1人脐带间充质干细胞分离培养条件的优化及其生物学特性显示文摘背景:由于大量获取骨髓细胞存在困难,并且骨髓间充质干细胞随年龄的增加出现数量及分化潜能下降,因此需要寻找其他间充质干细胞来源。目的:优化人脐带间充质干细胞的分离培养条件,进一步明确其生物学特性。设计、时间及地点:细胞学体外观察,于2006-09/2008-03在中国医学科学院血液学研究所国家重点实验室完成。材料:17份脐带标本取自健康足月新生儿,由天津市中心妇产医院提供。方法:脐带采集后在6h内分别用植块法、胶原酶消化法、胶原酶与胰酶联合消化法进行分离培养,于培养第14天计数集落数,超过50个细胞计为集落。当细胞达70%~80%融合时,胰蛋白酶-乙二胺四乙酸混合消化,以(2.5~5.0)×103/cm2密度传代培养。主要观察指标:比较不同培养方法所获贴壁细胞的形态学特征、细胞产率,应用流式细胞技术分析细胞增殖活性、免疫表型,特异性染色方法鉴定细胞的多向分化潜能。结果:植块法培养6~10d可见成纤维样细胞从植块边缘爬出,形态与骨髓间充质干细胞相似,散在分布或形成小克隆,原代可获得(5.2±1.7)×105个贴壁细胞/0.5cm脐带组织,至少可稳定传15代,平均每代扩增6.2倍,在原代细胞克隆数、细胞产率、传代时间及扩增倍数上与胶原酶消化法比较存在明显差异(P<0.05);而胶原酶与胰酶联合消化法接种后未见明显贴壁细胞。人脐带间充质干细胞的倍增时间为45h,72.09%细胞处于G0/G1期,不表达造血细胞及内皮细胞标记,高表达整合素和黏附分子CD44,CD90,CD95以及CD73,CD105,在特定诱导条件下,人脐带间充质干细胞能够向成骨及成脂肪细胞方向分化。结论:应用植块法可以从人脐带组织中简便高效地分离出一群具有高增殖活性及多向分化潜能的间充质干细胞,其免疫表型类似于骨髓间充质干细胞。徐燕 李长虹 孟恒星 郝牧 邱录贵 2009中国组织工程研究与临床康复2009,13,32:38
2人脐带间充质干细胞生物学特性及其研究进展显示文摘背景:人脐带间充质干细胞是一类具有自我更新和多向分化潜能的成体干细胞,具有来源丰富,对供者无影响,易于采集和运输,无异体排斥反应,避免伦理争议等诸多优点。目的:综述人脐带间充质干细胞的生物学特性及其应用进展。方法:以'人脐带间充质干细胞,生物学特征,基因分析,诱导分化,human umbilical cord mesenchymal stem cells(hUMSCs),biocharacteristics,gene analysis,induce differentiation'为关键词应用计算机检索CNKI数据库、万方数据库、PubMed数据库文章。结果与结论:人脐带间充质干细胞呈典型的成纤维状,其表达的表面标志抗原具有非单一性,高表达间质细胞标志、整合素受体,不表达造血系标志、协同刺激分子CD80、CD86和CD40、人白细胞抗原HLA-DR,HLA-G,HLA-DP,HLA-DQ、内皮标志CD31或CD33、CD14、CD56等。人脐带间充质干细胞与造血干细胞和胚胎干细胞类似,在体外可以分化为骨细胞、软骨细胞、肝细胞、心肌细胞等;在体内可以分化为多巴胺能神经元、骨骼肌细胞、内皮细胞、胰岛细胞等。但人脐带间充质干细胞的研究仍存在亟需解决的问题,如分离方法、培养条件的规范化,如何控制其生长和分化等。马锡慧 冯凯 石炳毅 2011中国组织工程研究与临床康复2011,15,32:23
3Effects of salvianolic acid B on proliferation, neurite outgrowth and differentiation of neural stem cells derived from the cerebral cortex of embryonic mice显示文摘Salvianolic acid B is isolated from Salvia miltiorrhiza,the root of which is widely used as a traditional Chinese medicine to treat stroke.However,little is known about how salvianolic acid B influences growth characteristics of neural stem cells (NSCs).The purpose of the present study was to evaluate the effects of salvianolic acid B on proliferation,neurite outgrowth and differentiation of NSCs derived from the cerebral cortex of embryonic mice using MTT,flow cytometry,immunofluorescence and RT-PCR.It was found that 20 μg mL·1 and 40 μg mL·1 salvianolic acid B had similar effects on proliferation of NSCs,and a suitable