3Identification of Ca2+ signaling components in neural stem/progenitor cells during differentiation into neurons and glia in intact and dissociated zebrafish neurospheres显示文摘The development of the CNS in vertebrate embryos involves the generation of different sub-types of neurons and glia in a complex but highly-ordered spatio-temporal manner. Zebrafish are commonly used for exploring the development, plasticity and regeneration of the CNS, and the recent development of reliable protocols for isolating and culturing neural stem/progenitor cells(NSCs/NPCs) from the brain of adult fish now enables the exploration of mechanisms underlying the induction/specification/differentiation of these cells. Here, we refined a protocol to generate proliferating and differentiating neurospheres from the entire brain of adult zebrafish. We demonstrated via RT-qPCR that some isoforms of ip3 r, ryr and stim are upregulated/downregulated significantly in differentiating neurospheres, and via immunolabelling that 1,4,5-inositol trisphosphate receptor(IP3 R) type-1 and ryanodine receptor(RyR) type-2 are differentially expressed in cells with neuron-or radial glial-like properties. Furthermore, ATP but not caffeine(IP3 R and RyR agonists, respectively), induced the generation of Ca^(2+) transients in cells exhibiting neuron-or glial-like morphology. These results indicate the differential expression of components of the Ca^(2+) -signaling toolkit in proliferating and differentiating cells. 4Nutrient supplemented serum-free medium increases cardiomyogenesis efficiency of human pluripotent stem cells显示文摘AIM: To development of an improved p38 MAPK inhibitor-based serum-free medium for embryoid body cardiomyocyte differentiation of human pluripotent stem cells. METHODS: Human embryonic stem cells (hESC) differentiated to cardiomyocytes (CM) using a p38 MAPK inhibitor (SB203580) based serum-free medium (SB media). Nutrient supplements known to increase cell viability were added to SB medium. The ability of these supplements to improve cardiomyogenesis was evaluated by measurements of cell viability, total cell count, and the expression of cardiac markers via flow cytometry. An improved medium containing Soy hydrolysate (HySoy) and bovine serum albumin (BSA) (SupSB media) was developed and tested on 2 additional cell lines (H1 and Siu-hiPSC). Characterization of the cardiomyocytes was done by immunohistochemistry, electrophysiology and quantitative real-time reverse transcriptionpolymerase chain reaction. RESULTS: hESC cell line, HES-3, differentiating in SB medium for 16 d resulted in a cardiomyocyte yield of 0.07 ± 0.03 CM/hESC. A new medium (SupSB media) was developed with the addition of HySoy and BSA to SB medium. This medium resulted in 2.6 fold increase in cardiomyocyte yield (0.21 ± 0.08 CM/hESC). The robustness of SupSB medium was further demonstrated using two additional pluripotent cell lines (H1, hESC and Siu1, hiPSC), showing a 15 and 9 fold increase in cardiomyocyte yield respectively. The age (passage number) of the pluripotent cells did not affect the cardiomyocyte yields. Embryoid body (EB) cardiomyocytes formed in SupSB medium expressed canonical cardiac markers (sarcomeric α-actinin, myosin heavy chain and troponin-T) and demonstrated all three major phenotypes: nodal-, atrial- and ventricular-like. Electrophysiological characteristics (maximum diastolic potentials and action potential durations) of cardiomyocytes derived from SB and SupSB media were similar. 5Health Emergency and Disaster Risk Management (Health-EDRM):Developing the Research Field within the Sendai Framework Paradigm显示文摘The intersection of health and disaster risk reduction(DRR) has emerged in recent years as a field of critical inquiry.Health is recognized as an outcome and a goal of DRR,and the integration of both fields is essential to ensure the implementation of the Sendai Framework for Disaster Risk Reduction 2015–2030.Health Emergency and Disaster Risk Management(Health-EDRM) has emerged as an umbrella field that encompasses emergency and disaster medicine,DRR,humanitarian response,community health resilience,and health systems resilience.In September 