1Scalable volumetric imaging for ultrahigh-speed brain mapping at synaptic resolution显示文摘The speed of high-resolution optical imaging has been a rate-limiting factor for meso-scale mapping of brain structures and functional circuits,which is of fundamental importance for neuroscience research.Here,we describe a new microscopy method of Volumetric Imaging with Synchronized on-the-fly-scan and Readout(VISo R)for high-throughput,high-quality brain mapping.Combining synchronized scanning beam illumination and oblique imaging over cleared tissue sections in smooth motion,the VISo R system effectively eliminates motion blur to obtain undistorted images.By continuously imaging moving samples without stopping,the system achieves high-speed 3D image acquisition of an entire mouse brain within1.5 hours,at a resolution capable of visualizing synaptic spines.A pipeline is developed for sample preparation,imaging,3D image reconstruction and quantification.Our approach is compatible with immunofluorescence methods,enabling flexible cell-type specific brain mapping and is readily scalable for large biological samples such as primate brains.Using this system,we examined behaviorally relevant whole-brain neuronal activation in 16 c-Fos-sh EGFP mice under resting or forced swimming conditions.Our results indicate the involvement of multiple subcortical areas in stress response.Intriguingly,neuronal activation in these areas exhibits striking individual variability among different animals,suggesting the necessity of sufficient cohort size for such studies.Hao Wang Qingyuan Zhu Lufeng Ding Yan Shen Chao-Yu Yang Fang Xu Chang Shu Yujie Guo Zhiwei Xiong Qinghong Shan Fan Jia Peng Su Qian-Ru Yang Bing Li Yuxiao Cheng Xiaobin He Xi Chen Feng Wu Jiang-Ning Zhou Fuqiang Xu Hua Han Pak-Ming Lau Guo-Qiang Bi 2019National Science Review2019,6,5:7
2Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. 4Divergent Projection Patterns Revealed by Reconstruction of Individual Neurons in Orbitofrontal Cortex显示文摘The orbitofrontal cortex(OFC)is involved in diverse brain functions via its extensive projections to multiple target regions.There is a growing understanding of the overall outputs of the OFC at the population level,but reports of the projection patterns of individual OFC neurons across different cortical layers remain rare.Here,by combining neuronal sparse and bright labeling with a whole-brain florescence imaging system(fMOST),we obtained an uninterrupted three-dimensional whole-brain dataset and achieved the full morphological reconstruction of 25 OFC pyramidal neurons.We compared the wholebrain projection targets of these individual OFC neurons in different cortical layers as well as in the same cortical layer.We found cortical layer-dependent projections characterized by divergent patterns for information delivery.Our study not only provides a structural basis for understanding the principles of laminar organizations in the OFC,but also provides clues for future functional and behavioral studies on OFC pyramidal neurons.Junjun Wang Pei Sun Xiaohua Lv Sen Jin Anan Li Jianxia Kuang Ning Li Yadong Gang Rui Guo Shaoqun Zeng Fuqiang Xu Yu-Hui Zhang 2021Neuroscience Bulletin2021,37,4:5
5Cortical Organization of Centrifugal Afferents to the Olfactory Bulb: Mono-and Trans-synaptic Tracing with Recombinant Neurotropic Viral Tracers显示文摘Sensory processing is strongly modulated by different brain and behavioral states,and this is based on the top-down modulation.In the olfactory system,local neural circuits in the olfactory bulb(OB)are innervated by centrifugal afferents in order to regulate the processing of olfactory information in the OB under different behavioral states.The purpose of the present study was to explore the organization of neural networks in olfactory-related cortices and modulatory nuclei that give rise to direct and indirect innervations to the glomerular layer(GL)of the OB at the whole-brain scale.Injection of different recombinant attenuated neurotropic viruses into the GL showed that it received direct inputs from each layer in the OB,centrifugal inputs from the ipsilateralanterior olfactory nucleus(AON),anterior piriform cortex(Pir),and horizontal limb of diagonal band of Broca(HDB),and various indirect inputs from bilateral cortical neurons in the AON,Pir,amygdala,entorhinal cortex,hippocampus,HDB,dorsal raphe,median raphe and locus coeruleus.These results provide a circuitry basis that will help further understand the mechanism by which olfactory informationprocessing in the OB is regulated.Pengjie Wen Xiaoping Rao Liuying Xu Zhijian Zhang Fan Jia Xiaobin He Fuqiang Xu 2019Neuroscience Bulletin2019,35,4:4

