| 2 | Ultrasound-triggered noninvasive regulation of blood glucose levels using microgels integrated with insulin nanocapsules显示文摘Diabetes is a serious public health problem affecting 422 million people worldwide. Traditional diabetes management often requires multiple daily insulin injections, associated with pain and inadequate glycemia control. Herein, we have developed an ultrasound-triggered insulin delivery system capable of pulsatile insulin release that can provide both long-term sustained and fast on-demand responses. In this system, insulin-loaded poly(lactic-co-glycolic acid) (PLGA) nanocapsules are encapsulated within chitosan microgels. The encapsulated insulin in nanocapsules can passively diffuse from the nanoparticle but remain restricted within the microgel. Upon ultrasound treatment, the stored insulin in microgels can be rapidly released to regulate blood glucose levels. | 3 | Mycorrhizal symbiosis modulates the rhizosphere microbiota to promote rhizobia–legume symbiosis显示文摘Plants establish symbioses with mutualistic fungi,such as arbuscular mycorrhizal(AM)fungi,and bacteria,such as rhizobia,to exchange key nutrients and thrive.Plants and symbionts have coevolved and represent vital components of terrestrial ecosystems.Plants employ an ancestral AM signaling pathway to establish intracellular symbioses,including the legume–rhizobia symbiosis,in their roots.Nevertheless,the relationship between the AM and rhizobial symbioses in native soil is poorly understood.Here,we examined how these distinct symbioses affect root-associated bacterial communities in Medicago truncatula by performing quantitative microbiota profiling(QMP)of 16S rRNA genes.We found that M.truncatula mutants that cannot establish AM or rhizobia symbiosis have an altered microbial load(quantitative abundance)in the rhizosphere and roots,and in particular that AM symbiosis is required to assemble a normal quantitative root-associated microbiota in native soil.Moreover,quantitative microbial co-abundance network analyses revealed that AM symbiosis affects Rhizobiales hubs among plant microbiota and benefits the plant holobiont.Through QMP of rhizobial rpoB and AM fungal SSU rRNA genes,we revealed a new layer of interaction whereby AM symbiosis promotes rhizobia accumulation in the rhizosphere of M.truncatula.We further showed that AM symbiosis-conditioned microbial communities within the M.truncatula rhizosphere could promote nodulation in different legume plants in native soil.Given that the AM and rhizobial symbioses are critical for crop growth,our findings might inform strategies to improve agricultural management.Moreover,our work sheds light on the co-evolution of these intracellular symbioses during plant adaptation to native soil conditions. | Xiaolin Wang Huan Feng Yayu Wang Mingxing Wang Xingguang Xie Huizhong Chang Like Wang Jicheng Qu Kai Sun Wei He Chunyan Wang Chuanchao Dai Zhaohui Chu Changfu Tian Nan Yu Xuebin Zhang Huan Liu Ertao Wang | 2021 | Molecular Plant2021,14,3: | 6 |
| 4 | Glucose-responsive oral insulin delivery for postprandial glycemic regulation显示文摘Controlling postprandial glucose levels for diabetic patients is critical to achieve the tight glycemic control that decreases the risk for developing long-term micro- and macrovascular complications.Herein,we report a glucose-responsive oral insulin delivery system based on Fc receptor (FcRn)-targeted liposomes with glucose-sensitive hyaluronic acid (HA) shell for postprandial glycemic regulation.After oral administration,the HA shell can quickly detach in the presence of increasing intestinal glucose concentration due to the competitive binding of glucose with the phenylboronic acid groups conjugated with HA.The exposed Fc groups on the surface of liposomes then facilitate enhanced intestinal absorption in an FcRn-mediated transport pathway.In vivo studies on chemically-induced type 1 diabetic mice show this oral glucose-responsive delivery approach can effectively reduce postprandial blood glucose excursions.This work is the first demonstration of an oral insulin delivery system directly triggered by increasing postprandial glucose concentrations in the intestine to provide an on-demand insulin release with ease of administration. | Jicheng Yu Yuqi Zhang Jinqiang Wang Di Wen Anna R. | 6 | Analysis of Aroma Volatile Compounds in Fuji Apple Using SPME with Different Fiber Coatings显示文摘The aroma volatile compounds in Fuji apple were isolated by solid-phase microextraction( SPME),and detected by gas chromatography-mass spectrometry( GC-MS). The results showed that the optimal retention time of the heating up of capillary columns was 5 min. Of the seven fibers used in this work,DVB/CAR/PDMS was found to be the most suitable to extract the aroma volatile compounds in Fuji apple. A total number of 43 volatile compounds were identified according to their retention time on capillary columns and their mass spectra,including eleven esters,ten alcohols,ten aldehydes,seven alkenes,two anhydrides,one ketone,one phenol and one ether. As reported, the risk of several serious diseases increases with weight gain, including type 2 diabetes,coronary heart disease, cancer, and respiratory diseases. In addition to lifestyle modifications, pharmacotherapy has become an important strategy to control weight gain. However, most of the anti-obesity drugs often show poor outcome for weight-loss and cause severe adverse effects. This review surveys recent advances in nanomedicine as an emerging strategy for obesity treatment with an emphasis on the enhanced therapeutic efficiency and minimized side effects.
