2Effect of dietary supplementation with protease on growth performance,nutrient digestibility, intestinal morphology, digestive enzymes and gene expression of weaned piglets显示文摘This study was conducted to investigate the effect of dietary protease supplementation on the growth performance, nutrient digestibility, intestinal morphology, digestive enzymes and gene expression in weaned piglets. A total of 300 weaned piglets(21 days of age Duroc x Large White x Landrace; initial BW = 6.27 ± 0.45 kg) were randomly divided into 5 groups. The 5 diets were: 1) positive control diet(PC), 2) negative control diet(NC), and 3) protease supplementations, which were 100, 200, and 300 mg per kg NC diet. Results indicated that final BW, ADG, ADFI, crude protein digestibility, enzyme activities of stomach pepsin, pancreatic amylase and trypsin, plasma total protein, and intestinal villus height were higher for the PC diet and the supplementations of 200 and 300 mg protease per kg NC diet than for the NC diet(P < 0.05). Supplementations of 200 and 300 mg protease per kg NC diet significantly increased the ratio of villus height to crypt depth(VH:CD) of duodenum, jejunum and ileum compared with NC diet(P < 0.05). Feed to gain ratio, diarrhea index, blood urea nitrogen, and diamine oxidase were lower for the PC diet and supplementations of 200 and 300 mg protease per kg NC diet than for the NC diet(P < 0.05). Piglets fed the PC diet had a higher peptide transporter 1(PepTl) mRNA abundance in duodenum than piglets fed the NC diet(P < 0.05), and supplementations of 100, 200 and 300 mg protease per kg NC diet increased the PepTl mRNA abundance in duodenum(P < 0.05) comparing with the NC diet. Piglets fed the PC diet had a higher b0,+AT mRNA abundance in jejunum than piglets fed the NC diet(P < 0.05), and supplementations of 200 and 300 mg protease per kg NC diet increased the b0,+AT mRNA abundance in jejunum and ileum comparing with the NC diet(P < 0.05). 3Numerical Simulation of Typhoon Muifa(2011) Using a Coupled Ocean-Atmosphere-Wave-Sediment Transport(COAWST) Modeling System显示文摘The newly developed Coupled Ocean-Atmosphere-Wave-Sediment Transport(COAWST) Modeling System is applied to investigate typhoon-ocean interactions in this study. The COAWST modeling system represents the state-of-the-art numerical simulation technique comprising several coupled models to study coastal and environmental processes. The modeling system is applied to simulate Typhoon Muifa(2011), which strengthened from a tropical storm to a super typhoon in the Northwestern Pacific, to explore the heat fluxes exchanged among the processes simulated using the atmosphere model WRF, ocean model ROMS and wave model SWAN. These three models adopted the same horizontal grid. Three numerical experiments with different coupling configurations are performed in order to investigate the impact of typhoon-ocean interaction on the intensity and ocean response to typhoon. The simulated typhoon tracks and intensities agree with observations. Comparisons of the simulated variables with available atmospheric and oceanic observations show the good performance of using the coupled modeling system for simulating the ocean and atmosphere processes during a typhoon event. The fully coupled simulation that includes a ocean model identifies a decreased SST as a result of the typhoon-forced entrainment. Typhoon intensity and wind speed are reduced due to the decrease of the sea surface temperature when using a coupled ocean model. The experiments with ocean coupled to atmosphere also results in decreased sea surface heat flux and air temperature. The heat flux decreases by about 29% compared to the WRF only case. The reduction of the energy induced by SST decreases, resulting in weakening of the typhoon. 4The influence of ENSO on sea surface temperature variations in the China seas显示文摘Positive SST anomalies usually appear in remote ocean such as the China seas during an ENSO event.By analyzing the monthly data of HadISST from 1950 to 2007,it shows that the interannual component of SST anomalies peak approximately 10 months after SST anomalies peak in the eastern equatorial Pacific.As the ENSO event progresses,the positive SST anomalies spread throughout the China seas and eastward along the Kuroshio extension.Atmospheric reanalysis data demonstrate that changes in the net surface heat flux entering into the China seas are responsible for the SST variability.During El Ni o,the western north Pacific anticyclone is generated,with anomalous southwester lies prevailing along the East Asian coast.This anticyclone reduces the mean surface wind speed which decreases the surface heat flux and then increases the SST.The delays between the developing of this anticyclone and the south Indian Ocean anticyclone with approximately 3–6 months cause the 2–3 months lag of the surface heat flux between the China seas and the Indian Ocean.The northwestern Pacific anticyclone is the key process bridging the warming in the eastern equatorial Pacific and that in the China seas.LIU Na WANG Hui LING Tiejun FENG Licheng 2013Acta Oceanologica Sinica2013,32,9:4
