| 1 | Validation of a serum microRNA panel as biomarkers for early diagnosis of hepatocellular carcinoma post-hepatitis C infection in Egyptian patients显示文摘AIM To investigate the prospective importance of serum micro(mi)RNAs(mi R-125 b, mi R-138 b, mi R-1269, mi R-214-5p, mi R-494, mi R375 and mi R-145) as early biomarkers for the diagnosis of hepatitis C virus(HCV)-related hepatocellular carcinoma(HCC).METHODS Two-hundred and fifty HCV4 a patients, 224 HCV4 aHCC patients, and 84 healthy controls were enrolled in the study. Expression levels of mi R214-5p, mi R-125 b, mi R-1269 and mi R-375 were quantified using quantitative real-time PCR.RESULTS Expression of the selected mi RNAs in serum wassignificantly lower in HCC patients than in the healthy controls, except for mi R-1269 and mi R-494. There was a significant difference between HCC and HCV patients, in particular for HCC and late stage fibrosis, rather than HCV patients and early fibrosis. It is obvious that mi R-1269 was significantly upregulated in HCC cases compared to hepatic fibrosis cases. Each mi RNA can show HCC progression. Multivariate logistic regression analysis indicated that the tested panel of mi RNAs(mi R214-5p, mi R-125 b, mi R-1269 and mi R-375) represent accurate and specific indictors of HCC development.CONCLUSION This study presents a panel of mi RNAs with strong power as putative diagnostic and prognostic biomarkers for HCV-induced HCC. | 2 | Monitoring the impact of climate change and human activities on grassland vegetation dynamics in the northeastern Qinghai-Tibet Plateau of China during 2000–2015显示文摘Climate change and human activities can influence vegetation net primary productivity(NPP), a key component of natural ecosystems. The Qinghai-Tibet Plateau of China, in spite of its significant natural and cultural values, is one of the most susceptible regions to climate change and human disturbances in the world. To assess the impact of climate change and human activities on vegetation dynamics in the grassland ecosystems of the northeastern Qinghai-Tibet Plateau, we applied a time-series trend analysis to normalized difference vegetation index(NDVI) datasets from 2000 to 2015 and compared these spatiotemporal variations with trends in climatic variables over the same time period. The constrained ordination approach(redundancy analysis) was used to determine which climatic variables or human-related factors mostly influenced the variation of NDVI. Furthermore, in order to determine whether current conservation measures and programs are effective in ecological protection and reconstruction, we divided the northeastern Qinghai-Tibet Plateau into two parts: the Three-River Headwater conservation area(TRH zone) in the south and the non-conservation area(NTRH zone) in the north. The results indicated an overall(73.32%) increasing trend of vegetation NPP in grasslands throughout the study area. During the period 2000–2015, NDVI in the TRH and NTRH zones increased at the rates of 0.0015/a and 0.0020/a, respectively. Specifically, precipitation accounted for 9.2% of the total variation in NDVI, while temperature accounted for 13.4%. In addition, variation in vegetation NPP of grasslands responded not only to long-and short-term changes in climate, as conceptualized in non-equilibrium theory, but also to the impact of human activities and their associated perturbations. The redundancy analysis successfully separated the relative contributions of climate change and human activities, of which village population and agricultural gross domestic product were the two most important contributors to the NDVI changes, explaining 17.8% and 17.1% of the total variation of NDVI(with the total contribution >30.0%), respectively. The total contribution percentages of climate change and human activities to the NDVI variation were 27.5% and 34.9%, respectively, in the northeastern Qinghai-Tibet Plateau. | 4 | Circulating tumor and cancer stem cells in hepatitis C virusassociated liver