| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 3 | Towards an Artificial Intelligence Framework for Data-Driven Prediction of Coronavirus Clinical Severity显示文摘The virus SARS-CoV2,which causes the Coronavirus disease COVID-19 has become a pandemic and has spread to every inhabited continent.Given the increasing caseload,there is an urgent need to augment clinical skills in order to identify from among the many mild cases the few that will progress to critical illness.We present a first step towards building an artificial intelligence(AI)framework,with predictive analytics(PA)capabilities applied to real patient data,to provide rapid clinical decision-making support.COVID-19 has presented a pressing need as a)clinicians are still developing clinical acumen given the disease’s novelty,and b)resource limitations in a rapidly expanding pandemic require difficult decisions relating to resource allocation.The objectives of this research are:(1)to algorithmically identify the combinations of clinical characteristics of COVID-19 that predict outcomes,and(2)to develop a tool with AI capabilities that will predict patients at risk for more severe illness on initial presentation.The predictive models learn from historical data to help predict specifically who will develop acute respiratory distress syndrome(ARDS),a severe outcome in COVID-19.Our experimental results based on two hospitals in Wenzhou,Zhejang,China identify features most predictive of ARDS in COVID-19 initial presentation which would not have stood out to clinicians.A mild increase in elevated alanine aminotransferase(ALT)(a liver enzyme)),a presence of myalgias(body aches),and an increase in hemoglobin,in this order,are the clinical features,on presentation,that are the most predictive.Those two centers’COVID-19 case series symptoms on initial presentation can help predict severe outcomes.Predictive models that learned from historical data of patients from two Chinese hospitals achieved 70%to 80%accuracy in predicting severe cases. | Xiangao Jiang Megan Coffee Anasse Bari Junzhang Wang Xinyue Jiang Jianping Huang Jichan Shi Jianyi Dai Jing Cai Tianxiao Zhang Zhengxing Wu Guiqing He Yitong Huang | 2020 | Computers, Materials & Continua2020,,4: | 15 |
| 4 | 3D Positional and Quantitative Prediction of the Xiaoqinling Gold Ore Belt in Tongguan, Shaanxi, China显示文摘Quantitative prediction of deep orebody based on 3D visualization technology is of great significance in mineral exploration. Based on the 2D traditional quantitative predicting method, the geoanomaly theory and the mineral exploration model idea, we constructed 3D models of the topography, strata, structure, magmatite and prospecting engineering of the study area using the commercial 3D modeling software Micromine, delineated eight prospective areas and estimated the gold resources amount with methods of Abundance Estimation and Volume Estimation. Then, we compared and counted the known ore blocks and the predicted blocks, which quantitatively explains this prediction's validity. | 5 | Dynamics analysis of chaotic circuit with two memristors显示文摘Based on Chua’s chaotic oscillation circuit, a fifth-order chaotic circuit with two memristors is designed and its corresponding dimensionless mathematic model is established. By using conventional dynamical analysis methods, stability analysis of the equilibrium set of the circuit is performed, the distribution of stable and unstable regions corresponding to the memristor initial states is achieved, and the complex dynamical behaviors of the circuit depending on the circuit parameters and the memristor initial states are investigated. The theoretical analysis and numerical simulation results demonstrate that the proposed chaotic circuit with two memristors has an equilibrium set located on the plane constituted by the inner state variables of two memristors. The stability of the equilibrium set depends on both the circuit parameters and the initial states of the two memristors. | 10 | Chemical vapor deposition of monolayer WS2 nano- sheets on Au foils toward direct application in hydrogen evolution显示文摘Monolayer tungsten disulfide (WS2), a typical member of the semiconducting transition metal dichalcogenide family has drawn considerable interest because of its unique properties. Intriguingly the edge of WS2 exhibits an ideal hydrogen binding energy which makes WS2 a potential alternative to Pt-based electrocatalysts for the hydrogen evolution reaction (HER). Here, we demonstrate for the first time the successful synthesis of uniform monolayer WS2 nanosheets on centimeter- scale Au foils using a facile, low-pressure chemical vapor deposition method. The edge lengths of the universally observed triangular WS2 nanosheets are tunable from -100 to N1,000 nm. The WS2 nanosheets on Au foils featuring abundant edges were then discovered to be efficient catalysts for the HER, exhibiting a rather high exchange current density of -30.20 μA/cm2 and a small onset potential of Nl10 mV. The effects of coverage and domain size (which correlate closely with the active edge density of WS2) on the electrocatalytic activity were investigated. | 14 | Sources and Accumulation Risk of Heavy Metal in Major Animal Manure显示文摘Feed and corresponding manure samples were collected from 27 scale pig farms and 7 scale chicken farms in Jiaxing City,Zhejiang Province,and their heavy metal contents were analyzed. The weighted average values of Cr,Cu,Zn,Pb and As in pig feed were 4. 