1Hsa-miR-1246, hsa-miR-320a and hsa-miR-196b-5p inhibitors can reduce the cytotoxicity of Ebola virus glycoprotein in vitro显示文摘Ebola virus(EBOV)causes a highly lethal hemorrhagic fever syndrome in humans and has been associated with mortality rates of up to 91%in Zaire,the most lethal strain.Though the viral envelope glycoprotein(GP)mediates widespread inflammation and cellular damage,these changes have mainly focused on alterations at the protein level,the role of microRNAs(miRNAs)in the molecular pathogenesis underlying this lethal disease is not fully understood.Here,we report that the miRNAs hsa-miR-1246,hsa-miR-320a and hsa-miR-196b-5p were induced in human umbilical vein endothelial cells(HUVECs)following expression of EBOV GP.Among the proteins encoded by predicted targets of these miRNAs,the adhesion-related molecules tissue factor pathway inhibitor(TFPI),dystroglycan1(DAG1)and the caspase 8 and FADD-like apoptosis regulator(CFLAR)were significantly downregulated in EBOV GP-expressing HUVECs.Moreover,inhibition of hsa-miR-1246,hsa-miR-320a and hsa-miR-196b-5p,or overexpression of TFPI,DAG1 and CFLAR rescued the cell viability that was induced by EBOV GP.Our results provide a novel molecular basis for EBOV pathogenesis and may contribute to the development of strategies to protect against future EBOV pandemics.SHENG MiaoMiao ZHONG Ying CHEN Yang DU JianChao JU XiangWu ZHAO Chen ZHANG GuiGen ZHANG LiFang LIU KangTai YANG Ning XIE Peng LI DangSheng ZHANG Michael Q. 2Research on the Damage of Porosityand Permeabilitydue to Perforation on Sandstone in the Compaction Zone显示文摘A perforating hole is a channel through which the oil and gas in a reservoir pass into the production well bore.During the process of perforating due to explosion,the surrounding sandstone will be damaged to a certain extent,which will increase the well bore skin and lead to the decrease of production consequently.In this work a mechanical model of perforating damage is developed to describe the influences of perforating due to explosion on the porosity and permeability of the surrounding sandstone near the compaction zone.Based on this developed model,the important data related to the damage of sandstone,such as matrix effective stress,plastic deformation,volumetric strain,and so on,can be numerically simulated.Especially the behaviors of plasticity kinematic hardening at high strain rate due to impact loads,which are the important characteristics in the sandstone,is taken into account in this developed model.Both numerical and testing results show that the damage due to perforation in the sandstone can be accurately predicted by the developed model together with the porosity and permeability evolving model of perforation in a compaction zone.As a practical application,a methodology for the analysis of damage of porosity and permeability around a perforation tunnel is supposed based on the developed model and the core flow efficiency test of interparticle pore spaced sandstone target in China Shengli Oilfield and the computed tomography test.Shifeng Xue Xiuxing Zhu Lin Zhang Shenghu Zhu Guigen Ye Xuejun Fan 2016Computers, Materials & Continua2016,,1:2

9Orientational Chirality,Its Asymmetric Control,and Computational Study显示文摘Orientational chirality was discovered and characterized by a C(sp)-C(sp3)axis-anchored chiral center and a remotely anchored blocker.X-ray structural analysis proved that orientatiomers are stabilized by through-space functional groups,making it possible for 1 R-or S-chiral center to exhibit 3 orientational isomers simply by rotating operations.A new model system was proposed,fundamentally different from the traditional Felkin-Ahn-type or Cram-type models.In these traditional models,chiral C(sp^(3))center and blocking C(sp^(3))carbons are connected adjacently,and there exist 6 energy barriers during rotating along the C(sp^(2))-C(sp^(3))axis.In comparison,the present orientational chirality model shows that a chiral C(sp)-C(sp^(3))carbon is remotely located from a blocking group.Thus,it is focused on the steric dialog between a chiral C(sp^(3))center and a remotely anchored functional group.There exist 3 energy barriers for either(R)-or(S)-C(sp)-C(sp^(3))stereogenicity in the new model.Chiral amide auxiliary was proven to be an excellent chiral auxiliary in controlling rotations of orientatiomers