2016,an international group of experts met in Hong Kong to assess the current status and potential of the Health-EDRM research field,a research area that these scholars characterized as underdeveloped and fragmented.Key challenges identified include research overlap,lack ofstrategic research agenda,absence of consensus regarding terminology,and limited coordination between stakeholders.The Sendai Framework provides a useful paradigm within which to shape the research field's strategic development.The WHO Thematic Platform for Health-EDRM Research Group was established to coordinate activities,promote information-sharing,develop partnerships,and provide technical advice to strengthen the Health-EDRM research field.This group will promote the generation of robust and scientific health research to support the meaningful implementation of the Sendai Framework.Sharon Tsoon Ting Lo Emily Ying Yang Chan Gloria Kwong Wai Chan Virginia Murray Jonathan Abrahams Ali Ardalan Ryoma Kayano Johnny Chung Wai Yau 2017International Journal of Disaster Risk Science2017,8,2:3
6Ethyl Acetate Fraction of Dicliptera chinensis(L.)Juss.Ameliorates Liver Fibrosis by Inducing Autophagy via PI3K/AKT/m TOR/p70S6K Signaling Pathways显示文摘Objective:To investigate the molecular mechanism underlying the anti-hepatic fibrosis activity of ethyl acetate fraction Dicliptera chinensis(L.)Juss.(EDC)in human hepatic stellate cells(HSCs)in vitro and in a carbon tetrachloride(CCl4)-induced hepatic fibrosis mouse model in vivo.Methods:For in vitro study,HSCs were pre-treated with platelet-derived growth factor(10 ng/mL)for 2 h to ensure activation and treated with EDC for 24 h and 48 h,respectively.The effect of EDC on HSCs was assessed using cell counting kit-8 assay,EdU staining,transmission electron microscopy,immunofluorescence staining,and Western blot,respectively.For in vivo experiments,mice were intraperitoneally injected with CCl4(2μL/g,adjusted to a 25%concentration in olive oil),3 times per week for 6 weeks,to develop a hepatic fibrosis model.Forty 8-week-old male C57 BL/6 mice were divided into 4 groups using a random number table(n=10),including control,model,positive control and EDC treatment groups.Mice in the EDC and colchicine groups were intragastrically administered EDC(0.5 g/kg)or colchicine(0.2 mg/kg)once per day for 6 weeks.Mice in the control and model groups received an equal volume of saline.Biochemical assays and histological examinations were used to assess liver damage.Protein expression levels ofα-smooth muscle actin(α-SMA)and microtubule-associated protein light chain 3 B(LC3 B)were measured by Western blot.Results:EDC reduced pathological damage associated with liver fibrosis,downregulated the expression ofα-SMA and upregulated the expression of LC3 B(P<0.05),both in HSCs and the CCl4-induced liver fibrosis mouse model.The intervention of bafilomycin A1 and rapamycin in HSCs strongly supported the notion that inhibition of autophagy enhancedα-SMA protein expression levels(P<0.01).The results also found that the levels of phosphoinositide(PI3 K),p-PI3 K,AKT,p-AKT,mammalian target of rapamycin(mTOR),p-mTOR,and p-p70 S6 K all decreased after EDC treatment(P<0.05).Conclusion:EDC has anti-hepatic fibrosis activity by inducing autophagy and might be a potential drug to be further developed for human liver fibrosis therapy.LIU Yuan BI Yan-meng PAN Ting ZENG Ting MO Chan SUN Bing GAO Lei LYU Zhi-ping 2022Chinese Journal of Integrative Medicine2022,28,1:3
7Ion channels/transporters as epigenetic regulators?——a microRNA perspective显示文摘MicroRNA (miRNA) alterations in response to changes in an extracellular microenvironment have been observed and considered as one of the major mechanisms for epigenetic modifications of the cell. While enormous efforts have been made in the understanding of the role of miRNAs in regulating cellular responses to the microenvironment, the mechanistic insight into how extracellular signals can be transduced into miRNA alterations in cells is still lacking. Interestingly, recent studies have shown that ion channels/transporters, which are known to conduct or transport ions across the cell membrane, also exhibit changes in levels of expression and activities in response to changes of extracellular microenvironment. More importantly, alterations in expression and function of ion channels/transporters have been shown to result in changes in miRNAs that are known to change in response to alteration of the microenvironment. In this review, we aim to summarize the recent data demonstrating the ability of ion channels/transporters to transduce extracellular signals into miRNA changes and propose a potential link between cells and their microenvironment through ion channels/transporters. 