disease显示文摘AIM:To assess the role of circulating tumor cells(CTCs)and cancer stem cells(CSCs)in hepatitis C virus(HCV)-associated liver disease.METHODS:Blood and/or tissue samples were obtained from HCV(genotype 4)-associated hepatocellularcarcinoma patients(HCC;n=120),chronic hepatitis C patients(CH;n=30)and 33 normal control subjects(n=33).Serum levels of alpha-fetoprotein(AFP),alkaline phosphatase,and alanine and aspartate aminotransferases were measured.Cytokeratin 19(CK19)monoclonal antibody was used to enumerate CTCs,and CD133 and CD90 were used to enumerate CSCs by flow cytometry.The expression levels of the CSCs markers(CD133 and CD90)as well as telomerase,melanoma antigen encoding gene 1(MAGE1)and MAGE3 were assessed by RTPCR and quantitative real-time polymerase chain reactions.The number of CTCs and/or the expression levels of CK19,CD133,telomerase,MAGE1 and MAGE3 were correlated to the standard clinicopathologic prognostic factors and disease progression.RESULTS:Levels of AFP,alkaline phosphatase and aspartate aminotransferase were significantly different among the HCC,CH and control groups(P<0.001),whereas alanine aminotransferase differed significantly between patient(HCC and CH)and control groups(P<0.001).At the specified cutoff values determine by flow cytometry,CK19(49.8),CD90(400)and CD133(73)were significantly higher in the blood of HCC patients compared to those in the CH and control groups(P<0.001).On the other hand,CD133 at a 69.5 cutoff was significantly higher in the CH compared to the control group(P≤0.001).Telomerase,MAGE1 and MAGE3RNA were expressed in 55.71%,60.00%and 62.86%of the HCC patients,respectively,but were not detected in patients in the CH or control groups,which were statistically significant(Ps<0.001).The expression levels of telomerase,CD90,MAGE3,CD133 and CK19 were all significantly associated with high tumor grade and advanced stage in HCC patients(all P s<0.05).CONCLUSION:CTC counts and AFP,CK19,telomerase,and MAGE1/MAGE3 expression predict disease progression in patients with HCV,whereas telomerase,MAGE3,CD90,CD133 and CK19 are prognostic mark- | Abeer A Bahnassy Abdel-Rahman N Zekri Ahmed El-Bastawisy Amal Fawzy Marwa Shetta Nehal Hussein Dalia Omran Abdallah A S Ahmed Samir S El-Labbody | 2014 | World Journal of Gastroenterology2014,20,48: | 9 |
| 5 | The adsorptive properties of UiO-66 towards organic dyes: A record adsorption capacity for the anionic dye Alizarin Red S.显示文摘In order to decisively determine the adsorption selectivity of zirconium MOF(UiO-66) towards anionic versus cationic species, the adsorptive removal of the anionic dyes(Alizarin Red S.(ARS), Eosin(E), Fuchsin Acid(FA)and Methyl Orange(MO)) and the cationic dyes(Neutral Red(NR), Fuchsin Basic(FB), Methylene Blue(MB),and Safranine T(ST)) has been evaluated. The results clearly reveal a significant selectivity towards anionic dyes. Such an observation agrees with a plethora of reports of UiO-66 superior affinity towards other anionic species(Floride, PO_4^(3-), Diclofenac sodium, Methylchlorophenoxy-propionic acid, Phenols, CrO_4^(2-), SeO_3^(2-), and AsO_4^-). The adsorption process of ARS as an example has been optimized using the central composite design(CCD). The resultant statistical model indicates a crucial effect of both pH and sorbent mass. The optimum conditions were determined to be initial dye concentration 11.82 mg.L^(-1), adsorbent amount 0.0248 g, shaking time of 36 min and pH 2. The adsorption process proceeds via pseudo-second order kinetics(R^2= 0.999). The equilibrium data were fit to Langmuir and Tempkin models(R^2= 0.999 and 0.997 respectively). The results reveal an exceptional removal for the anionic dye(Alizarin Red S.) with a record adsorption capacity of400 mg·g^(-1). | 8 | An Integrative Meta-analysis of MicroRNAs in Hepatocellular Carcinoma显示文摘We aimed to shed new light on the roles of microRNAs(miRNAs)in liver cancer using an integrative in silico bioinformatics analysis.A new protocol for target