4Differential effects of atrial and brain natriuretic peptides on human pulmonary artery:An in vitro study显示文摘BACKGROUND The prevalence of cardiovascular diseases,especially heart failure,continues to rise worldwide.In heart failure,increasing levels of circulating atrial natriuretic peptide(ANP)and brain natriuretic peptide(BNP)are associated with a worsening of heart failure and a poor prognosis.AIM To test whether a high concentration of BNP would inhibit relaxation to ANP.METHODS Pulmonary arteries were dissected from disease-free areas of lung resection,as well as pulmonary artery rings of internal diameter 2.5–3.5 mm and 2 mm long,were prepared.Pulmonary artery rings were mounted in a multiwire myograph,and a basal tension of 1.61gf was applied.After equilibration for 60 min,rings were pre-constricted with 11.21μmol/L PGF2α(EC80),and concentration response curves were constructed to vasodilators by cumulative addition to the myograph chambers.RESULTS Although both ANP and BNP were found to vasodilate the pulmonary vessels,ANP is more potent than BNP.pEC50 of ANP and BNP were 8.96±0.21 and 7.54±0.18,respectively,and the maximum efficacy(Emax)for ANP and BNP was-2.03 gf and-0.24 gf,respectively.After addition of BNP,the Emax of ANP reduced from-0.96gf to-0.675gf(P=0.28).CONCLUSION BNP could be acting as a partial agonist in small human pulmonary arteries,and inhibits relaxation to ANP.Elevated levels of circulating BNP could be responsible for the worsening of decompensated heart failure.This finding could also explain the disappointing results seen in clinical trials of ANP and BNP analogues for the treatment of heart failure.Azar Hussain Robert T Bennett Zaheer Tahir Emmanuel Isaac Mubarak A Chaudhry Syed S Qadri Mahmoud Loubani Alyn H Morice 2019World Journal of Cardiology2019,11,10:1

13The rediscovery of Strix butleri(Hume,1878)in Oman and Iran,with molecular resolution of the identity of Strix omanensis Robb,van den Berg and Constantine,2013显示文摘Background:Many species of owls(Strigidae) represent cryptic species and their taxonomic study is in flux.In recent years,two new species of owls of the genus Strix have been described from the Middle East by different research teams.It has been suggested that one of these species,S.omanensis,is not a valid species but taxonomic compari?sons have been hampered by the lack of voucher specimens of S.omanensis,and the poor state of the holotype of S.butleri.Methods:Here we use new DNA sequence data to clarify the taxonomy and nomenclature of the S.butleri complex.We also report the capture of a single S.butleri sensu stricto in Mashhad,Iran.Results:A cytochrome b sequence of S.omanensis was found to be identical to that of the holotype of S.butleri,indicating that the name S.omanensis is best regarded as a junior synonym of S.butleri.The identity of the S.butleri captured in Mashhad,Iran,was confirmed using DNA sequence data.This represents a major(1300 km) range exten?sion of this species.Conclusions:The population discovered in Oman in 2013 and originally named ‘S.omanensis' actually represents the rediscovery of S.butleri,which was known from a single specimen and had not been recorded since 1878.The range of S.butleri extends into northeast Iran.Our study augments the body of evidence for the recognition of S.butleri and S.hadorami as separate species and highlights the importance of using multiple evidence to study cryptic owl species.Magnus S.Robb George Sangster Mansour Aliabadian Arnoud B.van den Berg Mark Constantine Martin Irestedt Ali Khani Seyed Babak Musavi Joao M.G.Nunes Maia Sarrouf Willson Alyn J.Walsh 2016Chinese Birds2016,,2:0

