| 1 | Evaluating the reliability of microsatellite genotyping from low-quality DNA templates with a polynomial distribution model显示文摘Molecular studies using trace DNA, such as from museum specimens, ancient or forensic samples and samples obtained noninvasively, often have a common problem of low quality of DNA templates. Amplification errors, such as allelic dropout and false allele, may arise during polymerase chain reaction (PCR) using such samples. A mathematical model which treats homozygotes and heterozygotes discriminately has been developed to measure sample quality and compute the confidence level of using multipletube approaches. We use plucked hair samples collected from 26 individual Sichuan snub-nosed monkeys (Rhinopithecus roxel- lana) to test the model. In this case, a confidence level of 99% can be achieved by three positive PCRs. If the sample quality is very poor and requires many PCR replicates, an alternative multiplestep genotyping method is recommended. | 2 | Musk secretion in muskrats(Ondatra zibethicus L.):association with lipid and cholesterol metabolism-related pathways显示文摘Male muskrats(Ondatra zibethicus L.)secrete musk from their scent glands during musk secretion season.Musk plays an important role as a communication pheromone during the breeding season.In this study,gas chromatography–mass spectrometry(GC–MS)was used to analyze the main components of musk.The GC–MS results after methyl esterification showed that 71.55%of the musk is composed of fatty acids.The other components of muskrat musk include cholesterol(9.31%)and other organics.Transcriptome comparison between musk secretion and non-secretion seasons showed significant changes in the scent glands for 53 genes involved in fatty acid and cholesterol synthesis and metabolism regulatory pathways,which include fatty acid biosynthesis,elongation,and metabolism;steroid biosynthesis;steroid hormone biosynthesis pathways.A reverse transcription–polymerase chain reaction analysis confirmed these detected changes.Overall,our results indicated that lipid synthesis and metabolism play important roles in musk compound synthesis by providing energy for musk production,and the produced musk provides a mechanism for male muskrats to communicate with females during the breeding season. | TIANXIANG ZHANG MEISHAN ZHANG MINGHUI SHI WEIJIANG JIN SHUMIAO ZHANG MENGYUAN FAN YIMENG LI YUPING MENG XUELIN TIAN SHUQIANG LIU DEFU HU | 2021 | BIOCELL2021,45,2: | 2 |
| 3 | Using unmanned aerial vehicles with thermal-image acquisition cameras for animal surveys:a case study on the Sichuan snub-nosed monkey in the Qinling Mountains显示文摘Following significant developments in technology,alternative devices have been applied in fieldwork for animal and plant surveys.Thermal-image acquisition cameras installed on unmanned aerial vehicles(UAVs)have been used in animal surveys in the wilderness.This article demonstrates an example of how UAVs can be used in high mountainous regions,presenting a case study on the Sichuan snub-nosed monkey with a detection rate of 65.19%for positive individual identification.It also presents a model that can prospectively predict population size for a given animal species,which is based on combined initial work using UAVs and traditional surveys on the ground.A great potential advantage of UAVs is significantly shortening survey procedures,particularly for areas with high mountains and plateaus,such as the Himalayas,the Qinghai-Tibet Plateau,Hengduan Mountains,the Yunnan-Gui Plateau and Qinling Mountains in China,where carrying out a traditional survey is extremely difficult,so that species and population surveys,particularly for critically endangered animals,are largely absent.This lack of data has impacted the management of endangered animals as well as the formulation and amendment of conservation strategies. | Gang HE Haitao YANG Ruliang PAN Yewen SUN Pengbin ZHENG Jinghua WANG Xuelin JIN Jingjie ZHANG Baoguo LI Songtao GUO | 2020 | Integrative Zoology2020,15,1: | 2 |
