| 1 | Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection,histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection,tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection,and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys).In 30 of them,both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern,tumor staging,lymph node metastasis,grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma,andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. | 2 | Effects of four regulating-intestine prescriptions on pathology and ultrastructure of colon tissue in rats with ulcerative colitis显示文摘AIM: To observe different histomorphologic changes of ulcerative colitis (UC) rats that were treated with four regulating-intestine prescriptions (FRIP), to investigate the curative effects of FRIP and to analyze their treatment mechanism.METHODS: The UC rat model was made by the method of 2,4-dinitro chloro benzene (DNCB) immunity and acetic acid local enema. Ninety-eight SD rats were randomly divided into seven groups, namely, the normal control group, model group, salicylazosulfapyridine (SASP) group,Wumeiwan (WMW) group, Baitouwengtang (BTWT) group,Senglingbaishusan (SLBSS) group, and Tongxieyaofang (TXYF) group. Each group had 14 rats (with equal ratio of male and female). The six animal model groups of UC-SASP, TXYF, WMW, BTWT, SLBSS, TXYF-were treated by distilled water except the normal control group. Changes of the rat's general conditions after treatment were respectively observed, the colon tissue damage scores were given out, the pathology of colonic mucosa and changes of ultrastructure were analyzed.RESULTS: Different pathological changes on histology were shown after treatment by FRIP. The colon tissue damage score in model group was higher than that of FRIP groups and SASP group (q = 4.59, 4.77, P<0.05 or q = 5.48,6.25, 5.97, P<0.01). The scores of WMW group, BTWT group and SLBSS group were lower than that of SASP (q = 4.13, P<0.05 or q = 5.31, 5.12, P<0.01). There was no remarkable difference between the damage score of TXYF group and SASP group (q = 3.75, P>0.05). | 4 | Characterization of the aerosol chemical composition during the COVID-19 lockdown period in Suzhou in the Yangtze River Delta,China显示文摘To control the spread of COVID-19,rigorous restrictions have been implemented in China,resulting in a great reduction in pollutant emissions.In this study,we evaluated the air quality in the Yangtze River Delta during the COVID-19 lockdown period using satellite and ground-based data,including particle matter(PM),trace gases,water-soluble ions(WSIs) and black carbon(BC).We found that the impacts of lockdown policy on air quality cannot be accurately assessed using MODIS aerosol optical depth(AOD) data,whereas the tropospheric nitrogen dioxide(NO_(2)) vertical column density can well reflect the influences of these restrictions on human activities.Compared to the pre-COVID period,the PM_(2.5),PM_(10),NO_(2),carbon monoxide(CO),BC and WSIs during the lockdown in Suzhou were observed to decrease by 37.2%,38.3%,64.5%,26.1%,53.3% and 58.6%,respectively,while the sulfur dioxide(SO_(2)) and ozone(O_(3)) increased by 1.5% and 104.7%.The WSIs ranked in the order of NO_(3)~->NH_(4)^(+)>SO_(4)^(2-)>Cl~->Ca^(2+)>K^(+)>Mg^(2+)>Na + during the lockdown period.By comparisons with the ion concentrations during the pre-COVID period,we found that the ions NO 3-,NH 4 +,SO_(4)^(2-),Cl~-,Ca^(2+),K^(+) and Na^(+) decreased by 66.3%,48.8%,52.9%,56.9%,57.9% and 76.3%,respectively,during the lockdown,in contrast to Mg^(2+),which increased by 30.2%.The lockdown policy was found to have great impacts on the diurnal variations of Cl~-,SO_(4)^(2-),Na^(+) and Ca^(2+). | Honglei Wang Qing Miao Lijuan Shen Qian Yang Yezheng Wu Heng Wei Yan Yin Tianliang Zhao Bin Zhu Wen Lu | 2021 | Journal of Environmental Sciences2021,33,4: | 8 |