prediction and functional analysis is presented and applied to the 26 highly differentially deregulated miRNAs in hepatocellular carcinoma.This framework comprises:(1)the overlap of prediction results by four out of fve target prediction tools,including TargetScan,PicTar,miRanda,DIANA-microT and miRDB(combining machine-learning,alignment,interaction energy and statistical tests in order to minimize false positives),(2)evidence from previous microarray analysis on the expression of these targets,(3)gene ontology(GO)and pathway enrichment analysis of the miRNA targets and their pathways and(4)linking these results to oncogenesis and cancer hallmarks.This yielded new insights into the roles of miRNAs in cancer hallmarks.Here we presented several key targets and hundreds of new targets that are signifcantly enriched in many new cancer-related hallmarks.In addition,we also revealed some known and new oncogenic pathways for liver cancer.These included the famous MAPK,TGFb and cell cycle pathways.New insights were also provided into Wnt signaling,prostate cancer,axon guidance and oocyte meiosis pathways.These signaling and developmental pathways crosstalk to regulate stem cell transformation and implicate a role of miRNAs in hepatic stem cell deregulation and cancer development.By analyzing their complete interactome,we proposed new categorization for some of these miRNAs as either tumor-suppressors or oncomiRs with dual roles.Therefore some of these miRNAs may be addressed as therapeutic targets or used as therapeutic agents.Such dual roles thus expand the view of miRNAs as active maintainers of cellular homeostasis. | Mahmoud ElHefnawi Bangli Soliman Nourhan Abu-Shahba Marwa Amer | 2013 | Genomics, Proteomics & Bioinformatics2013,11,6: | 5 |

Nevertheless, in digitally controlled systems, the PWM and the calculating delays modify the system characteristics in terms of frequency and phase, thus destabilizing the system and degrading the VR-AD performances, mainly in low switching frequencies. Moreover, the stability of the system is greatly affected under weak grid operation characterized by large grid impedance variation. This paper solves these problems by proposing a systematic, robust and optimized design procedure of voltage oriented PI control(VOC) with VRAD. The considered design procedure ensures robust control(sufficient stability margins) and high quality of grid current(reduced steady-state error and minimized THD value) despite the negative impact of digital time delay, grid impedance variation and filter parameters change. | 12 | Antimycobacterial natural products from Moroccan medicinal plants: Chemical composition, bacteriostatic and bactericidal profile of Thymus satureioides and Mentha pulegium essential oils显示文摘Objective: To evaluate the susceptibility of Mycobacterium aurum and Mycobacterium smegmatis in vitro to the essential oils obtained from two medicinal plants: Thymus satureioides(T. satureioides) and Mentha pulegium(M. pulegium), and to study their chemical composition.Methods: The aerial parts of T. satureioides and M. pulegium(leaves and stems) were hydro-distillated using a Clevenger-type apparatus and essential oils were analyzed and identified by gas chromatography-mass spectrometry. Antimycobacterial screening of essential oils was performed on the basis of the inhibition zone diameter by disc diffusion method against two mycobacterial strains whereas the minimal inhibitory concentration and minimal bactericidal concentration were determined by using the micro-dilution method.Results: Chemical analysis of their aerial part's essential oils gave as major compounds,borneol(34.26%), carvacrol(31.21%) and thymol(3.71%) for T. satureioides and R(+)-pulegone(75.48%), carvone(6.66%) and dihydrocarvone(4.64%) for M. pulegium.Thereafter their antimycobacterial effect evaluation, using the micro-dilution method,indicated that minimal inhibitory concentration values of T. satureioides essential oil ranged from 0.062% to 0.015%(v/v) and