| 1 | Passive-source ocean bottom seismograph(OBS) array experiment in South China Sea and data quality analyses显示文摘Long-term passive source ocean bottom seismograph(OBS) observatory is challenging due to various technical difficulties. In order to gain experience in this field, and to reveal the lithospheric structure beneath the extinct ridge in the central South China Sea(SCS), we carried out a passive source OBS array experiment, which includes 18 OBSs, in the deep portion of SCS. Here we present the instrumentation, the OBS deployment and recovery of this experiment, and more importantly, the data quality evaluated by a number of approaches. Through processing and inspecting waveforms from global, regional and local earthquakes, we find that most of recovered OBSs have good data quality with discernible main phases. The ambient noise analyses of OBS recordings show that their noise is higher than the global average, and the horizontal component is noisier than the vertical, indicating current impacts on horizontal components are more severe. In the period range of 5–10 s, there is a noise notch for the SCS OBSs, and noise levels of horizontal components are comparable to the vertical. This feature, which is not seen at OBS stations in open ocean, suggests the distant sources for double frequency microseism in this marginal sea are not significant. In addition, we successfully determined the orientations for 7 OBSs by investigating their Rayleigh wave polarizations; and we demonstrated the dispersion feature of Rayleigh waves through the frequency-time analysis. | 2 | Key nodes of a micro RNA network associated with the integrated mesenchymal subtype of high-grade serous ovarian cancer显示文摘Metastasis is the main cause of cancer mortality. One of the initiating events of cancer metastasis of epithelial tumors is epithelial-to-mesenchymal transition(EMT), during which cells dedifferentiate from a relatively rigid cell structure/morphology to a flexible and changeable structure/morphology often associated with mesenchymal cells. The presence of EMT in human epithelial tumors is reflected by the increased expression of genes and levels of proteins that are preferentially present in mesenchymal cells. The combined presence of these genes forms the basis of mesenchymal gene signatures, which are the foundation for classifying a mesenchymal subtype of tumors. Indeed, tumor classification schemes that use clustering analysis of large genomic characterizations, like The Cancer Genome Atlas(TCGA), have defined mesenchymal subtype in a number of cancer types, such as high-grade serous ovarian cancer and glioblastoma. However, recent analyses have shown that gene expression-based classifications of mesenchymal subtypes often do not associate with poor survival. This 'paradox' can be ameliorated using integrated analysis that combines multiple data types. We recently found that integrating m RNA and micro RNA(mi RNA) data revealed an integrated mesenchymal subtype that is consistently associated with poor survival in multiple cohorts of patients with serous ovarian cancer. This network consists of 8 major mi RNAs and 214 m RNAs. Among the 8 mi RNAs, 4 are known to be regulators of EMT. | 3 | Prognostic factors and therapeutic effects of different treatment modalities for colorectal cancer liver metastases显示文摘BACKGROUND Colorectal cancer(CRC)is one of the most common malignant tumors in China,and the liver is the most common metastatic site in patients with advanced CRC.Hepatectomy is the gold standard treatment for colorectal liver metastases.For patients who cannot undergo radical resection of liver metastases for various reasons,ablation therapy,interventional therapy,and systemic chemotherapy can be used to improve their quality of life and prolong their survival time.AIM To explore the prognostic factors and treatments of liver metastases of CRC.METHODS A retrospective analysis was conducted on 87 patients with liver metastases from CRC treated at the Liaoning Cancer Hospital and Institute between January 2005 and March 2011.According to different treatments,the patients were divided into the following four groups:Surgical resection group(36 patients);ablation group(23 patients);intervention group(15 patients);and drug group(13 patients).The clinicopathological data and postoperative survival of the four groups were analyzed.The Kaplan-Meier method was used for survival analysis,and the Cox proportional hazards regression model was used for multivariate analysis.RESULTS The median survival time of the 87 patients was 38.747±3.062 