10Longitudinal Measurement of Hemodynamic Changes within the Posterior Optic Nerve Head in Rodent Nonarteritic Anterior Ischemic Optic Neuropathy显示文摘ObjectiveTo assess the in vivo dynamic blood flow features of posterior optic nerve head (ONH) in rat model of nonarteritic anterior ischemic optic neuropathy (rNAION). MethodsrNAION was established with Rose Bengal and argon green laser in Sprague-Dawley rats. Fundus photography and fundus fluorescein angiography (FFA) were performed to assess the dynamic changes of optic disc in morphology in 90 days and in blood perfusion in 3 hours after the induction of disease. Histological examinations were performed to evaluate the success of modeling. Thedynamic blood flow kinetics of posterior ONH in rNAION were measured by Laser Doppler Flowmetry (LDF) on the day 3, 7, 14, 21, and 40 after the disease induction. One-way ANOVA, Student'st-test and Bonferroni adjustment were used for multiple comparisons of kinetic measurements of blood flow. ResultsOptic disc edema and subsequent resolution associated with the development of optic disc pallor were observed in rNAION. FFA showed that the optic disc was hypofluorescence in the early phase and hyperfluorescence in the late phase. Histological studies suggested edema and loosened tissues of ONH, loss of retinal ganglion cells (RGCs), optic nerve substance and gliosis. Compared to the naive rats, the blood flow kinetics of posterior ONH in rNAION significant reduced at each time point after modeling (F=175.06,P<0.0001). The reductions were specifically remarkable in 14 days after the disease induction (AllP<0.01). Conclusions Continuous blood perfusion reduction was found in rNAION, with significant alteration in 14 days after disease induction. 14Pulse oximeter as a home assessment tool:Knowledge and user experience among the community in Malaysia during the COVID-19 pandemic显示文摘Objective:To determine the degree of knowledge in the usage of pulse oximeter as a home assessment tool among the community in Malaysia.Methods:A cross-sectional survey was conducted in November 2021.The questionnaire assessed the knowledge in using pulse oximeters,user experience and opinions in using pulse oximeter as a home assessment tool during the pandemic.Results:A total of 504 respondents were included in the study,and the mean score in knowledge related to application of pulse oximetry was 73.00%,while the mean score in knowledge related to factors affecting pulse oximetry readings was only 38.51%.A total of 90.5% of the respondents recognised normal pulse rate and 88.5% knew the blood oxygen saturation levels of a healthy adult,while 69.4% recognised the definition of silent hypoxia.In addition,the majority of the respondents agreed that factors such as poor blood circulation(71.2%),excessive movements(69.8%),and hand position(60.7%)affected oximetry readings.However,61.7%,81.7%,77.2% and 76.8% of the respondents could not identify nail polish,skin colour,skin thickness and tattoos as factors that may affect oximetry readings respectively.Conclusions:The respondents showed a satisfactory level of knowledge related to application of pulse oximetry,but a poor level of knowledge related to factors affecting pulse oximetry readings among the community in Malaysia.Continuous efforts in educating the community on the correct use of pulse oximeters are crucial for appropriate home assessment and avoiding unnecessary stress.Siew-Keah Lee Jin Wei Ma Khai Ying Chan Yi Song Cheong Wan Ting Hong Regine Chew Yi Ong Wei Cheng Tay Ang-Lim Chua 2022Asian Pacific Journal of Tropical Medicine2022,15,5:1
