1Concurrent infections of dengue viruses serotype 2 and 3 in patient with severe dengue from Jakarta, Indonesia显示文摘Objective: To describe the clinical manifestation of patient with severe dengue, to identify the serotypes and genotypes of dengue viruses(DENV) which concurrently infecting the patient, and to explore the possible relationship of severe dengue with the concurrent infection of DENV. Methods: Dengue diagnosis was performed using NS1 antigen detection and Ig G/Ig M ELISA. Standard clinical and laboratory examinations were performed to obtain the clinical and hematological data. DENV concurrent infections were detected and confirmed using RT-PCR and DENV Envelope gene sequencing. Phylogenetic analyses were performed to determine the genotypes of the viruses. Results: The patient was classified as having severe dengue characterized by severe plasma leakage, hemorrhage, and organ damage involving lung, liver, and kidney. Concurrent infection of DENV serotype 2 and 3 was observed. The infecting DENV-2 virus was grouped into Cosmopolitan genotype while DENV-3 virus was classified into Genotype Ⅰ. Both viruses were closely related to isolates that were endemic in Jakarta. Viremia measurement was conducted and revealed a significantly higher virus titer of DENV-3 compared to DENV-2. Conclusions: The occurrence of multi-serotype DENV infections was presented in a patient with severe clinical manifestation in Indonesia. 2Anti-diabetic potential of Urena lobata leaf extract through inhibition of dipeptidyl peptidase IV activity显示文摘Objective: To evaluate the anti-diabetic potential of leaf extract from Urena lobata(U. lobata) through dipeptidyl peptidase IV(DPP-IV) inhibitory activity.Methods: U. lobata leaf was extracted in hot water and ethanol. The activity of DPPIV inhibitor was tested by in vitro study using gly-pro-p-nitroanilide as substrat of DPPIV and vildagliptin, as standard reference. A product of the reactions between gly-pro-pnitroanilide and DPP-IV, was observed by microplate readers with λ = 405 nm. All data were expressed as mean ± SD and the IC50 value was determined by non linear regression curve fit. Active substances in leaf extract of U. lobata was analyzed by liquid chromatography-mass spectrometry. DPP-IV inhibitory activity of active compounds was evaluated in silico using docking server. Results: The ethanolic extract of U. lobata showed stronger DPP-IV inhibitor activity than water extract with the IC50 values of 1 654.64 and 6 489.88 μg/mL, respectively. Vildagliptin, based on standard reference for DPP-IV inhibitor activity, has IC50 value of 57.44 μg/mL. Based on in silico analysis, mangiferin, stigmasterol and β-sitosterol in U. lobata extract have a strong inhibitory activity on DPP-IV. 9The effectiveness of dalethyne dressings for reducing bacteria in diabetic foot ulcers显示文摘Objective:This study evaluates the effectiveness of a dalethyne dressing for decreasing bacteria in diabetic patients with infected foot ulcers.Methods:This study was conducted from March to September 2018 with a sample of 30 participants from the outpatient Kitamura Wound Clinic in Pontianak City,Indonesia.A quasi-experimental non-equivalent pretest-posttest control group design was used for the study.Participants were divided into two groups:an intervention group(treated with a dalethyne dressing)and a control group(treated with a standard dressing).Two trained research assistants collected the data using the Wagner wound classification system and a bacteria counter.The assistants swabbed each wound surface with sterile cotton,and the swabs were used to conduct a bacteria culture and count.Results:The study population was 50%female and 50%male with no significant differences between each other in age,HbA1c,blood pressure,or ankle-brachial index(ABI;P>0.05).Both groups had a significant reduction in the number of bacteria from the pretest to posttest(P<0.05).Mann-Whitney analysis of posttest data indicated a significant difference in bacteria reduction between the control group(median=2.25)and the intervention group(median=7.6;P=0.018).It was noted that Staphylococcus aureus was found in the control group at posttest,but not in the intervention group.Conclusions:This study provides evidence that a dalethyne dressing is effective for killing S.aureus in the infected foot ulcers of diabetic patients.Suriadi Jais Djoko Priyono 2021Frontiers of Nursing2021,8,2:1