from 0.125% to 0.031%(v/v) for M. pulegium respectively against Mycobacterium aurum and Mycobacterium smegmatis.Conclusions: It is clearly evident from the results obtained that the Moroccan medicinal plants have great potential to be used as anti-tuberculosis agents. | 13 | MicroRNA signature in patients with hepatocellular carcinoma associated with type 2 diabetes显示文摘BACKGROUND Nonalcoholic steatohepatitis-related cirrhosis is one of the liver complications in type 2 diabetes mellitus(T2DM)and reported to be a risk factor for developing hepatocellular carcinoma(HCC).A reliable screening biomarker of liver cirrhosis(LC)and HCC among T2DM patients is important to reduce the morbidity and mortality of this disease.MicroRNA(miRNA)is considered a key player in HCC and T2DM,and it might be a hidden culprit in diabetes-associated HCC,making it a promising reliable prognostic tool.AIM To investigate the signature of serum miRNAs as early biomarkers for the screening of HCC among diabetic patients.METHODS Expression profiles of miRNAs in serum samples of diabetic LC and diabetic HCC patients were assessed using Illumina sequencing;then,RT-qPCR was used to validate significantly altered miRNAs between the two groups.Candidate miRNAs were tested in serum samples of 200 T2DM patients,270 LC patients,200 HCC patients,and 225 healthy control subjects.Additionally,receiver operating characteristic(ROC)analysis,with area under the curve(AUC),was performed to assess the diagnostic performance of the screened miRNAs for discriminating HCC from LC and nonmalignant patients(LC+T2DM).RESULTS Expression of the sequenced miRNAs in serum was different in HCC vs LCpositive T2DM patients.Two miRNAs(miR-34a,miR-221)were significantly upregulated and five miRNAs(miR-16,miR-23-3p,miR-122-5p,miR-198,miR-199a-3p)were significantly down-regulated in HCC compared to LC patients.Analysis of ROC curve demonstrated that the combination of these seven miRNAs can be used as a reliable biomarker for detection of HCC in diabetic patients,as it could identify HCC with high diagnostic accuracy in diabetic LC patients(AUC=0.993)and in diabetic nonmalignant patients(AUC=0.961).CONCLUSION This study validates a panel of serum miRNAs that can be used as a reliable noninvasive screening biomarker of HCC among T2DM cirrhotic and noncirrhotic patients.The study recommends further research to shed light on a possible role of c-Met in T2DM-associated HCC via the miRNA regulatory pathway. | Moustafa Nouh Elemeery Marwa Anwar Mohamed Marwa Ahmed Madkour Mohammed Mohammed Shamseya Noha Mahmoud Issa Ahmed Noah Badr Doaa Ahmed Ghareeb Cheol-Ho Pan | 2019 | World Journal of Gastroenterology2019,25,42: | 3 |

| 15 | Tumor necrosis factor-α-G308A polymorphism is associated with liver pathological changes in hepatitis C virus patients显示文摘AIM To investigate the association of tumor necrosis factor alpha(TNFα)-G308 A polymorphism with different liver pathological changes in treatment-na?ve Egyptian patients infected with hepatitis C virus(HCV) genotype 4.METHODS This study included 180 subjects,composed of 120 treatment-na?ve chronic HCV patients with different fibrosis grades(F0-F4) and 60 healthy controls. The TNFα-G308 A region was amplified by PCR and the different genotypes were detected by restriction fragment length polymorphism analysis. The TNFα protein was detected by enzyme-linked immunosorbent assay. The influence of different TNFα-G308 A genotypes on TNFα expression and liver disease progression were statistically analyzed. The OR and 95%CI were calculated to assess the relative risk confidence.RESULTS Current data showed that the TNFα-G308 A SNP frequency was significantly different between controls and HCV infected patients(P = 0.001). Both the AA genotype and A allele were significantly higher in late fibrosis patients(F2-F4,n = 60) than in early fibrosis patients(F0-F1,n = 60)(P = 0.05,0.04 respectively). Moreover,the GA or AA genotypes increased the TNFα serum level greater than