1以柑橘多胚性二倍体母本倍性杂交培育三倍体显示文摘2009—2012年,以柑橘8个多胚性二倍体品种为母本与近年始花的5个异源四倍体体细胞杂种和1个同源四倍体分别杂交,培育三倍体新种质。连续4年共配置15个杂交组合,授粉4 442朵花,坐果1 484个,平均坐果率33.4%;用于幼胚挽救的果实1 075个,共培养幼嫩种子12 578颗,经生芽、生根诱导获得再生植株2 832个;以流式细胞仪对所有再生植株倍性检测,15个组合共获得三倍体401个,四倍体121个;移栽成活三倍体349个,四倍体98个。对W.默科特×NH组合89个株系的三倍体的SSR分子鉴定表明,所有三倍体后代均为双亲的有性杂种。解凯东 王晓培 王惠芹 梁武军 谢宗周 郭大勇 伊华林 邓秀新 Grosser Jude W. 郭文武 2014园艺学报2014,41,4:16
2单胚性二倍体为母本与异源四倍体杂交大规模创制柑橘三倍体显示文摘【目的】以单胚性的二倍体柑橘品种为母本与异源四倍体体细胞杂种杂交培育三倍体植株。【方法】选择亲本配置杂交组合进行人工授粉,授粉后70—100 d采幼果进行胚挽救,以流式细胞仪结合根尖染色体压片对再生植株进行倍性检测,最后进行SSR分子标记鉴定。【结果】2009—2012连续4年,以8个二倍体为母本、以4个异源四倍体体细胞杂种为父本,共配置14个倍性杂交组合,授粉花数3 347朵,坐果数678个,平均坐果率20.26%。实施胚挽救的果实数505个,培养种子12 357粒,经生芽、生根诱导培养共获得1 022个株系。通过对再生植株的倍性检测,共获得三倍体植株755个株系,四倍体19个株系。对华农红柚×NH组合的三倍体、四倍体后代进行分子鉴定,表明三倍体和四倍体全部为双亲的有性杂种后代。【结论】本研究通过杂交获得的三倍体为我国无籽柑橘品种选育奠定了材料基础;同时获得的四倍体后代也为未来的柑橘三倍体育种提供了优良的亲本材料。解凯东 王惠芹 王晓培 梁武军 谢宗周 伊华林 邓秀新 Grosser Jude W 郭文武 2013中国农业科学2013,46,21:15
3以本地早橘和槾橘为母本倍性杂交创制柑橘三倍体显示文摘【目的】基于二倍体与四倍体倍性杂交策略创制柑橘三倍体新种质。【方法】以二倍体为母本,四倍体为父本进行人工授粉,授粉后85 d采摘幼果并对未成熟种子实施幼胚离体挽救培养;再生植株后,用流式细胞仪和根尖染色体计数法对再生植株进行倍性鉴定;并用SSR标记对三倍体后代进行分子鉴定。【结果】以本地早橘和槾橘为母本,8个异源四倍体体细胞杂种和1个双二倍体为父本,配置9个倍性杂交组合,共授粉2749朵花,坐果489个,平均坐果率17.8%;培养幼嫩种子2239粒,经幼胚离体挽救培养,共再生植株260株;用流式细胞仪和根尖染色体计数检测再生植株倍性,获得三倍体141株;SSR分子鉴定表明从槾橘×NS组合随机选取的50株三倍体后代全部为双亲的有性后代;三倍体后代在温室生长一年后,采用嫁接(枳砧)将这些三倍体定植于田间。【结论】这些三倍体新种质为本地早橘和槾橘无核新品种培育奠定了宝贵的材料基础。解凯东 彭珺 袁东亚 强瑞瑞 谢善鹏 周锐 夏强明 伍小萌 柯甫志 刘高平 GROSSER Jude W 郭文武 2020中国农业科学2020,53,23:9
4Molecular mechanism and functional consequences of lansoprazole-mediated heme oxygenase-1 induction显示文摘AIM:To investigate the molecular mechanism and functional consequences of heme oxygenase-1(HO-1) activation by lansoprazole in endothelial cells and macrophages. METHODS:Expression of HO-1 mRNA was analyzed by Northern blotting.Western blotting was used to determine the HO-1 and ferritin protein levels. NADPH-dependent reactive oxygen species(ROS) formation was measured with lucigenin-enhanced chemiluminescence.HO-1 promoter activity in mouse fibroblasts,stably transfected with a 15-kb HO-1 gene that drives expression of the reporter gene luciferase, was assessed using in vivo bioluminescence imaging. RESULTS:Lansoprazole increased HO-1 mRNA levels in endothelial cells and HO-1 protein levelsin macrophages.In addition,lansoprazole-induced ferritin protein levels in both cell systems.Moreover, induction of the antioxidant proteins HO-1 and ferritin by lansoprazole was followed by a decrease in NADPH- mediated ROS formation.The radical scavenging properties of lansoprazole were diminished in the presence of the HO inhibitor,chromium mesoporphyrin IX.Induction of HO-1 gene expression by lansoprazole was not related to oxidative stress or to the activation of the mitogen-activated protein kinase pathway. However,the phosphatidylinositol 3-kinase inhibitor LY294002 showed a concentration-dependent inhibition of HO-1 mRNA and promoter activity. 5柑桔原生质体融合再生出叶肉亲本类型植株显示文摘粗柠檬(Citrus jambhiri Lush)叶肉原生质体与哈姆林甜橙(C.sinensis Osbeck)悬浮细胞系原生质经聚乙二醇(PEG)诱导融合、再生的植株中,除体细胞杂种外,发现两棵为二倍体,与其亲本一样,2n=2x=18。这两棵植株叶片形态似粗柠檬。GOT、SOD及POX 3种同工酶分析结果显示,其酶带与粗柠檬基本相同,但个别带纹有差异。本文就这类植株的来源问题作了讨论。邓秀新 肖顺元 章文才 J. W. Grosser 1993中国柑桔1993,22,2:7
