| 5 | Optimization of the Assembly Efficiency for Lidamycin Chromophore Bound to its Apoprotein:A Case Study using Orthogonal Array显示文摘Objective Lidamycin (LDM) can be dissociated to an apoprotein (LDP) and an active enediyne chromophore (AE).The detached AE can reassemble with its LDP‐containing fusion protein to endow the latter with potent antitumor activity.However,the reassembly of AE with LDP is affected by several factors.Our aim was to optimize the assembly efficiency of the AE with a LDP‐containing fusion protein and investigate the influence of several factors on the assembly efficacy.Methods A method based on RP‐HPLC was developed to analyze the assembly rate,and an orthogonal experimental design L9 (3 4 ) was used to investigate the effects of temperature,assembly time,pH and molecular ratio of LDP‐containing fusion protein to AE on the assembly rate.Furthermore,the determined optimum conditions for the assembly rate of the LDP‐containing fusion protein with AE were applied and evaluated.Results A calibration curve based on the LDM micromolar concentration against the peak‐area of AE by HPLC was obtained.The order in which individual factors in the orthogonal experiment affected the assembly rate were temperature>time>pH>molar ratio of AE to protein and all were statistically significant (P<0.01).The optimal assembly conditions were temperature at 10°C,time of 12 h,pH 7.0,and the molar ratio of AE: protein of 5:1.The assembly rate of AE with a LDP‐containing fusion protein was improved by 23% after condition optimization.Conclusion The assembly rate of chromophore of lidamycin with its LDP‐containing fusion protein was improved after condition optimization by orthogonal design,and the optimal conditions described herein should prove useful for the development of this type of LDP‐containing fusion protein. | ZHONG Gen Shen GUO Xiao Fang ZHANG Sheng Hua ZHEN Yong Su | 2011 | Biomedical and Environmental Sciences2011,24,6: | 9 |
| 6 | Research on the Anticorrosion Coating Under the Paved Layer for Highway Steel Box Bridge Deck显示文摘The corrosion of the anticorrosion coating and the defects of the asphalt concrete paved layer have been investigated on long-span steel box bridge decks. The anticorrosion coating lies in the middle of two entirely different materials: a highway steel box bridge deck and a paved layer, which is used as anticorrosion and waterproof coating for the steel bridge deck. For our study, electrochemical corrosion and pull strength experiments have been selected for the investigation of the corrosion properties of inorganic zinc rich coating, epoxy zinc rich coating and arc sprayed zinc coating. The adhesive strength between the coatings and the panel, and the effect of the coating corrosion on the shear properties of the paved layers including cast asphalt, thermal asphalt mortar, epoxy asphalt and modified asphalt concrete have been investigated. The results show that the adhesive strength between the coatings and the bridge panel is controlled by the method of pre-processing rust removal. Coating by sandblasting has stronger adhesive strength than coating by shot peening. The results also reveal that shear strength of the paved layer is affected by the corrosion product of zinc coating. | 9 | Assessment of dairy cow feed intake based on BP neural network with polynomial decay learning rate显示文摘To overcome the shortcomings of traditional dairy cow feed intake assessment model andBP neural network, this paper proposes a method of optimizing BP neural network usingpolynomial decay learning rate, taking the cow’s body weight, lying duration, lying times,walking steps, foraging duration and concentrate-roughage ratio as input variables andtaking the actual feed intake is the output variable to establish a dairy cow feed intakeassessment model, and the model is trained and verified by experimental data collectedon site. For the sake of comparative study, feed intake is simultaneously assessed by SVRmodel, KNN logistic regression model, traditional BP neural network model, and multilayerBP neural network model. The results show that the established BP model using the polynomial decay learning rate has the highest assessment accuracy, the MSPE, RMSE, MAE,MAPE and R2 are 0.043 kg2/d and 0.208 kg/d, 0.173 kg/d, 1.37% and 0.94 respectively. Compared with SVR model and KNN mode, the RMSE value reduced by 43.9% and 26.5%, it isalso found that the model designed in this paper has many advantages in comparison withthe BP model and multilayer BP model in terms of precision and generalization. | 13 | N-doped graphene quantum dot-decorated N-TiO2/P-doped porous hollow g-C_(3)N_(4) nanotube composite photocatalysts for antibiotic photodegradation and H2 production显示文摘Exclusive responsiveness to ultraviolet light (~3.2 eV) and high photogenerated charge recombination rate are the two primary drawbacks of pure TiO_(2). We combined N-doped graphene quantum dots (N-GQDs), morphology regulation, and heterojunction construction strategies to synthesize N-GQD/N-doped TiO_(2)/P-doped porous hollow g-C_(3)N_(4) nanotube (PCN) composite photocatalysts (denoted as G-TPCN). The optimal sample (G-TPCN doped with 0.1wt% N-GQD, denoted as 0.1% G-TPCN) exhibits significantly enhanced photoabsorption, which is attributed to the change in bandgap caused by elemental doping (P and N), the improved light-harvesting resulting from the tube structure, and the upconversion effect of N-GQDs. In addition, the internal charge separation and transfer capability of0.1% G-TPCN are dramatically boosted, and its carrier concentration is 3.7, 2.3, and 1.9 times that of N-TiO_(2), PCN, and N-TiO_(2)/PCN(TPCN-1), respectively. This phenomenon is attributed to the formation of Z-scheme heterojunction between N-TiO_(2) and PCNs, the excellent electron conduction ability of N-GQDs, and the short transfer distance caused by the porous nanotube structure. Compared with those of N-TiO_(2), PCNs, and TPCN-1, the H2 production activity of 0.1%G-TPCN under visible light is enhanced by 12.4, 2.3, and 1.4times, respectively, and its ciprofloxacin (CIP) degradation rate is increased by 7.9, 5.7, and 2.9 times, respectively. The optimized performance benefits from excellent photoresponsiveness and improved carrier separation and migration efficiencies. Finally, the photocatalytic mechanism of 0.1% G-TPCN and five possible degradation pathways of CIP are proposed.
