| 2 | Human urokinase-type plasminogen activator gene-modifiedbone marrow-derived mesenchymal stem cells attenuateliver fibrosis in rats by down-regulating the Wnt signalingpathway显示文摘AIM: To evaluate the therapeutic effects of bone marrow-derived mesenchymal stem cells(BMSCs) with human urokinase-type plasminogen activator(u PA) on liver fibrosis, and to investigate the mechanism of gene therapy.METHODS: BMSCs transfected with adenovirusmediated human urokinase plasminogen activator(Adu PA) were transplanted into rats with CCl4-induced liver fibrosis. All rats were sacrificed after 8 wk, and their serum and liver tissue were collected for biochemical, histopathologic, and molecular analyzes. The degree of liver fibrosis was assessed by hematoxylin and eosin or Masson's staining. Western blot and quantitative reverse transcription-polymerase chain reaction were used to determine protein and m RNA expression levels.RESULTS: Serum levels of alanine aminotransferase, aminotransferase, total bilirubin, hyaluronic acid, laminin, and procollagen type Ⅲ were markedly decreased, whereas the levels of serum albumin were increased by u PA gene modified BMSCs treatment. Histopathology revealed that chronic CCl4-treatment resulted in significant fibrosis while u PA gene modified BMSCs treatment significantly reversed fibrosis. By quantitatively analysing the fibrosis area of liver tissue using Masson staining in different groups of animals, we found that model animals with CCl4-induced liver fibrosis had the largest fibrotic area(16.69% ± 1.30%), while fibrotic area was significantly decreased by BMSCs treatment(12.38% ± 2.27%) and was further reduced by u PA-BMSCs treatment(8.31% ± 1.21%). Both protein and m RNA expression of β-catenin, Wnt4 and Wnt5 a was down-regulated in liver tissues following u PA gene modified BMSCs treatment when compared with the model animals.CONCLUSION: Transplantation of u PA gene modified BMSCs suppressed liver fibrosis and ameliorated liver function and may be a new approach to treating liver fibrosis. | 5 | Tracing the first hematopoietic stem cell generation in human embryo by single-cell RNA sequencing显示文摘Tracing the emergence of the first hematopoietic stem cells(HSCs)in human embryos,particularly the scarce and transient precursors thereof,is so far challenging,largely due to the technical limitations and the material rarity.Here,using single-cell RNA sequencing,we constructed the first genome-scale gene expression landscape covering the entire course of endothelial-to-HSC transition during human embryogenesis.The transcriptomically defined HSC-primed hemogenic endothelial cells(HECs)were captured at Carnegie stage(CS)12-14 in an unbiased way,showing an unambiguous feature of arterial endothelial cells(ECs)with the up-regulation of RUNX1,MYB and ANGPTL Importantly,subcategorizing CD34+CD45-ECs into a CD44+population strikingly enriched HECs by over 10-fold.We further mapped the developmental path from arterial ECs via HSC-primed HECs to hematopoietic stem progenitor cells,and revealed a distinct expression pattern of genes that were transiently over-represented upon the hemogenic fate choice of arterial ECs#including EMCN,PROCR and RUNX1T1.We also uncovered another temporally and molecularly distinct intra-embryonic HEC population,which was detected mainly at earlier CS 10 and lacked the arterial feature.Finally,we revealed the cellular components of the putative aortic niche and potential cellular interactions acting on the HSCprimed HECs.The cellular and molecular programs that underlie the generation of the first HSCs from HECs in human embryos,together with the ability to distinguish the HSC-primed HECs from others,will shed light on the strategies for the production of clinically useful HSCs from pluripotent stem cells. | Yang Zeng Jian He Zhijie Bai Zongcheng Li Yandong Gong Chen Liu Yanli Ni Junjie Du Chunyu Ma Lihong Bian Yu Lan Bing Liu | 2019 | Cell Research2019,29,11: | 14 |
