| 1 | High expression of ACE2 receptor of 2019-nCoV on the epithelial cells of oral mucosa显示文摘It has been reported that ACE2 is the main host cell receptor of 2019-nCoV and plays a crucial role in the entry of virus into the cell to cause the final infection. To investigate the potential route of 2019-n Cov infection on the mucosa of oral cavity, bulk RNA-seq profiles from two public databases including The Cancer Genome Atlas(TCGA) and Functional Annotation of The Mammalian Genome Cap Analysis of Gene Expression(FANTOM5 CAGE) dataset were collected. RNA-seq profiling data of 13 organ types with para-carcinoma normal tissues from TCGA and 14 organ types with normal tissues from FANTOM5 CAGE were analyzed in order to explore and validate the expression of ACE2 on the mucosa of oral cavity. Further, single-cell transcriptomes from an independent data generated in-house were used to identify and confirm the ACE2-expressing cell composition and proportion in oral cavity. The results demonstrated that the ACE2 expressed on the mucosa of oral cavity. Interestingly, this receptor was highly enriched in epithelial cells of tongue. | 3 | Estimation of inbreeding and identification of regions under heavy selection based on runs of homozygosity in a Large White pig population显示文摘Background: Runs of homozygosity(ROHs) are homozygous segments of the genome where the two haplotypes inherited from the parents are identical. The current availability of genotypes for a very large number of single nucleotide polymorphisms(SNPs) is leading to more accurate characterization of ROHs in the whole genome. Here,we investigated the occurrence and distribution of ROHs in 3,692 Large White pigs and compared estimates of inbreeding coefficients calculated based on ROHs(FROH), homozygosity(FHOM), genomic relationship matrix(FGRM)and pedigree(FPED). Furthermore, we identified genomic regions with high ROH frequencies and annotated their candidate genes.Results: In total, 176,182 ROHs were identified from 3,569 animals, and all individuals displayed at least one ROH longer than 1 Mb. The ROHs identified were unevenly distributed on the autosomes. The highest and lowest coverages of Sus scrofa chromosomes(SSC) by ROH were on SSC14 and SSC13, respectively. The highest pairwise correlation among the different inbreeding coefficient estimates was 0.95 between FROH_totaland FHOM, while the lowest was-0.083 between FGRMand FPED. The correlations between FPEDand FROHusing four classes of ROH lengths ranged from 0.18 to 0.37 and increased with increasing ROH length, except for ROH > 10 Mb. Twelve ROH islands were located on four chromosomes(SSC1, 4, 6 and 14). These ROH islands harboured genes associated with reproduction, muscular development, fat deposition and adaptation, such as SIRT1, MYPN, SETDB1 and PSMD4.Conclusion: FROHcan be used to accurately assess individual inbreeding levels compared to other inbreeding coefficient estimators. | 6 | Isolation and characterization of 49 polymorphic microsatellite loci for Decapterus maruadsi using SLAF-seq, and cross-amplifi cation to related species显示文摘Decapterus maruadsi is a commercially important species in China, but has been heavily exploited in some areas. There is a growing need to develop microsatellites promoting its genetic research for the adequate management of this ?shery resources. The recently developed speci?c-locus ampli?ed fragment sequencing(SLAF-seq) is an effcient and high-resolution method for genome-wide microsatellite markers discovery. In this study, 28 905 microsatellites(mono-to hexa-nucleotide repeats) were identi?ed using SLAF-seq technology, of which di-nucleotide was the most frequent(13 590, 47.02%), followed by mono-nucleotide(8 138, 28.15%), tri-nucleotide(5 727, 19.81%), tetra-nucleotide(1 104, 3.82%), pentanucleotide(234, 0.81%), and hexa-nucleotide(112, 0.39%). One hundred and thirty-two microsatellite loci(di-and tri-nucleotide) were randomly selected for ampli?cation and polymorphism, of which 49 were highly polymorphic and well-resolved. The average number of alleles per locus was 13.63, ranging from 4 to 25, and allele sizes varied between 110 bp and 309 bp. The observed heterozygosity( Ho) and expected heterozygosity( He) ranged from 0.233 to 1.000 and from 0.374 to 0.959, with mean values of 0.738 and0.836, respectively. The polymorphism information content(PIC) ranged from 0.341 to 0.941(mean=0.806).However, 12 loci deviated from Hardy-Weinberg equilibrium. Furthermore, transferability tests were also successful in validating the utility of the developed markers in ?ve phylogenetically related species of family Carangidae. | 10 | Fucoidan-functionalized activated platelet-hitchhiking micelles simultaneously track tumor cells and remodel the immunosuppressive microenvironment for efficient metastatic cancer treatment显示文摘Tumor metastasis is responsible for most mortality in cancer patients, and remains a challenge in clinical cancer treatment. Platelets can be recruited and activated by tumor cells, then adhere to circulating tumor cells(CTCs) and assist tumor cells extravasate in distant organs. Therefore, nanoparticles specially hitchhiking on activated platelets are considered to have excellent targeting ability for primary tumor, CTCs and metastasis in distant organs. However, the activated tumor-homing platelets will release transforming growth factor-β(TGF-β), which promotes tumor metastasis and forms immunosuppressive microenvironment. Therefore, a multitalent strategy is needed to