concentration of salvianolic acid B increased the number of NSCs and their derivative neurospheres.The growth-promoting activity of salvianolic acid B was dependent on and associated with an accumulation in the G2/S-phase cell population.Salvianolic acid B also promoted the neurite outgrowth of NSCs and their differentiation into neurons.The mRNA for tau,GFAP and nestin were present in differentiating neurospheres induced by salvianolic acid B.However,high-level expression of tau mRNA and low-level expression of GFAP mRNA was detected in differentiated cells,in contrast to the control conditions.This collective evidence indicates that exogenous salvianolic acid B is capable of promoting proliferation of neurospheres and differentiation towards the neuronal lineage in vitro and may act in the proliferation of NSCs and may promote NSC differentiation into neuronal cells.GUO GuoQing1,LI Bin1,WANG YuanYuan1,SHAN AiJing1,SHEN WeiZai1,YUAN Lin2 & ZHONG ShiZhen2 1Department of Anatomy,Medical College of Jinan University,Guangzhou 510630,China 2Institute of Clinical Anatomy,Key Laboratory of Tissue Construction and Detection of Guangdong Province,Southern Medical University,Guangzhou 510150,China 2010Science China(Life Sciences)2010,53,6:21

6Ultrastructure of neuronal-like cells differentiated from adult adipose-derived stromal cells显示文摘β-mercaptoethanol induces in vitro adult adipose-derived stromal cells(ADSCs) to differentiate into neurons.However,the ultrastructural features of the differentiated neuronal-like cells remain unknown.In the present study,inverted phase contrast microscopy was utilized to observe β-mercaptoethanol-induced differentiation of neuronal-like cells from human ADSCs,and immunocytochemistry and real-time polymerase chain reaction were employed to detect expression of a neural stem cells marker(nestin),a neuronal marker(neuron-specific enolase),and a glial marker(glial fibrillary acidic protein).In addition,ultrastructure of neuronal-like cells was observed by transmission election microscopy.Results revealed highest expression rate of nestin and neuron-specific enolase at 3 and 5 hours following induced differentiation;cells in the 5-hour induction group exhibited a neuronal-specific