to give complete stereoselectivity.The asymmetric synthesis of individual orientatiomers was conducted via multistep synthesis by taking advantage of the Suzuki-Miyaura cross-coupling and Sonogashira coupling reactions.Density functional theory computational study presented optimized conformers and relative energies for individual orientatiomers.This discovery would be anticipated to result in a new stereochemistry topic and have a broad impact on chemical,biomedical,and material sciences in the future.Shengzhou Jin Yu Wang Yao Tang Jia-Yin Wang Ting Xu Junyi Pan Sai Zhang Qiankai Yuan Anis Ur Rahman James D.McDonald Guo-Qiang Wang Shuhua Li Guigen Li 2023Research2023,,1:0
10Timing noise study of four pulsars显示文摘Analyzing up to ten years of pulsar timing data observed by the 25-m Nanshan telescope at Urumqi,China,we studied power spectra of timing residuals for four pulsars based on orthogonal polynomials.Our work reveals that an index of 4 is consistent with a power law model and reveals random walk noise in the rotating frequency.Stability with 8 is investigated and found to be consistent with previous results.A new method called the morphology filter is applied to denoise the pulsar timing residuals and to perform further spectrum analysis.The spectra obtained with this method are improved in accuracy compared with those from timing residuals of pulsars directly.ZHANG CaiHong WANG Na YUAN JianPing LIU ZhiYong LIU JingNan NIE GuiGen 2012Science China(Physics,Mechanics & Astronomy)2012,55,2:0

12Controlled generation of cell–laden hydrogel microspheres with core–shell scaffold mimicking microenvironment of tumor显示文摘Development of an in vitro three-dimensional(3D) model that closely mimics actual environment of tissue has become extraordinarily important for anti-cancer study. In recent years, various 3D cell culture systems have been developed,with multicellular tumor spheroids being the most popular and effective model. In this work, we present a microfluidic device used as a robust platform for generating core–shell hydrogel microspheres with precisely controlled sizes and varied components of hydrogel matrix. To gain a better understanding of the governing mechanism of microsphere formation,computational models based on multiphase flow were developed to numerically model the droplet generation and velocity field evolution process with COMSOL Multiphysics software. Our modeling results show good agreement with experiments in size dependence on flow rate as well as effect of vortex flow on microsphere formation. With real-time tuning of the flow rates of aqueous phase and oil phase, tumor cells were encapsulated into the microspheres with controllable core–shell structure and different volume ratios of core(comprised of alginate, Matrigel, and/or Collagen) and shell(comprised of alginate). Viability of cells in four different hydrogel matrices were evaluated by standard acridine orange(AO) and propidium iodide(PI) staining. 15Catalyst-Dependent Direct and Deoxygenative Coupling of Alcohols by Convergent Paired Electrolysis显示文摘Developing a general and mild approach to upgrade alcohols into high value products is a hot topic in synthetic chemistry because alcohol is one of the most abundant raw chemicals.Specifically,direct coupling and deoxygenative coupling of alcohols are the two main approaches for the functionalization of alcohols to afford structurally diverse products,and it receives considerable attention.Despite significant advances in the field,there still remains a great challenge to develop a general approach accommodating both coupling reactions,as they commonly involve distinct pathways.Herein,we report an electrochemical approach for the direct coupling and deoxygenative coupling of alcohols with fluorenones.Under paired electrolysis,this catalyst-dependent protocol gives divergent access to diols and tertiary alcohols.Moreover,the synthetic utility of 9H-fluoren-9-ol products has been demonstrated in the synthesis of organic luminophores,phenanthrol,phenanthridine,and amino alcohol.The present approach exhibits some impressive features:(a)catalystdependent selectivity;(b)excellentfunctional-group tolerance(156 examples);(c)mild conditions;and(d)good scalability(∼20 gram scale).Sheng Zhang Jianxue Shi Jingjing Li Man-Bo Li Guigen Li Michael Findlater 2022CCS Chemistry2022,4,6:0