the GG genotype(P = 0.002). The results showed a clear association between severe liver pathological conditions(inflammation,steatosis and fibrosis) and(GA + AA) genotypes(P = 0.035,0.03,0.04 respectively). The stepwise logistic regression analysis showed that the TNFα genotypes(GA + AA) were significantly associated with liver inflammation(OR = 3.776,95%CI: 1.399-10.194,P = 0.009),severe steatosis(OR = 4.49,95%CI: 1.441-14.0,P = 0.010) and fibrosis progression(OR = 2.84,95%CI: 1.080-7.472,P = 0.034). Also,the A allele was an independent risk factor for liver inflammation(P = 0.003),steatosis(P = 0.003) and fibrosis(P = 0.014). | 16 | Assessment of lnc RNA GAS5, lnc RNA HEIH, lnc RNA BISPR and its m RNA BST2 as serum innovative non-invasive biomarkers: Recent insights into Egyptian patients with hepatitis C virus type 4显示文摘BACKGROUND Hepatitis C virus(HCV)infection and its consequent complications are undeniably a public health burden worldwide,particularly in Egypt.Emerging evidence suggests that many lncRNAs have relevant roles in viral infections and antiviral responses.AIM To investigate the expression profiles of circulating lncRNAGAS5,lncRNAHEIH,lncRNABISPR and mRNABST2 in naïve,treated and relapsed HCV Egyptian patients,to elucidate relation to HCV infection and their efficacy as innovative biomarkers for the diagnosis and prognosis of HCV GT4.METHODS One hundred and thirty HCV-infected Egyptian patients and 20 healthy controls were included in this study.Serum lncRNAs and mRNABST2 were measured using quantitative real-time polymerase chain reaction(qRT-PCR).RESULTS Our results indicated that serum lncRNAGAS5 and LncRNABISPR were upregulated,whereas mRNA BST2 and LncRNA HEIH were downregulated in naïve patients.In contrast,HCV patients treated with sofosbuvir and simeprevir;with sofosbuvir and daclatasvir;or with sofosbuvir,daclatasvir and ribavirin exhibited lower levels of lncRNAGAS5 and lncRNABISPR with higher mRNABST2 compared to naïve patients.Notably,patients relapsed from sofosbuvir and simeprevir showed higher levels of these lncRNAs with lower mRNABST2 compared to treated patients.LncRNAGAS5 and lncRNABISPR were positively correlated with viral load and ALT at P<0.001,whereas mRNABST2 was negatively correlated with viral load at P<0.001 and ALT at P<0.05.Interestingly,a significant positive correlation between lncRNA HEIH and AFP was observed at P<0.001.CONCLUSION Differential expression of these RNAs suggests their involvement in HCV pathogenesis or antiviral response and highlights their promising roles in diagnosis and prognosis of HCV. | Nourhan M El Samaloty Marwa I Shabayek Ramy S Ghait Shohda A El-Maraghy Sherine M Rizk Maha M El-Sawalhi | 2020 | World Journal of Gastroenterology2020,26,2: | 2 |
| 17 | Climate change impact and adaptation on wheat yield, water use and water use efficiency at North Nile Delta显示文摘Investigation of climate change impacts on food security has become a global hot spot.Even so,efforts to mitigate these issues in arid regions have been insufficient.Thus,in this paper,further research is discussed based on data obtained from various crop and climate models.Two DSSATcrop models(CMs)(CERESWheat and N-Wheat)were calibrated with two wheat cultivars(Gemiza9 and Misr1).A baseline simulation(1981-2010)was compared with different scenarios of simulations using three Global Climate Models(GCMs)for the 2030s,2050s and 2080s.Probable impacts of climate change were assessed using the GCMs and CMs under the high emission Representative Concentration Pathway(RCP8.5).Results predicted decreased wheat grain yields by a mean of 8.7%,11.4%and 13.2%in the 2030s,2050s and 2080s,respectively,relative to the baseline yield.Negative impacts of climatic change are probable,despite some uncertainties within the GCMs(i.e.,2.1%,5.0%and 8.0%)and CMs(i.e.,2.2%,6.0%and 9.2%).Changing the planting date with a scenario of plus or minus 5 or 10 days from the common practice was assessed as a potentially effective adaptation