structure,i.e.,Nissl bodies.However,when induction solution was replaced by complete culture medium after 8-hour induction,the differentiated cells reverted to the fibroblast-like morphology from day 1.These results demonstrate that β-mercaptoethanol-induced ADSCs induced differentiation into neural stem cells,followed by morphology of neuronal-like cells.However,this differentiation state was not stable.Changqing Ye Xiaodong Yuan Hui Liu Yanan Cai 2010Neural Regeneration Research2010,5,19:16
7人脐带间充质干细胞分化为胰岛β细胞的实验研究显示文摘目的探讨人脐带华尔通胶质来源的间充质干细胞(MSCs)分化为胰岛素分泌细胞的可能性。方法从足月剖宫产的新生儿脐带华尔通胶质中分离培养出MSCs,传至第5代用含尼克酰胺和β巯基乙醇的低糖达尔伯克必需基本培养基预诱导,以含尼克酰胺和β巯基乙醇的高糖达尔伯克必需培养基诱导其向胰岛素分泌细胞分化,应用免疫细胞化学法检测其胰岛素的表达,以双硫腙染色鉴定胰岛β细胞,并用放射免疫法检测诱导后细胞培养液中胰岛素水平,检测均以未诱导细胞作为实验对照。结果免疫细胞化学法检测结果显示,经尼克酰胺和β巯基乙醇联合诱导后的人脐带MSCs表达胰岛β细胞的标记胰岛素。双硫腙染色发现脐带华尔通胶质来源的MSCs经尼克酰胺和β巯基乙醇诱导后细胞内锌离子增加。通过胰岛素放射免疫法检测诱导前后细胞培养液中胰岛素水平,实验组和对照组分别为(6.63±1.80)U/L和(3.39±0.21)U/L,差异具有统计学意义(P<0.05)。结论人脐带MSCs经尼克酰胺和β巯基乙醇联合诱导后可表达胰岛素β细胞的表面标记,增加细胞内锌离子水平,使之具备胰岛β细胞的特点,诱导后的细胞也可分泌胰岛素。用尼克酰胺和β巯基乙醇可诱导人脐带MSCs分化成为胰岛素分泌细胞,从而为1型糖尿病的细胞移植治疗提供新的细胞来源。马桂霞 何红燕 罗敏洁 黄天华 肖升平 崔冰琳 马廉 2008实用儿科临床杂志2008,23,8:15
8Transplantation of mesenchymal stem cells from human umbilical cord versus human umbilical cord blood for peripheral nerve regeneration显示文摘BACKGROUND: Mesenchymal stem cells (MSCs) appear to be a good alternative to Schwann cells in the treatment of peripheral nerve injury. Fetal stem cells, like umbilical cord blood (UCB) and umbilical cord (UC) stem cells, have several advantages over adult stem cells. OBJECTIVE: To assess the effects of UC-derived MSCs (UCMSCs) and UCB-derived MSCs (UCBMSCs) in repair of sciatic nerve defects. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at the laboratory of Department of Oral and Maxillofacial Surgery, Seoul National University Dental Hospital, from July to December 2009. MATERIALS: UCMSCs were provided by the Research Institute of Biotechnology, Dongguk University. UCBMSCs were provided by the Laboratory of Stem Cells and Tumor Biology, College of Veterinary Medicine, Seoul National University. Dulbecco's modified Eagle's medium (DMEM) was purchased from Gibco-BRL, USA. METHODS: Seven-week-old Sprague-Dawley rats were randomly and evenly divided into three groups: DMEM, UCBMSCs, and