1Regression analysis of major parameters affecting the intensity of coal and gas outbursts in laboratory显示文摘Estimating the intensity of outbursts of coal and gas is important as the intensity and frequency of outbursts of coal and gas tend to increase in deep mining. Fully understanding the major factors contributing to coal and gas outbursts is significant in the evaluation of the intensity of the outburst. In this paper, we discuss the correlation between these major factors and the intensity of the outburst using Analysis of Variance(ANOVA) and Contingency Table Analysis(CTA). Regression analysis is used to evaluate the impact of these major factors on the intensity of outbursts based on physical experiments. Based on the evaluation, two simple models in terms of multiple linear and nonlinear regression were constructed for the prediction of the intensity of the outburst. The results show that the gas pressure and initial moisture in the coal mass could be the most significant factors compared to the weakest factor-porosity. The P values from Fisher's exact test in CTA are: moisture(0.019), geostress(0.290), porosity(0.650), and gas pressure(0.031). P values from ANOVA are moisture(0.094), geostress(0.077), porosity(0.420), and gas pressure(0.051). 2Antidiabetic effect of an engineered bacterium Lactobacillus plantarum-pMG36e-GLP-1 in monkey model显示文摘Glucagon-like peptide-1(GLP-1)reduces postprandial hyperglycaemia,but its short half-life inhibits clinical application.The aim of the current study was to evaluate the treatment efforts of an engineered strain,Lactobacillus plantarum-pMG36e-GLP-1(L.plantarum-pMG36e-GLP-1),that continuously expresses GLP-1 in spontaneous type 2 diabetes mellitus(T2DM)monkeys.After 7 weeks of oral supplementation with L.plantarum-pMG36e-GLP-1,the fasting blood glucose(FPG)of monkeys was significantly(p<0.05)reduced to a normal level and only a small amount of weight was lost.The results of metagenomic sequencing showed that L.plantarum-pMG36e-GLP-1 caused a substantial(p<0.05)reduction in the intestinal pathogen Prevotella and marked enhancement of butyrate-producing Alistipes genera.According to the functional analysis using Kyoto Encyclopaedia of Genes and Genomes(KEGG)pathways,19 metabolism-related pathways were significantly enriched in T2DM monkeys after treatment with L.plantarum-pMG36e-GLP-1.LC-MS faecal metabolomics analysis found 41 significant differential metabolites(11 higher and 30 lower)in monkeys after treatment pathways linked to the metabolism of cofactors and vitamins were the most relevant.The present study suggests that L.plantarum-pMG36e-GLP-1 had an impact on the gut microbial composition and faecal metabolomic profile in spontaneous T2DM monkeys and may be a novel candidate for diabetes treatment.Jie Luo Hongfei Zhang Jiachen Lu ChaoLin Ma Tingtao Chen 2021Synthetic and Systems Biotechnology2021,6,4:3
3Oral PLGA-based DNA vaccines using interferons as adjuvants remarkably promote the immune protection of grass carp(Ctenopharyngodon idella)against GCRV infection显示文摘Grass carp hemorrhagic disease caused by grass carp reovirus(GCRV)results in significant economic losses to the global grass carp aquaculture industry.Oral vaccination is an ideal choice for disease precaution in aquaculture.However,oral vaccine can be degraded in the gut.Therefore,the selection of loading materials is essential.In this study,the S6 and S7 fragments(encoding the outer capsid protein VP4 and fibronectin VP56 of GCRV)and grass carp interferons(IFNs),including IFN1,IFN3,and IFNγ2 were used to create DNA vaccines and adjuvants based on pcDNA3.1,respectively.The oral DNA vaccine was encapsulated in poly(lactic-co-glycolic acid)(PLGA)and polyvinyl alcohol(PVA)with IFNs.The PLGA-PVA(PP)nano-microspheres were prepared by double emulsionsolvent evaporation technique.Using transmission electron microscopy and dynamic light scattering assays,it was determined the vaccines had a spherical structure with uniform particle size(643.5±35.3 nm).The nanomicrospheres possessed excellent encapsulation efficiency(81.6±2.6%)and loading rate(0.54±0.02%),and simultaneously exhibited negligible hemolytic activity and cell toxicity.The protection rate and tissue viral loads post-GCRV challenge in grass carp were assessed.The oral PP nano-microsphere with pVP4 t pIFN1(PP41)vaccine increased protection rate by 44%compared with the control group and was correlated with relatively low viral loads in the spleen,head kidney,and hindgut.Further,three crucial serum biochemical indexes,total superoxide dismutase(TSOD),complement C3(C3),and lysozyme(LZM),were also dramatically increased.Furthermore,mRNA expressions of representative immune-related genes(IgM,IFN1,IFNγ2,MHC-I,and CD8α)in the head kidney and spleen were significantly enhanced.In addition,mRNA expression of IgT was significantly boosted in the hindgut.The results indicate that DNA vaccine capsulated with PP is effective against GCRV infection.The present study provides insights into a prospective strategy for oral vaccine development in aquaculture.Chaolin Jiang Xingchen Huo Lingjie Tang Meidi Hu Chunrong Yang Daji Luo Jianguo Su 2023Water Biology and Security2023,2,2:0
4Automatic Extraction Method of Weld Weak Defect Features for Ultra-High Voltage Equipment显示文摘To solve the problems of low precision of weak feature extraction,heavy reliance on labor and low efficiency of weak feature extraction in X-ray weld detection image of ultra-high voltage(UHV)equipment key parts,an automatic feature extraction algorithm is proposed.Firstly,the original weld image is denoised while retaining the characteristic information of weak defects by the proposed monostable stochastic resonance method.Then,binarization is achieved by combining Laplacian edge detection and Otsu threshold segmentation.Finally,the automatic identification of weld defect area is realized based on the sequential traversal of binary tree.Several characteristic analysis dimensions are established for weld defects of UHV key parts,including defect area,perimeter,slenderness ratio,duty cycle,etc.The experiment using theweld detection image of the actual production site shows that the proposedmethod can effectively extract theweak feature information ofweld defects and further provide reference for decision-making.Guanghua Zheng Chaolin Luo Mengen Shen Wanzhong Lv Wenbo Jiang Weibo Yang 2023Energy Engineering2023,120,4:0
