1Human ESC-derived expandable hepatic organoids enable therapeutic liver repopulation and pathophysiological modeling of alcoholic liver injury显示文摘We report the generation of human ESC-derived,expandable hepatic organoids(hEHOs)using our newly established method with wholly defined(serum-free,feeder free)media.The hEHOs stably maintain phenotypic features of bipotential liver stem/progenitor cells that can differentiate into functional hepatocytes or cholangiocytes.The hEHOs can expand for 20 passages enabling large scale expansion to cell numbers requisite for industry or clinical programs.The cells from hEHOs display remarkable repopulation capacity in injured livers of FRG mice following transplantation,and they differentiate in vivo into mature hepatocytes.If implanted into the epididymal fat pads of immune-deficient mice,they do not generate non-hepatic lineages and have no tendency to form teratomas.We further develop a derivative model by incorporating human fetal liver mesenchymal cells(hFLMCs)into the hEHOs,referred to as hFLMC/hEHO,which can model alcoholic liver disease-associated pathophysiologic changes,including oxidative stress generation,steatosis,inflammatory mediators release and fibrosis,under ethanol treatment.Our work demonstrates that the hEHOs have considerable potential to be a novel,ex vivo pathophysiological model for studying alcoholic liver disease as well as a promising cellular source for treating human liver diseases.Shuyong Wang Xuan Wang Zuolong Tan Yuxin Su Juan Liu Mingyang Chang Fang Yan Jie Chen Tao Chen Chuanjiang Li Jie Hu Yunfang Wang 2019Cell Research2019,29,12:20
2Three-dimensional finite-time cooperative guidance for multiple missiles without radial velocity measurements显示文摘In order to simultaneously attack a target with impact angle constraint in threedimensional(3-D) space, a novel distributed cooperative guidance law for multiple missiles under directed communication topologies is proposed without radial velocity measurements. First, based on missiles-target 3-D relative motion equations, the multiple missiles cooperative guidance model with impact angle constraint is constructed. Then, in Line-of-Sight(LOS) direction, based on multiagent system cooperative control theory, one guidance law with directed topologies is designed with strict proof, which can guarantee finite time consensus of multiple missiles' impact times. Next, in elevation direction and azimuth direction of LOS, based on homogeneous system stability theory and integral sliding mode control theory, two guidance laws are proposed respectively with strict proof, which can guarantee LOS angles converge to desired values and LOS angular rates converge to zero in finite time. 3Effects of fumaric acid supplementation on methane production and rumen fermentation in goats fed diets varying in forage and concentrate particle size显示文摘Background: In rumen fermentation, fumaric acid(FA) could competitively utilize hydrogen with methanogenesis to enhance propionate production and suppress methane emission, but both effects were diet-dependent. This study aimed to explore the effects of FA supplementation on methanogenesis and rumen fermentation in goats fed diets varying in forage and concentrate particle size.Methods: Four rumen-cannulated goats were used in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments: low or high ratio of forage particle size: concentrate particle size(Fps:Cps), without or with FA supplementation(24 g/d). Fps:Cps was higher in the diet with chopped alfalfa hay plus ground corn than in that with ground alfalfa hay plus crushed corn.Results: Both increasing dietary Fps:Cps and FA supplementation shifted ruminal volatile fatty acid(VFA) patterns toward more propionate and less acetate in goats. An interaction between dietary Fps:Cps and FA supplementation was observed for the ratio of acetate to propionate(A:P), which was more predominant when FA was supplemented in the low-Fps:Cps diet. Methane production was reduced by FA, and the reduction was larger in the low-Fps:Cps diet(31.72%) than in the high-Fps:Cps diet(17.91%). Fumaric acid decreased ruminal total VFA concentration and increased ruminal p H. No difference was found in ruminal DM degradation of concentrate or alfalfa hay by dietary Fps:Cps or FA. Goats presented a lower ruminal methanogen abundance with FA supplementation and a higher B. fibrisolvens abundance with high dietary Fps:Cps.Conclusions: Adjusting dietary Fps:Cps is an alternative dietary model for studying diet-dependent effects without changing dietary chemical composition. 4The effect of the wave-induced mixing on the upper ocean temperature in a climate model显示文摘The significant underestimation of sea surface temperature(SST)and the temperature in the upper ocean is one of common problems in present climate models.The influence of the wave-induced mixing on SST and the temperature in the upper ocean was examined based on a global climate model.The results from the model coupled with wave-induced mixing showed a significant improvement in the simulation of SST and the temperature in the upper ocean compared with those of the original model without wave effects.Although there has still a cold bias,the new simulation is much closer to the climatology,especially in the northern ocean and tropical ocean.This study indicates that some important physical processes in the accurate simulation of the ocean may be ignored in present climate models,and the wave-induced mixing is one of those factors.Thus,the wave-induced mixing(or the effect of surface waves)should be incorporated properly into climate models in order to simulate or forecast the ocean,then climate system,more accurately.HUANG Chuanjiang QIAO Fangli SONG Zhenya 2008Acta Oceanologica Sinica2008,27,3:5