| 6 | Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. CCL5/CCR5 combination is known for facilitating inflammatory responses, as well as inducing the adhesion and migration of different T cell subsets in immune responses. In addition, recent studies have shown that the interaction between CCL5 and CCR5 is involved in various pathological processes including inflammation, chronic diseases, cancers as well as the infection of COVID-19. This review focuses on how CCL5/CCR5 axis participates in the pathological processes of different diseases and their relevant signaling pathways for the regulation of the axis. Moreover, we highlighted the gene therapy and chemotherapy studies for treating CCR5-related diseases, including the ongoing clinical trials. | 9 | ABNORMAL FLOWER AND GRAIN 1 encodes OsMADS6 and determines palea identity and affects rice grain yield and quality显示文摘The palea and lemma are floral organ structures unique to grasses;these structures form the hull and directly affect grain size and quality. However, the molecular mechanisms controlling the development of the hull are not well understood. In this study, we characterized the rice(Oryza sativa) abnormal flower and grain1(afg1) mutant, a new allele of OsMADS6. Similar to previously characterized osmads6 alleles, in the afg1 floret, the palea lost its marginal region and acquired the lemma identity. However, in contrast to other osmads6 alleles, the afg1 mutant showed altered grain size and grain quality, with decreased total starch and amylose contents, and increased protein and soluble sugar contents. The analysis of transcriptional activity suggested that AFG1 is a transcriptional activator and may affect grain size by regulating the expression levels of several genes related to cell expansion and proliferation in the afg1 mutant. | 10 | Design and test of operation parameters for rice air broadcasting by unmanned aerial vehicle显示文摘Considering the difficulty of broadcasting in the small plots and complex terrain in South China,this research aimed to explore a new efficient broadcasting way and figure out advisable operation parameters by using a hollow 12-axis,rotor-wing Unmanned Aerial Vehicle(UAV)which is typically made up of four groups of solid support structure,with each consisting of three axes and two rotor wings.A 3.7 L reverse pyramid-shape seed hopper with a 60 mm×13 mm rectangular outlet at the bottom was designed to realize self-gravity seeding.Rice seed firstly directly falls on the rotating disc driven by direct-current dynamo before being sown.The disc was located 120 mm above the ground,with a diameter of 350 mm.Under constant flight conditions,parameters of the on-board broadcasting devices(dropping speed referring to speed for the outlet and the broadcasting speed for the disc’s rotation speed)determine the uniformity of air broadcasting.The FUTABA T8FG transmitter and receiver system were employed as the remote control device.When the UAV flies at 3 m/s and 2 m above the ground with an expected seeding of 180 grain/m^(2),the RD(Right Down)knob marked 29 and LD(Left Down)knob marked-24 with a disc rotation speed of 900 r/min,the broadcasting lasts for 15 seconds with a 25%opening(1.95 cm^(2))of rectangular outlet.In order to verify the feasibility of air broadcasting parameters,the project team carried out a field broadcasting test by using a hollow multi-rotor UAV at Zhongluotan Test Base in Guangzhou in July,2014.In the square field plots of 0.09 hm^(2),flying path of air broadcasting operation was designed.The collected sampling data of broadcasting quantity per unit area after the test showed that there were,on average,187.4 grains/m^(2)of the five sampling points with a standard deviation of 22.77,and a coefficient of variation of 12.15%which was far smaller than that of artificial broadcasting.The average yield of field broadcasted by UAV is 7705.5 kg/hm^(2)in 2014,implying that rice air broadcasting by UAV is feasible. | Li Jiyu Lan Yubin Zhou Zhiyan Zeng Shan Huang Cong Yao Weixiang Zhang Yang Zhu Qiuyang | 2016 | International Journal of Agricultural and Biological Engineering2016,9,5: | 10 |

