| 1 | Relationship between expression of Smac and Survivin and apoptosis of primary hepatocellular carcinoma显示文摘BACKGROUND: T he second mitochondria-derived activator of caspase/direct inhibitor of apoptosis-binding protein with low pI (Smac/DIABLO) was recently identified as a protein that is released from mitochondria in response to apoptotic stimuli and promotes apoptosis by antagonizing inhibitor of apoptosis proteins. Furthermore, Smac/DIABLO plays an important regulatory role in the sensitization of cancer cells to both immune- and drug-induced apoptosis. However, little is known about the clinical significance of Smac/DIABLO in various cancers including hepatocellular carcinoma (HCC). This study was undertaken to investigate the expression of Smac and Survivin and their relationship with the apoptosis in primary HCC. METHODS: The expression of Smac and Survivin proteins was evaluated by immunohistochemistry. The mRNA expression of Smac and Survivin was evaluated by reverse transcription-polymerase chain reaction (RT-PCR) in HCC tissues of 50 patients, para-carcinoma tissues of 20 patients, and normal liver tissues of 15 patients. RESULTS: Smac mRNA was detected by RT-PCR in HCC tissues of 21 (42.0%) of the 50 patients, para-carcinoma tissues of 19 (95.0%) of the 20 patients, and normal liver tissues of 15 (100%) of the 15 patients. Survivin mRNA was found in HCC tissues of 46 of the 50 patients, para- carcinoma tissues of 2 of the 20 patients, and normal liver tissues of 0 of 15 patients. Immunohistochemistry revealed Smac protein in HCC tissues of 20 patients (40.0%), in para-carcinoma tissues of 18 patients (90.0%), and normal liver tissues of 15 patients (100.0%). The expression of Smac was significantly different in HCC tissues and non- HCC tissues. Survivin protein was found in HCC tissuesin 45 patients, para-carcinoma tissues in 2 patients, and normal liver tissues in none of the patients. The expression of Survivin was significantly different in HCC tissues and non-HCC tissues. | 2 | Efficacy of intramuscular matrine in the treatment of chronic hepatitis B显示文摘BACKGROUND: Hepatitis B virus (HBV) infection, a glo- bal public health problem, is the leading cause of cirrhosis and hepatocellular carcinoma (HCC) worldwide. There are more than 350 million HBV carriers in the world and up to one million die annually due to hepatitis B associated liv- er disease. So far no optimal treatment is available for pa- tients with chronic hepatitis B. In the paper we investigated the efficacy of intramuscular matrine in the treatment of chronic hepatitis B. METHODS: One hundred and twenty patients with chronic hepatitis B were randomly divided into matrine treatment group (n =60) and control group (n =60). The patients of the matrine group were given intramuscularly with matrine (an alkaloid extracted from a traditional Chinese herb Radix Sophorae Flavescentis by Guangzhou Ming Xing Pharmaceu cal Factory, Guangzhou, China) of 100 mg daily for 90 days in addition to conventional liver-protective drugs in- cluding glucurone, inosine, compound vitamin B and caryophyllin. The control group received conventional liv- er-protective drugs alone. Clinical manifestations and labo- ratory parameters including liver biochemistry and serum hepatitis B virus markers were monitored before and after treatment in the two groups. RESULTS: Significant differences were seen between the two groups in terms of improvement of clinical symptoms and signs, recovery of liver functions, and serum conver- sion from hepatitis Be antigen to HBe antibody and from positive to negative serum HBV DNA (P <0.05-0.01). The result of the matrine group was more marked than that of the control group. Serious side-effects were not observed except mild pain at the site of injection of matrine in a few patients. | 3 | Effect of Protein Kinase C on Proliferation and Apoptosis of T Lymphocytes in Idiopathic Thrombocytopenic Purpura Children显示文摘It is well-documented that T lymphocyte proliferation and apoptosis are abnormal in idiopathic thrombocytopenic purpura (ITP) children. However, the underlying regulation mechanisms especially in terms of signal transduction remain unknown. In this paper, we reported the changes of protein kinase C (PKC) activity in peripheral blood T lymphocytes and the effect