UCMSCs. A 10-mm defect in the left sciatic nerve was constructed in all rats. DMEM (15 μL) containing 1 × 106 UCBMSCs or UCMSCs was injected into the gap between nerve stumps, with the surrounding epineurium as a natural conduit. For the DMEM group, simple DMEM was injected. MAIN OUTCOME MEASURES: At 7 weeks after sciatic nerve dissection, dorsal root ganglia neurons were labeled by fluorogold retrograde labeling. At 8 weeks, electrophysiology and histomorphometry were performed. At 2, 4, 6, and 8 weeks after surgery, sciatic nerve function was evaluated using gait analysis. RESULTS: The UCBMSCs group and the UCMSCs group exhibited similar sciatic nerve function and electrophysiological indices, which were better than the DMEM group, as measured by gait analysis (P < 0.05). Fluorogold retrograde labeling of sciatic nerve revealed that the UCBMSCs group demonstrated a higher number of labeled neurons; however, the differences were not significant. Histomorphometric indices were similar in the UCBMSCs and UCMSCs groups, and total axon counts, particularly axon density (P < 0.05), were significantly greater in the UCBMSCs and UCMSCs groups than in the DMEM group. CONCLUSION: Transplanting either UCBMSCs or UCMSCs into axotomized sciatic nerves could accelerate and promote sciatic nerve regeneration over 8 weeks. 10异种脐带间充质干细胞静脉应用的安全性显示文摘背景:人脐带间充质干细胞因其来源丰富,具有较强增殖分化能力,不涉及伦理道德问题,有望成为组织修复与再生工程的首选细胞。但是,对人脐带间充质干细胞异种静脉移植的安全性研究较少。目的:观察Wistar大鼠静脉输注人脐带间充质干细胞后的安全性。方法:SPF级Wistar大鼠30只,随机分为空白对照组、阴性对照组、细胞移植组,每组10只,雌雄各半。体外分离扩增人脐带间充质干细胞,细胞移植组大鼠尾静脉一次注射5×106间充质干细胞,注射体积为1mL;阴性对照组大鼠尾静脉一次注射相同体积50g/L葡萄糖注射液。注射后每天观察大鼠一般症状,14d后解剖动物,进行血常规、血生化和脏器病理学检查及脏器质量测定。结果与结论:细胞移植组大鼠血常规、血生化与对照组比较差异无显著性意义(P>0.05)。细胞移植组大鼠脏器组织病理学观察与对照组大鼠无明显光镜下形态学差别。结果提示Wistar大鼠一次性静脉移植人脐带间充质干细胞是安全可行的,人脐带间充质干细胞异种移植对受者无不良影响。张学峰 张美荣 胡建霞 王丽 张桂芝 邵磊 李晟刚 高宏 苗志敏 王颜刚 2010中国组织工程研究与临床康复2010,14,32:14
11人脐带间充质干细胞向胰岛样细胞的分化显示文摘目的:前期实验采用药物诱导的方法将人脐带间充质干细胞诱导分化为胰岛样细胞,将人脐带间充质干细胞与大鼠胰岛细胞共同培养及移植入糖尿病大鼠体内,观察其在胰岛细胞生长的微环境及在体环境中向胰岛样细胞分化的潜能。方法:实验于2006-10/2007-09在汕头大学医学院生殖遗传实验室完成,属于广东省科技厅重点实验室。①实验材料:SD大鼠由广州中医药大学实验动物中心提供。对动物处置符合动物伦理学标准。脐带取自汕大附二妇产科健康足月妊娠剖宫产的胎儿,产妇及家属对实验知情同意,并经医院伦理委员会批准。②实验方法:分离培养人脐带间充质干细胞,并采用酶消法分离大鼠胰岛细胞。将人脐带间充质干细胞与大鼠胰岛细胞以半透膜相隔共同培养,在共培养的第3,7,14天采用放射免疫法检测胰岛素水平,RT-PCR方法检测PDX-1基因的表达,以单独培养的脐带间充质干细胞为对照。