| 4 | Efficient production of cyclic adenosine monophosphate from adenosine triphosphate by the N-terminal half of adenylate cyclase from Escherichia coli显示文摘In this study,we aimed at developing an efficient biocatalytic process for bio-production of cyclic adenosine monophosphate(c AMP)from adenosine triphosphate(ATP).First,adenylate cyclase from Escherichia coli MG1655(EAC)and Bordetella Pertussis(BAC)were expressed in E.coli BL21(DE3)and comparatively analyzed for their activities.As a result,EAC from E.coli MG1655 exhibited a higher activity.However,amount of EAC were obtained in an insoluble form.Therefore,we expressed the first 446 amino acids of EAC(EAC446)to avoid the inclusion body.The effects of induction temperature,incubation time,and incubation p H were further evaluated to improve the expression of EAC446.Subsequently,the reaction process for the production of c AMP with ATP as a starting material was investigated.As none of c AMP was detected in the whole-cell based biocatalytic process,the reaction catalyzed by the crude enzyme was determined for c AMP production.What's more,the reaction temperature,reaction p H,metal ion additives and substrate concentration was optimized,and the maximum c AMP production of 18.45 g·L^-1was achieved with a yield of 95.4%after bioconversion of 6 h. | Chen Ma Jing Wang Xuelin Wang Dandan Mai Yuqi Jin Kequan Chen Xin Wang Pingkai Ouyang | 2020 | Chinese Journal of Chemical Engineering2020,28,8: | 1 |
| 5 | Astragaloside IV derivative HHQ16 ameliorates infarctioninduced hypertrophy and heart failure through degradation of lncRNA4012/9456显示文摘Reversing ventricular remodeling represents a promising treatment for the post-myocardial infarction(MI)heart failure(HF).Here,we report a novel small molecule HHQ16,an optimized derivative of astragaloside IV,which effectively reversed infarction-induced myocardial remodeling and improved cardiac function by directly acting on the cardiomyocyte to reverse hypertrophy.The effect of HHQ16 was associated with a strong inhibition of a newly discovered Egr2-affliated transcript Inc9456 in the heart.While minimally expressed in normal mouse heart,Inc9456 was dramatically upregulated in the heart subjected to left anterior descending coronary artery ligation(LADL)and in cardiomyocytes subjected to hypertrophic stimulation.The critical role of Inc9456 in cardiomyocyte hypertrophy was confirmed by specific overexpression and knockout in vitro.A physical interaction between Inc9456 and G3BP2 increased NF-kB nuclear translocation,triggering hypertrophy-related cascades.HHQ16 physically bound to Inc9456 with a high-affinity and induced its degradation.Cardiomyocyte-specific Inc9456 overexpression induced,but knockout prevented LADL-induced,cardiac hypertrophy and dysfunction.HHQ16 reversed the effect of Inc9456 overexpression while lost its protective role when Inc9456 was deleted,further confirming Inc9456 as the bona fide target of HHQ16.We further identified the human ortholog of Inc9456,also an Egr2-affliated transcript,Inc4012.Similarly,Inc4012 was significantly upregulated in hypertrophied failing hearts of patients with dilated cardiomyopathy.HHQ16 also specifically bound to Inc4012 and caused its degradation and antagonized its hypertrophic effects.Targeted degradation of pathological increased Inc4012/lnc9456 by small molecules might serve as a novel promising strategy to regress infarction-induced cardiac hypertrophy and HF. | Jingjing Wan Zhen Zhang hennan Wu Saisai Tianbei Zang Ge Jin Qingyan Sun Pin Wang Xin Luan Yili Yang Xuelin Zhan Lingyu Linda Ye Dayue Darrel Duan Xia Liu Weidong Zhang | 2023 | Signal Transduction and Targeted Therapy2023,8,11: | 0 |
| 6 | Prim-O-glucosylcimifugin ameliorates aging-impaired endogenous tendon regeneration by rejuvenating senescent tendon stem/progenitor cells显示文摘Adult tendon stem/progenitor cells(TSPCs)are essential for tendon maintenance,regeneration,and repair,yet they become susceptible to senescence with age,impairing the self-healing capacity of tendons.In this study,we employ a recently developed deep-learning-based efficacy prediction system to screen potential stemness-promoting and senescence-inhibiting drugs from natural products using the transcriptional signatures of stemness.The top-ranked candidate,prim-O-glucosylcimifugin(POG),a saposhnikovia root extract,could ameliorate TPSC senescent phenotypes caused by long-term passage and natural aging in rats and humans,as well as restore the self-renewal and proliferative capacities and tenogenic potential of aged TSPCs.In vivo,the systematic administration of POG or the local delivery of POG nanoparticles functionally rescued endogenous tendon regeneration and repair in