1Undulated 2D Covalent Organic Frameworks Based on Bowl-Shaped Cyclotricatechylene显示文摘Two 2D wavy hexagonal hexahydroxyl cyclotricatechylene (CTC) based COFs, CTC-COF-2 and CTC-COF-3were synthesized through solvothermal reaction. The bowl-shaped conformation caused CTC skeletons packed in acolumnar manner with the same oriented units, thus forming an undulated structure. 13Derivation of totipotent-like stem cells with blastocyst-like structure forming potential显示文摘It is challenging to derive totipotent stem cells in vitro that functionally and molecularly resemble cells from totipotent embryos.Here,we report that a chemical cocktail enables the derivation of totipotent-like stem cells,designated as totipotent potential stem(TPS)cells,from 2-cell mouse embryos and extended pluripotent stem cells,and that these TPS cells can be stably maintained long term in vitro.TPS cells shared features with 2-cell mouse embryos in terms of totipotency markers,transcriptome,chromatin accessibility and DNA methylation patterns.In vivo chimera formation assays show that these cells have embryonic and extraembryonic developmental potentials at the single-cell level.Moreover,TPS cells can be induced into blastocyst-like structures resembling preimplantation mouse blastocysts.Mechanistically,inhibition of HDAC1/2 and DOT1L activity and activation of RARγsignaling are important for inducing and maintaining totipotent features of TPS cells.Our study opens up a new path toward fully capturing totipotent stem cells in vitro.Yaxing Xu Jingru Zhao Yixuan Ren Xuyang Wang Yulin Lyu Bingqing Xie Yiming Sun Xiandun Yuan Haiyin Liu Weifeng Yang Yenan Fu Yu Yu Yinan Liu Rong Mu Cheng Li Jun Xu Hongkui Deng 2022Cell Research2022,32,6:1

17Chlamydia trachomatis infection in the genital tractis associated with inflammation and hypospermia inthe infertile male of China显示文摘Chlamydia trachomatis (CT) infection is the most prevalent sexually transmitted bacterial disease worldwide. However, unlikethat in female infertility, the role of CT infection in male infertility remains controversial. The objective of this retrospective studywas to explore the impacts of CT infection in the genital tract on sperm quality, sperm acrosin activity, antisperm antibody levels,and inflammation in a large cohort of infertile males in China. A total of 7154 semen samples were collected from infertile malesubjects, 416 of whom were CT positive (CT+ group) and 6738 of whom were CT negative (CT− group), in our hospital betweenJanuary 2016 and December 2018. Routine semen parameters (semen volume, pH, sperm concentration, viability, motility,morphology, etc.), granulocyte elastase levels, antisperm antibody levels, and sperm acrosin activity were compared between theCT+ and CT− groups. Our results showed that CT infection was significantly correlated with an abnormally low semen volume, aswell as an increased white blood cell count and granulocyte elastase level (all P < 0.05) in the semen of infertile males;otherroutine semen parameters were not negatively impacted. The antisperm antibody level and sperm acrosin activity were not affectedby CT infection. 18Radioiodinated tyrosine based carbon dots with efficient renal clearance for single photon emission computed tomography of tumor显示文摘Nan oparticles with effective tumor accumulation and efficient renal clearance have attracted significant interests for clinical applications.Weprepared 2.5 nm tyrosine based carbon dots(TCDs)with phenolic hydroxyl groups on the surface for directly 125I labeling.The 125I labeledpolyethylene glycol(PEG)functionalized TCDs(125I-TCDPEGs)showed excellent radiochemical stability both in vitro and in vivo.Due to theenhanced permeability and retentio n effect,these 125I-TCDPEGs demonstrated a tumor accumulatio n around 4%-5%of the injected doseper gram(ID/g)for U87MG,4T1,HepG2 and MCF7 tumor-bearing mice at 1 h post-injection.Meanwhile,the 125I-TCDPEGs also could befast renally excreted,with less than 0.6%ID/g left in the liver and spleen within 24 h.These radioactive carb on dots not only can be used forcellular fluorescence imaging due to their intrinsic optical property,but are also effective single photon emission computed tomography(SPECT)imaging agents for tumor.Together with their excellent biocompatibility and stability,we anticipate these 125I-TCDPEGs of great potential for earlytumor diagnosis in clinic.Whafs more,our TCDPEGs are also proved to be feasible carriers for other iodine isotopes such as 127I and 131I fordifferent biomedical application.Nian Liu Yiyue Shi Jingru Guo Hai Li Qiang Wang Menglin Song Zhiyuan Shi Le He Xinhui Su Jin Xie Xiaolian Sun 2019Nano Research2019,12,12:1
19The role of CYP1A1/2 in cholesterol ester accumulation provides a new perspective for the treatment of hypercholesterolemia显示文摘Cholesterol is an important precursor of many endogenous molecules.Disruption of cholesterol homeostasis can cause many pathological changes,leading to liver and cardiovascular diseases.CYP1A is widely involved in cholesterol metabolic network,but its exact function has not been fully elucidated.Here,we aim to explore how CYP1A regulates cholesterol homeostasis.Our data showed that CYP1A1/2 knockout(KO)rats presented cholesterol deposition in blood and liver.The serum levels of low-density lipoprotein cholesterol,high-density lipoprotein cholesterol and total cholesterol were significantly increased in KO rats.Further studies found that the lipogenesis pathway(LXRa-SREBP1-SCD1)of KO rats was activated,and the key protein of cholesterol ester hydrolysis(CES1)was inhibited.Importantly,lansoprazole can significantly alleviate rat hepatic lipid deposition in hypercholesterolemia models by inducing CYP1A.Our findings reveal the role of CYP1A as a potential regulator of cholesterol homeostasis and provide a new perspective for the treatment of hypercholesterolemia.Jian Lu Xuyang Shang Bingyi Yao Dongyi Sun Jie Liu Yuanjin Zhang He Wang Jingru Shi Huaqing Chen Tieliu Shi Mingyao Liu Xin Wang 2023Acta Pharmaceutica Sinica B2023,13,2:0
20Color-tunable Ca10Na(PO4)7:Ce^3+/Tb^3+/Mn^2+ phosphor via energy transfer显示文摘A series of Ca_(10)Na(PO_4)_7:Ce^(3+)/Tb^(3+)/Mn^(2+)(CNPO:Ce^(3+)/Tb^(3+)/Mn^(2+)) phosphors with high brightness were synthesized by high-temperature solid-state method. X-ray diffraction(XRD), scanning electron microscopy(SEM), diffuse reflectance spectra(DRS), photo luminescence(PL) spectra, luminescence decay curves and thermally stability were performed to characterize the as-prepared samples. For Ce^(3+)-doped samples, an intense and broad band emission is present under 265 nm excitation. When Ce^(3+) and Tb^(3+)are codoped, energy transfer(ET) process from Ce3+ to Tb3+ is demonstrated with electric dipole-dipole interaction. The internal and external quantum efficiencies(QEs) of CNPO:0.15 Ce^(3+), 0.04 Tb^(3+), 0.005 Mn^(2+)are measured to 76.79% and 54.11% under 265 nm excitation and temperature-dependent PL intensity can remain 51.78% at 150 ℃ of its initial intensity at 25 ℃.