经尾静脉将人脐带间充质干细胞移植入糖尿病模型大鼠体内,采用半定量RT-PCR方法检测人insulin基因表达。结果:①共培养条件下的人脐带间充质干细胞由长梭形逐渐变圆,并聚集成团。②放射免疫法结果表明,共培养组人脐带间充质干细胞随葡萄糖浓度的增高而分泌的胰岛素量也增加,与对照组相比差异显著(P<0.05)。③未经共培养诱导的人脐带间充质干细胞不表达PDX-1,共培养3d的人脐带间充质干细胞表达PDX-1,共培养7d的人脐带间充质干细胞仍表达PDX-1,共培养14d的人脐带间充质干细胞未表达PDX-1。④未经移植人脐带间充质干细胞的大鼠胰腺不表达人insulin基因,而移植人脐带间充质干细胞的大鼠胰腺在第60天时能够检测出人insulin基因。结论:人脐带间充质干细胞具有在体内外的微环境中向胰岛样细胞分化的潜能。王爱红 何红燕 罗敏洁 黄天华 马廉 马桂霞 林丽敏 林广裕 史雪川 2008中国组织工程研究与临床康复2008,12,21:12
12丹酚酸B促进胎鼠大脑皮质神经干细胞增殖、突起生长和分化显示文摘丹参是一种常用于治疗脑卒中的传统中草药,丹酚酸B是其有效成份之一.但人们对丹酚酸B如何影响神经干细胞的生长还了解甚少.本研究用MTT、流式细胞术、免疫荧光和RT-PCR技术检测丹酚酸B对胎鼠大脑皮质神经干细胞增殖、突起生长和分化的作用.结果显示,20和40μg/mL丹酚酸B对促进神经干细胞增殖的作用相似,适量的丹酚酸B可促进神经干细胞增殖并形成较多的神经干细胞球,其促增殖作用通过提高G2/S期细胞的数量而实现.丹酚酸B还可促进神经干细胞球发出较多的突起并分化出较多的成熟神经元.分化开始时,tau,GFAP和nestin mRNA均有表达.但分化后的细胞与对照组比较,tau mRNA表达增多,而GFAP mRNA表达减少.说明外源性的丹酚酸B具有促进神经干细胞增殖并向神经元分化的作用,有望成为一种获取更多神经干细胞和促进神经干细胞分化的药物.郭国庆 李斌 王圆圆 山爱景 沈伟哉 原林 钟世镇 2009中国科学(C辑)2009,39,8:12
13Effect of allogeneic umbilical cord mesenchymal stem cell transplantation in a rat model of hepatic cirrhosis显示文摘OBJECTIVE: To determine the effects of human umbilical cord mesenchymal stem cell(UCMSC) transplantation, alone or in combination with tanshinone IIA(Tan IIA) on hepatic cirrhosis in rats.METHODS: A rat model of cirrhosis was established. Rats were divided into control, UCMSC, and UCSMC plus Tan IIA groups. Rats in the UCMSC group were injected via the tail vein with 0.2 m L Di I-labeled UCMSC suspension. Intraperitoneal Tan IIA injections(20 mg/kg) were started on the day ofUCMSC transplantation in the UCMSC plus Tan IIA group, and continued for 7 consecutive days thereafter. Rats were sacrificed 1 day, 3 days, 1 month,and 3 months after transplantation and the numbers of Di I-labeled UCMSCs colonizing the liver were determined. Albumin(ALB) and alanine aminotransferase(ALT) levels were measured in venous blood, and m RNA and protein expression levels of human ALB and cytokeratin(CK)-18 in liver tissues were determined by reverse transcription-polymerase chain reaction and western blotting, respectively.RESULTS: Serum ALT levels were significantly lower and serum ALB levels significantly higher in rats in the UCMSC group compared with the control group(P < 0.05). Hepatic CK-18 and ALB m RNA and protein expression levels