5Sea level rise projection in the South China Sea from CMIP5 models显示文摘Future potential sea level change in the South China Sea(SCS) is estimated by using 24 CMIP5 models under different representative concentration pathway(RCP) scenarios. By the end of the 21 st century(2081–2100 relative to 1986–2005), the multimodel ensemble mean dynamic sea level(DSL) is projected to rise 0.9, 1.6, and 1.1 cm under RCP2.6, RCP4.5, and RCP8.5 scenarios, respectively, resulting in a total sea level rise(SLR) of 40.9, 48.6, and 64.1 cm in the SCS. It indicates that the SCS will experience a substantial SLR over the 21 st century, and the rise is only marginal larger than the global mean SLR. During the same period, the steric sea level(SSL) rise is estimated to be 6.7, 10.0, and 15.3 cm under the three scenarios, respectively, which accounts only for 16%, 21% and 24% of the total SLR in this region. The changes of the SSL in the SCS are almost out of phase with those of the DSL for the three scenarios. 6Diversification of Sisorid catfishes(Teleostei: Siluriformes)in relation to the orogeny of the Himalayan Plateau显示文摘Sisorid catfishes are primarily limited in distribution to rivers of the Himalayan region and Tibetan Plateau. These species have external morphologies that are adapted for extremely fast-flowing riverine systems. Given the diversity of the group and the above qualities of these catfishes, this lineage serves as an ideal group for inferring the geological history of this region based on their phylogenetic relationships reflecting evolutionary history. We sequenced the complete mitochondrial genome and four nuclear genes of representative sisorids distributed across river systems in China. Phylogenetic analyses strongly support the monophyly of the Sisoridae and the glyptosternoids. An analysis of the reconstructed ancestral states derived from inferred genealogical relationships suggests that the evolution of this lineage was accompanied by convergent evolution in morphological traits that were presumably in response to environmental pressure involving the rapid flowing river system that were generated during the uplift of the Tibetan Plateau(UTP).Molecular dating indicates that the Chinese sisorids and the glyptosternoids originated at the later Miocene(*10.9–9.8Mya), and with further biogeographic analyses indicates that the species of Sisoridae likely originated from a widely distributed ancestor. Moreover, the divergence of the Sisoridae in China can be divided into two phases consistent with the UTP. 7Backstepping-based distributed coordinated tracking formultiple uncertain Euler-Lagrange systems显示文摘Based on the idea of backstepping design, distributed coordinated tracking problems under directed topology are discussed for multiple Euler-Lagrange(EL) systems. The dynamic leader case is considered. First, with the parameter-linearity property, a distributed coordinated adaptive control scheme is proposed for EL systems in the presence of parametric uncertainties.Then, subject to nonlinear uncertainties and external disturbances,an improved adaptive control algorithm is developed by using neural-network(NN) approximation of nonlinear functions. Both proposed algorithms can make tracking errors for each follower ultimately bounded. The closed-loop systems are investigated by using the combination of graph theory, Lyapunov theory, and Barbalat Lemma. 8Nonlinear dynamics and station-keeping control of a rotating tethered satellite system in halo orbits显示文摘The dynamics of a rotating tethered satellite system(TSS) in the vicinity of libration points are highly nonlinear and inherently unstable.In order to fulfll the station-keep control of the rotating TSS along halo orbits,a nonlinear output tracking control scheme based on the h–D technique is proposed.Compared with the popular time-variant linear quadratic regulator(LQR) controller,this approach overcomes some limitations such as on-line computations of the algebraic Riccati equation.Besides,the obtained nonlinear suboptimal controller is in a closed form and easy to implement.Numerical simulations show that the TTS trajectories track the periodic reference orbit with low energy consumption in the presence of both tether and initial injection errors.The axis of rotation can keep pointing to an inertial specifc object to fulfll an observation mission.In addition,the thrusts required by the controller are in an acceptable range and can be implemented through some low-thrust propulsion devices.Liu Gang Huang Jing Ma Guangfu Li Chuanjiang 2013Chinese Journal of Aeronautics2013,26,5:3
9The relationship between sea surface temperature anomaly and wind energy input in the Pacific Ocean显示文摘The relationship between sea surface temperature anomaly(SSTA) and wind energy input in the Pacific Ocean over the period of 1949-2003 is studied by using daily-mean NOAA/NCEP wind stress and monthly mean Reynolds SST data.The results indicate the strong negative correlation between SSTA and local wind energy input to surface waves in most of the domain at low and middle latitudes.The SST is low(high) during the years with more(less) wind energy input.The correlation coefficients are high in the central and eastern tropical Pacific and the central midlatitude North Pacific at the decadal scale,and in the central tropical Pacific at the interannual scale.Vertical mixing processes in the upper ocean are closely associated with wind energy input,indicating that wind energy input may play an important role in interannual and decadal variability in the Pacific Ocean via regulating vertical mixing.Chuanjiang Huang,Fangli Qiao Key Laboratory of Marine Sciences and Numerical Modeling,The First Institute of Oceanography,State Oceanic Administration,Qingdao 266061,China 2009Progress in Natural Science:Materials International2009,19,10:2

12Water-soluble phosphate-functionalized polyfluorene as fluorescence biosensors toward cytochrome c显示文摘An anionic water-soluble polyfluorene derivative, poly(9,9-bis(6′-phosphatehexyl)fluorene-alt-1,4-pheny lene) sodium salt (PFHPNa), was synthesized by Suzuki coupling reaction in DMF/water. Polymer PFHPNa was well soluble in water with a strong blue fluorescence emission. Effect of the side chain length on fluorescence sensory properties was studied by comparing quenching efficiencies toward different quenchers of PFHPNa with a reported polymer poly(9,9-bis(3′-phosphatepropyl)fluorene-alt-1,4-phenylene) sodium salt (PFPPNa), which have different side chains in length. For small molecular quenchers (methylviologen, MV2+) and meso-5,10,15,20-tetrakis-(N-methyl-4-pyridyl)porphine (TMPyP4), polymer PFHPNa had lower sensitivity due to the much longer side chain length. The positively charged metalloprotein cytochrome c could quench fluorescence of conjugated polymers via energy transfer and electron transfer. Moreover, polymer PFHPNa showed higher fluorescence quenching toward large biomolecules than PFPPNa. The corresponding Stern-Volmer (Ksv) value of polymer PFHPNa was determined to be 2.1×108 M-1 for cytochrome c.
