1Selective dopamine D3 receptor antagonist YQA14 inhibits morphine-induced behavioral sensitization in wild type,but not in dopamine D3 receptor knockout mice显示文摘Increasing preclinical evidence demonstrates that dopamine D3 receptor (D3R) antagonists are a potential option for the treatment of drug addiction. The reinstatement of the addiction can be triggered by environmental stimuli that acquire motivational salience through repeated associations with the drug’s effects. YQA14 is a novel D3R antagonist that has exhibited pharmacotherapeutic efficacy in reducing cocaine and amphetamine reward and relapse to drug seeking in mice. In this study we investigated the effects of YQA14 on morphine-induced context-specific locomotor sensitization in mice. We showed that repeated injection of YQA14 (6.25–25mg/kg every day ip) prior to morphine (10mg/kg every day sc) not only inhibited the acquisition, but also significantly attenuated the expression of morphine-induced locomotor sensitization. Furthermore, in the expression phase, one single injection of YQA14 (6.25-25mg/kg, ip) dose-dependently inhibited the expression of morphine-induced behavioral sensitization. Moreover, YQA14 inhibited the expression of morphine-induced behavioral sensitization in wild mice (WT), but not in D3R knockout (D3R^-/-) mice in the expression phase. In addition, D3R^-/- mice also displayed the reduction in the expression phase compared with WT mice. 3Tire Dynamics Modeling Method Based on Rapid Test Method显示文摘Combined with the tire dynamics theoretical model,a rapid test method to obtain tire lateral and longitudinal both steady-state and transient characteristics only based on the tire quasi-steady-state test results is proposed.For steady state data extraction,the test time of the rapid test method is half that of the conventional test method.For transient tire characteristics the rapid test method omits the traditional tire test totally.At the mean time the accuracy of the two method is much closed.The rapid test method is explained theoretically and the test process is designed.The key parameters of tire are extracted and the comparison is made between rapid test and traditional test method.The result show that the identification accuracy based on the rapid test method is almost equal to the accuracy of the conventional one.Then,the heat generated during the rapid test method and that generated during the conventional test are calculated separately.The comparison shows that the heat generated during the rapid test is much smaller than the heat generated during the conventional test process.This benefits to the reduction of tire wear and the consistency of test results.Finally,it can be concluded that the fast test method can efficiently,accurately and energy-efficiently measure the steady-state and transient characteristics of the tire.Dang Lu Lei Lu Haidong Wu Wei Wang Manyi Lv 2020Chinese Journal of Mechanical Engineering2020,33,6:1

5Photon-counting distributed free-space spectroscopy显示文摘Spectroscopy is a well-established nonintrusive tool that has played an important role in identifying and quantifying substances,from quantum descriptions to chemical and biomedical diagnostics.Challenges exist in accurate spectrum analysis in free space,which hinders us from understanding the composition of multiple gases and the chemical processes in the atmosphere.A photon-counting distributed free-space spectroscopy is proposed and demonstrated using lidar technique,incorporating a comb-referenced frequency-scanning laser and a superconducting nanowire single-photon detector.It is suitable for remote spectrum analysis with a range resolution over a wide band.As an example,a continuous field experiment is carried out over 72 h to obtain the spectra of carbon dioxide(CO_(2))and semi-heavy water(HDO,isotopic water vapor)in 6 km,with a range resolution of 60 m and a time resolution of 10 min.Compared to the methods that obtain only column-integrated spectra over kilometer-scale,the range resolution is improved by 2-3 orders of magnitude in this work.The CO_(2)and HDO concentrations are retrieved from the spectra acquired with uncertainties as low as±1.2%and±14.3%,respectively.This method holds much promise for increasing knowledge of atmospheric environment and chemistry researches,especially in terms of the evolution of complex molecular spectra in open areas.Saifen Yu Zhen Zhang Haiyun Xia Xiankang Dou Tengfei Wu Yihua Hu Manyi Li Mingjia Shangguan Tianwen Wei Lijie Zhao Lu Wang Pu Jiang Chengjun Zhang Lixing You Leigang Tao Jiawei Qiu 2021Light(Science & Applications)2021,10,11:1