The yield strength(YS) and elongation to failure of the composite were dramatically increased by 44.9% and 38.6%, respectively, compared to its alloy counterpart. The significantly enhanced mechanical properties of the as-synthesized composite are mainly ascribed to an improved interfacial bond, grain refinement and good dispersion of CNTs in the matrix via. coating NiO on CNTs.It is shown that the NiO-nanolayer on the CNTs significantly enhances the interfacial bonding strength and effectively prevents the agglomeration of CNTs. | 17 | Establishment of in Vitro Regeneration System of Bitter Melon (Momordica Charantia L. )显示文摘Explants of bitter melon can produce three types of callus: green, yellow green and fragile yellow callus. During the period of inducing callus, the explants are not sensitive to the kinds and proportions of phytohormone. The rate of callus inductivity is about 90.0%. However, in the process of callus differentiating adventitious bud, the kind, proportion and quantity of phytohormone and the type of callus made different result. The adventitious buds were induced successfully on MS medium+6-BA (N6-benzyladenine) 4.0 mg l-1+KT(kinetin) 2.0 mg l-1) by yellow green callus. The frequency was about 66.7 %. Yellow callus did not differentiated adventitious bud. The frequency of green callus differentiation was very low (<15.0 %) even under the most suitable conditions. The MS + ZT (zeatin) 5.0 mgl-1 +KT 0.5 mgl-1 medium was suitable for the proliferation of bud and three weeks later, the coefficient of proliferation was about 5-6. The 1/2 MS + ZT 0.02 mgl-1 or 1/2 MS media were suitable for in vitro rooting of shoot, the shoot on them could produce 6-7 new roots in three weeks. | 18 | A novel thioredoxin reductase inhibitor inhibits cell growth and induces apoptosis in HL-60 and K562 cells显示文摘Human thioredoxin reductase (TrxR) system is associated with cancer cell growth and anti-apoptosis process. Effects of 1,2-bis(1,2-benzisoselenazolone-3(2H)-ketone)ethane (BBSKE),a novel TrxR inhibitor,were investigated on human leu-kemia cell lines HL-60 and K562. BBSKE treatment induced cell growth inhibition and apoptosis in both cell lines. Apoptosis induced by BBSKE is through Bcl-2/Bax and caspase-3 pathways. Ehrlich's ascites carcinoma-bearing mice were used to inves-tigate the anti-tumor effect of BBSKE in vivo. Tumor-bearing mice treated with BBSKE showed an increase of life span with a comparable effect to cyclophosphamide (CTX). | 19 | The molecular characterization of maize B chromosome specific AFLPs显示文摘The origin and evolution of B chromosomes could be explained by the specific DNA sequence on them. But the specific sequences known were quite limited. To investigate maize B chromosome sqicific DNA sequeces, maize genomes with and without B chromosomes were analyzed by AFLP. Only 5 markers were found specific to genomes with B chromosomes among about 2000 AFLP markers. Southern hybridization and sequence analysis revealed that only the sequence of M8-2D was a B chromosome specific sequence. This sequence contained the telomeric repeat unit AGGGTTT conserved in plant chromosome telomeres. In addition, the sequence of M8-2D shared low homology to clones from maize chromosome 4 centromere as well. | 20 | A Small-Scale Medication of Leflunomide as a Treatment of COVID-19 in an Open-Label Blank-Controlled Clinical Trial显示文摘We recently reported that inhibitors against human dihydroorotate dehydrogenase(DHODH)have broad-spectrum antiviral activities including their inhibitory efficacies on SARS-CoV-2 replication in infected cells.However,there are limited data from clinical studies to prove the application of DHODH inhibitors in Coronavirus disease 2019(COVID-19)patients.In the present study,we evaluated Leflunomide,an approved DHODH inhibitor widely used as a modest immune regulator to treat autoimmune diseases,in treating COVID-19 disease with a small-scale of patients.Cases of 10 laboratory-confirmed COVID-19 patients of moderate type with obvious opacity in the lung were included.Five of the patients were treated with Leflunomide,and another five were treated as blank controls without a placebo.All the patients accepted standard supportive treatment for COVID-19.The patients given Leflunomide had a shorter viral shedding time(median of5 days)than the controls(median of 11 days,P=0.046).The patients given Leflunomide also showed a significant reduction in C-reactive protein levels,indicating that immunopathological inflammation was well controlled.No obvious adverse effects were observed in Leflunomide-treated patients,and they all discharged from the hospital faster than controls.This preliminary study on a small-scale compassionate use of Leflunomide provides clues for further understanding of Leflunomide as a potential antiviral drug against COVID-19. | Ke Hu Mengmei Wang Yang Zhao Yunting Zhang Tao Wang Zhishui Zheng Xiaochen Li Shaolin Zeng Dong Zhao Honglin Li Ke Xu Ke Lan | 2020 | Virologica Sinica2020,35,6: | 7 |