5A 31-year Global Diurnal Sea Surface Temperature Dataset Created by an Ocean Mixed-Layer Model显示文摘A dataset of hourly sea surface temperature(SST) from the period 1 January 1982 to 31 December 2012, and covering the global ocean at a resolution of 0.3°× 0.3°, was created using a validated ocean mixed-layer model(MLSST). The model inputs were heat flux and surface wind speed obtained from the Coupled Forecast System Reanalysis dataset. Comparisons with in-situ data from the Tropical Atmosphere Ocean array and the National Data Buoy Center showed that the MLSST fitted very well with observations, with a mean bias of 0.07℃, and a root-mean-square error(RMSE) and correlation coefficient of 0.37℃ and 0.98, respectively. Also, the MLSST fields successfully reproduced the diurnal cycle of SST in the in-situ data, with a mean bias of -0.005℃ and RMSE of 0.26℃. The 31-year climatology revealed that the diurnal range was small across most regions, with higher values in the eastern and western equatorial Pacific, northern Indian Ocean, western Central America, northwestern Australia, and several coastal regions. Significant seasonal variation of diurnal SST existed in all basins. In the Atlantic and Pacific basins, this seasonal pattern was oriented north–south, following the variation in solar insolation, whereas in the Indian basin it was dominated by monsoonal variability. 14Soil and fine root-associated microbial communities are niche dependent and influenced by copper fungicide treatment during tea plant cultivation显示文摘Dear Editor,Fungicide treatment has a profound effect on controlling plant pathogens in modern agriculture,however,it also carries the risk of undesirable outcomes.For decades,scientists have been concerned about the harmful impacts of heavy metals like copper(Cu)on crop performance and soil microorganisms.Use of various copper fungicides,like Bordeaux mixture,have been a component of conventional agricultural practices to control fungal and bacterial pathogens,especially in vineyards,tea gardens,or fruit tree orchards[9,10].This treatment increases the accumulation of high levels of Cu in surface soils,and despite the critical role of Cu as an essential trace element in wide biological and metabolic processes,it becomes toxic to plants when applied at high levels[4].The regular application of copper fungicides has also been linked to affecting microbial communities at the levels of diversity[8],population structure[2],abundance,and growth[1,3].Understanding the undesired effects of fungicides on microorganisms’beneficial activities is therefore important for evaluating the hazards associated with the fungicide used in agriculture.Yet,the effects of copper fungicide on full microbial communities remains relatively understudied,especially in tea plants.Thus,we herein explored the inf luence of Bordeaux mixture under different management regimes(raking or without raking leaf litter)on microbial communities of root,bulk soil,and rhizosphere compartments of tea plants planted in a ten-year-old tea garden.We provided insights into the ecological consequences of tea management practices that might help to identify specific fungicide treatment regimens,environmental characteristics,and microbial community members to minimize the negative environmental outcomes and optimize the positive anti-pathogen aspects of fungicide treatment.Ali Inayat Mallano Jie Yu Tabys Dina Fangdong Li Tiejun Ling Naveed Ahmad Jeffrey Bennetzen Wei Tong 2023Horticulture Research2023,10,3:0

17microR-1294-5p inhibits glycolytic metabolism of non-small cell lung cancer cells via targeting TMPRSS11B显示文摘Non-smallcell lung cancer(NSCLC)cells intake and consume glucose at high efficiency by aerobic glycolysis to maintain robust cell growth and resist cell death.MicroRNAs(miRNAs)have been known to play pivotal roles in NSCLC development partly through mediating glycolysis.However,only a few miRNAs have been experimentally confirmed as critical regulators of glycolysis in NSCLC.TCGA datasets were analyzed to screen for differentially expressed miRNAs between NSCLC and normal tissues.The function of miR-1294-5p was determined in NSCLC cells by cell proliferation,glucose uptake,lactate release,and Extracellular Acidification Rate(ECAR)assays.The target of miR-1294-5p was predicted by TargetScan and miRDB,which was further validated by flow cytometry analysis,RT-qPCR,western blotting,a dual-luciferase reporter assay,and RNA immunoprecipitation(RIP)assay.In the present study,it was found that miR-1294-5p was a significantly downregulated miRNA in lung adenocarcinoma(LUAD)and lung squamous cell carcinoma(LUSC).The overexpression of miR-1294-5p inhibited glycolysis,lactate export,ECAR,and cell proliferation in NSCLC cells.Analysis with bioinformatic tools,Western Blotting,RT-qPCR,flow cytometry analysis,dual-luciferase reporter assay,and RIP assay showed that miR-1294-5p directly bound to complementary sites in the 3’-Untranslated Region(UTR)of TMPRSS11B resulted in downregulation of TMPRSS11B expression.In addition,transfection of recombinant TMPRSS11B rescued the functions of miR-1294-5p on glycolysis and proliferation of NSCLC cells.The findings provided novel insights for understanding the regulation of glycolytic metabolism in NSCLC.JI ZHU XIYING BO GENGXI JIANG SHIHUA YAO TIEJUN ZHAO LING CHEN 2021BIOCELL2021,45,3:0