of PKC on T lymphocyte proliferation and apoptosis. We demonstrated that in ITP children, the activator (PMA) and inhibitor (H-7) of PKC affected on T lymphocyte proliferation and apoptosis dramatically, but they altered little in healthy children. PKC activity was significantly enhanced in ITP children together with an increased expression of FasL on CD3+ T, CD4+ T and CD8+T cells, resulting in a positive correlation between PKC activity and the expression of FasL on T cells. While the PKC activity and the platelet count were negatively correlated. In the recent fifty years, there were many papers concerned with the equation written by Ljunggren, Nagell, Brown, Toyoizumi and Cohn. In 1986, ref. | 8 | Geochemical characteristics of organic matter-rich strata of lower Cambrian in Tarim Basin and its origin显示文摘The organic matter-rich strata of the Yurtusi Formation of Lower Cambrian are wildly spread and steady in the Tarim Basin, in which cherts are developed, companied with phos-phorite. Al/(Al+Fe+Mn) ratio and Si/(Si+Al+Fe) ratio of cherts range from 0.0023 to 0.0046 and 0.965 to 0.98, respectively, suggesting that cherts are formed in submarine hydrothermal activity and far away from terrestrial. Trace elements, such as As, Hg, Pb, Zn, Cu, Co, P, V, Ba, etc., are markedly rich in organic matter-rich sedimentary rocks of which Ba/Sr ratios are between 21.2 and 158.1. Compared with modern hydrothermal sediments, their Ba/Sr ratios are similar, it in-dicates that the organic matter-rich strata have the geochemical feature of submarine hydro-thermal sediments. The total content of rare earth elements (SREE) in cherts is from 8.81 to 56.682 mg/g, on average 31.41 mg/g and the SREE of cherts is between continental margin cherts and abyssal cherts. The LREE/HREE ratio of cherts varies from 1.01 to 3.56, which re-veals the characteristics of hydrothermal sediments. | 10 | The noble gas isotope geochemical composition of chert at the bottom of Cambrian in Tarim Basin,China显示文摘In the Tarim Basin,black shale series at the bottom of Cambrian is one of the important marine facies hydrocarbon source rocks. This research focuses on the analysis of the isotope of noble gas of 11 cherts. The R/Ra ratio of chert in the Keping area is 0.032―0.319,and 40Ar/36Ar is 338―430. In Quruqtagh the R/Ra ratio is 0.44―10.21,and 40Ar/36Ar is 360―765. The R/Ra ratio of chert increases with 40Ar/36Ar from the west to the east accordingly. They have evolved from the crust source area to the mantle source area in a direct proportion. Surplus argon 40ArE in chert is in direct proportion to the R/Ra ratio,indicating that it has the same origin of excess argon as in fluid and mantle source helium. Comparison of the R/Ra ratios between the west and the east shows that the chert in the eastern part formed from the activity system of the bottom hydrothermal venting driven by the mantle source,where the material and energy of crust and mantle had a strong interaction in exchange;whereas in the western part,chert deposited from the floating of hydrothermal plume undersea bottom,which is far away from the centre of activities of the hydrothermal fluid of ocean bottom. | 11 | Cu_2ZnSnS_4 thin films prepared by sulfurizing different multilayer metal precursors显示文摘Cu2ZnSnS4 (CZTS) thin films were successfully fabricated on glass substrates by sulfurizing Cu-Sn-Zn multilayer precursors, which were deposited by ion beam sputtering and RF magnetron sputtering, respectively. The structural, electrical and optical properties of the prepared films under various processing conditions were investigated in detail. Results showed that the as-deposited CZTS thin films with the precursors by both ion beam sputtering and RF magnetron sputtering have a composition near stoichiometric. The crystallization of the samples, however, has a strong dependence on the atomic percent of constituents of the prepared CZTS films. A single phase stannite-type structure CZTS with a large absorption coefficient of 104/cm in the visible range could be obtained after sulfurization at 520℃ for 2 h. The samples relative to the RF magnetron sputtering showed a low resistivity of 0.073 ?cm and band gap energy of about 1.53 eV. | 12 | Polarization and Apoptosis of T Cell Subsets in Idiopathic Thrombocytopenic Purpura显示文摘It is well-known