| 1 | 猪细小病毒AV30 VP2基因在昆虫细胞中表达及其免疫原性鉴定显示文摘本研究选用Bac-to-Bac昆虫杆状病毒表达系统构建含有猪细小病毒AV30未优化、优化后衣壳蛋白VP2基因的重组杆状病毒,感染sf9、HF细胞表达VP2蛋白。首先,扩增PPV AV30得到VP2基因,测序后由公司根据昆虫细胞嗜性进行基因优化。同时构建并收获未优化、优化重组杆状病毒r BacVP2。其感染HF细胞后,进行Western-blot及IFA实验分析表明,未优化、优化VP2蛋白在昆虫细胞中正确表达,分子质量约为64 ku,并具有良好的反应原性。电镜观察,未优化、优化VP2蛋白均自我组装成VLP,与PPV AV30病毒粒子大小形状相似。最后,用VP2-VLP免疫BALB/c小鼠,能刺激小鼠产生抗PPV的特异性抗体。免疫后的小鼠脾脏淋巴细胞在PPV AV30病毒的刺激后,出现显著的增殖情况,同时产生了Th1和Th2型相关细胞因子。本研究成功构建了VP2-VLP,并在小鼠实验中显示了良好的免疫效果,为以VP2为表位载体的嵌合型病毒样颗粒疫苗进一步研制奠定了基础。 | 李茜 代军飞 丁耀忠 ASHENAFI Kiros Wubshe 李国秀 马炳 张杰 | 2020 | 中国兽医科学2020,50,7: | 4 |
| 2 | 南非2型口蹄疫病毒结构蛋白抗原表位研究进展显示文摘口蹄疫病毒(FMDV)感染会引起偶蹄动物发生口蹄疫(FMD),产生急性系统性传染性水疱。其分为7个血清型,包括A、O、C型,亚州1型(Asia 1)及南非1、2、3型(SAT1、2、3)。其中,SAT FMDV具有明显的地域性,主要集中在撒哈拉沙漠以南,侵害水牛及野生动物,容易发生抗原变异,疫苗交叉保护效果低。近几年的报告表明,中东地区暴发了SAT2FMDV,跨越了长久以来的地域界限,也对我国的养殖业造成了潜在的威胁。我国在SAT FMDV方面的研究较少,有必要建立特异性和灵敏性高的SAT FMDV监测方法作为战略储备,以防其跨境传播。论文主要介绍了SAT2FMDV结构蛋白抗原表位国内外研究进展,以期为今后研发有关SAT2FMDV战略储备表位疫苗提供理论依据。 | 李茜 代军飞 丁耀忠 李国秀 侯谦 Ashenafi Kiros Wubshet 马炳 张永光 张杰 | 2020 | 动物医学进展2020,41,6: | 3 |
| 3 | Beyond just a tight fortress:contribution of stroma to epithelial-mesenchymal transition in pancreatic cancer显示文摘Novel effective treatment is direly needed for patients with pancreatic ductal adenocarcinoma(PDAC).Therapeutics that target the driver mutations,especially the KRAS oncoprotein and its effector cascades,have been ineffective.It is increasing clear that the extensive fibro-inflammatory stroma(or desmoplasia)of PDAC plays an active role in the progression and therapeutic resistance of PDAC.The desmoplastic stroma is composed of dense extracellular matrix(ECM)deposited mainly by the cancer-associatedfibroblasts(CAFs)and infiltrated with various types of immune cells.The dense ECM functions as a physical barrier that limits tumor vasculatures and distribution of therapeutics to PDAC cells.In addition,mounting evidence have demonstrated that both CAFs and ECM promote PDAC cells aggressiveness through multiple mechanisms,particularly engagement of the epithelial-mesenchymal transition(EMT)program.Acquisition of a mesenchymal-like phenotype renders PDAC cells more invasive and resistant to therapyinduced apoptosis.Here,we critically review seminal and recent articles on the signaling mechanisms by which each stromal element promotes EMT in PDAC.We discussed the experimental models that are currently employed and best suited to study EMT in PDAC,which are instrumental in increasing the chance of successful clinical translation. | Ashenafi Bulle Kian-Huat Lim | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 2 |