increased after transplantation, and were significantly higher in the UCMSC plus Tan IIA group compared with the UCMSC group(P < 0.05).CONCLUSION: Human UCMSCs transplanted into rats with liver cirrhosis can grow and differentiate into hepatocyte-like cells resulting in improved liver function in vivo. 14人脐带间充质干细胞分化为神经细胞的形态学改变显示文摘目的研究人脐带华尔通胶来源的间充质干细胞(MSCs)分化为神经细胞的形态学特征,为神经细胞移植探索细胞来源。方法用复方丹参注射液诱导人脐带来源的MSCs向神经细胞分化,用光学和电子显微镜对分化和未分化的细胞进行观察;了解细胞分化同形态学改变的关系。结果丹参可诱导人脐带MSCs向神经样细胞分化,光镜下细胞变短,体积变小,细胞形成双极或多极的细胞体,细胞呈星型,部分细胞的胞质收缩形成细胞的突起;扫描电镜下观察,复方丹参注射液诱导后,细胞胞体形成多样的神经样轴突,细胞之间形成网络样的连接;透射电镜下,丹参诱导后,细胞内结构、细胞器发生明显变化,细胞成熟,胞质内可见大量的线粒体,粗面内质网和游离的核糖体等,诱导后的细胞可见尼氏体样结构——大量排列的粗面内质网和广泛分布的游离核糖体和发达的线粒体;诱导后的细胞也表达神经细胞的标记。结论复方丹参注射液不仅诱导人脐带来源的MSCs发生神经样细胞的形态学改变,同时伴发有细胞分化的过程和神经细胞杯记表达,证明人脐带来源的MSCs具有分化为神经细胞的能力。何红燕 崔冰琳 冯学永 肖平 林晓波 蒋学武 罗敏洁 马廉 2006国际输血及血液学杂志2006,29,6:11
15长期培养人脐带间充质干细胞的生物活性及其限制性显示文摘背景:间充质干细胞具有多向分化能力、免疫调节能力,并可在体外进行长期培养扩增,但其体外长期培养的生物学活性变化特点及其限制性仍不十分清晰。目的:观察体外长期培养人脐带间充质干细胞的生物活性变化特点及其限制性。方法:无菌收集剖宫产新生儿脐带,经胶原酶Ⅱ消化后,用黏附生长选择法获取人脐带间充质干细胞,胰酶消化传代;取第3代细胞进行流式分析,脂肪和成骨诱导鉴定;收集不同代的细胞分别用MTT法、流式细胞仪和爬片分析法检测细胞增殖活性、细胞周期、凋亡率及黏附能力。结果与结论:人脐带间充质干细胞的细胞表型CD105+/CD29+/CD44+/CD31-/CD34-/CD45-/HLADR-,可诱导分化为脂肪细胞和成骨细胞;传代培养第3~23代细胞形态无明显变化,生长曲线基本一致,第28代后细胞增长缓慢;流式分析发现第33代细胞较第3代G0/G1减少17.9%、S+G2/M增加103.4%、细胞凋亡率增加316.7%,差异均有显著性意义(P<0.01),第3~18代细胞间G0/G1、S+G2/M、细胞凋亡率和黏附细胞数差异均无显著性意义(P>0.05)。提示在体外长期培养环境中,随着传代次数的递增,人脐带间充质干细胞的增殖及黏附能力下降,细胞凋亡率增加,总的生物活性呈逐渐下降趋势,其最佳生物活性期出现在培养第3~18代之间。许超 廖继东 柳菁 毛文哲 李扬秋 2011中国组织工程研究与临床康复2011,15,10:11
16人脐带间充质干细胞经体内定植并向心肌样细胞分化的研究显示文摘目的探讨人脐带间充质干细胞(MSCs)移植到大鼠心肌后能否存活并向心肌样细胞分化,为心肌细胞的移植探索新的细胞来源。方法从人脐带华尔通氏胶分离、培养MSCs,检测人脐带来源的MSCs的细胞表面标记。用5-溴-2-脱氧尿苷(BrdU)标记人脐带间充质干细胞,标记细胞移植到梗塞大鼠心肌,观察移植后2周细胞能否存活,移植细胞能否向心肌样细胞分化,表达心肌细胞标记物troponinI及troponinT。结果人脐带间充质干细胞移植到心肌梗塞模型大鼠心肌后细胞能存活,并表达心肌细胞标记物troponinI及troponinT。结论人脐带间充质干细胞移植至体内能存活并分化为心肌样细胞,人脐带间充质干细胞可能作为心肌细胞移植的来源。何红燕 林晓波 应文娟 林丽敏 冯学永 王鸿武 马廉 2009中国输血杂志2009,22,3:11

18人脐带间充质干细胞在组织工程中的研究进展显示文摘人脐带间充质干细胞是一类具有自我更新、增殖和多向分化潜能的干细胞,具有来源广泛、易于采集、保存和运输、无异体排斥、避免伦理争议等诸多优点。流式细胞仪分析发现人脐带间充质干细胞高表达间质细胞标志(CD44、CD105)、整合素受体(CD29、CD49b、CD49c、CD51),不表达造血系标志(CD34、CD45)人白细胞抗原HLA-DR和内皮细胞标志CD31。人脐带间充质干细胞在体内外可以分化为骨细胞、软骨细胞、肝细胞、心肌细胞、骨骼肌细胞以及神经元细胞等。目前人脐带间充质干细胞在组织工程骨、人工血管以及基因治疗等临床应用研究中已逐渐深入,并已显示出广阔的应用前景。本文就人脐带间充质干细胞的生物学特性及其在组织工程中的研究作一综述。孙国栋 李志忠 2009现代生物医学进展2009,9,12:11