7Numerical study on the thermal performance of packed-bed latent heat thermal energy storage system with biomimetic alveoli structure capsule显示文摘Bionics provides a positive and beneficial impact on the development of various materials and systems, which has been widely used in energy storage, heat transfer enhancement, and solar thermochemical reactions. In this paper, the idea of heat storage unit with biomimetic alveoli structure is proposed and introduced to increase the heat transfer area and improve the thermal performance of the packed-bed latent heat thermal energy storage(LHTES) system. The effects of heat storage unit structure on temperature distribution, liquid fraction, thermal storage capacity, and other performance indexes of the conventional model and biomimetic model are analyzed. The numerical results indicate that the biomimetic alveoli structure can improve the thermal response of heat storage unit, increase the specific surface area, and achieve a faster heat storage process of the packed-bed compared with the conventional structure. Among various existing models, the double-cell model has a significant improvement.Compared with the conventional model, the maximum increase of liquid fraction and charging efficiency are 9.8% and 6.4%,respectively. 9Numerical Study of Effect of Compressor Swirling Flow on Combustor Design in a MTE显示文摘An effect of the swirling flow on the combustion performance is studied by the computational fluid dynamics(CFD) in a micro-gas turbine with a centrifugal compressor, dump diffuser and forward-flow combustor. The distributions of air mass and the Temperature Pattern Factor(as: Overall Temperature Distribution Factor-OTDF) in outlet are investigated with two different swirling angles of compressed air as 0° and 15° in three combustors. The results show that the influences of swirling flow on the air distribution and OTDF cannot be neglected. Compared with no-swirling flow, the air through outer liner is more, and the air through the inner liner is less, and the pressure loss is bigger under the swirling condition in the same combustor. 10Rapid and Sparse Labeling of Neurons Based on the Mutant Virus-Like Particle of Semliki Forest Virus显示文摘Sparse labeling of neurons contributes to uncovering their morphology, and rapid expression of a fluorescent protein reduces the experiment range. To achieve the goal of rapid and sparse labeling of neurons in vivo, we established a rapid method for depicting the fine structure of neurons at 24 h post-infection based on a mutant viruslike particle of Semliki Forest virus. Approximately 0.014 fluorescent focus-forming units of the mutant virus-like particle transferred enhanced green fluorescent protein into neurons in vivo, and its affinity for neurons in vivo was stronger than for neurons in vitro and BHK21(baby hamster kidney) cells. Collectively, the mutant virus-likeparticle provides a robust and convenient way to reveal the fine structure of neurons and is expected to be a helper virus for combining with other tools to determine their connectivity. 11Regional Metabolic Patterns of Abnormal Postoperative Behavioral Performance in Aged Mice Assessed by XH-NMR Dynamic Mapping Method显示文摘Abnormal postoperative neurobehavioral performance(APNP)is a common phenomenon in the early postoperative period.The disturbed homeostatic status of metabolites in the brain after anesthesia and surgery might make a significant contribution to APNP.The dynamic changes of metabolites in different brain regions after anesthesia and surgery,as well as their potential association with APNP are still not well understood.Here,we used a battery of behavioral tests to assess the effects of laparotomy under isoflurane anesthesia in aged mice,and investigated the metabolites in 12 different sub-regions of the brain at different time points using proton nuclear magnetic resonance('H-NMR)spectroscopy.The abnormal neurobehavioral performance occurred at 6 h and/or 9 h,and recovered at 24 h after anesthesia/surgery.Compared with the control group,the altered metabolite of the model group at 6 h was aspartate(Asp),and the difference was mainly displayed in the cortex;while significant changes at 9 h occurred predominantly in the cortex and hippocampus,and the corresponding metabolites were Asp and glutamate(Glu).All changes returned to baseline at 24 h.The altered metabolic changes could have occurred as a result of the acute APNP,and the metabolites Asp and Glu in the cortex and hippocampus could provide preliminary evidence for understanding the APNP process.Taotao Liu Zhengqian Li Jindan He Ning Yang Dengyang Han Yue Li Xuebi Tian Huili Liu Anne Manyande Hongbing Xiang Fuqiang Xu Jie Wang Xiangyang Guo 2020Neuroscience Bulletin2020,36,1:2

