1Challenges and strategies for next-generation bispecific antibody-based antitumor therapeutics显示文摘Bispecific antibodies(bsAbs)refer to a large family of molecules that recognize two different epitopes or antigens.Although a series of challenges,especially immunogenicity and chain mispairing issues,once hindered the development of bsAbs,they have been gradually overcome with the help of rapidly developing technologies in the past 5 decades.In the meantime,an increasing number of bsAb platforms have been designed to satisfy different clinical demands.Currently,numerous preclinical and clinical trials are underway,portraying a promising future for bsAb-based cancer treatment.Nevertheless,bsAb drugs still face enormous challenges in their application as cancer therapeutics,including tumor heterogeneity and mutational burden,intractable tumor microenvironment(TME),insufficient costimulatory signals to activate T cells,the necessity for continuous injection,fatal systemic side effects,and off-target toxicities to adjacent normal cells.Therefore,we provide several strategies as solutions to these issues,which comprise generating multispecific bsAbs,discovering neoantigens,combining bsAbs with other anticancer therapies,exploiting natural killer(NK)-cell-based bsAbs and producing bsAbs in situ.In this review,we mainly discuss previous and current challenges in bsAb development and underscore corresponding strategies,with a brief introduction of several typical bsAb formats.Heliang Li Phei Er Saw Erwei Song 2020Cellular & Molecular Immunology2020,17,5:6
2Effects of Low Protein Diets Supplemented with Ami-no Acids on Production Performance,Carcass Qual-ity and Input-output Ratio of Pigs显示文摘[Objectives] To study the effects of low-protein diets supplemented with amino acids on the production performance,carcass quality and input-output ratio of fattening pigs and growing-finishing pigs. [Methods] Two studies were conducted on fattening pigs and growing pigs respectively. Pigs in two experimental groupswere fed with low-protein diets supplemented with amino acids and normal diets respectively,and the initial weight,final weight,feed consumption and carcass index of the tested pigs were measured.[Results]The average final weight of fattening pigs in test group was 2. 4 kg lower than that in control group,while the difference was not significant( P > 0. 05). The average daily gain of fattening pigs in test group was 60 g lower than that in control group,indicating that low protein diets had a tendency to decrease the growth rate( P > 0. 05),and reduction of daily weight gain in low protein diet pigs reached 7. 6%. The development of pigs in low protein diet group was poor,however,the difference was not significant. Carcass backfat thickness in low-protein diet group was slightly higher than that in control group,the difference was significant( P < 0. 05). There was no significant difference in input-output ratio between low-protein diet group and control group.Finalweight of growing-finishing pigs in low-protein diet group was 3. 22 % lower than that in control group,the average daily weight gainwas 6. 91% lower than that in control group,feed intake in test group was 5. 48% lower than that in control group,and difference between the two treatments was significant( P < 0. 05). Feed meat ratio in low-protein diet group improved,however,the difference was not significant( P > 0. 05).[Conclusions]Production performance of the pigs would be decreasedand the input-output ratio could not be improvedwhen the adding contents of crude protein in diets was reduced 3%-4%. 5New insight into scale inhibition during tea brewing: Ca^(2+)/Mg^(2+) complexing and alkalinity consumption显示文摘Scale not only affects the taste and color ofwater,but also increases the risks of osteoporosis and cardiovascular diseases associated with drinking it.As a popular beverage,tea is rich many substances that have considerable potential for scale inhibition,including protein,tea polyphenols and organic acids.In this study,the effect of tea brewing on scale formationwas explored.It was found that the proteins,catechins and organic acids in tea leaves could be released when the green tea was brewed in water with sufficient hardness and alkalinity.The tea-released protein was able to provide carboxyl groups to chelate with calcium ions(Ca^(2+)),preventing the Ca^(2+)from reacting with the carbonate ions(CO_(3)^(2-)).The B rings of catechins were another important structure in the complexation of Ca^(2+)and magnesium ions(Mg2+).The carboxyl and hydroxyl groups on the organic acids was able to form fivemembered chelating rings with Ca^(2+)and Mg^(2+),resulting in a significant decrease in Ca^(2+)from 100.0 to 60.0 mg/L.Additionally,the hydrogen ions(H^(+))provided by the organic acids consumed and decreased the alkalinity of the water from 250.0 to 131.4 mg/L,leading to a remarkable reduction in pH from 8.93 to 7.73.It further prevented the bicarbonate(HCO_(3)^(-))from producing CO_(3)^(2−)when the water was heated.The reaction of the tea constituents with the hardness and alkalinity inhibited the formation of scale,leading to a significant decrease in turbidity from 10.6 to 1.4 NTU.Overall,this study provides information to help build towards an understanding of the scale inhibition properties of tea and the prospects of tea for anti-scaling in industrial applications.Linjun Li Jinsuo Lu Heliang Pang Zhiqiang Zhang Jing Yang Pengpeng Li Xiaoyu Yan Miaomiao Fan 2023Journal of Environmental Sciences2023,,2:0
