1Differentiation of rat marrow mesenchymal stem cells into pancreatic islet beta-cells显示文摘AIM: To explore the possibility of marrow mesenchymal stem cells (MSC) in vitro differentiating into functional isletlike cells and to test the diabetes therapeutic potency of Islet-like cells. METHODS: Rat MSCs were isolated from Wistar rats and cultured. Passaged MSCs were induced to differentiate into islet-like cells under following conditions: pre-induction with L-DMEM including 10 mmol/L nicotinamide+l mmol/L β-mercaptoethanol+200 mL/L fetal calf serum (FSC) for 24 h, followed by induction with serum free H-DMEM solution including 10 mmol/L nicotinamide+1 mmol/L,β-mercaptoethanol for 10 h. Differentiated cells were observed under inverse microscopy, insulin and nestin expressed in differentiated cells were detected with immunocytochemistry. Insulin excreted from differentiated cells was tested with radioimmunoassay. Rat diabetic models were made to test in vivo function of differentiated MSCs. RESULTS: Typical islet-like clustered cells were observed. Insulin mRNA and protein expressions were positive in differentiated cells, and nestin could be detected in predifferentiated cells. Insulin excreted from differentiated MSCs (446.93±102.28 IU/L) was much higher than that from pre-differentiated MSCs (2.45+0.81 IU/L (P<0.01). Injected differentiated MSCs cells could down-regulate glucose level in diabetic rats. CONCLUSION: Islet-like functional cells can be differentiated from marrow mesenchymal stem cells, which may be a new procedure for clinical diabetes stem -cell therapy, these cells can control blood glucose level in diabetic rats. 2A modified method of discontinuity trace mapping using three-dimensional point clouds of rock mass surfaces显示文摘This paper presents an automated method for discontinuity trace mapping using three-dimensional point clouds of rock mass surfaces.Specifically,the method consists of five steps:(1)detection of trace feature points by normal tensor voting theory,(2)co ntraction of trace feature points,(3)connection of trace feature points,(4)linearization of trace segments,and(5)connection of trace segments.A sensitivity analysis was then conducted to identify the optimal parameters of the proposed method.Three field cases,a natural rock mass outcrop and two excavated rock tunnel surfaces,were analyzed using the proposed method to evaluate its validity and efficiency.The results show that the proposed method is more efficient and accurate than the traditional trace mapping method,and the efficiency enhancement is more robust as the number of feature points increases.Keshen Zhang Wei Wu Hehua Zhu Lianyang Zhang Xiaojun Li Hong Zhang 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,3:6
3Three-dimensional distinct element modeling of fault reactivation and induced seismicity due to hydraulic fracturing injection and backflow显示文摘This paper presents a three-dimensional fully hydro-mechanical coupled distinct element study on fault reactivation and induced seismicity due to hydraulic fracturing injection and subsequent backflow process,based on the geological data in Horn River Basin,Northeast British Columbia,Canada.The modeling results indicate that the maximum magnitude of seismic events appears at the fracturing stage.The increment of fluid volume in the fault determines the cumulative moment and maximum fault slippage,both of which are essentially proportional to the fluid volume.After backflow starts,the fluid near the joint intersection keeps flowing into the critically stressed fault,rather than backflows to the wellbore.Although fault slippage is affected by the changes of both pore pressure and ambient rock stress,their contributions are different at fracturing and backflow stages.At fracturing stage,pore pressure change shows a dominant effect on induced fault slippage.While at backflow stage,because the fault plane is under a critical stress state,any minor disturbance would trigger a fault slippage.The energy analysis indicates that aseismic deformation takes up a majority of the total deformation energy during hydraulic fracturing.A common regularity is found in both fracturing-and backflow-induced seismicity that the cumulative moment and maximum fault slippage are nearly proportional to the injected fluid volume.This study shows some novel insights into interpreting fracturing-and backflowinduced seismicity,and provides useful information for controlling and mitigating seismic hazards due to hydraulic fracturing.Zirui Yin Hongwei Huang Fengshou Zhang Lianyang Zhang Shawn Maxwell 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,4:4