option,which may partially offset the negative impacts of climate change.Delaying the sowing date by 10 days(from 20 November to 30 November)proved the optimum scenario and decreased further reduction in wheat yields resulting from climate change to 5.2%,6.8%and 8.5%in the 2030s,2050s and 2080s,respectively,compared with the 20 November scenario.The planting 5-days earlier scenario showed a decreased impact on climate change adaptation.However,the 10-days early planting scenario increased yield reduction under projected climate change.The cultivar Misr1 was more resistant to rising temperature than Gemiza9.Despite the negative impacts of projected climate change on wheat production,water use efficiency would slightly increase.The ensemble of multi-model estimated impacts and adaptation uncertainties of climate change can assist decision-makers in planning climate adaptation strategies. | Marwa Gamal Mohamed Ali Mahmoud Mohamed Ibrahim Ahmed El Baroudy Michael Fullen El-Said Hamad Omar Zheli Ding Ahmed Mohammed Saad Kheir | 2020 | Frontiers of Earth Science2020,14,3: | 2 |

| 19 | Rosmarinic acid attenuates hepatic fibrogenesis via suppression of hepatic stellate cell activation/proliferation and induction of apoptosis显示文摘Objective:To investigate the antifibrotic role of rosmarinic acid(RA),a natural polyphenolic compound,on HSCs activation/proliferation and apoptosis in vitro and in vivo. Methods:The impact of RA on stellate cell line(HSC-T6) proliferation,activation and apoptosis was assessed along with its safety on primary hepatocytes. In vivo,rats were divided into:(i) normal;(ii) thioacetamide(TAA)-intoxicated rats for 12 weeks;(iii) TAA+silymarin or(iv) TAA+RA. At the end of experiment,liver functions,oxidative stress,inflammatory and profibrogenic markers,tissue inhibitor metalloproteinases type-1(TIMP-1) and hydroxyproline(HP) levels were evaluated. Additionally,liver histopathology and immunohistochemical examinations of alpha-smooth muscle actin(α-SMA),caspase-3 and proliferation cellular nuclear antigen(PCNA) were determined. Results:RA exhibited anti-proliferative effects on cultured HSCs in a time and concentration dependent manner showing an IC50 of 276 μg/mL and 171 μg/mL for 24 h and 48 h,respectively,with morphological reversion of activated stellate cell morphology to quiescent form. It significantly improved ALT,AST,oxidative stress markers and reduced TIMP-1,HP levels,inflammatory markers and fibrosis score(S1 vs S4). Furthermore,reduction in α-SMA plus elevation in caspase-3 expressions of HSCs in vitro and in vivo associated with an inhibition in proliferation of damaged hepatocytes were recorded. | 20 | Exploring the role of molecular biomarkers as a potential weapon against gastric cancer: a review of the literature显示文摘Gastric cancer(GC) is a global health problem and a major cause of cancer-related death with high recurrence rates ranging from 25% to 40% for GC patients staging Ⅱ-Ⅳ. Unfortunately, while the majority of GC patients usually present with advanced tumor stage; there is still limited evidence-based therapeutic options. Current approach to GC management consists mainly of; endoscopy followed by, gastrectomy and chemotherapy or chemo-radiotherapy. Recent studies in GC have confirmed that it is a heterogeneous disease. Many molecular characterization studies have been performed in GC. Recent discoveries of the molecular pathways underlying the disease have opened the door to more personalized treatment and better predictable outcome. The identification of molecular markers is a useful tool for clinical managementin GC patients, assisting in diagnosis, evaluation of response to treatment and development of novel therapeutic modalities. While chemotherapeutic agents have certain physiological effects on the tumor cells, the prediction of the response is different from one type of tumor to the other. The specificity of molecular biomarkers is a principal feature driving their application in anticancer therapies.