6A Novel Fluorescent Probe for ATP Detection Based on Synergetic Effect of Aggregation-induced Emission and Counterion Displacement显示文摘In this work,a fluorescent probe(TPEBe-I)was developed for adenosine triphosphate(ATP)detection based on the synergetic effect of aggregation-induced emission and counterion displacement.TPEBe-I gave weak emission in aqueous solution due to the heavy-atom effect of counter iodide ion.However,upon the addition of ATP,the new aggregate complex(TPEBe-ATP)was formed between the cationic unit of TPEBe-I and ATP through electrostatic interactions,which not only restricted the intramolecular motion of luminogen but also eliminated the quenching effect of iodide ion.As a result,the fluorescent light-up detection for ATP was successfully achieved.Moreover,TPEBe-I exhibited high selectivity towards ATP and showed a wide linear detection region towards the logarithm of ATP concentration(5—600µmol/L)with a detection limit of 1.0µmol/L,enabling TPEBe-I as a promising probe for ATP quantitative analysis.PAN Quan MA Feiyan PU Xinqing ZHAO Manyi WU Qiling ZHAO Na YANG Jun TANG Ben Zhong 2021Chemical Research in Chinese Universities2021,37,1:1
7Genomic selection of eight fruit traits in pear显示文摘Genomic selection (GS) has the potential to improve selection efficiency and shorten the breeding cycle in fruit tree breeding. In this study,we evaluated the effect of prediction methods, marker density and the training population (TP) size on pear GS for improving its performance and reducing cost. We evaluated GS under two scenarios:(1) five-fold cross-validation in an interspecific pear family;(2) independent validation. Based on the cross-validation scheme, the prediction accuracy (PA) of eight fruit traits varied between 0.33 (fruit core vertical diameter)and 0.65 (stone cell content). Except for single fruit weight, a slightly better prediction accuracy (PA) was observed for the five parametrical methods compared with the two non-parametrical methods. In our TP of 310 individuals, 2 000 single nucleotide polymorphism (SNP) markers were sufficient to make reasonably accurate predictions. PAs for different traits increased by 18.21%-46.98%when the TP size increased from 50to 100, but the increment was smaller (-4.13%-33.91%) when the TP size increased from 200 to 250. For independent validation, the PAs ranged from 0.11 to 0.45 using rrBLUP method. In summary, our results showed that the TP size and SNP numbers had a greater impact on the PA than prediction methods. 8Telomere-to-telomere pear (Pyrus pyrifolia) reference genome reveals segmental and whole genome duplication driving genome evolution显示文摘Previously released pear genomes contain a plethora of gaps and unanchored genetic regions.Here,we report a telomere-to-telomere(T2T)gap-free genome for the red-skinned pear,‘Yunhong No.1’(YH1;Pyrus pyrifolia),which is mainly cultivated in Yunnan Province(southwest China),the pear’s primary region of origin.The YH1 genome is 501.20 Mb long with a contig N50 length of 29.26 Mb.All 17 chromosomes were assembled to the T2T level with 34 characterized telomeres.The 17 centromeres were predicted and mainly consist of centromeric-specific monomers(CEN198)and long terminal repeat(LTR)Gypsy elements(≥74.73%).By filling all unclosed gaps,the integrity of YH1 is markedly improved over previous P.pyrifolia genomes(‘Cuiguan’and‘Nijisseiki’).A total of 1531 segmental duplication(SD)driven duplicated genes were identified and enriched in stress response pathways.Intrachromosomal SDs drove the expansion of disease resistance genes,suggesting the potential of SDs in adaptive pear evolution.A large proportion of duplicated gene pairs exhibit dosage effects or sub-/neo-functionalization,whichmay affect agronomic traits like stone cell content,sugar content,and fruit skin russet.Furthermore,as core regulators of anthocyanin biosynthesis,we found that MYB10 and MYB114 underwent various gene duplication events.Multiple copies of MYB10 and MYB114 displayed obvious dosage effects,indicating role differentiation in the formation of red-skinned pear fruit.In summary,the T2T gap-free pear genome provides invaluable resources for genome evolution and functional genomics.Manyi Sun Chenjie Yao Qun Shu Yingyun He Guosong Chen Guangyan Yang Shaozhuo Xu Yueyuan Liu Zhaolong Xue Jun Wu 2023Horticulture Research2023,10,11:0