| 5 | Expanded endoscopic therapy criteria should be cautiously used in intramucosal gastric cancer显示文摘Objective: Accurate estimation of lymph node metastasis(LNM) in intramucosal gastric cancer is essential to select less invasive treatment options and even avoid surgery. The aim of this study was to evaluate combined clinicopathological features to predict the presence of LNM.Methods: A retrospective review of data from 386 intramucosal gastric cancer patients who underwent gastrectomy with extended lymphadenectomy from 2003 to 2010 was conducted. The mutual relation between clinicopathological characteristics and LNM was analyzed.Results: LNM was detected in 40(10.4%) of the 386 patients. Histological type and vascular or lymphatic invasion presence showed a positive correlation with LNM occurrence by univariate analysis. Multivariate analysis revealed that histological type was the only factor associated with LNM. Combined clinicopathologic characteristics would be more predictable for LNM. We found no LNM when we used combined clinicopathological characteristics conforming to Japanese absolute indications for endoscopic therapy. The LNM rate was as high as 8.7% when Japanese expanded criteria were used. Univariate analysis in cancer conformity to expand endoscopic submucosal dissection(ESD) indication also revealed that the undifferential type was the only significant factor for LNM.Conclusions: It was possible to predict intramucosal gastric cancer cases without LNM using combined clinicopathological characteristic analysis. The biological functions of these gaseous molecules, which were first discovered and identified as gasotransmitters in animals, have received unprecedented attention from plant scientists in recent decades. Researchers have arrived at the consensus that H_2S is synthesized endogenously and serves as a signaling molecule throughout the plant life cycle.However, the mechanisms of H_2S action in redox biology is still largely unexplored. This review highlights what we currently know about the characteristics and biosynthesis of H_2S in plants. Additionally,we summarize the role of H_2S in plant resistance to abiotic stress. | 11 | Spatio-temporal variation in rock exhumation linked to large-scale shear zones in the southeastern Tibetan Plateau显示文摘Crustal-scale shear zones are believed to have played an important role in the tectonic and landscape evolution of orogens. However, the variation of long-term rock exhumation between the interior of shear zones and adjacent regions has not been documented in detail. In this study, we obtained new zircon U-Pb, biotite ^(40)Ar/^(39)Ar, zircon and apatite(U-Th)/He data, and conducted inverse thermal history modeling from two age-elevation profiles(the Pianma and Tu'er profiles) in the southeastern Tibetan Plateau. Our goal is to constrain the exhumation history of the Gaoligong and Chongshan shear zones and adjacent regions, so as to explore the effect of the shear zones on exhumation and their thermal effect on cooling that should not be ignored. Our results suggest that during the interval of 18–11 Ma the exhumation rates of rocks within the Gaoligong shear zone are anomalously high compared with those outside of. The rapid cooling during 18–11 Ma appears to be restricted to the shear zone, likely due to localized thermal effects of shearing and exhumation. After 11 Ma, both the areas within and outside of the shear zones experienced a similar two-stage exhumation history: slower cooling until the early Pliocene, and then a rapid increase in cooling rate since the early Pliocene. Our results indicate a synchronized exhumation but with spatially varied exhumation rates. | 12 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 14 | Advanced primary peritoneal carcinoma:clinicopathological and prognostic factor analyses显示文摘Objective: To investigate the factors favoring a positive prognosis for advanced primary peritoneal carcinoma (PPC). Methods: Twenty-four cases meeting the criteria for PPC were analyzed retrospectively for the clinicopathologic profiles. Im- munohistochemistry was used to determine the expressions of p53, Top2α, Ki-67 and Her-2/neu. Then all these clinicopa- thological factors and molecular markers were correlated with the prognosis. Results: There were 15 cases of primary peritoneal serous papillary carcinoma (PPSPC), 6 cases of mixed epithelial carcinoma (MEC) and 3 cases of malignant mixed Mullerian tumor (MMMT). All patients underwent cytoreductive surgery with optimal debulking achieved in 3 cases. Among those re- ceiving first-line chemotherapy, 13 patients received the TP regimen (paclitaxel-cisplatin or carboplatin) and 7 patients received the PAC regimen (cisplatin-doxorubicin-cyclophosphamide). The median overall survival of all patients was 42 months, while the breakdown for survival time for patients with PPSPC, MMT and MEC was 44, 13 and 19 months, respectively. The expressions of p53, Top2α and Ki-67 were all demonstrated in 11 cases respectively. None showed the expression of Her-2/neu. There were significant differences in the