| 1 | PKS5,a SNF1-related kinase,interacts with and phosphorylates NPR1,and modulates expression of WRKY38 and WRKY62显示文摘NPR1 (Nonexpressor of Pathogenesis-Related gene 1) is a major co-activator of plant defense. Phosphorylations of NPR1 play impor-tant roles in fine-tuning its activity, however a kinase corresponding to such modification remains uncharacterized. Here, we report that NPR1 interacts with PKS5 (SOS2-like Protein Kinase 5). The AKR (AnKyrin Repeats) motif of NPR1 is required for this interaction. PKS5 phosphorylates NPR1 at the C-terminal region. Expression of PKS5 is induced quickly by Pseudomonas syringae pv. tomato DC3000. Expression level of two NPR1 target genes, WRKY38 and WRKY62, is reduced and/or delayed in pks5 mutants. Moreover, the expression of WRKY38 and WRKY62 displays a similar pattern in npr1-1pks5-1 double mutant comparing to that in npr1-1. | 3 | DJ-1 is involved in the multidrug resistance of SGC7901 gastric cancer cells through PTEN/PI3K/Akt/Nrf2 pathway显示文摘Gastric cancer is a common malignancy worldwide.The occurrence of multidrug resistance(MDR)is the major obstacle for effective gastric cancer chemotherapy.In this study,the in-depth molec-ular mechanism of the DJ-1-induced MDR in SGC7901 gastric cancer cells was investigated.The results showed that DJ-1 expression level was higher in MDR variant SGC7901NCR cells than that in its parental SGC7901 cells.Moreover,DJ-1 overexpression conferred the MDR phenotype to SGC7901 cells,while DJ-1 knockdown in SGC7901NCR cells induced re-sensitization to adriamycin,vincristine,cisplatin,and 5-fluorouracil.These results suggested that DJ-1 mediated the develop-ment of MDR in SGC7901 gastric cancer cells.Importantly,further data revealed that the activation of PI3k/Akt and Nrf2 signaling pathway were required for the DJ-1-induced MDR phenotype in SGC7901 gastric cancer cells.Meanwhile,we found that Pl3k/Akt pathway was activated probably through DJ-1 directly binding to and negatively regulating PTEN,consequently resulting in Nrf2 phosphorylation and activation,and thereby inducing Nrf2-dependent P-glycoprotein(P-gp)and Bcl-2 expressions in the DJ-1-mediated MDR of SGC7901 gastric cancer cells.Overall,these results revealed that activating PTEN/PI3K/Akt/Nrf2 pathway and subsequently upregulating P-gp and Bcl-2 expression could be a critical mechanism by which DJ-1 mediates the development of MDR in SGC7901 gastric cancer cells.The new findings may be helpful for understanding the mechanisms of MDR in gastric cancer cells,prompting its further investigation as a molecular target to overcome MDR. | Lejia Qiu Zhaoxia Ma Xiaoran Li Yizhang Deng Guangling Duan Le Zhao Xingwang Xu Lin Xiao Haoyue Liu Zhengming Zhu Heping Chen | 2020 | Acta Biochimica et Biophysica Sinica2020,52,11: | 5 |

| 7 | Exploring the genetic characteristics of 93-11 and Nipponbare recombination inbred lines based on the Golden Gate SNP assay显示文摘Understanding genetic characteristics in rice populations will facilitate exploring evolutionary mechanisms and gene cloning. Numerous molecular markers have been utilized in linkage map construction and quantitative trait locus(QTL) mappings. However, segregation-distorted markers were rarely considered, which prevented understanding genetic characteristics in many populations. In this study, we designed a 384-marker Golden Gate SNP array to genotype 283 recombination inbred lines(RILs) derived from 93-11 and Nipponbare Oryza sativa crosses. Using 294 markers that were highly polymorphic between parents, a linkage map with a total genetic distance of 1,583.2 c M was constructed, including 231 segregation-distorted markers. This linkage map was consistent with maps generated by other methods in previous studies. In total, 85 significant quantitative trait loci(QTLs) with phenotypic variation explained(PVE) values?5% were identified. Among them, 34 QTLs were overlapped with reported genes/QTLs relevant to corresponding traits, and 17 QTLs were overlapped with reported sterility-related genes/QTLs. Our study provides evidence that segregation-distorted markers can be used in linkage map construction and QTL mapping. Moreover, genetic information resulting from this study will help us to understand recombination events and segregation distortion. | 9 | Selenium-layered nanoparticles serving for oral delivery of phytomedicines with hypoglycemic activity to synergistically potentiate the antidiabetic effect显示文摘Diabetes mellitus(DM) remains a great challenge in treatment due to pathological complexity.It has been proven that phytomedicines and natural medicines have prominent antidiabetic effects. This work aimed to develop selenium-layered nanoparticles(SeNPs) for oral delivery of mulberry leaf and Pueraria Lobata extracts(MPE), a group of phytomedicines with significant hypoglycemic activities, to achieve a synergic antidiabetic effect. MPE-loaded SeNPs(MPE-SeNPs) were prepared through a solvent diffusion/in situ reduction technique and characterized by particle size, ζ potential, morphology,entrapment efficiency(EE) and drug loading(DL). The resulting MPE-SeNPs were 120 nm around in particle size with EE of 89.38% for rutin and 90.59% for puerarin, two