| 1 | Long-term bone and lung consequences associated with hospital-acquired severe acute respiratory syndrome:a 15-year follow-up from a prospective cohort study显示文摘The most severe sequelae after rehabilitation from SARS are femoral head necrosis and pulmonary fibrosis. We performed a 15-year follow-up on the lung and bone conditions of SARS patients. We evaluated the recovery from lung damage and femoral head necrosis in an observational cohort study of SARS patients using pulmonary CT scans, hip joint MRI examinations, pulmonary function tests and hip joint function questionnaires. Eighty medical staff contracted SARS in 2003. Two patients died of SARS, and78 were enrolled in this study from August 2003 to March 2018. Seventy-one patients completed the 15-year follow-up. The percentage of pulmonary lesions on CT scans diminished from 2003(9.40 ± 7.83)% to 2004(3.20 ± 4.78)%(P < 0.001) and remained stable thereafter until 2018(4.60 ± 6.37)%. Between 2006 and 2018, the proportion of patients with interstitial changes who had improved pulmonary function was lower than that of patients without lesions, as demonstrated by the one-second ratio(FEV1/FVC%, t = 2.21, P = 0.04) and mid-flow of maximum expiration(FEF25%–75%, t = 2.76, P = 0.01). The volume of femoral head necrosis decreased significantly from 2003(38.83 ± 21.01)% to 2005(30.38 ± 20.23)%(P = 0.000 2), then declined slowly from 2005 to 2013(28.99 ± 20.59)% and plateaued until 2018(25.52 ± 15.51)%. Pulmonary interstitial damage and functional decline caused by SARS mostly recovered, with a greater extent of recovery within 2 years after rehabilitation. | 3 | Severity of early allograft dysfunction following donation after circulatory death liver transplantation:a multicentre study显示文摘Background:Early allograft dysfunction(EAD)is associated with decreased graft and patient survival rates.This study aimed to identify the severity of EAD and develop a predictive model for EAD after donation after circulatory death(DCD)liver transplantation(LT).Furthermore,the influence of operative time on EAD incidence was also evaluated.Methods:In this retrospective,multicentre cohort study,nomograms were established based on a single-centre training cohort(n=321)and validated in a 3-center validation cohort(n=501).Results:The incidence rate of EAD was 46.4%(149/321)in the training cohort and 40.5%(203/501)in the validation cohort.Of the 149 EAD patients in the training cohort,77 patients with either elevated alanine aminotransferase(ALT)or aspartate aminotransferase(AST)were classified as having EAD type A,and the rest of the EAD patients were classified as having EAD type B.Recipients with EAD type B had lower graft and patient survival rates than recipients with EAD type A(P=0.043 and 0.044,respectively).We further developed a nomogram to predict EAD(graft weight,cold ischemia time,donor age,model for end-stage liver disease(MELD)score)and another nomogram to predict EAD type B(graft weight,cold ischemia time,MELD score).The nomograms for the prediction of EAD and EAD type B had good discrimination[concordance index(C-index)=0.712(0.666-0.758),0.707(0.641-0.773)]and calibration[Hosmer-Lemeshow(HL)P=0.384,P=0.425]in the validation cohort.An increased operative time(>6 h)was associated with increased EAD and EAD type B incidence in the high-risk group(P=0.005,P=0.020,respectively).Conclusions:EAD type B was associated with decreased graft and patient survival rates.The novel nomograms effectively predicted the incidence of EAD and EAD type B in DCD LT patients. | Kun Wang Di Lu Yuhui Liu Wangyao Li Li Zhuang Zhenyu Ma Qinfen Xie Binhua Pan Yichao Wu Junli Chen Lidan Lin Xiaowen Feng Qiang Wei Xuyong Wei Haiyang Xie Zhengxin Wang Shusen Zheng Xiao Xu | 2021 | Hepatobiliary Surgery and Nutrition2021,10,1: | 4 |
| 4 | Non-contact tensile viscoelastic characterization of microscale biological materials显示文摘Many structures and materials in nature and physiology have important 'meso-scale' structures at the micron lengthscale whose tensile responses have proven difficult to characterize mechanically. Although techniques such as atomic force microscopy and micro-and nano-identation are mature for compression and indentation testing at the nano-scale, and standard uniaxial and shear rheometry techniques exist for the macroscale, few techniques are applicable for tensile-testing at the micrometre-scale, leaving a gap in our understanding of hierarchical biomaterials. Here, we present a novel magnetic mechanical testing(MMT) system that enables viscoelastic tensile testing at this critical length scale. The MMT system applies non-contact loading, avoiding gripping and surface interaction effects. | 9 | Construction of a CaHP04-PGUSl hybrid nanoflower through protein-inorganic self-assembly, and its application in glycyrrhetinic acid 3-0-mono-β-D-glucuronide