aged rats to levels similar to those of normal animals.Mechanistically,POG protects TSPCs against functional impairment during both passage-induced and natural aging by simultaneously suppressing nuclear factor-κB and decreasing mTOR signaling with the induction of autophagy.Thus,the strategy of pharmacological intervention with the deep learning-predicted compound POG could rejuvenate aged TSPCs and improve the regenerative capacity of aged tendons. | Yu Wang Shanshan Jin Dan Luo Danqing He Min Yu Lisha Zhu Zixin Li Liyuan Chen Chengye Ding Xiaolan Wu Tianhao Wu Weiran Huang Xuelin Zhao Meng Xu Zhengwei Xie Yan Liu | 2023 | Bone Research2023,11,4: | 0 |
| 7 | Influence of band microstructure on carbide precipitation behavior and toughness of 1 GPa-grade ultra-heavy gauge low-alloy steel显示文摘This study investigated the influence of band microstructure induced by centerline segregation on carbide precipitation behavior and toughness in an 80 mm-thick 1 GPa low-carbon low-alloy steel plate.The quarter-thickness(1/4t)and half-thickness(1/2t)regions of the plate exhibited similar ductility and toughness after quenching.After tempering,the 1/4t region exhibited~50%and~25%enhancements in both the total elongation and low-temperature toughness at-40°C,respectively,without a decrease in yield strength,whereas the toughness of the 1/2t region decreased by~46%.After quenching,both the 1/4t and 1/2t regions exhibited lower bainite and lath martensite concentrations,but only the 1/2t region exhibited microstructure bands.Moreover,the tempered 1/4t region featured uniformly dispersed short rod-like M_(23)C_(6)carbides,and spherical MC precipitates with diameters of~20–100 nm and<20 nm,respectively.The uniformly dispersed nanosized M_(23)C_(6)carbides and MC precipitates contributed to the balance of high strength and high toughness.The band microstructure of the tempered 1/2t region featured a high density of large needle-like M3C carbides.The length and width of the large M3C carbides were~200–500 nm and~20–50 nm,respectively.Fractography analysis revealed that the high density of large carbides led to delamination cleavage fracture,which significantly deteriorated toughness. | Peng Han Zhipeng Liu Zhenjia Xie Hua Wang Yaohui Jin Xuelin Wang Chengjia Shang | 2023 | International Journal of Minerals,Metallurgy and Materials2023,30,7: | 0 |
| 8 | Mechanosynthesis of polymer-stabilized lead bromide perovskites:Insight into the formation and phase conversion of nanoparticles显示文摘The application of polymers to replace oleylamine(OLA)and oleic acid(OA)as ligands for perovskite nanocrystals is an effective strategy to improve their stability and durability especially for the solution-based processing.Herein,we report a mechanosynthesis of lead bromide perovskite nanoparticles(NPs)stabilized by partially hydrolyzed poly(methyl methacrylate)(h-PMMA)and highmolecular-weight highly-branched poly(ethylenimine)(PEI-25K).The as-synthesized NP solutions exhibited green emission centered at 516 nm,possessing a narrow full-width at half-maximum of 17 nm and as high photoluminescence quantum yield(PL QY)as 85%,while showing excellent durability and resistance to polar solvents,e.g.,methanol.The colloids of polymer-stabilized NPs were directly processable toform stable and strongly-emitting thin films and solids,making them attractive as gain media.Furthermore,the roles of h-PMMA and PEI-25K in the grinding process were studied in depth.The h-PMMA can form micelles in the grinding solvent of dichloromethane to act as size-regulating templates for the growth of NPs.The PEI-25K with large amounts of amino groups induced significant enrichment of PbBr_(2)in the reaction mixture,which in turn caused the formation of CsPb_(2)Br_(5)-mPbBr_(2)and CsPbBr_(3)-Cs_(4)PbBr_(6)-nCsBr NPs.The presence of CsPbBr_(3)-Cs_(4)PbBr_(6)-nCsBr NPs was responsible for the high PL QY,as the Cs_(4)PbBr_(6)phase with a wide energy bandgap can passivate the surface defects of the CsPbBr_(3)phase.This work describes a direct and facile mechanosynthesis of polymer-coordinated perovskite NPs and promotes in-depth understanding of the formation and phase conversion for perovskite NPs in the grinding process. | Guocan Jiang Onur Erdem Rene Hubner Maximilian Georgi Wei Wei Xuelin Fan Jin Wang Hilmi Volkan Demir Nikolai Gaponik | 2021 | Nano Research2021,14,4: | 0 |
