Based on the principle of 'user friendly',it reasonably arranged spatial position of information management system according to spatial distribution of tractor cab. Then,it analyzed operation habits and thinking ways of drivers,and formulated design principle meeting demands of drivers. Besides,it used LabView software to design user interface,including interface layout and interface design. User interface includes basic information interface,job information interface,camera monitoring interface,and fault diagnosis interface. Finally,it made evaluation of the user interface from color,indicator lamp,dial,and pointer. | 8 | A chromosome-scale genome assembly of Dasypyrum villosum provides insights into its application as a broad-spectrum disease resistance resource for wheat improvement显示文摘Dasypyrum villosum is one of the most valuable gene resources in wheat improvement,especially for disease resistance.The mining of favorable genes from D.villosum is frustrated by the lack of a whole genome sequence.In this study,we generated a doubled-haploid line,91C43^(DH),using microspore culture and obtained a 4.05-GB high-quality,chromosome-scale genome assembly for D.villosum.The assembly contains39727 high-confidence genes,and 85.31% of the sequences are repetitive.Two reciprocal translocation events were detected,and 7VS-4VL is a unique translocation in D.villosum.The prolamin seed storage protein-coding genes were found to be duplicated;in particular,the genes encoding low-molecular-weight glutenin at the Glu-V3 locus were significantly expanded.RNA sequencing(RNA-seq)analysis indicated that,after Blumeria graminearum f.sp tritici(Bgt)inoculation,there were more upregulated genes involved in the pattern-triggered immunity and effector-triggered immunity defense pathways in D.villosum than in Triticum urartu.MNase hypersensitive sequencing(MH-seq)identified two Bgt-inducible MH sites(MHSs),one in the promoter and one in the 3'terminal region of the powdery mildew resistance(Pm)gene NLR1-V.Each site had two subpeaks and they were termed MHS1(MHS1.1/1.2)and MHS2(MHS2.1/2.2).Bgt-inducible MHS2.2 was uniquely present in D.villosum,and MHS1.1 was more inducible in D.villosum than in wheat,suggesting that MHSs may be critical for regulation of NLR1-V expression and plant defense.In summary,this study provides a valuable genome resource for functional genomics studies and wheat-D.villosum introgression breeding.The identified regulatory mechanisms may also be exploited to develop new strategies for enhancing Pm resistance by optimizing gene expression in wheat. | Xu Zhang Haiyan Wang Haojie Sun Yingbo Li Yilong Feng Chengzhi Jiao Mengli Li Xinying Song Tong Wang Zongkuan Wang Chunxia Yuan Li Sun Ruju Lu Wenli Zhang Jin Xiao Xiue Wang | 2023 | Molecular Plant2023,16,2: | 1 |
| 9 | Combined cell grafting and VPA administration facilitates neural repair through axonal regeneration and synaptogenesis in traumatic brain injury显示文摘Neuronal regeneration and functional recovery are severely compromised following traumatic brain injury(TBl).Treatment options,including cell transplantation and drug therapy,have been shown to benefit TBl,although the underlying mechanisms remain elusive.In this study,neural stem cells(NSCs)are transplanted into TBl-challenged mice,together with olfactory ensheathing cells(OECs)or followed by valproic acid(VPA)treatment.Both OEC grafting and VPA treatment facilitate the differentiation of NSCs into neurons(including endogenous and exogenous neurons)and significantly attenuate neurological functional defects in TBl mice.Combination of NSCs with OECs or VPA administration leads to overt improvement in axonal regeneration,synaptogenesis,and synaptic plasticity in the cerebral cortex in TBl-challenged mice,as shown by retrograde corticospinal tract tracing,electron microscopy,growth-associated protein 43(GAP43),and synaptophysin(SYN)analyses.However,these beneficial effects of VPA are reversed by local delivery of N-methyl-D-aspartate(NMDA)into tissues surrounding the injury epicenter in the cerebral cortex,accompanied by a pronounced drop in axons and synapses in the brain.Our findings reveal that increased axonal regeneration and synaptogenesis evoked by cell grafting and VPA fosters neural repair in a murine model of TBl.Moreover,VPA-induced neuroprotective roles are antagonized by exogenous NMDA administration and its concomitant decrease in the number of neurons of local brain,indicating that increased neurons induced by VPA treatment mediate axonal regeneration and synaptogenesis in mice after TBl operation.Collectively,this study provides new insights into NSC transplantation therapy for TBI. | Sujuan Liu Haili Tian Yanmei Niu Chunxia Yu Lingjian Xie Zhe Jin Wenyan Niu Jun Ren Li Fu Zhi Yao | 2022 | Acta Biochimica et Biophysica Sinica2022,54,9: | 1 |

| 13 | A simple and sensitive assay for apurinic/apyrimidinic endonuclease 1activity based on host-guest interaction of β-cyclodextrin polymer and pyrene显示文摘Apurinic/apyrimidinic endonuclease 1(APE1) plays an important role in protecting the fidelity of genetic material and is also a promising biomarker of cancers. Herein, a simple and sensitive fluorescent method for APE1 activity detection was designed based on host-guest interaction between β-cyclodextrin polymer(β-CDP) and pyrene. In this method, pyrene-labelled DNA probes(AP-S1 S2) and β-CDP functioned as fluorescent signal producer and enhancer, respectively. When APE1 was absent, pyrene on the AP-S1 S2 could not enter the cavity of β-CDP because of steric hindrance, leading to a weak fluorescence intensity/anisotropy. When APE1 was present, it would recognize and cleave AP site in APS1 S2, thus producing a dissociated pyrene-labelled oligonucleotide segment. Pyrene labelled at the 50 end of oligonucleotide segment could be easily trapped into the cavity of β-CDP via host-guest interaction, leading to a significant enhancement of fluorescence intensity/anisotropy. This assay offered a dynamic range of 0.05 U/m L–5 U/m L and an estimated detection limit of 0.05 U/m L.
