| 1 | Short-chain fatty acids act as antiinflammatory mediators by regulating prostaglandin E_2 and cytokines显示文摘AIM:To investigate the effect of short-chain fatty ac-ids (SCFAs) on production of prostaglandin E2 (PGE2),cytokines and chemokines in human monocytes.METHODS:Human neutrophils and monocytes were isolated from human whole blood by using 1-Step Polymorph and RosetteSep Human Monocyte Enrich-ment Cocktail,respectively. Human GPR41 and GPR43 mRNA expression was examined by quantitative real-time polymerase chain reaction. The calcium flux assay was used to examine the biological activities of SCFAs in human neutrophils and monocytes. The effect ofSCFAs on human monocytes and peripheral blood mononuclear cells (PBMC) was studied by measuring PGE2,cytokines and chemokines in the supernatant. The effect of SCFAs in vivo was examined by intraplantar injection into rat paws.RESULTS:Human GPR43 is highly expressed in human neutrophils and monocytes. SCFAs induce robust calcium flux in human neutrophils,but not in human monocytes. In this study,we show that SCFAs can in-duce human monocyte release of PGE2 and that this effect can be enhanced in the presence of lipopolysac-charide (LPS). In addition,we demonstrate that PGE2 production induced by SCFA was inhibited by pertussis toxin,suggesting the involvement of a receptor-mediated mechanism. Furthermore,SCFAs can specifi cally inhibit constitutive monocyte chemotactic protein-1 (MCP-1) production and LPS-induced interleukin-10 (IL-10) production in human monocytes without affecting the secretion of other cytokines and chemokines examined. Similar activities were observed in human PBMC for the release of PGE2,MCP-1 and IL-10 after SCFA treatment. In addition,SCFAs inhibit LPS-induced production of tumor necrosis factor-α and interferon-γ in human PBMC. Finally,we show that SCFAs and LPS can induce PGE2 production in vivo by intraplantar injection into rat paws (P < 0.01). | 2 | High-density SNP-based genetic maps for the parents of an outcrossed and a selfed tetraploid garden rose cross, inferred from admixed progeny using the 68k rose SNP array显示文摘Dense genetic maps create a base for QTL analysis of important traits and future implementation of marker-assisted breeding.In tetraploid rose,the existing linkage maps include<300 markers to cover 28 linkage groups(4 homologous sets of 7 chromosomes).Here we used the 68k WagRhSNP Axiom single-nucleotide polymorphism(SNP)array for rose,in combination with SNP dosage calling at the tetraploid level,to genotype offspring from the garden rose cultivar‘Red New Dawn’.The offspring proved to be not from a single bi-parental cross.In rose breeding,crosses with unintended parents occur regularly.We developed a strategy to separate progeny into putative populations,even while one of the parents was unknown,using principle component analysis on pairwise genetic distances based on sets of selected SNP markers that were homozygous,and therefore uninformative for one parent.One of the inferred populations was consistent with self-fertilization of‘Red New Dawn’.Subsequently,linkage maps were generated for a bi-parental and a self-pollinated population with‘Red New Dawn’as the common maternal parent.The densest map,for the selfed parent,had 1929 SNP markers on 25 linkage groups,covering 1765.5 cM at an average marker distance of 0.9 cM.Synteny with the strawberry(Fragaria vesca)genome was extensive.Rose ICM1 corresponded to F.vesca pseudochromosome 7(Fv7),ICM4 to Fv4,ICM5 to Fv3,ICM6 to Fv2 and ICM7 to Fv5.Rose ICM2 corresponded to parts of F.vesca pseudochromosomes 1 and 6,whereas ICM3 is syntenic to the remainder of Fv6. | Mirjana Vukosavljev Paul Arens Roeland E Voorrips Wendy P C van't Westende G D Esselink Peter M Bourke Peter Cox W Eric van de Weg Richard G F Visser Chris Maliepaard Marinus J M Smulders | 2016 | Horticulture Research2016,3,1: | 6 |

