1MicroRNA-23a promotes colorectal cancer cell migration and proliferation by targeting at MARK1显示文摘The functional role of microRNA-23a in tumorigenesis has been investigated;however, the exact mechanism of microRNA-23a (miR-23a) in colorectal cancer development has not been fully explored. In the present study, we aimed to investigate the molecular functional role of miR-23a in colorectal carcinogenesis. Quantitative real-time polymerase chain reaction was conducted to investigate the expression level of miR-23a in tissue samples and cell lines (HCT116 and SW480). CCK-8, colony formation and Transwell assay were used to explore the role of miR-23a in cell proliferation and migration. Dual luciferase reporter assay was used to identify the direct binding of miR-23a with its target, MARK1. Western blot analysis was used to analyze the expression level of MARK1, as well as a confirmed miR-23a target gene, MTSS1, in miR-23a-mimic and miR-23a-inhibit groups. Rescue experiments were conducted by overexpression of MARK1 in miR-23a-mimic-transfected cell lines. The results showed that miR-23a was highly expressed in colorectal cancer tissue and cell lines. MiR-23a could promote proliferation and migration of colorectal cancer cell lines. MARK1 was a direct target of miR-23a and the expression level of MARK1 was down-regulated in miR-23a-mimic-transfected cell lines but up-regulated in miR-23a-inhibit-transfected cells. Overexpression of MARK1 could partly reverse the cancer-promoting function of miR-23a. Our results suggested that miR-23a promotes colorectal cancer cell proliferation and migration by mediating the expression of MARK1. 2Retardant effect of dihydroartemisinin on ulcerative colitis in a JAK2/STAT3-dependent manner显示文摘Dihydroartemisinin(DHA)is a semi-synthetic derivative and the main active metabolite of artemisinin.The purpose of this study was to investigate the effect of DHA on the ulcerative colitis(UC)in both in vivo and in vitro models.Weight,survival rate,colon length,and Disease Activity Index score were used to evaluate the severity of colitis.Reverse transcription quantitative polymerase chain reaction and enzyme-linked immunosorbent assay were used to detect the expressions of cytokines interleukin(IL)-1,IL-1β,IL-4,IL-6,IL-10,IL-12,and tumor necrosis factor-α(TNF-α).The expressions of Janus kinase 2(JAK2)and signal transducer and activator of transcription 3(STAT3),and the phosphorylation of JAK2(p-JAK2)and STAT3(p-STAT3),were measured by western blot analysis.Western blot analysis and immunohistochemistry were used to detect the expressions of tight junction proteins.We found that the weights and colon lengths of mice in dextran sodium sulfate(DSS)+DHA group were significantly lower and longer than those in the DSS group,respectively.Compared with those in the DSS group,the expressions of IL-1β,IL-6,IL-17,and TNF-αin the DSS+DHA and DSS+5-aminosalicylic acid(5-ASA)groups were decreased,while the expressions of IL-4 and IL-10 were significantly upregulated.DHA largely increased the expressions of zonula occludens-1 and occludin.Western blot analysis and/or immunohistochemical staining analysis showed that the expressions of JAK2,STAT3,p-JAK2,and p-STAT3 in DSS+DHA and DSS+5-ASA groups were significantly lower than those in DSS group.DHA has a specific therapeutic effect on UC.The anti-inflammatory mechanism of DHA is related to the blockage of the JAK2/STAT3 signaling pathway.These findings provide evidence that DHA may be a useful drug and is expected to become a promising new treatment for human UC.Mingrui Jiang Guangjun Zhong Yichao Zhu Liuhua Wang Yuzhe He Qiannan Sun Xiaoqing Wu Xiaolan You Sujun Gao Dong Tang Daorong Wang 2021Acta Biochimica et Biophysica Sinica2021,53,9:6
3Syndecan-2, negatively regulated by miR-20b-5p, contributes to 5-fluorouracil resistance of colorectal cancer cells via the JNK/ERK signaling pathway显示文摘5-Fluorouracil (5-FU) resistance has been long considered as an obstacle to the efficacy of chemotherapy in colorectal cancer (CRC). In this study, we demonstrated the role of miR-20b-5p-regulated syndecan-2 (SDC2) in 5-FU resistance of CRC cells. 