6,123. 4,370. 7,0. 69 and 6. 9 mg/kg,respectively. However,referred to Safe Use Specification of Feed Additives and MOA Announcement No. 1224,the exceeding standards rates of Cr,Cu,Zn,Pb and As in pig feed were 5. 36%,62. 5%,19. 64%,3. 57% and 37. 5%,respectively. The sequences of heavy metal content in pig feed successively were piglet > growing pig > big pig > sow,but the exceeding standard rates of heavy metal in feed were big pig > sow > growing pig > piglet,and the exceeding standard rate of big pig feed was more than 95%. The exceeding standard rates of Cu,Zn and As in chicken feed were 46. 7%,11. 1% and 33. 3% respectively. Based on Organic Fertilizer( NY525-2012) and Sludge Agricultural Pollution Control Standard( GB4284-84),Cu,Zn and As in pig manure and chicken manure exceeded the standard; the exceeding standard rates of pig manure were 71. 9%,78. 9% and 42. 1%,while that of chicken manure were 12. 5%,25. 0% and 44. 4%,respectively. The exceeding standard rates of Cu and Zn in piglet manure reached 100%. There were very significant positive correlations in Cu,Zn and As between pig feed and corresponding manure,or Hg,Cu and As between chicken feed and corresponding manure,and the correlation coefficients r^2 were greater than 0. 8. | 16 | Depression accelerates gastric cancer invasion and metastasis by inducing a neuroendocrine phenotype via the catecholamine/β_(2)-AR/MACC1 axis显示文摘Background:Depression is a common,easily ignored,accompanied disease of gastric cancer(GC)patients and is often observed with elevated plasma catecholamine levels.Depression frequently promotes GC progression and leads to poor clinical outcomes;however,the molecular mechanisms underlying depression-induced GC progression remain poorly understood.We aimed to study the effects of depression on GC progression and explore possible mechanisms mediating the action of depression-associated catecholamines on GC.Methods:Depression states of GC patients were graded using the Patient Health Questionnaire-9,and plasma catecholamine levels were examined by high performance liquid chromatography coupled with tandem mass spectrometry.Migrative and invasive GC cells were examined using transwell assays,and metastatic GC niches were imaged using bioluminescence technology in a depression mouse model established with chronic unpredictable mild stress.Mouse depression-like behaviors were assessed through sucrose preference,forced swimming,and tail suspension tests.Characteristics of the neuroendocrine phenotype were observed via RT-PCR,Western blotting,flow cytometry,and transmission electron microscopy.Results:Fifty-one GC patients(age:53.61±1.79 years;cancer duration:3.71±0.33 months;depression duration:2.37±0.38 months;male-to-female ratio:1.55:1)were enrolled in the study.Depression grade was significantly higher in GC patients showing higher plasma levels of catecholamines(epinephrine:P=0.018;noradrenaline:P=0.009),higher oncogene metastasis-associated in colon cancer-1(MACC1)level(P=0.018),and metastasis(P<0.001).Further,depression-associated catecholamine specifically bound to the beta-2 adrenergic receptor(β_(2)-AR)and upregulated MACC1 expression,and thus promoting neuroendocrine phenotypic transformation through direct binding betweenMACC1 and synaptophysin.Eventually,the neuroendocrine phenotypic transformation accelerated GC invasion in vitro and metastasis in vivo.However,β_(2)-AR antagonist ICI-118,551 or MACC1 silencing effectively blocked the catecholamineinduced neuroendocrine phenotypic transformation and eliminated depressionenhanced GC migration and invasion.Moreover,β_(2)-AR blocking or MACC1 silencing prevented GC metastasis attributed to a neuroendocrine phenotype in a depression mouse model.Conclusions:Catecholamine-induced neuroendocrine phenotypes of GC cells led to depression-accelerated GC invasion and metastasis via the β_(2)-AR/MACC1 axis,while β_(2)-AR antagonist or MACC1 silencing could reverse it,showing promising potential therapeutic strategies for improving the outcome of GC patients with comorbid depression. | Changqie Pan Jianhua Wu Siting Zheng Huiying Sun Yisheng Fang Zhenhua Huang Min Shi Li Liang Jianping Bin Yulin Liao Jinzhang Chen Wangjun Liao | 2021 | Cancer Communications2021,41,10: | 4 |
| 17 | Prediction and systematic study of protein-protein interaction networks of Leptospira interrogans显示文摘Leptospira interrogans serovar Lai is a pathogenic bacterium that causes a spirochetal zoonosis in humans and some animals. With its complete genome sequence available, it is possible to analyze protein-protein interactions from a whole- genome standpoint. Here we combine four recently developed computational approaches (gene fusion method, gene neighbor method, phylogenetic profiles method, and operon method) to predict protein-pro- tein interaction networks of Leptospira interrogans strain Lai. Through comprehensive analysis on in- teractions among proteins of motility and chemotaxis system, signal transduction, lipopolysaccaride bio- synthesis and a series of proteins related to adhesion and invasion, we provided information for further studying on its pathogenic mechanism. In addition, we also assigned 203 previously uncharacterized proteins with possible functions based on the known functions of its interacting partners.