marker components in MPE. MPESeNPs exhibited a slow drug release and good physiological stability in the simulated digestive fluid.After oral administration, MPE-SeNPs produced significant hypoglycemic effects both in the normal and diabetic rats. Ex vivo intestinal imaging and cellular examinations demonstrated that MPE-SeNPs were provided with outstanding intestinal permeability and transepithelial transport aptness. It was also revealed that MPE-SeNPs could alleviate the oxidative stress, improve the pancreatic function, and promote the glucose utilization by adipocytes. | 14 | Rumen microbiota-host transcriptome interaction mediates the protective effects of trans-10,cis-12 CLA on facilitating weaning transition of lambs显示文摘Developing alternatives to antibiotics for prevention of gastrointestinal dysbiosis in early-weaning farmed animals is urgently needed.This study was to explore the potential effects of trans-10,cis-12 conjugated linoleic acid(CLA)on maintaining ruminal homeostasis of young ruminants during the weaning transition period.Thirty neonatal lambs were selected(6 lambs per group)and euthanized for rumen microbial and epithelial analysis.The lambs were weaned at 28 d and experienced the following 5 treatments:euthanized on d 28 as the pre-weaning control(CON0),fed starter feed for 5(CON5)or 21(CON21)d,fed starter feed with 1%of CLA supplemented for 5(CLA5)or 21(CLA21)d.Results showed that the average daily weight gain and dry matter intake were significantly higher in CLA5 than CON5 group.As compared with the CON5 and CON21 group,the relative abundances of volatile fatty acid(VFA)producing bacteria including Bacteroides,Treponema,Parabacteroides and Anaerovibrio,as well as the concentrations of acetate,butyrate and total VFA were significantly increased in CLA5 and CLA21 group,respectively.Integrating microbial profiling and epithelial transcriptome results showed that 7 downregulated inflammatory signaling-related host genes IL2RA,CXCL9,CD4,CCR4,LTB,SPP1,and BCL2A1 with CLA supplementation were significantly negatively correlated with both VFA concentration and VFA producing bacteria,while 3(GPX2,SLC27A2 and ALDH3A1)and 2(GSTM3 and GSTA1)upregulated metabolism-related genes,significantly positively correlated with either VFA concentration or VFA producing bacteria,respectively.To confirm the effects of CLA on epithelial signal transduction,in vitro experiment was further conducted by treating rumen epithelial cells without or with IL-17A+TNF-αfor 12 h after pretreatment of 100μM CLA or not(6 replicates per treatment).The results demonstrated the anti-inflammatory effect of CLA via suppressing the protein expression of NF-кB p-p65/p65 with the activation of peroxisome proliferator-activated receptor gamma(PPARγ).In conclusion,CLA supplementation enhanced the ruminal microbiota-driven transcriptional regulation in healthy rumen epithelial development via rumen VFA production,and CLA may therefore serve as an alternative way to alleviate early-weaning stress and improve physiological and metabolic conditions of young ruminants. | Chunlei Yang Xiangfei Deng Peter Lund Haixia Liu Xingwang Ding Zhengwei Fu Naifeng Zhang Jinjun Li Lifeng Dong | 2023 | Animal Nutrition2023,,1: | 0 |
| 15 | Methyl cinnamate protects against dextran sulfate sodium-induced colitis in mice by inhibiting the MAPK signaling pathway显示文摘Effective and non-toxic therapeutic agents are lacking for the prevention and treatment of colitis.Previous studies found that methyl cinnamate(MC),extracted from galangal(Alpinia officinarum Hance),has anti-inflammatory properties.However,whether MC is effective as anti-colitis therapy remains unknown.In this study,we investigate the therapeutic effects of MC on dextran sulfate sodium(DSS)-induced colitis in mice and further explore its potential mechanism of action.MC treatment relieves symptoms associated with DSS-induced colitis,including the recovery of DSS-induced weight loss,decreases the disease activity index score,and increases the colon length without toxic side effects.MC treatment protects the integrity of the intestinal barrier in mice with DSS-induced colitis and inhibits the overexpression of pro-inflammatory cytokines in vivo and in vitro.Moreover,the MAPK signaling pathway is found to be closely related to the treatment with MC of colitis.Western blot analysis show that phosphorylation of the p38 protein in colon tissues treated with MC is markedly reduced and phosphorylation levels of the p38,JNK and ERK proteins are significantly decreased in RAW 264.7 cells treated with MC,indicating that the mechanism of MC in treating DSS-induced colitis could be achieved by inhibiting the MAPK signaling pathway.Furthermore,16S RNA sequencing analysis show that MC can improve intestinal microbial dysbiosis in mice with DSS-induced colitis.Altogether,these findings suggest that MC may be a novel therapeutic candidate with anti-colitis efficacy.Furthermore,MC treatment relieves the symptoms of colitis by inhibiting the MAPK signaling pathway and improving the intestinal microbiota. | Lilin E Wenjie Li Yuanjia Hu Lijuan Deng Jianping Yao Xingwang Zhou | 2023 | Acta Biochimica et Biophysica Sinica2023,55,11: | 0 |