preparation显示文摘Glycyrrhetinic acid 3-0-mono-β-D-glucuronide (GAMG), an important pharmaceutical intermediate and functional sweetener, has broad applications in the food and medical industries. A green and cost-effective method for its preparation is highly desired. Using sitedirected mutagenesis, we previously obtained a variant of β-glucuronidase from Aspergillus oryzae Li-3 (PGUS1), which can specifically transform glycyrrhizin (GL) into GAMG. In this study, a facile method was established to prepare a CaHP04-PGUSl hybrid nanoflower for enzyme immobilization, based on protein-inorganic hybrid selfassembly. Under optimal conditions, 1.2 mg of a CaHP04- PGUS1 hybrid nanoflower precipitate with 71.2% immobilization efficiency, 35.60 mg·g^-1 loading capacity, and 118% relative activity was obtained. Confocal laser scanning microscope and scanning electron microscope results showed that the enzyme was encapsulated in the CaHP04-PGUSl hybrid nanoflower. Moreover, the thermostability of the CaHP04-PGUSl hybrid nanoflower at 55°C was improved, and its half-life increased by 1.3 folds. | 16 | The Association Between H3K4me3 and Antisense Transcription显示文摘Histone H3 lysine 4 trimethylation(H3K4me3) is well known to occur in the promoter region of genes for transcription activation.However,when investigating the H3K4me3 profiles in the mouse cerebrum and testis,we discovered that H3K4me3 also has a significant enrichment at the 3 0 end of actively transcribed(sense) genes,named as 3 0-H3K4me3.3 0-H3K4me3 is associated with 15% of protein-coding genes in both tissues.In addition,we examined the transcriptional initiation signals including RNA polymerase II(RNAPII) binding sites and 5 0-CAGE-tag that marks transcriptional start sites.Interestingly,we found that 3 0-H3K4me3 is associated with the initiation of antisense transcription.Furthermore,3 0-H3K4me3 modification levels correlate positively with the antisense expression levels of the associated sense genes,implying that 3 0-H3K4me3 is involved in the activation of antisense transcription.Taken together,our findings suggest that H3K4me3 may be involved in the regulation of antisense transcription that initiates from the 3 0 end of sense genes.In addition,a positive correlation was also observed between the expression of antisense and the associated sense genes with 3 0-H3K4me3 modification.More importantly,we observed the 3 0-H3K4me3 enrichment among genes in human,fruitfly and Arabidopsis,and found that the sequences of 3 0-H3K4me3-marked regions are highly conserved and essentially indistinguishable from known promoters in vertebrate.Therefore,we speculate that these 3 0-H3K4me3-marked regions may serve as potential promoters for antisense transcription and 3 0-H3K4me3 appear to be a universal epigenetic feature in eukaryotes.Our results provide a novel insight into the epigenetic roles of H3K4me3 and the regulatory mechanism of antisense transcription. | Peng Cui Wanfei Liu Yuhui Zhao Qiang Lin Feng Ding Chengqi Xin Jianing Geng Shuhui Song Fanglin Sun Songnian Hu Jun Yu | 2012 | Genomics, Proteomics & Bioinformatics2012,10,2: | 1 |

| 20 | Novel photoelectric material of perovskite-like(CH_(3))_(3)SPbI_(3) nanorod arrays with high stability显示文摘Organometallic halide perovskite materials make great achievements in optoelectronic fields,especially in solar cells,in which the organic cations contain amine components.However,the amine with NàH bonds is easily hydrolyzed with moisture in the air,weakening the perovskite materials stability.It is desirable to develop other non-amine stable perovskite materials.In this work,sulfur-based perovskite-like(CH_(3))_(3)SPbI_(3) nanorod arrays were fabricated by a solution-processed method,which can be indexed hexagonal crystal structure in the space group P63 mc.The binding force is exceptionally strong between the non-amine(CH_(3))_(3) S+and[PbI_(6)]_(4)-octahedral,leading to high stability of(CH_(3))_(3)SPbI_(3).The(CH_(3))_(3)SPbI_(3) nanorod arrays can keep the morphology and crystal structure in an ambient atmosphere over 60 days.In addition,the(CH_(3))_(3)SPbI_(3) nanorod arrays can offer direct charge transfer channels,which show excellent optoelectronic properties.The(CH_(3))_(3)SPbI_(3) nanorod arrays-based solar cells with VOx hole transfer layers achieved a power conversion efficiency of 2.07%with negligible hysteresis.And the(CH_(3))_(3)SPbI_(3) nanorod arrays were also effectively applied in photodetectors with interdigitated gold electrodes.This work demonstrates that sulfur-based perovskite-like(CH_(3))_(3)SPbI_(3) is a novel promising stable compound with great potential for practical optoelectronic applications. | Ruiyuan Hu Chuangye Ge Liang Chu Yifei Feng Shanshan Xiao Yuhui Ma Wei Liu Xing'ao Li Mohammad Khaja Nazeeruddin | 2021 | Journal of Energy Chemistry2021,30,8: | 0 |