that idiopathic thrombocytopenic purpura (ITP) is an acquired organ-specific autoimmune hemorrhagic disease and dysfunctional cellular immunity is considered important in the pathophysiology of ITP. However, polarization patterns and apoptosis profiles of T lymphocytes remain unclear. In this study, we investigated the polarization of T cell subsets, the expressions of apoptotic proteins Fas/FasL on the subsets and the level of anti-apoptotic gene bcl-2 and bax mRNA. It was demonstrated that the ratios of Th1/Th2 and Tc1/Tc2 in ITP children were increased obviously and that the average percentages were increased clearly for Th1 and Th2,but not for Tc1 and Tc2. In ITP children, the enhancing expressions were detected for FasL on Th1 and Tc1 and for Fas on Th2 and Tc2. With increasing level of bcl-2 mRNA and decreasing expression of bax mRNA in ITP children, the ratio of bcl-2/bax mRNA was improved obviously, which was positive correlated with the ratio of Th1/Th2. Taken together, our findings indicate that ITP is a Th1 predominant disease. This polarization pattern of T cell subsets might be related to the high ratio of bcl-2/bax mRNA and the abnormal expressions of Fas and FasL on T cell subsets. | 14 | Assessment of Heavy Metals in Grain Fields with Long-term Treatment of Agrochemicals and Organic Manure in Hailun City of Northeast China显示文摘Heavy metals were evaluated in the soils of grain fields in Hailun City Experimental Research Station--Chinese Academy of Sciences (CAS). Four categories of the soil samples were chosen from the fields after 19 years of treatment with chemicals, no fertilizer, organic manure, and organic manure combined with chemical fertilizer. The study revealed a prevalence of significantly high concentrations of Cu, Zn and Pb in organic manure combined with chemical fertilizer in the grain fields of black soils. The concentrations of Cu and Zn in no fertilizer treatment, chemical treatment, and organic treatment also exceeded the permissible limits. | 15 | Investigation of thermoluminescence in Li_2B_4O_7 phosphors doped with Cu, Ag and Mg显示文摘Li2B4O7 (LBO):Cu,Ag,Mg phosphors have been prepared by the sintering technique. The roles of the Ag and Mg dopants in the phosphors have been studied using the methods of thermoluminescence (TL) glow curves and TL 3D spectra. The results indicated that proper concentrations of Ag and Mg can enhance the TL of LBO:Cu. It was also indicated that the intensity of TL peak at 130℃ is reduced with the in- creasing Ag concentration, and enhanced with the increasing Mg concentration. | 17 | Significance of MD-2 and MD-2B expression in rat liver during acute cholangitis显示文摘AIM: To investigate the expression of myeloid differentiation protein-2 (MD-2), MD-2B (a splicing isoform of MD-2 that can block Toll-like receptor 4 (TLR4)/MD-2 LPS-mediated signal transduction) and TLR4 in the liver of acute cholangitis rats. METHODS: Male Sprague-Dawley rats (SPF level) were randomly divided into four groups: (A) sham-operated group; (B) simple common bile duct ligation group; (C)acute cholangitis group; and (D) acute cholangitis anti- TLR4 intervention group (n = 25 per group). Rat liver tissue samples were used to detect TLR4, MD-2 and MD-2B mRNA expression by fluorescence quantitative PCR in parallel with pathological changes. RESULTS: In acute cholangitis, liver TLR4 and MD-2 mRNA expression levels at 6, 12, 24, 48 and 72 h were gradually up-regulated but MD-2B mRNA expression gradually down-regulated (P < 0.05). After TLR4 antibody treatment, TLR4 and MD-2 mRNA expression were lower compared with the acute cholangitis group (P < 0.05). However, MD-2B mRNA expression was higher than in the acute cholangitis group (P < 0.05). MD-2 and TLR4 mRNA expressions were positively correlated (r = 0.94981, P < 0.05) and MD-2B mRNA expression was negatively correlated with MD-2 and TLR4 mRNA (r = -0.89031, -0.88997, P< 0.05). Theplay conditions of Kela-2 field and Ya21-l structure featuresin the Qiongdongnan Basin on the South China Sea shelf hasbeen studied, and a gas accumulation under deep overpres-sure must have the following two conditions (besides otherfavourable conditions for gas accumulation): one is that thetime of gas charge matches with overpressure occurrencetime, and the other is a good quality caprock.