balance the accurate tumor tracking and alleviate the immunosuppressive signals. In this study, a fucoidan-functionalized micelle(FD/DOX) was constructed, which could efficiently adhere to activated platelets through P-selectin.Compared with the micelle without P-selectin targeting effect, FD/DOX had increased distribution in both tumor tissue and metastasis niche, and exhibited excellent anti-tumor and anti-metastasis efficacy on 4 T1 spontaneous metastasis model. In addition, due to the contribution of fucoidan, FD/DOXtreatment was confirmed to inhibit the expression of TGF-β, thereby stimulating anti-tumor immune response and reversing the immunosuppressive microenvironment. | 18 | Comparative Study of 3D-Printed Porous Titanium Alloy with Rod Designs of Three Different Geometric Structures for Orthopaedic Implantation显示文摘Porous titanium alloy is currently widely used in clinical treatment of orthopaedic diseases for its lower elastic modulus and ability to integrate with bone tissue.At the micro-level,cells can respond to different geometries,and at the macro-level,the geometric design of implants will also affect the biological function of cells.In this study,three kinds of porous scaffolds with square,triangular and circle rod shapes were designed and 3D printed.This study observed the proliferation and differentiation of MC3T3-E1 cells during surface culture of the three types of scaffolds.It also evaluated the characteristics of the three scaffolds by means of compression tests and scanning electron microscopy to provide a reference for the design of porous titanium alloy implants for clinical applications.The trends of cell proliferation and gene expression between the three types of scaffolds were observed after treatment with two inhibitors.The results show that the square rod porous scaffolds have the best proliferative and osteogenic activities,and these findings may be due to differences in piezo-type mechanosensitive ion channel component 1(Piezo1)and Yes-associated protein(YAP)expression caused by the macro-geometric topography. | Jiaxin Li Haozhang Zhong Bojun Cao Zhaoyang Ran Jia Tan Liang Deng Yongqiang Hao Jinglong Yan | 2024 | Acta Metallurgica Sinica(English Letters)2024,37,1: | 0 |
| 19 | Deep-ultraviolet photonics for the disinfection of SARS-CoV-2 and its variants(Delta and Omicron)in the cryogenic environment显示文摘Deep-ultraviolet(DUV)disinfection technology provides an expeditious and efficient way to suppress the transmission of coronavirus disease 2019(COVID-19).However,the influences of viral variants(Delta and Omicron)and low temperatures on the DUV virucidal efficacy are still unknown.Here,we developed a reliable and uniform planar light source comprised of 275-nm light-emitting diodes(LEDs)to investigate the effects of these two unknown factors and delineated the principle behind different disinfection performances.We found the lethal effect of DUV at the same radiation dose was reduced by the cryogenic environment,and a negative-U large-relaxation model was used to explain the difference in view of the photoelectronic nature.The chances were higher in the cryogenic environment for the capture of excited electrons within active genetic molecules back to the initial photo-ionised positions.Additionally,the variant of Omicron required a significantly higher DUV dose to achieve the same virucidal efficacy,and this was thanks to the genetic and proteinic characteristics of the Omicron.The findings in this study are important for human society using DUV disinfection in cold conditions(e.g.,the food cold chain logistics and the open air in winter),and the relevant DUV disinfection suggestion against COVID-19 is provided. | Wenyu Kang Jing Zheng Jiaxin Huang Lina Jiang Qingna Wang Zhinan Guo Jun Yin Xianming Deng Ye Wang Junyong Kang | 2023 | Opto-Electronic Advances2023,6,9: | 0 |
| 20 | Hyperspectral and multispectral remote sensing image fusion using SwinGAN with joint adaptive spatial-spectral gradient loss function显示文摘Hyperspectral remote sensing image(HSI)fusion with multispectral remote sensing images(MSI)improves data resolution.However,current fusion algorithms focus on local information and overlook long-range dependencies.The parameter of network tuning prioritizes global optimization,neglecting spatial and spectral constraints,and limiting spatial and spectral reconstruction capabilities.This study introduces SwinGAN,a fusion network combining Swin Transformer,CNN,and GAN architectures.SwinGAN’s generator employs a detail injection framework to separately extract HSI and MSI features,fusing them to generate spatial residuals.These residuals are injected into the supersampled HSI to produce thefinal image,while a pure CNN architecture acts as the discriminator,enhancing the fusion quality.Additionally,we introduce a new adaptive loss function that improves image fusion accuracy.The loss function uses L1 loss as the content loss,and spatial and spectral gradient loss functions are introduced to improve the spatial representation and spectralfidelity of the fused images.Our experimental results on several datasets demonstrate that SwinGAN outperforms current popular algorithms in both spatial and spectral reconstruction capabilities.The ablation experiments also demonstrate the rationality of the various components of the proposed loss function. | Chunyu Zhu Shangqi Deng Jiaxin Li Ying Zhang Liwei Gong Liangbo Gao Na Ta Shengbo Chen Qiong Wu | 2023 | International Journal of Digital Earth2023,16,1: | 0 |