mo,and the 1-and 3-year survival rates were 87.5%and 53.1%,respectively.The Cox proportional hazards model showed that the following factors were independent factors affecting prognosis:The degree of tumor differentiation,the number of metastases,the size of metastases,and whether the metastases are close to great vessels.The results of treatment factor analysis showed that the effect of surgical treatment was better than that of drugs,intervention,or ablation alone,and the median survival time was 48.83±4.36 mo.The drug group had the worst prognosis,with a median survival time of only 13.5±0.7 mo(P<0.05).For patients with liver metastases of CRC near the great vessels,the median survival time(27.3 mo)of patients undergoing surgical resection was better than that of patients using other treatments(20.6 mo)(P<0.05).CONCLUSION Patients with a low degree of primary tumor differentiation,multiple liver metastases(number of tumors>4),and maximum diameter of liver metastases>5 cm have a poor prognosis.Among drug therapy,intervention,ablation,and surgical treatment options,surgical treatment is the first choice for liver metastases.When liver metastases are close to great vessels,surgical treatment is significantly better than drug therapy,intervention,and ablation alone. | Zuo-Hong Ma Yong-Peng Wang Wen-Heng Zheng Ji Ma Xue Bai Yong Zhang Yuan-He Wang Da Chi Xi-Bo Fu Xiang-Dong Hua | 2020 | World Journal of Gastrointestinal Oncology2020,12,10: | 7 |
| 4 | Trimethylamine N-oxide attenuates high-fat high-cholesterol dietinduced steatohepatitis by reducing hepatic cholesterol overload in rats显示文摘BACKGROUND Trimethylamine N-oxide (TMAO) has been shown to be involved in cardiovascular disease (CVD). However, its role in nonalcoholic steatohepatitis (NASH) is unknown. AIM To determine the effect of TMAO on the progression of NASH. METHODS A rat model was induced by 16-wk high-fat high-cholesterol (HFHC) diet feeding and TMAO was administrated by daily oral gavage for 8 wk. RESULTS Oral TMAO intervention attenuated HFHC diet-induced steatohepatitis in rats. Histological evaluation showed that TMAO treatment significantly alleviated lobular inflammation and hepatocyte ballooning in the livers of rats fed a HFHC diet. Serum levels of alanine aminotransferase and aspartate aminotransferase were also decreased by TMAO treatment. Moreover, hepatic endoplasmic reticulum (ER) stress and cell death were mitigated in HFHC diet-fed TMAOtreated rats. Hepatic and serum levels of cholesterol were both decreased by TMAO treatment in rats fed a HFHC diet. Furthermore, the expression levels of intestinal cholesterol transporters were detected. Interestingly, cholesterol influxrelated Niemann-Pick C1-like 1 was downregulated and cholesterol efflux-related ABCG5/8 were upregulated by TMAO treatment in the small intestine. Gut microbiota analysis showed that TMAO could alter the gut microbial profile and restore the diversity of gut flora. CONCLUSION These data suggest that TMAO may modulate the gut microbiota, inhibit intestinal cholesterol absorption, and ameliorate hepatic ER stress and cell death under cholesterol overload, thereby attenuating HFHC diet-induced steatohepatitis in rats. | 5 | CDH13 Genetic Polymorphisms, Adiponectin and Ischemic Stroke: a Chinese Family-based Sib-pair Study显示文摘Objective To understand the relationships between CDH13(T-cadherin) genetic polymorphisms, adiponectin levels and ischemic stroke, and possible interactions between CDH13 polymorphisms and other risk factors. Methods We recruited 342 Chinese ischemic stroke sib pairs. We genotyped rs4783244 and rs7193788 on CDH13 using time-of-flight mass spectrometry genotyping technology and measured total and high-molecular weight(HMW) adiponectin levels. We investigated associations between SNPs and ischemic stroke, and interactions between SNPs and other risk factors using multi-level mixed-effects regression model. Results In individuals without ischemic stroke, CDH13 rs4783244 was associated with total adiponectin levels(per T: Coef =-0.257, P = 0.001). CDH13 rs7193788 was associated with total adiponectin levels(per A: Coef =-0.221, P = 0.001) and HMW adiponectin levels(per A: Coef =-0.163, P = 0.003). rs7193788 was significantly associated with ischemic stroke(GA/AA vs. GG: OR = 1.55, 95% CI: 1.07 to 2.24, P = 0.020) after Bonferroni correction(α = 0.025). There was an interaction between rs7193788 and diabetes(P = 0.036). Compared to diabetes-free individuals with rs7193788 GG genotype, diabetes patients with rs7193788 GA/AA genotypes had higher risks for ischemic stroke(OR = 2.64, 95% CI: 1.58-4.40, P < 0.001). Conclusion CDH13 genetic polymorphisms are associated with adiponectin levels and ischemic stroke. | 7 | Insulin-like growth factor 1-induced enolase 2 deacetylation by HDAC3 promotes metastasis of pancreatic cancer显示文摘Enolase 2(ENO2)is a key glycolytic enzyme in the metabolic process of glycolysis,but its potential function in pancreatic ductal adenocarcinoma(PDAC)is unclear.In this study,we observed a significant overexpression of ENO2 in PDAC tissues,and its expression was correlated with metastasis and poor prognosis in PDAC patients.K394 was identified as a major acetylation site in ENO2 that regulates its enzymatic activity,cell metabolism and PDAC progression.Knockdown of ENO2 suppressed tumor growth and liver metastasis in PDAC.Re-expression of wild-type(WT)ENO2,but not the K394 acetylation mimetic mutant,could reverse the decreased tumor malignancy.We further characterized histone deacetylase 3(HDAC3)and P300/CBP-associated factor(PCAF)as the potential deacetylase and acetyltransferase for ENO2,respectively.HDAC3-mediated deacetylation was shown to lead to ENO2 activation and enhancement of glycolysis.Importantly,insulin-like growth factor-1(IGF-1)was found to decrease K394 acetylation and stimulate ENO2 activity in a dose-and time-dependent manner.The PI3K/AKT/mTOR pathway facilitated the phosphorylation of HDAC3 on S424,which promoted K394 deacetylation and activation of ENO2.Linsitinib,an oral small-molecule inhibitor of IGF-1R,could inhibit IGF-1-induced ENO2 deacetylation by HDAC3 and the PI3K/AKT/mTOR pathway.Furthermore,linsitinib showed a different effect on the growth and metastasis of PDAC depending on the overexpression of WT versus K394-mutant ENO2.Our results reveal a novel mechanism by which acetylation negatively regulates ENO2 activity in the metastasis of PDAC by modulating glycolysis.Blockade of IGF-1-induced ENO2 deacetylation represents a promising strategy to prevent the development of PDAC. | Yan Zheng Chao Wu Jimeng Yang Yue Zhao Huliang Jia Min Xue Da Xu Feng Yang Deliang Fu Chaoqun Wang Beiyuan Hu Ze Zhang Tianen Li Shican Yan Xuan Wang Peter J.Nelson Christiane Bruns Lunxiu Qin Qiongzhu Dong | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3 |
| 8 | Novel Kriging-Based Decomposed-Coordinated Approach for Estimating the Clearance Reliability of Assembled Structures显示文摘Turbine blisks are assembled using blades,disks and casings.They can endure complex loads at a high temperature,high pressure and high speed.The safe operation of assembled structures depends on the reliability of each component.Monte Carlo(MC)simulation is commonly used to analyze structural reliability,but this method needs to run thousands of computations.In order to assess the clearance reliability of assembled structures in an efficient and precise manner,the novel Kriging-based decomposed-coordinated(DC)(DCNK)approach is proposed by integrating the DC strategy,the Kriging model and the importance sampling-based Markov chain(MCIS)technique.In this method,the DC strategy is used to decompose a multi-objective problem into many single-objective problems.The relationships between these many single-objectives and the overall objective are then coordinated.The Kriging model is applied to establish the limit state functions of the single-objectives and multi-objective problems,while the MCIS method is used to assess the structural assembled clearance reliability.Moreover,a highly nonlinear complex compound function is first utilized to verify the DCNK model from a mathematical perspective.Then,the reliability of an aeroengine high-pressure turbine(HPT)blade-tip radial running clearance(BTRRC)is analyzed to validate the DCNK approach by considering thermo-structural interaction.The analytical results show that the reliability is 0.9976 when the allowance value of the BTTRC is 1.7650×10^(−3)m.Compared with different methodologies(including direct simulation,the classical Kriging model,and the weighted response surface method(WRSM)),the proposed method holds obvious advantages in computing time and precision,as well as simulation efficiency and precision.The efforts of this paper provide a useful approach to analyzing assembled clearance reliability and contribute to the development of structural reliability theory. | Da Teng Yunwen Feng Cheng Lu Chengwei Fei Jiaqi Liu Xiaofeng Xue | 2021 | Computer Modeling in Engineering & Sciences2021,,11: | 2 |