median survival between patients with PPSPC and those with MMMT (44 months vs 13 months, P<0.05), also between patients receiving TP combination and those receiving the PAC regimen (75 months vs 28 months, P<0.05). Another significant difference in the median progression-free survival (PFS) was identified between patients with positive p53 immunostaining and those with negative p53 immunostaining (15 months vs 47 months, P<0.05), whereas age, menopausal status, residual tumor size and the other molecular factors did not significantly impact survival. Conclusion: Patients with PPC should be treated with a comprehensive management plan including appropriate cytoreductive surgery and responsive chemotherapy. Overestimating an optimal debulking surgery may not benefit survival. | 19 | Anisotropic,Wrinkled,and Crack-Bridging Structure for Ultrasensitive,Highly Selective Multidirectional Strain Sensors显示文摘Flexible multidirectional strain sensors are crucial to accurately determining the complex strain states involved in emerging sensing applications.Although considerable efforts have been made to construct anisotropic structures for improved selective sensing capabilities,existing anisotropic sensors suffer from a trade-off between high sensitivity and high stretchability with acceptable linearity.Here,an ultrasensitive,highly selective multidirectional sensor is developed by rational design of functionally different anisotropic layers.The bilayer sensor consists of an aligned carbon nanotube(CNT)array assembled on top of a periodically wrinkled and cracked CNT-graphene oxide film.The transversely aligned CNT layer bridge the underlying longitudinal microcracks to effectively discourage their propagation even when highly stretched,leading to superior sensitivity with a gauge factor of 287.6 across a broad linear working range of up to 100%strain.The wrinkles generated through a pre-straining/releasing routine in the direction transverse to CNT alignment is responsible for exceptional selectivity of 6.3,to the benefit of accurate detection of loading directions by the multidirectional sensor.This work proposes a unique approach to leveraging the inherent merits of two cross-influential anisotropic structures to resolve the trade-off among sensitivity,selectivity,and stretchability,demonstrating promising applications in full-range,multi-axis human motion detection for wearable electronics and smart robotics. | Heng Zhang Dan Liu Jeng-Hun Lee Haomin Chen Eunyoung Kim Xi Shen Qingbin Zheng Jinglei Yang Jang-Kyo Kim | 2021 | Nano-Micro Letters2021,13,8: | 3 |
| 20 | Association of Polygenic Risk Score with Age at Onset and Cerebrospinal Fluid Biomarkers of Alzheimer’s Disease in a Chinese Cohort显示文摘To evaluate whether the polygenic profile modifies the development of sporadic Alzheimer’s disease(sAD)and pathological biomarkers in cerebrospinal fluid(CSF),462 sAD patients and 463 age-matched cognitively normal(CN)controls were genotyped for 35 singlenucleotide polymorphisms(SNPs)that are significantly associated with sAD.Then,the alleles found to be associated with sAD were used to build polygenic risk score(PRS)models to represent the genetic risk.Receiver operating characteristic(ROC)analyses and the Cox proportional hazards model were used to evaluate the predictive value of PRS for the sAD risk and age at onset.We measured the CSF levels of Aβ42,Aβ42/Aβ40,total tau(T-tau),and phosphorylated tau(P-tau)in a subgroup(60 sAD and 200 CN participants),and analyzed their relationships with the PRSs.We found that 14 SNPs,including SNPs in the APOE,BIN1,CD33,EPHA1,SORL1,and TOMM40 genes,were associated with sAD risk in our cohort.The PRS models built with these SNPs showed potential for discriminating sAD patients from CN controls,and were able to predict the incidence rate of sAD and age at onset.Furthermore,the PRSs were correlated with the CSF levels of Aβ42,Aβ42/Aβ40,T-tau,and P-tau.Our study suggests that PRS models hold promise for assessing the genetic risk and development of AD.As genetic risk profiles vary among populations,large-scale genome-wide sequencing studies are urgently needed to identify the genetic risk loci of sAD in Chinese populations to build accurate PRS models for clinical practice. | Wei-Wei Li Zhen Wang Dong-Yu Fan Ying-Ying Shen Dong-Wan Chen Hui-Yun Li Ling Li Heng Yang Yu-Hui Liu Xian-Le Bu Wang-Sheng Jin Fan Zeng Zhi-Qiang Xu Jin-Tai Yu Li-Yong Chen Yan-Jiang Wang | 2020 | Neuroscience Bulletin2020,36,7: | 3 |
