| 1 | Biomass-Based Rice (Oryza sativa L.) Aboveground Architectural Parameter Models显示文摘To quantify the relationships between rice plant architecture parameters and the corresponding organ biomass, and to research on functional structural plant models of rice plant, this paper presented a biomass-based model of aboveground architectural parameters of rice (Oryza sativa L.) in the young seedling stage, designed to explain effects of cultivars and environmental conditions on rice aboveground morphogenesis at the individual leaf level. Various model variables, including biomass of blade and blade length, were parameterized for rice based on data derived from an outdoor experiment with rice cv. Liangyou 108, 86You 8, Nanjing 43, and Yangdao 6. The organ dimensions of rice aboveground were modelled taking corresponding organ biomass as an independent variable. Various variables in rice showed marked consistency in observation and simulation, suggesting possibilities for a general rice architectural model in the young seedling stage. Our descriptive model was suitable for our objective. However, they can set the stage for connection to physiological model via biomass and development of functional structural rice models (FSRM), and start with the localized production and partitioning of assimilates as affected by abiotic growth factors. | 3 | Impact of diabetes on promoting the growth of breast cancer显示文摘Background:Type Ⅱ diabetes mellitus(DM2)is a significant risk factor for cancers,including breast cancer.However,a proper diabetic breast cancer mouse model is notwell-established for treatment strategy design.Additionally,the precise diabetic signaling pathways that regulate cancer growth remain unresolved.In the present study,we established a suitable mouse model and demonstrated the pathogenic role of diabetes on breast cancer progression.Methods:We successfully generated a transgenic mouse model of human epidermal growth factor receptor 2 positive(Her2^(+) or ERBB2)breast cancer with DM2 by crossing leptin receptor mutant(Lepr^(db/+))mice with (MMTV-ErbB2/neu)mice.Themousemodelswere administrated with antidiabetic drugs to assess the impacts of controlling DM2 in affecting tumor growth.Magnetic resonance spectroscopic imaging was employed to analyze the tumor metabolism.Results:Treatment with metformin/rosiglitazone in MMTV-ErbB2/Lepr^(db/db) mousemodel reduced serum insulin levels,prolonged overall survival,decreased cumulative tumor incidence,and inhibited tumor progression.Anti-insulin resistance medications also inhibited glycolytic metabolism in tumors in vivo as indicated by the reduced metabolic flux of hyperpolarized ^(13)C pyruvate-to-lactate reaction.The tumor cells from MMTV-ErbB2/Lepr^(db/db) transgenic mice treated with metformin had reprogrammed metabolism by reducing levels of both oxygen consumption and lactate production.Metformin decreased the expression of Myc and pyruvate kinase isozyme 2(PKM2),leading to metabolism reprogramming.Moreover,metformin attenuated the mTOR/AKT signaling pathway and altered adipokine profiles.Conclusions:MMTV-ErbB2/Lepr^(db/db) mouse model was able to recapitulate diabetic HER2^(+) human breast cancer.Additionally,our results defined the signaling pathways deregulated in HER2^(+) breast cancer under diabetic condition,which can be intervened by anti-insulin resistance therapy. | Ping-Chieh Chou Hyun Ho Choi Yizhi Huang Enrique Fuentes-Mattei Guermarie Velazquez-Torres Fanmao Zhang Liem Phan Jaehyuk Lee Yanxia Shi James A.Bankson Yun Wu Huamin Wang Ruiying Zhao Sai-Ching Jim Yeung Mong-Hong Lee | 2021 | Cancer Communications2021,41,5: | 4 |
| 4 | Comparison of Crop Model Validation Methods显示文摘In this paper, the many indices used in validation of crop models, such as RMSE (root mean square errors), Sd (standard error of absolute difference), da (mean absolute difference), dap (ratio of da to the mean observation), r (correlation), and R2 (determination coefficient), are compared for the same rice architectural parameter model, and their advantages and disadvantages are analyzed. A new index for validation of crop models, dap between the observed and the simulated values, is proposed, with dap<5% as the suggested standard for precision of crop models. | 16 | Spatial distribution and network morphology of epicardial,endocardial,interstitial,and Purkinje cell-associated elastin fibers in porcine left ventricle显示文摘Cardiac extracellular matrices(ECM)play crucial functional roles in cardiac biomechanics.Previous studies have mainly focused on collagen,the major structural ECM in heart wall.The role of elastin in cardiac mechanics,however,is poorly understood.In this study,we investigated the spatial distribution and microstructural morphologies of cardiac elastin in porcine left ventricles.We demonstrated that the epicardial elastin network had location-and depth-dependency,and the overall epicardial elastin fiber mapping showed certain correlation with the helical heart muscle fiber architecture.When compared to the epicardial layer,the endocardial layer was thicker and has a higher elastin-collagen ratio and a denser elastin fiber network;moreover,the endocardial elastin fibers were finer and more wavy than the epicardial elastin fibers,all suggesting various interface mechanics.The myocardial interstitial elastin fibers co-exist with the perimysial collagen to bind the cardiomyocyte bundles;some of the interstitial elastin fibers showed a locally aligned,hinge-like structure to connect the adjacent cardiomyocyte bundles.This collagen-elastin combination reflects an optimal design in which the collagen provides mechanical strength and elastin fibers facilitate recoiling during systole.Moreover,cardiac elastin fibers,along with collagen network,closely associated with the Purkinje cells,indicating that this ECM association could be essential in organizing cardiac Purkinje cells into“fibrous”and“branching”morphologies and serving as a protective feature when Purkinje fibers experience large deformations in vivo.In short,our observations provide a structural basis for future in-depth biomechanical investigations and biomimicking of this long-overlooked cardiac ECM component. | Xiaodan Shi Song Zhang Yue Liu Bryn Brazile Jim Cooley J.Ryan Butler Sara R.McMahan Karla L.Perez Jiazhu Xu Timothy Eastep Kytai T.Nguyen Pietro Bajona Matthias Peltz Huajian Gao Yi Hong Jun Liao | 2023 | Bioactive Materials2023,,1: | 0 |