15Characterization of optimal resting tension in human pulmonary arteries显示文摘AIM To determine the optimum resting tension(ORT) for in vitro human pulmonary artery(PA) ring preparations.METHODS Pulmonary arteries were dissected from disease free sections of the resected lung in the operating theatre and tissue samples were directly sent to the laboratory in Krebs-Henseleit solution(Krebs).The pulmonary arteries were then cut into 2 mm long rings.PA rings were mounted in 25 m L organ baths or 8 m L myograph chambers containing Krebs compound(37 ℃,bubbled with 21% O_2:5% CO_2) to measure changes in isometric tension.The resting tension was set at 1-gram force(gf) with vessels being left static to equilibrate for duration of one hour.Baseline contractile reactions to 40 mmol/L KCl were obtained from a resting tension of 1 gf.Contractile reactions to 40 mmol/L KCl were then obtained from stepwise increases in resting tension(1.2,1.4,1.6,1.8 and 2.0 gf).RESULTS Twenty PA rings of internal diameter between 2-4 mmwere prepared from 4 patients.In human PA rings incrementing the tension during rest stance by 0.6 gf,up to 1.6 gf significantly augmented the 40 mmol/L KCl stimulated tension.Further enhancement of active tension by 0.4 gf,up to 2.0 gf mitigate the 40 mmol/L KCl stimulated reaction.Both Myograph and the organ bath demonstrated identical conclusions,supporting that the radial optimal resting tension for human PA ring was 1.61 g.CONCLUSION The radial optimal resting tension in our experiment is 1.61 gf(15.78 m N) for human PA rings.Azar Hussain Robert T Bennett Mubarak A Chaudhry Syed S Qadri Mike Cowen Alyn H Morice Mahmoud Loubani 2016World Journal of Cardiology2016,8,9:0
16Biofilm analyses and exoproduct release by clinical and environmental isolates of Burkholderia pseudomallei from Brazil显示文摘Objective:To characterize biofilm production by clinical(n=21)and environmental(n=11)isolates of Burkholderia pseudomallei and evaluate the production of proteases,hemolysins and siderophores.Methods:Initially,the 32 strains were evaluated for biofilm production in Müller-Hinton broth-1%glucose(MH-1%glucose)and BHI broth-1%glucose,using the crystal violet staining technique.Subsequently,growing(48 h)and mature(72 h)biofilms were evaluated by confocal microscopy.Finally,the production of proteases,hemolysins and siderophores by planktonic aggregates,growing biofilms and mature biofilms was evaluated.Results:All isolates produced biofilms,but clinical isolates had significantly higher biomass in both MH-1%glucose(P<0.001)and BHI-glucose 1%(P=0.005).The structural analyses by confocal microscopy showed thick biofilms,composed of multiple layers of cells,homogeneously arranged,with mature biofilms of clinical isolates presenting higher biomass(P=0.019)and thickness of the entire area(P=0.029),and lower roughness coefficient(P=0.007)than those of environmental isolates.Protease production by growing biofilms was significantly greater than that of planktonic(P<0.001)and mature biofilms(P<0.001).Hemolysin release by planktonic aggregates was higher than that of biofilms(P<0.001).Regarding siderophores,mature biofilms presented higher production than growing biofilms(P<0.001)and planktonic aggregates(P<0.001).Conclusions:Clinical isolates have higher production of biofilms than their environmental counterparts;protease and siderophores seem important for growth and maintenance of Burkholderia pseu­domallei biofilms.Glaucia Morgana de Melo Guedes Crister JoséOcadaque Alyne Soares Freitas Rodrigo Machado Pinheiro Giovanna Barbosa Riello Silviane Praciano Bandeira Rossana de Aguiar Cordeiro Marcos Fábio Gadelha Rocha JoséJúlio Costa Sidrim Débora de Souza Collares Maia Castelo-Branco 2023Asian Pacific Journal of Tropical Medicine2023,16,7:0

18Partitioning between primary and secondary metabolism of carbon allocated to roots in four maize genotypes under water deficit and its effects on productivity显示文摘Plants may respond to drought by altering biomass allocation to shoots and roots or by changing the metabolic activities in these organs. To determine how drought changes the partitioning of carbon allocated to growth and secondary metabolism in maize roots and how it affects photosynthesis(A) and productivity in maize, we evaluated leaf gas exchange, yield componentes, root morphology, and primary and secondary metabolites including total soluble sugars(TSS), starch(S), phenolics(PHE), and lignin(LIG). Data were collected from pot-grown plants of four maize genotypes: BRS 1010 and 2B710(sensitive genotypes) and DKB390 and BRS1055(tolerant genotypes) under two soil water tensions: field capacity(FC,-18 kP a) and water deficit(WD,-138 kP a). WD was applied at the pre-flowering stage for 12 days and then the water supply was restored and maintained at optimum levels until the end of the cycle. For genotype BRS 1055 under FC, the greatest A did not result in greater grain biomass(DGB) because the accumulated photoassimilates had already filled the cells, and thus the excessive TSS synthesized in leaves was allocated to roots in large amounts. However, the sharp decrease in A caused by WD imposition in this genotype did not affect the influx pressure of leaf TSS, which was due largely to conversion of primary metabolites to PHE compounds to increase the length of fine roots. In leaves of DKB390 under WD, both S and TSS were reduced, whereas PHE were increased to prevent excessive water loss and xylem cavitation. Under WD, both BRS1010 and2B710 genotypes displayed reduced allocation of biomass to shoots and roots and LIG content in leaves, as well as lower A and DGB values.