13SOD1^(G93A) Induces a Unique PSAP-Dependent Mitochondrial Apoptosis Pathway via Bax-Bak Interaction显示文摘Amyotrophic lateral syndrome(ALS)is a progressive degenerative disorder characterized by motor neuron death and axon degeneration.Mitochondrial dysfunction plays a key role in the pathogenesis of ALS,the mechanism of which remains poorly understood.The B-cell lymphoma-2(Bcl-2)family of proteins that control and mediate mitochondrial function and apoptosis,including the pro-apoptotic members Bcl2-Associated X(Bax),are involved in ALS development.The death receptor 6(DR6)regulates motor neuron death in ALS,and DR6 antibodies can prevent axon degeneration and motor neuron damage by blocking DR6.Previous studies demonstrated that PSAP localized to mitochondria and was required for DR6-induced apoptosis.In this study,SOD1^(G93A) was transfected into the motor neuron cell line NSC-34 to serve as an ALS cell model in vitro.The data assessed the role of PSAP in SOD1^(G93A)induced apoptosis and demonstrated that the overexpression of SOD1^(G93A),but not wtSOD1,induced PARP cleavage,caspase-3 activation,cytochrome c release,and Bax translocation.PSAP,Bax,and Bak were necessary for SOD1^(G93A)induced apoptosis,as silencing PSAP inhibited SOD1^(G93A)-mediated cell death that was dependent on Bax-Bak interaction.HAN NIU XIN CHEN XUEQI FU JINGTIAN ZHANG GUODONG LI YUXIANG WANG JIAYUE SONG XUETING MA CHEN HU XUEMIN XU FUQIANG ZHANG LINLIN ZENG 2021BIOCELL2021,45,4:2
14Experimental Investigation on Ignition and Lean Blow-out Performance of a Multi-sector Centrally Staged Combustor显示文摘Improvement on extinction and pollution emission have become one of the most prominent research topics in gas turbine.It is widely recognized that the fuel/air mixture distribution in the recirculation zone is a critical factor in improving lean blow-out(LBO) and ignition.This paper proposed a new low emission scheme with fuel staged centrally and hybrid injector to improve flameout and emission.A relative small amount of fuel enters into central pilot airblast atomizer burner and then atomized by inner swirl air.The remaining majority of fuel is directly injected into vane channels of the primary swirler through a series of holes located on the sidewall of the main stage.Only pilot stage is fueled under ignition and lean flameout condition.The uniformity of fuel/air mixture distribution in the primary zone of the new design decreases NOX emission,meanwhile the fuel air mixture in pilot recirculation zone is locally rich to improve flameout and ignition.Experimental investigation was conducted to compare the new scheme with baseline design of dual-swirler in terms of LBO and ignition characteristics under the same condition in a multi-sector combustor.It is found that the fuel-air ratio of ignition limit and LBO decrease with the reference velocity increasing.The experimental results also show that the new scheme successfully improve lean blow-out and broaden the operation range of the combustor.The experimental results indicated that the centrally staged scheme can widen the operation boundary of the combustor and can provide guidance for design and optimization of combustion chamber.LIU Fuqiang ZHANG Kaiyu MU Yong LIU Cunxi YANG Jinhu Xu Gang ZHU Junqiang 2014Journal of Thermal Science2014,23,5:2
15Rabies Virus Pseudotyped with CVS-N2C Glycoprotein as a Powerful Tool for Retrograde Neuronal Network Tracing显示文摘Efficient viral vectors for mapping and manipulating long-projection neuronal circuits are crucial in structural and functional studies of the brain. The SAD strain rabies virus with the glycoprotein gene deleted pseudotyped with the N2 C glycoprotein(SAD-RV(DG)-N2 C(G)) shows strong neuro-tropism in cell culture, but its in vivo efficiency for retrograde gene transduction and neuro-tropism have not been systematically characterized.We compared these features in different mouse brain regions for SAD-RV-N2 C(G) and two other widely-used retrograde tracers, SAD-RV(DG)-B19(G) and r AAV2-retro. We found that SAD-RV(DG)-N2 C(G) enhanced the infection efficiency of long-projecting neurons by^10 times but with very similar neuro-tropism, compared with SAD-RV(DG)-B19(G). On the other hand, SAD-RV(DG)-N2 C(G) had an infection efficiency comparable with r AAV2-retro, but a more restricted diffusion range, and broader tropism to different types and regions of longprojecting neuronal populations. 