4Calibration after bootstrap for accurate uncertainty quantification in regression models显示文摘Obtaining accurate estimates of machine learning model uncertainties on newly predicted data is essential for understanding the accuracy of the model and whether its predictions can be trusted.A common approach to such uncertainty quantification is to estimate the variance from an ensemble of models,which are often generated by the generally applicable bootstrap method.In this work,we demonstrate that the direct bootstrap ensemble standard deviation is not an accurate estimate of uncertainty but that it can be simply calibrated to dramatically improve its accuracy.We demonstrate the effectiveness of this calibration method for both synthetic data and numerous physical datasets from the field of Materials Science and Engineering.The approach is motivated by applications in physical and biological science but is quite general and should be applicable for uncertainty quantification in a wide range of machine learning regression models.Glenn Palmer Siqi Du Alexander Politowicz Joshua Paul Emory Xiyu Yang Anupraas Gautam Grishma Gupta Zhelong Li Ryan Jacobs Dane Morgan 2022npj Computational Materials2022,,1:0
5Crystal structure of Arabidopsis thaliana HPPK/DHPS,a bifunctional enzyme and target of the herbicide asulam显示文摘Herbicides are vital formodern agriculture,but their utility is threatened by genetic or metabolic resistance in weeds,as well as regulatory barriers.Of the known herbicide modes of action,7,8-dihydropterin synthase(DHPS),which is involved in folate biosynthesis,is targeted by just one commercial herbicide,asulam.A mimic of the substrate para-aminobenzoic acid,asulam is chemically similar to sulfonamide antibiotics,and although it is still in widespread use,asulam has faced regulatory scrutiny.With an entire mode of action represented by just one commercial agrochemical,we sought to improve the understanding of its plant target.Here we solve a 2.3A°resolution crystal structure for Arabidopsis thaliana DHPS that is conjoined to 6-hydroxymethyl-7,8-dihydropterin pyrophosphokinase(HPPK),and we reveal a strong structural conservation with bacterial counterparts at the sulfonamide-bindingpocket of DHPS.We demonstrate that asulamand the antibiotic sulfamethoxazole have herbicidal as well as antibacterial activity,andwe explore the structural basis of their potency by modeling these compounds in mitochondrial HPPK/DHPS.Our findings suggest limited opportunity for the rational design of plant selectivity fromasulamand indicate that pharmacokinetic or delivery differences between plants andmicrobesmight be the bestways to safeguard thismode of action.Grishma Vadlamani Kirill V.Sukhoverkov Joel Haywood Karen J.Breese Mark F.Fisher Keith A.Stubbs Charles S.Bond Joshua S.Mylne 2022Plant Communications2022,3,4:0
6A case report of acute pediatric bacterial meningitis due to the rare isolate,Pseudomonas putida显示文摘Acute bacterial meningitis(ABM)is the medical emergency which warrants an early diagnosis and an aggressive therapy.Despite the availability of the potent newer antibiotics,the mortality caused by ABM and its complications remain high in India,ranging from 16%to 32%.The aim of this case report is to present the rare isolation of Pseudomonas putida from cerebrospinal fluid sample.Besides this,the author also emphasizes the importance of correctly identifying the organism and thus the selection of the most accurate antibiotic from the susceptibility profile to allow for early recovery and to improve the patient outcome and survival.Grishma V.Kulkarni 2016Neuroimmunology and Neuroinflammation2016,3,1:0