4The Genomes of the Allohexaploid Echinochloa crus-galli and Its Progenitors Provide Insights into Polyploidization-Driven Adaptation显示文摘The hexaploid species Echinochloa crus-galli is one of the most detrimental weeds in crop fields,especially in rice paddies.Its evolutionary history is similar to that of bread wheat,arising through polyploidization after hybridization between a tetraploid and a diploid species.In this study,we generated and analyzed high-quality genome sequences of diploid(E.haploclada),tetraploid(E.oryzicola),and hexaploid(E.crus-galli)Echinochloa species.Gene family analysis showed a significant loss of disease-resistance genes such as those encoding NB-ARC domain-containing proteins during Echinochloa polyploidization,contrary to their significant expansionduring wheat polyploidization,suggesting that natural selection might favor reduced investment in resistance in this weed to maximize its growth and reproduction.In contrast to the asymmetric patterns of genome evolution observed in wheat and other crops,no significant differences in selection pressure were detected between the subgenomes in E.oryzicola and E.crus-galli.In addition,distinctive differences in subgenome transcriptome dynamics during hexaploidization were observed between E.crus-galli and bread wheat.Collectively,our study documents genomic mechanisms underlying the adaptation of a major agricultural weed during polyploidization.The genomic and transcriptomic resources of three Echinochloa species and new insights into the polyploidization-driven adaptive evolution would be useful for future breeding cereal crops.Chu-Yu Ye Dongya Wu Lingfeng Mao Lei Jia Jie Qiu Sangting Lao Meihong Chen Bowen Jiang Wei Tang Qiong Peng Lang Pan Lifeng Wang Xiaoxiao Feng Longbiao Guo Chulong Zhang Elizabeth A.Kellogg Kenneth M.Olsen Lianyang Bai Longjiang Fan 2020Molecular Plant2020,13,9:4
5Quinclorac Resistance in Echinochloa crus-galli from China显示文摘Echinochloa crus-galli is a major weed in rice fields in China,and quinclorac has been long used for its control.Over-reliance of quinclorac has resulted in quinclorac resistance in E.crus-galli.Two resistant(R)E.crus-galli populations from Hunan,China were confirmed to be at least 78-fold more resistant to quinclorac than the susceptible(S)population.No difference in foliar uptake of 14C-labelled quinclorac was detected between the R and S plants.However,a higher level of 14C translocation and a lower level of quinclorac metabolism were found in the R plants.Basal and induced expression levels ofβ-cyanoalanine synthase(β-CAS)gene andβ-CAS activity were not significantly different between the R and S plants.However,the induction expression of 1-aminocyclopropane-1-carboxylic acid oxidase(ACO1)gene by quinclorac treatment was evident in the S plants but not in the R plants.Quinclorac resistance in the two resistant E.crus-galli populations was not likely to be related to foliar uptake,translocation or metabolism of quinclorac,nor to cyanide detoxification viaβ-CAS.Thus,target-site based quinclorac signal reception and transduction and regulation of the ethylene synthesis pathway should be the focus for further research.PENG Qiong HAN Heping YANG Xia BAI Lianyang YU Qin Stephen BPOWLES 2019Rice science2019,26,5:4
6Determination and applications of rock quality designation(RQD)显示文摘Characterization of rock masses and evaluation of their mechanical properties are important and challenging tasks in rock mechanics and rock engineering.Since in many cases rock quality designation(RQD)is the only rock mass classification index available,this paper outlines the key aspects on determination of RQD and evaluates the empirical methods based on RQD for determining the deformation modulus and unconfined compressive strength of rock masses.First,various methods for determining RQD are presented and the effects of different factors on determination of RQD are highlighted.Then,the empirical methods based on RQD for determining the deformation modulus and unconfined compressive strength of rock masses are briefly reviewed.Finally,the empirical methods based on RQD are used to determine the deformation modulus and unconfined compressive strength of rock masses at five different sites including 13 cases,and the results are compared with those obtained by other empirical methods based on rock mass classification indices such as rock mass rating(RMR),Q-system(Q) and geological strength index(GSI).It is shown that the empirical methods based on RQD tend to give deformation modulus values close to the lower bound(conservative) and unconfined compressive strength values in the middle of the corresponding values from different empirical methods based on RMR,Q and GSI.The empirical methods based on RQD provide a convenient way for estimating the mechanical properties of rock masses but,whenever possible,they should be used together with other empirical methods based on RMR,Q and GSI.Lianyang Zhang 2016Journal of Rock Mechanics and Geotechnical Engineering2016,8,3:4

