1Targeting of the AKT/m-TOR Pathway: Biomarkers of Resistance to Cancer Therapy——AKT/m-TOR Pathway and Resistance to Cancer Therapy显示文摘Resistance to cancer therapy continues to be a major limitation for the successful treatment of cancer. There are many published studies on therapy resistance in breast and prostate cancers; however, there are currently no data on molecular markers associated with resistance. The conflicting data were reported regarding the AKT/m-TOR signaling pathway components as markers predicting resistance. The AKT/m-TOR signaling pathway is involved in the development of many human cancers; its activation is related to cell proliferation, angiogenesis, apoptosis, as well as to therapy resistance. 2Immunopathogenesis of chronic hepatitis B显示文摘Chronic hepatitis B(CHB) is a widespread infectious disease with unfavorable outcomes and life-threatening consequences for patients, in spite of modern vaccination and antiviral treatment modalities. Cutting-edge experimental approaches have demonstrated key pathways that involve cross-talk between viral particles and host immune cells. All events, including penetration of hepatitis B virus(HBV) particles into host cells, establishing persistence, and chronization of CHB infection, and possibility of complete elimination of HBV particles are controlled by the immune system. Researchers have paid special attention to the replication capacity of HBV in host cells, which is associated with cellular changes that reflect presentation of viral antigens and variability of HBV antigen features. In addition, specific HBV proteins have an immune-modulating ability to initiate molecular mechanisms that 'avoid' control by the immune system. The relationship between immunological shifts and chronic infection stages has been intensively studied since it was recognized that the immune system is a direct participant in the recurrent(cyclic) nature of CHB. 4Biomineralization of organic matter in cobalt-rich crusts from the Marcus–Wake Seamounts of the western Pacific Ocean显示文摘Organic matter in cobalt-rich crust(CRC) from the Marcus–Wake Seamounts of the western Pacific Ocean, Sample CM1D03, has been analyzed to understand the source, geochemistry and mineralization of organic matter, and the mineralization environment. Biomarkers, including n-alkanes, isoprenoids, terpanes and sterols, have been detected in various layers of the CRC sample, using gas chromatography(GC) and gas chromatography–mass spectrometry(GC–MS). The content of organic carbon(OC) and its stable isotope(δ13C), and the combined features of the biomarkers show that the mineralized organic matter in CM1D03 CRC was mainly derived from microorganisms and lower plankton(e.g., bacteria and algae, respectively) from marine surface water, with some terrestrial higher plant components. The ratio of chloroform bitumen 'A': OC was high in the CRC, between 10.51 and 20.66, showing significant migration characteristics of n-alkanes. Four mineralization categories of organic matter were recognized based on GC chromatograms of n-alkane molecules:(1) primitive type(bacteria and algae), which is characterized by moderately mature of n-alkanes preserving the original characteristics of the organic matter from microorganisms and lower plankton;(2) microbial degradation type, which is characterized by low contents of n-alkanes and rising baseline in the chromatogram, with the 'bulge' being the products of organic matter by biodegradation;(3) organic matter migration type, which is characterized by low carbon number of n-alkanes with n C18 as the main peak carbon, without odd even predominance, and low concentrations of isoprenoids and hydrocarbons with high carbon number; and(4) organic matter hydrothermal type, which is characterized by relatively low concentration of small molecular weight n-alkanes, pristane, and phytane, accompanied by higher concentration of n-alkanes with carbon number greater than n C18. 