fernando Fernandes Paiva Jaime Araújo Oliveira Neto Amanda Almeida Miranda Cláudia Cravo Moreira Flávia Ferreira Fernandes Carlos Frederico FerreiraCampos Nathalie Carvalho Leite Renata de Mello Perez Rosana Souza Rodrigues | 2020 | World Journal of Hepatology2020,12,9: | 0 |
| 12 | Synthesis and structure of a praseodymium (Ⅲ) complex with carboxylate ligand:A thermal and spectroscopic study显示文摘One Pr(Ⅲ) lanthanide ion complex was initially synthesized and characterized by TGA-DSC in air atmosphere, as well as characterized by CHN elemental analysis, defining the stoichiometric ratio as Pr(DMBz)_3. The gaseous products evolved during the thermal decomposition were also monitored in N_2 atmosphere employing TGA/FT-IR system. A crystal structure is obtained by state-of-the-art powder X-rays diffraction methods measured in conventional laboratory equipment and refined by the Rietveld method, which defined it as a monoclinic system of the space group P2_1/C with a polymeric crystal structure, [Pr(DMB_Z)_3]_n. FT-IR theoretical spectrum and time-dependent density functional theory(TD-DFT) were calculated from TGA-DSC and crystalline system data. | 13 | Melatonin restores zinc levels,activates the Keap1/Nrf2 pathway,and modulates endoplasmic reticular stress and HSP in rats with chronic hepatotoxicity显示文摘BACKGROUND Melatonin(MLT)is a potent antioxidant molecule that is shown to have a beneficial effect in various pathological situations,due to its action against free radicals.AIM To evaluate the effect of MLT on carbon tetrachloride(CCl_(4))induced liver injury in rats in terms of oxidative stress,reticular stress,and cell damage.METHODS Twenty male Wistar rats(230-250 g)were divided into four groups:Control rats,rats treated with MLT alone,rats treated with CCl_(4)alone,and rats treated with CCl_(4)plus MLT.CCl_(4)was administered as follows:Ten doses every 5 d,ten every 4 d,and seven every 3 d.MLT was administered intraperitoneally at a dose of 20 mg/kg from the 10th wk to the end of the experiment(16th wk).RESULTS MLT was able to reduce the release of liver enzymes in the bloodstream and to decrease oxidative stress in CCl_(4)treated rats by decreasing the level of thiobarbituric acid reactive substances and increasing superoxide dismutase activity,with a lower reduction in serum zinc levels,guaranteeing a reduction in liver damage;additionally,it increased the expression of nuclear factor(erythroid-derived 2)-like 2 and decreased the expression of Kelch-like ECH-associated protein 1.MLT also decreased the expression of the proteins associated with endoplasmic reticulum stress,i.e.,glucose-regulated protein 78 and activating transcription factor 6,as well as of heat shock factor 1 and heat shock protein 70.CONCLUSION MLT has a hepatoprotective effect in an experimental model of CCl_(4)-induced liver injury,since it reduces oxidative stress,restores zinc levels,and modulates endoplasmic reticulum stress. | Silvia Bona Sabrina Alves Fernandes Andrea C Janz Moreira Graziella Rodrigues Elizângela G Schemitt Fabio Cangeri Di Naso Cláudio A Marroni Norma P Marroni | 2022 | World Journal of Gastrointestinal Pharmacology and Therapeutics2022,13,2: | 0 |
| 14 | Immediate effects of Tuina techniques on working-related musculoskeletal disorder of professional orchestra musicians显示文摘BACKGROUND:Musicians are a prone group to suffer from working-related musculoskeletal disorder(WRMD).Conventional solutions to control musculoskeletal pain include pharmacological treatment and rehabilitation programs but their effi ciency is sometimes disappointing.OBJECTIVE:The aim of this research is to study the immediate effects of Tuina techniques on WRMD of professional orchestra musicians from the north of Portugal.DESIGN,SETTING,PARTICIPANTS AND INTERVENTIONS:We performed a prospective,controlled,single-blinded,randomized study.Professional orchestra musicians with a diagnosis of WRMD were randomly distributed into the experimental group(n=39)and the control group(n=30).During an individual interview,Chinese diagnosis took place and treatment points were chosen.Real acupoints were treated by Tuina techniques into the experimental group and non-specifi c skin points were treated into the control group.Pain was measured by verbal numerical scale before and immediately after intervention.RESULTS:After one treatment session,pain was reduced in 91.8%of the cases for the experimental group and 7.9%for the control group.CONCLUSION:Although results showed that Tuina techniques are effectively reducing WRMD in professional orchestra musicians of the north of Portugal,further investigations with stronger measurements,double-blinding designs and bigger simple sizes are needed. | Cláudia Maria Sousa Luis Moreira Daniela Coimbra Jorge Machado Henry J.Greten | 2015 | Journal of Integrative Medicine2015,13,4: | 0 |
| 15 | Can small-scale altitudinal gradients predict spatial and temporal patterns in tropical forests?显示文摘In tropical montane forests,compositional and structural changes are commonly driven by broad-scale altitudinal variation.Here,given the lack of knowledge on small-scale vegetation changes and temporal dynamics,we address the effects of small-scale variations in soil and altitude on tree community structure,temporal dynamics and phylogenetic diversity in a semi-deciduous tropical forest of the Atlantic Forest Domain,southeastern Brazil.In 2010 and 2015 we sampled thirty plots of 400 m^(2),set up along an altitudinal gradient between 1000 and 1500 m a.s.I..In each plot,we collected soil samples for chemical and textural analyses.We fitted linear models to test the effects of altitude and soil on community dynamics and phylogenetic parameters.Altitude and soil explained the spatial variation in number of individuals and phylogenetic diversity metrics.From lower to higher altitudes,we found decreasing fertility,increasing tree density and decreasing phylogenetic diversity.Altitude significantly influenced the increases in total biomass(from 240.9 to 255.4 t ha^(-1))and individual biomass(from 0.15 to 0.17 t)recorded in the interval.And while community temporal dynamics had rates of 1.96%for mortality,1.02%for recruitment,1.61%for biomass loss and 2.81%for biomass gain,none of them were explained by altitude or soil.Temporal species substitution averaged0.1 in the interval.Altogether,these results suggest that the small-scale variations in altitude and soil likely determine the conditions and resources that drive community assembly and structure,which are expressed by spatial variations along the altitudinal gradient.At the same time,temporal patterns were not influenced by altitude-related environmental variation,resulting in a similar dynamic behaviour across the gradient,suggesting that broad-scale factors may play a more important role than local ones. | Mariana Caroline Moreira Morelli Cléber Rodrigo de Souza Jean Daniel Morel Vinícius Andrade Maia Alisson Borges Miranda Santos Kaline Fernandes Miranda Rubens Manoel dos Santos | 2021 | Journal of Forestry Research2021,32,5: | 0 |