8Evolution and modeling of mine water inflow and hazard characteristics in southern coalfields of China:A case of Meitanba mine显示文摘In this paper,the hydrogeological characteristics in the southern coalfields of China are first briefly outlined.Then,taking the Meitanba mine as an example,the evolution and modeling of mine water inflow are studied.Finally,the hazard characteristics related to mine water and mud inrush are analyzed.The results show that the main mine water sources in the Meitanba mine area are groundwater,surface water and precipitation.The evolution of mine water inflow with time indicates that the water inflow is closely related to the development of karst structures,the amount of water from rainfall infiltration,and the scope of groundwater depression cone.The mine water inflow increases with time due to the increase in mining depth and the expansion of groundwater depression cone.Using the big well method and following the potential superposition principle,a hydrogeological model considering multi-well interactions has been developed to predict the mine water inflow.Based on the monitored data in the Meitanba mine area over a period of nearly 60 years,it is found that with increasing mining depth,the number of water and mud inrush points tended to decrease.However,the average water and mud flow rate per point tended to increase.Jinhai Liu Yanlin Zhao Tao Tan Lianyang Zhang Sitao Zhu Fangyan Xu 2022International Journal of Mining Science and Technology2022,32,3:2
9Aspects of rock permeability显示文摘Effective evaluation of rock permeability is required in different energy,engineering and environmental projects.Although much research has been conducted on rock permeability,it is still one of the most difficult tasks for practicing rock engineers to accurately determine rock permeability.Based on a comprehensive literature review,this paper outlines the key aspects of rock permeability by presenting the representative values of the permeability of different rocks,describing the empirical and semi-empirical correlations for estimating the permeability of rocks,and discussing the main factors affecting the permeability of rocks.The factors discussed include stress,depth,temperature,and discontinuity intensity and aperture.This paper also highlights the scale effect on rock permeability,interconnectivity of discontinuities,and anisotropy of rock permeability.This paper provides the fundamental and essential information required for effective evaluation of rock permeability.Lianyang ZHANG 2013Frontiers of Structural and Civil Engineering2013,7,2:2
10Cultivar-dependent rhizobacteria community and cadmium accumulation in rice: Effects on cadmium availability in soils and iron-plaque formation显示文摘The association between the rhizospheric microbial community and Cd accumulation in rice is poorly understood.A field trial was conducted to investigate the different rhizobacterial communities of two rice cultivars with high Cd accumulation(HA)and low Cd accumulation(LA)at four growth stages.Results showed that the Cd content in the roots of the HA cultivar was 1.23-27.53 higher than that of the LA cultivar(0.08-10.5μg/plant)at four stages.The LA cultivar had a significantly lower Cd availability in rhizosphere and a higher quantity of iron plaque(IP)on the root surface than the HA cultivar at four stages.This resulted in the reduction of Cd concentration in IPs and Cd translocation from IP-to-root.Microbial analysis indicated that the LA cultivar formed a distinct rhizobacterial community from the HA cultivar and had lessα-diversity.The rhizosphere of the LA cultivar was enriched in specific bacterial taxa(e.g.,Massilia and Bacillus)involved in Cd immobilization by phosphate precipitation and IP formation by iron oxidization.However,the rhizosphere in the HA cultivar assembled abundant sulfur-oxidizing bacteria(e.g.,Sulfuricurvum)and iron reduction bacteria(Geobacter).They promoted Cd mobilization and reduced IP formation via the metal redox process.This study reveals a potential approach in which specific rhizobacteria decrease or increase Cd accumulation in rice on contaminated soil and provides a new perspective for secure rice production.Feng Zhang Di Peng Lu Liu Huidan Jiang Lianyang Bai 2022Journal of Environmental Sciences2022,34,6:2
