| 1 | Short-term effectiveness of golimumab for ulcerative colitis: Observational multicenter study显示文摘AIM To evaluate the real-world effectiveness of golimumab in ulcerative colitis(UC) and to identify predictors of response.METHODS We conducted an observational, prospective and multi-center study in UC patients treated with golimumab, from September 2014 to September 2015. Clinical activity was assessed at wk 0 and 14 with the physician's global clinical assessment(PGA) and the partial Mayo score. Colonoscopies and blood tests were performed, following daily-practice clinical criteria, and the results were recorded in an SPSS database.RESULTS Thirty-three consecutive patients with moderately to severely active UC were included. Among them, 54.5% were female and 42 years was the average age. Thirty percent had left-sided UC(E2) and 70% had extensive UC(E3). All patients had an endoscopic Mayo score of 2 or 3 at baseline. Twenty-seven point three percent were anti-tumor necrosis factor(TNF) treatment na?ve, whereas 72.7% had previously received infliximab and/or adalimumab. Sixty-nine point seven percent showed clinical response and were steroid-free at wk 14(a decrease from baseline in the partial Mayo score of at least 3 points). Based on PGA, the clinical remission and clinical response rates were 24% and 55% respectively. Withdrawal of corticosteroids was observed in 70.8% of steroid-dependent patients at the end of the study. Three out of 10 clinical non-responders needed a colectomy. Mean fecal calprotectin value at baseline was 300 μg/g, and 170.5 μg/g at wk 14. Being anti-TNF treatment na?ve was a protection factor, which was related to better chances of reaching clinical remission. Twenty-seven point three percent of the patients required treatment intensification at 14 wk of followup. | 3 | Arsenite provides a selective signal that coordinates arsenate uptake and detoxification through the regulation of PHR1 stability in Arabidopsis显示文摘In nature,plants acquire nutrients from soils to sustain growth,and at the same time,they need to avoid the uptake of toxic compounds and/or possess tolerance systems to cope with them.This is particularly challenging when the toxic compound and the nutrient are chemically similar,as in the case of phosphate and arsenate.In this study,we demonstrated that regulatory elements of the phosphate starvation response(PSR)coordinate the arsenate detoxification machinery in the cell.We showed that arsenate repression of the phosphate transporter PHT1;1 is associated with the degradation of the PSR master regulator PHR1.Once arsenic is sequestered into the vacuole,PHR1 stability is restored and PHT1;1 expression is recovered.Furthermore,we identified an arsenite responsive SKP1-like protein and a PHR1 interactor F-box(PHIF1)as constituents of the SCF complex responsible for PHR1 degradation.We found that arsenite,the form to which arsenate is reduced for compartmentalization in vacuoles,represses PHT1;1 expression,providing a highly selective signal versus phosphate to control PHT1;1 expression in response to arsenate.Collectively,our results provide molecular insights into a sensing mechanism that regulates arsenate/phosphate uptake depending on the plant’s detoxification capacity. | Cristina Navarro Cristian Mateo-Elizalde Thotegowdanapalya C.Mohan Eduardo Sanchez-Bermejo Oscar Urrutia Maria Nieves Fernandez-Muniz Jose M.Garcia-Mina Riansares Munoz Javier Paz-Ares Gabriel Castrillo Antonio Leyva | 2021 | Molecular Plant2021，14，9： | 3 |
| 4 | DmpIRFs and DmpST:DAMPE instrument response functions and science tools for gamma-ray data analysis显示文摘Observing GeV gamma-rays is an important goal of the DArk Matter Particle Explorer(DAMPE)for indirect dark matter searching and high energy astrophysics. In this work, we present a set of accurate instrument response functions for DAMPE(DmpIRFs) including the effective area, point-spread function and energy dispersion, which are crucial for gamma-ray data analysis based on statistics from simulation data. A dedicated software named DmpST is developed to facilitate the scientific analyses of DAMPE gamma-ray data. Considering the limited number of photons and angular resolution of DAMPE, the maximum likelihood method is adopted in DmpST to better disentangle different source components. | 5 | Trypanosoma rangeli:growth in mammalian cells in vitro and action of a repositioned drug(17-AAG)and a natural extract(Artemisia sp.essential oil)显示文摘Trypanosoma rangeli and T.cruzi are both parasitic unicellular species that infect humans.Unlike T.cruzi,the causative agent of Chagas disease,T.rangeli is an infective and non-pathogenic parasite for humans,but pathogenic for vectors from the Rhodnius genus.Because both species can coexist in different hosts and overlap their infective cycles but very little is known about the infection of T.rangeli in mammalian cells,we decided to characterize both the development of this parasite in cell culture and the effect of therapeutic agents with potential trypanocidal action on it.We found that T.rangeli exhibits a cycle of infection in Vero cells similar to that for T.cruzi and that the repurposed drug,17-AAG,and the natural extract Artemisia sp.essential oil produce a toxic effect on epimastigotes showing a trypanocidal action from the fifth day of culture.Both treatments also affected the infection of trypomastigotes and reduced the capacity of replication of amastigotes of T.rangeli.Since T.cruzi/T.rangeli coinfection cases have been reported,the finding of drugs with potential activity against both species could be significant in the future.Furthermore,studies of susceptibility of both species to drugs could also help to know the different mechanisms of pathogenicity in humans displayed by T.cruzi that are absent in T.rangeli. | Ana Laura CIMADOR Emeli Luciana GALANTE Lucila Ibel MUNOZ Patricia Silvia ROMANO Antonella Denisse LOSINNO Maria Cristina VANRELL | 2019 | BIOCELL2019，43，1： | 1 |
| 6 | An algorithm to resolve γ-rays from charged cosmic rays with DAMPE显示文摘The DArk Matter Particle Explorer(DAMPE),also known as Wukong in China,which was launched on 2015 December 17,is a new high energy cosmic ray and γ-ray satellite-borne observatory.One of the main scientific goals of DAMPE is to observe Ge V-Te V high energy γ-rays with accurate energy,angular and time resolution,to indirectly search for dark matter particles and for the study of high energy astrophysics. Due to the comparatively higher fluxes of charged cosmic rays with respect to γ-rays,it is challenging to identify γ-rays with sufficiently high efficiency,minimizing the amount of charged cosmic ray contamination. In this work we present a method to identify γ-rays in DAMPE data based on Monte Carlo simulations,using the powerful electromagnetic/hadronic shower discrimination provided by the calorimeter and the veto detection of charged particles provided by the plastic scintillation detector.