5Linagliptin alleviates fatty liver disease in diabetic db/db mice显示文摘AIM To study the effects of linagliptin on the structural signs of non-alcoholic fatty liver disease(NAFLD) in db/db mice. METHODS Male diabetic db /db mice(BKS.Cg-Dock7m+/+Leprdb/J) aged 10 wk received the dipeptidyl peptidase 4(DPP4) inhibitor linagliptin(10 mg/kg) or saline as a placebo once per day by gavage for 8 wk. Intact db/db mice served as controls. Structural changes in the liver were analyzed from light and electron microscopic images of sections from intact, placebo-treated and linagliptin-treated animals. We estimated the changes in hepatocytes, sinusoidal cells, liver microvasculature and lymphatic roots. Hepatic staining for lymphatic vessel endothelial hyaluronan receptor-1(LYVE-1) was assessed by immunohistochemistry. RESULTS In 18-wk-old diabetic mice, liver steatosis(predominantly microvesicular and mediovesicular steatosis) was accompanied by dilation of the roots of the lymphatic system, interlobular blood vessels and bile canaliculi. Compared to saline-treated mice, linagliptin-treated mice exhibited a reduction in the mean numeral densities of hepatocytes with lipid droplets(92.4% ± 1.7% vs 64.9% ± 5.8% per field of view, P = 0.0002) and a lower proportion of hepatocytes with a high density of lipid droplets(20.7% ± 3.6% vs 50.4% ± 3.1%, P = 0.0007). We observed heterogeneous hepatocytes and relatively preserved cell structures in the linagliptin group. Dilation of blood and lymphatic vessels, as well as ultrastructural changes in the hepatocyte endoplasmic reticulum and mitochondria, were alleviated by linagliptin treatment. In intact and placebo-treated mice, immunohistochemical staining for LYVE-1 was observed in the endothelial cells of interlobular lymphatic vessels and on the membranes of some endothelial sinusoidal cells. We observed an enlarged LYVE-1 reaction area in linagliptin-treated mice compared to intact and placebo-treated mice. The improvement in the structural parameters of the liver in linagliptin-treated mice was independent to changes in the plasma glucose levels. 6Bifunctional chimeric SuperCD suicide gene -YCD: YUPRT fusion is highly effective in a rat hepatoma model显示文摘AIM: To investigate the effects of catalytically superior gene-directed enzyme prodrug therapy systems on a rat hepatoma model.METHODS: To increase hepatoma cell chemosensitivity for the prodrug 5-fluorocytosine (5-FC), we generated a chimeric bifunctional SuperCD suicide gene, a fusion of the yeast cytosine deaminase (YCD) and the yeast uracil phosphoribosyltransferase (YUPRT) gene.RESULTS: In vitro stably transduced Morris rat hepatoma cells (MH) expressing the bifunctional SuperCD suicide gene (MH SuperCD) showed a clearly marked enhancement in cell killing when incubated with 5-FC as compared with MH ceils stably expressing YCD solely (MH YCD) or the cytosine deaminase gene of bacterial origin(MH BCD), respectively. In vivo, MH SuperCD tumors implanted both subcutaneously as well as orthotopically into the livers of syngeneic ACI rats demonstrated significant tumor regressions (P<0.01) under both high dose as well as low dose systemic 5-FC application,whereas MH tumors without transgene expression (MH naive) showed rapid progression. 7西太平洋海山富钴结壳钙质超微化石变化与E/O界限的地质记录显示文摘选择马尔库斯-威克海山区和麦哲伦海山区的2块壳结纹层记录,进行生物地层学的精细研究,并利用钙质超微化石的生物印痕分出时代:马尔库斯-威克海山CM1D03年代为晚古新世到更新世,而麦哲伦海山CM3D06结壳年代则更久远些——白垩纪晚期(约大于70.0 Ma),这表明不同海山的富钴结壳最初形成年代和富集特征具有显著的时空差异性,而不同海山结壳层内部的古微体化石组合面貌、分布特征上的差别表明有关属种对大洋不同环境的适应性差异所致.本文还利用结壳纹层中所保存的钙质超微化石种群数进行生态学研究,寻找钙质超微化石变化与古新世末至渐新世初(E/O)界限地质事件的联系,研究发现在E/O界线之交(对应CM1D03结壳的25 mm和CM3D06结壳的58mm附近)生物群落发生明显演替和重组,这些生物群落在E/O界限上下的改变所形成独特的生态结构,无疑反映了西太平洋古生物群落对全球冷事件存在明显响应,也说明结壳生长过程与全球气候变化有着紧密联系.张海生 胡佶 赵军 韩正兵 于培松 武光海 雷吉江 卢冰 PULYAEVA Irina A 2015中国科学:地球科学2015,45,4:2