5-FU-resistant SW480 CRC cells were established by treatment of SW480 cells with stepwise increase of 5-FU concentration. The results showed that SDC2 was expressed significantly higher in SW480/5-FU cells than in SW480/WT cells as revealed by quantitative real-time polymerase chain reaction and western blot analysis. MTT assay and BrdU assay showed that SDC2 overexpression led to increased cell survival rate, while SDC2 knockdown reversed the drug resistance of SW480/5-FU cells. Wound healing and transwell invasion assays revealed that knockdown of SDC2 inhibited the migratory and invasive ability of SW480/5-FU cells. Moreover, animal experiments indicated that si-SDC2 plays a suppressive role in tumor growth in vivo. We also confirmed that miR-20b-5p interacted with SDC2, which reversed the effect of SDC2 in SW480/5-FU cells via the c-Jun N-terminal kinase (JNK)/extracellular regulated protein kinases (ERK) signaling pathway. 4Combination analysis of hypermethylated SFRP1 and SFRP2 gene in fecal as a novel epigenetic biomarker panel for colorectal cancer screening显示文摘Objective:To investigate the feasibility of the combination of detecting hypermethylated secreted frizzled-related protein 1(SFRP1)and secreted frizzled-related protein 2(SFRP2)in feces as a panel of biomarkers for colorectal cancer(CRC)screening.Methods:Methylation-specific PCR(MSP)was performed to analyze methylation status of SFRP1 and SFRP2 in a blinded fashion in tumor tissues and in matched stool samples from 39 patients with primary CRC,34 patients with adenomas,17 patients with hyperplastic polyps and 20 endoscopically normal subjects as normal controls.Simultaneously we analyzed the correlation of hypermethylated SFRP1 and SFRP2 with the clinicopathological features of CRC.Results:Hypermethylated SFRP1 was detected in 92.3%,76.5%,47.1% of tissue samples and in 89.7%,64.7%,35.3% of matched fecal samples from CRC,adenoma and hyperplastic polyp,respectively.Hypermethylated SFRP2 was detected in 87.2%,67.6%,35.3% of tissue samples and in 82.1%,55.9%,29.4% of matched fecal samples from CRC,adenoma and hyperplastic polyp,respectively.Of these two genes,at least one hypermethylated was 94.9%,82.4%,52.9% in tissue samples and 92.3%,73.5%,47.1% in matched fecal samples from CRC,adenoma and hyperplastic polyp,respectively.In contrast,no hypermethylated SFRP1 and SFRP2 were detected in mucosa tissues of normal controls,only 2 cases of fecal samples was detected with hypermethylated SFRP1 and another 1 case was detected with hypermethylated SFRP2.Moreover,no significant associations were observed between hypermethylated SFRP1,SFRP2 and clinicopathological features of CRC.Conclusion:Hypermethylation of SFRP1 and SFRP2 in feces are novel epigenetic biomarkers of CRC and carried high potential for the remote detection of CRC as non-invasive screening method,and combined analysis of hypermethylated SFRP1 and SFRP2 in fecal could further increase the detection rate of CRC and premalignant lesions.Dong Tang Daorong Wang Hongbo Li 2008Journal of Nanjing Medical University2008,22,2:4
5Inheritance and expression of multiple disease and insect resistance genes in transgenic rice显示文摘2-3 anti-fungal disease genes are coinserted with hygromycin phosphotransferase in the same vector. Two insecticidal genes and PPT acetyl transferase genes are placed in another one. The vectors are co-delivered to rice embryonic cellus tissue at a molar ratio of 1:1 using the particle gun method. 55 independent regenerated lines have been obtained through screening for hygromycin resistance. Of these, 70% transgenic plants harbor 6-7 foreign genes. The genes on the same vectors are always co-delivered to rice plant. Northern blot analysis has indicated that the multiple foreign genes give stable expression. In the 6 transgenic plants carrying 6-7 foreign genes, multiple foreign genes tend to integrate in 1 or 2 genetic loci. 6STAM2 knockdown inhibits proliferation,migration,and invasion by affecting the JAK2/STAT3 signaling pathway in gastric cancer显示文摘Signal transducing adaptor molecule 2(STAM2)is a phosphotyrosine protein,which regulates receptor signaling and trafficking of mammalian cells.However,its role in gastric cancer(GC)remains undiscovered.In this study,we aimed to investigate the functions of STAM2 in GC.The mRNA and protein expression levels of STAM2 were measured by quantitative real-time PCR,western blot analysis,and immunohistochemistry.STAM2 was stably silenced in AGS and HGC-27 cells using small interfering RNA.The function of STAM2 in GC cells was further investigated by CCK-8 assay,EdU incorporation assay,flow cytometry,and scratch wound healing and Boyden chamber assays.Additionally,we conducted biological pathway enrichment analysis and rescue assays to explore the effects of STAM2 on JAK/STAT signaling pathway.Our results showed that STAM2 is remarkably highly expressed in GC tissues and cells,and overexpressed STAM2 is correlated with tumor size,advanced tumor node metastasis stage,and poor prognosis.In addition,STAM2 knockdown could significantly inhibit proliferation,block cell cycle,and restrain migration and invasion capabilities of GC cells.Mechanistically,we found that STAM2 knockdown effectively decreased the expressions of MMP2 and MMP9 and the phosphorylation levels of JAK2 and STAT3.Taken together,this study revealed that STAM2 knockdown could suppress malignant process by targeting the JAK2/STAT3 signaling pathway in GC.Yang Yang Qi Zhang Jiakui Liang Meiyuan Yang Zheng Wang Dong Tang Daorong Wang 2021Acta Biochimica et Biophysica Sinica2021,53,6:2