| 12 | Supplementation of oligosaccharide-based polymer enhanced growth and disease resistance of weaned pigs by modulating intestinal integrity and systemic immunity显示文摘Background:There is a great demand for antibiotic alternatives to maintain animal health and productivity.The objective of this experiment was to determine the efficacy of dietary supplementation of a blood group A6 type 1antigen oligosaccharides-based polymer(Coligo)on growth performance,diarrhea severity,intestinal health,and systemic immunity of weaned pigs experimentally infected with an enterotoxigenic Escherichia coli(ETEC),when compared with antibiotics.Results:Pigs in antibiotic carbadox or Coligo treatment groups had greater(P<0.05)body weight on d 5 or d 11post-inoculation(PI)than pigs in the control group,respectively.Supplementation of antibiotics or Coligo enhanced(P<0.05)feed efficiency from d 0 to 5 PI and reduced(P<0.05)frequency of diarrhea throughout the experiment,compared with pigs in the control group.Supplementation of antibiotics reduced(P<0.05)fecalβ-hemolytic coliforms on d 2,5,and 8 PI.Pigs in antibiotics or Coligo groups had reduced(P<0.05)neutrophil counts and serum haptoglobin concentration compared to pigs in the control group on d 2 and 5 PI.Pigs in Coligo had reduced(P<0.05)total coliforms in mesenteric lymph nodes on d 5 and 11 PI,whereas pigs in antibiotics or Coligo groups had reduced(P<0.05)total coliforms in spleen on d 11 PI compared with pigs in the control group.On d 5 PI,pigs in the Coligo group had greater(P<0.05)gene expression of ZO1 in jejunal mucosa,but less(P<0.05)m RNA expression of IL1B,IL6,and TNF in ileal mucosa,in comparison with pigs in the control group.Supplementation of antibiotics enhanced(P<0.05)the gene expression of OCLN in jejunal mucosa but decreased(P<0.05)IL1B and IL6 gene expression in ileal mucosa,compared with the control.On d 11 PI,supplementation of antibiotics or Coligo up-regulated(P<0.05)gene expression of CLDN1 in jejunal mucosa,but Coligo reduced(P<0.05)IL6 gene expression in ileal mucosa compared to pigs in the control group.Conclusions:Supplementation of Coligo improved growth performance,alleviated diarrhea severity,and enhanced gut health in weaned pigs infected with ETEC F18 in a manner similar to in-feed antibiotics. | Kwangwook Kim Yijie He Cynthia Jinno Lauren Kovanda Xunde Li David Bravo Eric Cox Yanhong Liu | 2022 | Journal of Animal Science and Biotechnology2022,13,3: | 0 |
| 13 | Impact of an oligosaccharide-based polymer on the metabolic profiles and microbial ecology of weanling pigs experimentally infected with a pathogenic E.coli显示文摘Background Our previous study has reported that supplementation of oligosaccharide-based polymer enhances gut health and disease resistance of pigs infected with enterotoxigenic E.coli(ETEC)F18 in a manner similar to carbadox.The objective of this study was to investigate the impacts of oligosaccharide-based polymer or antibiotic on the host metabolic profiles and colon microbiota of weaned pigs experimentally infected with ETEC F18.Results Multivariate analysis highlighted the differences in the metabolic profiles of serum and colon digesta which were predominantly found between pigs supplemented with oligosaccharide-based polymer and antibiotic.The relative abundance of metabolic markers of immune responses and nutrient metabolisms,such as amino acids and carbohydrates,were significantly differentiated between the oligosaccharide-based polymer and antibiotic groups(q<0.2 and fold change>2.0).In addition,pigs in antibiotic had a reduced(P<0.05)relative abundance of Lachnospiraceae and Lactobacillaceae,whereas had greater(P<0.05)Clostridiaceae and Streptococcaceae in the colon digesta on d 11 post-inoculation(PI)compared with d 5 PI.Conclusions The impact of oligosaccharide-based polymer on the metabolic and microbial profiles of pigs is not fully understood,and further exploration is needed.However,current research suggest that various mechanisms are involved in the enhanced disease resistance and performance in ETEC-challenged pigs by supplementing this polymer. | Kwangwook Kim Cynthia Jinno Xunde Li David Bravo Eric Cox Peng Ji Yanhong Liu | 2024 | Journal of Animal Science and Biotechnology2024,15,2: | 0 |