18Overexpression of Sirt6 ameliorates sleep deprivation induced-cognitive impairment by modulating glutamatergic neuron function显示文摘Sleep benefits the restoration of energy metabolism and thereby suppo rts neuronal plasticity and cognitive behaviors.Sirt6 is a NAD+-dependent protein deacetylase that has been recognized as an essential regulator of energy metabolism because it modulates various transcriptional regulators and metabolic enzymes.The aim of this study was to investigate the influence of Sirt6 on cerebral function after chronic sleep deprivation(CSD).We assigned C57BL/6J mice to control or two CSD groups and subjected them to AAV2/9-CMV-EGFP or AAV2/9-CMV-Sirt6-EGFP infection in the prelimbic cortex(PrL).We then assessed cerebral functional connectivity(FC) using resting-state functional MRI,neuron/astrocyte metabolism using a metabolic kinetics analysis;dendritic spine densities using sparse-labeling;and miniature excitato ry postsynaptic currents(mEPSCs) and action potential(AP) firing rates using whole-cell patchclamp recordings.In addition,we evaluated cognition via a comprehensive set of behavioral tests.Compared with controls,Sirt6 was significantly decreased(P<0.05) in the PrL after CSD,accompanied by cognitive deficits and decreased FC between the PrL and accumbens nucleus,piriform cortex,motor co rtex,somatosensory co rtex,olfactory tubercle,insular cortex,and cerebellum.Sirt6 ove rexpression reve rsed CSD-induced cognitive impairment and reduced FC.Our analysis of metabolic kinetics using [1-13C] glucose and [2-13C] acetate showed that CSD reduced neuronal Glu4and GABA2synthesis,which could be fully restored via forced Sirt6 expression.Furthermore,Sirt6 ove rexpression reversed CSD-induced decreases in AP firing rates as well as the frequency and amplitude of mEPSCs in PrL pyramidal neurons.These data indicate that Sirt6 can improve cognitive impairment after CSD by regulating the PrL-associated FC network,neuronal glucose metabolism,and glutamatergic neurotransmission.Thus,Sirt6 activation may have potential as a novel strategy for treating sleep disorder-related diseases.Jinpiao Zhu Chang Chen Zhen Li Xiaodong Liu Jingang He Ziyue Zhao Mengying He Binbin Nie Zili Liu Yingying Chen Kuanpin Su Xiang Li Juxiang Chen Hongbing Xiang Fuqiang Xu Kangguang Lin Zongze Zhang Jie Wang 2023Neural Regeneration Research2023,18,11:1

20Experimental Studies on Fuel Spray Characteristics of Pressure-Swirl Atomizer and Air-Blast Atomizer显示文摘In this work,the effects of fuel temperatures and pressure drops on the flow field and spray characteristics of a pressure-swirl atomizer were discussed using the Particle Imaging Velocimetry(PIV),Planar Laser Induced Fluorescence(PLIF)and Laser Particle Size Analyzer(LPSA)methods.Then the air-blast atomizer was selected to study the interaction of initial atomization and flow field.The effect of fuel-air ratio on the air-blast atomizer were also considered,where the fuel-air ratio was varied by adjusting mass flow rate of the air and fuel respectively.The results show that the spray angle of the pressure-swirl atomizer increases first and changes a little after the pressure drop higher than 0.5 MPa.However,more fuel concentrate on the central region,which is mainly caused by the increase of the proportion of small droplets with lower centrifugal force.The fuel temperature can improve the spray angle only in lower pressure drop,and it has a little effect under higher pressure drops.In addition,the fuel pressure drop has an obvious influence on the fuel distribution and flow field near the nozzle exit compared with the downstream.For the air-blast atomizer,the spray angle increases compared with the pressure-swirl atomizer for the introduction of swirl air.Furthermore,the spray angle decreases with the air mass rate increasing,and it increases with the fuel mass rate increasing.The distribution of velocity and droplet near the nozzle exit is influenced by the air mass rate,and the fuel mass rate mainly affects the distribution in the downstream.The fuel accumulates in the annular area below the nozzle,and the distribution of it changes little with the development along the axial direction.WANG Kaixing FAN Xiongjie LIU Fuqiang LIU Cunxi LU Haitao XU Gang 2021Journal of Thermal Science2021,30,2:1