9The study on quantitative expression of CD44v6mRNA by real-time RT-PCR with the micro-metastases of gastric cancer显示文摘Objective: To study the expression of CD44 correlation and the ability of metastasis of tumor cells in gastric carcinoma, and to find the correlation of the quantitative of CD44V6mRNA and the histology expression of CD44v6 in tumors with the clinic-pathologic features, and to make the quantitative expression of CD44v6mRNA. Methods: Twenty patients with gastric carcinoma, 4 patients with gastritis, and 10 apparently healthy controls were recruited. Blood samples were obtained before surgery. 10 days after surgery, the blood samples were obtained again. Serum CD44v6mRNA in all cases was measured by real-time quantitative PCR. Results: Serum CD44V6mRNA was detectable in 20 of 20(100%) gastric carcinoma cases, The expression level ranged from 4.9×102 copies/μg RNA to 3.2×105 copies/μg RNA, the average levels of peripheral blood was 3.9×104 copies/μg RNA, The expression level of peripheral blood of gastric cancer after curative operation ranged from 5.5×100 copies/μg RNA to 7.6×103 copies/μg RNA. After curative operation the expression level was decreased markedly. Conclusion: Serum CD44v6mRNA is expressed in the peripheral blood of gastric carcinoma patients. The expression level of CD44V6mRNA is obviously decreased after curative operation. 19Enhanced photocatalytic ammonia oxidation activity and nitrogen selectivity over Ag/AgCl/N-TiO_(2) photocatalyst显示文摘The removal of ammonia(NH_(3))emitted from agricultural and industrial activities is of great significance to protect human health and ecological environment.Photocatalytic NH_(3)oxidation to N2under mild conditions is a promising strategy.However,developing visible light photocatalysts for NH_(3)oxidation is still in its infancy.Here,we fabricate N-TiO_(2)and Ag/AgCl/N-TiO_(2)photocatalysts by sol-gel and photodeposition methods,respectively.The introduction of N not only endows TiO_(2)with visible light response(absorption edge at460 nm)but also results in the formation of heterophase junction(anatase and rutile).Thus,N-TiO_(2)shows 2.0 and 1.8 times higher than those over anatase TiO_(2)and commercial TiO_(2)for NH_(3)oxidation under full spectrum irradiation.Meanwhile,surface modification of Ag can simultaneously enhance visible light absorption(generating localized surface plasmon resonance effect)and charge separation efficiency.Therefore,the photocatalytic activity of Ag/AgCl/N-TiO_(2)is further improved.Furthermore,the presence of N and Ag also enhances the selectivity of N2product owing to the change of reaction pathway.This work simultaneously regulates photocatalytic conversion efficiency and product selectivity,providing some guidance for developing highly efficient photocatalysts for NH_(3)elimination.Jiaojiao Guan Daorong Li Jiahui Feng Peng Xu Zhaonian Li Siqi Ge Hongxia Chen Kunfeng Zhang 2024Journal of Environmental Sciences2024,,4:0
20Expression and Clinical Significance of CD44vin Hum an Gastric Cancers显示文摘Expression of CD44v mRNA in 20 patients with gastric tumor was investigated to identify the correlation of the CD44 gene splice and abnormal expression with the clinicopathological changes. The results showed that CD44v mRNA was expressed in 16 of 20 gastric cancers. In 5 out of the 10 mucosa tissues adjacent to gastric cancer, However, no expression of CD44v mRNA was found in all 10 normal mucosa specimens; Positive expression was found in 15 of 16 patients with lymph node metastasis, and 1 out of 4 patients without lymph node metastasis, as well as 5 cases with distant metastasis (P<0.05). Positive expression of CD44v was found in 2 out of 5 well differentiated type gastric cancers, and 14 out of 15 poorly differentiated gastric cancers. Differences were found in the expression between the two types of gastric mucosa (P<0.05). All the mucosa samples including normal gastric mucosa, gastric cancer and its adjacent tissues presented the standard CD44v mRNA.