| 11 | Effect of six-stitch pancreaticojejunostomy on pancreatic fistula:A propensity score-matched comparative cohort study显示文摘Background:Clinically relevant postoperative pancreatic fistula(CR-POPF)is the most common and severe complication after pancreaticoduodenectomy(PD).Despite the development of numerous anastomotic surgical techniques to minimize CR-POPF,more than 30%of patients who undergo PD develop CR-POPF.Herein,we propose a novel pancreaticojejunostomy(PJ)technique and evaluate its efficacy and safety compared to traditional PJ.Methods:This retrospective study enrolled 164 consecutive patients who underwent PJ after PD between January 2012 and June 2017.Of them,78(47.6%)underwent traditional PJ and 86(52.4%)underwent sixstitch PJ.The primary outcome was CR-POPF at 1-month follow-up defined according to the revised 2016 International Study Group on Pancreatic Fistula definition.To adjust for baseline differences and selection bias,patients were matched by propensity scores,which left 63 patients with traditional PJ and 63 with six-stitch PJ.Results:Compared to patients who underwent traditional PJ(mean age 56.2±9.4 years),patients who underwent six-stitch PJ(mean age 57.4±11.4 years)had a lower CR-POPF rate.The risk of CR-POPF among patients who underwent six-stitch PJ was decreased by 81.7%after adjustment for age,sex,body mass index,and disease severity compared to patients who underwent traditional PJ.Additionally,the surgery time was reduced from 29 min for traditional PJ to 15 min for six-stitch PJ(P<0.001).Adverse effects such as abdominal fluid collection,abdominal bleeding,and wound infection were similar between two groups.Conclusion:Six-stitch PJ may be an effective and efficient PJ technique for patients who undergo PD surgery. | Zhao-Lin Zeng Yan Sun Da Xue Pi-Li Liu Wang-Ming Chen Lei Zhang | 2020 | Hepatobiliary & Pancreatic Diseases International2020,19,3: | 2 |

| 13 | Intranasal Immunization Using CTA1-DD as a Mucosal Adjuvant for an Inactivated Influenza Vaccine显示文摘Objective To evaluate the effect of intranasal immunization with CTA1-DD as mucosal adjuvant combined with H3N2 split vaccine. Methods Mice were immunized intranasally with PBS(negative control), or H3N2 split vaccine(3 μg/mouse) alone, or CTA1-DD(5 μg/mouse) alone, or H3N2 split vaccine(3 μg/mouse) plus CTA1-DD(5 μg/mouse). Positive control mice were immunized intramuscularly with H3N2 split vaccine(3 μg/mouse) and alum adjuvant. All the mice were immunized twice, two weeks apart. Then sera and mucosal lavages were collected. The specific HI titers, IgM, IgG, IgA, and IgG subtypes were examined by ELISA. IFN-γ and IL-4 were test by ELISpot. In addition, two weeks after the last immunization, surivival after H3N2 virus lethal challenge was measured. Results H3N2 split vaccine formulated with CTA1-DD could elicit higher Ig M, Ig G and hemagglutination inhibition titers in sera. Furthermore, using CTA1-DD as adjuvant significantly improved mucosal secretory Ig A titers in bronchoalveolar lavages and vaginal lavages. Meanwhile this mucosal adjuvant could enhance Th-1-type responses and induce protective hemagglutination inhibition titers. Notably, the addition of CTA1-DD to split vaccine provided 100% protection against lethal infection by the H3N2 virus. | 20 | Comparative proteomics analysis of pomegranate seeds on fruit maturation period(Punica granatum L.)显示文摘Seeds play a central role in the life cycle of plants. Seed hardness in pomegranates is of economic relevance, yet scarcely studied and poorly understood in China. In this study, we compared the proteomic differences between Zhongnonghong(soft-seeded) and Sanbai(hard-seeded) pomegranates. A total of 892 protein spots from both varieties were detected on two-dimensional electrophoresis gels(2-DE); 76 spots showed greater than a 1.5-fold or less than a 0.66-fold difference(P<0.05) in Zhongnonghong compared to Sanbai, of which 24 exhibited greater than a 2-fold change. Compared with Sanbai, Zhongnonghong possessed 14 up-regulated, and 10 down-regulated proteins. We identified and annotated 5 of these by using MALDI-TOF-TOF MS: pyruvate dehydrogenase(PDH) E1-β family protein(spot 4 609); alanine aminotransferase 2-like(ALT2L); mitochondrial glycine decarboxylase complex P-protein(spot 5 803); phosphofructokinase B(Pfk B)-type family of carbohydrate kinase(spot 8 411); and putative dna K-type molecular chaperone heat shock cognate protein 70(Hsc70)(spot 9 006). Of these, 3 proteins(spots 4 609, 5 608, 5 803) were hypothesized to play a role in the formation of seed hardness. The other two proteins(spots 8 411, 9 006) were theorized to play a role in protecting the seeds from adverse stress during periods of fruit maturation.
