| 1 | Mnk kinase pathway: Cellular functions and biological outcomes显示文摘The mitogen-activated protein kinase(MAPK) interacting protein kinases 1 and 2(Mnk1 and Mnk2) play important roles in controlling signals involved in mRNA translation. In addition to the MAPKs(p38 or Erk), multiple studies suggest that the Mnk kinases can be regulated by other known kinases such as Pak2 and/or other unidentified kinases by phosphorylation of residues distinct from the sites phosphorylated by the MAPKs. Several studies have established multiple Mnk protein targets, including PSF, heterogenous nuclear ribonucleoprotein A1, Sprouty 2 and have lead to the identification of distinct biological functions and substrate specificity for the Mnk kinases. In this review we discuss the pathways regulating the Mnk kinases, their known substrates as well as the functional consequences of engagement of pathways controlled by Mnk kinases. These kinases play an important role in mRNA translation via their regulation of eukaryotic initiation factor 4E(eIF4E) and their functions have important implications in tumor biology as well as the regulation of drug resistance to anti-oncogenic therapies. Other studies have identified a role for the Mnk kinases in cap-independent mRNA translation, suggesting that the Mnk kinases can exert important functional effects independently of the phosphorylation of eIF4 E. | 2 | Plasma chitinase 3-like 1 is persistently elevated during first month after minimally invasive colorectal cancer resection显示文摘AIM: To assess blood chitinase 3-like 1(CHi3L1) levels for 2 mo after minimally invasive colorectal resection(MICR) for colorectal cancer(CRC). METHODS: CRC patients in an Institutional Review Board approved data/plasma bank who underwent elective MICR for whom preoperative(PreO p), early postoperative(PostO p), and 1 or more late PostO p samples [postoperative day(POD) 7-27] available were included. Plasma CHi3L1 levels(ng/m L) were determined in duplicate by enzyme linked immunosorbent assay. RESULTS: PreOp and PostOp plasma sample were available for 80 MICR cancer patients for the study. The median PreOp CHi3L1 level was 56.8 CI: 41.9-78.6 ng/mL(n = 80). Significantly elevated(P < 0.001) median plasma levels(ng/mL) over PreOp levels were detected on POD1(667.7 CI: 495.7, 771.7; n = 79), POD 3(132.6 CI: 95.5, 173.7; n = 76), POD7-13(96.4 CI: 67.7, 136.9; n = 62), POD14-20(101.4 CI: 80.7, 287.4; n = 22), and POD 21-27(98.1 CI: 66.8, 137.4; n = 20, P = 0.001). No significant difference in plasma levels were noted on POD27-41. CONCLUSION: Plasma CHi3L1 levels were significantly elevated for one month after MICR. | 3 | Thrombocytopenia in cirrhosis:Impact of fibrinogen on bleeding risk显示文摘AIM To investigate the relationship between baseline platelet count,clauss fibrinogen,maximum amplitude(MA) on thromboelastography,and blood loss in orthotopic liver transplantation(OLT).METHODS A retrospective analysis of our OLT Database(2006-2015) was performed.Baseline haematological indices and intraoperative blood transfusion requirements,as a combination of cell salvage return and estimation of 300 mls/unit of allogenic blood,was noted as a surrogate for intraoperative bleeding.Two groups:Excessive transfusion(>1200 mL returned) and No excessive transfusion(<1200 m L returned) were analysed.All data analyses were conducted using IBM SPSS Statistics version 23.RESULTS Of 322 OLT patients,77 were excluded due to fulminant disease;redo transplant or baseline haemoglobin(Hb) of<80 g/L.One hundred and fourteen(46.3%) were classified into the excessive transfusion group,132(53.7%) in the no excessive transfusion group.Mean age and gender distribution were similar in both groups.Baseline Hb(P≤0.001),platelet count(P=0.005),clauss fibrinogen(P=0.004) and heparinase MA(P=0.001) were all statistically significantly different.Univariate logistic regression with a cut-off of platelets<50×10~9/L as the predictor and Haemorrhage as the outcome showed an odds ratio of 1.393(95%CI:0.758-2.563;P=0.286).Review of receiver operating characteristic curves showed an area under the curve(AUC) for platelet count of 0.604(95%CI:0.534-0.675;P=0.005) as compared with AUC for fibrinogen level,0.678(95%CI:0.612-0.744;P≤0.001).A multivariate logistic regression shows United Kingdom model for End Stage Liver Disease(P=0.006),Hb(P=0.022) and Fibrinogen(P=0.026) to be statistically significant,whereas Platelet count was not statistically significant.CONCLUSION Platelet count alone does not predict excessive transfusion.Additional investigations,e.g.,clauss fibrinogen and viscoelastic tests,provide more robust assessment of bleeding-risk in thrombocytopenia and cirrhosis. | Sonali V Thakrar Susan V Mallett | 2017 | World Journal of Hepatology2017,9,6: | 4 |
| 4 | Dysfunctional stem and progenitor cells impair fracture healing with age显示文摘Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. The power conversion efficiency(PCE) of perovskite solar cells(PSCs) is currently reaching parity(>25 percent), an accomplishment attained over past decades. PSCs are seen as perovskites sandwiched between an electron transporting material(ETM) and a hole transporting material(HTM). As a primary component of PSCs, HTM has been shown to have a considerable effect on solar energy harvesting, carrier extraction and transport, crystallization of perovskite, stability, and price. In PSCs, it is still necessary to use a HTM.While perovskites are capable of conducting holes, they are present in trace amounts, necessitating the use of an HTM layer for efficient charge extraction. In this review, we provide an understanding of the significant forms of HTM accessible(inorganic, polymeric and small molecule-based HTMs), to motivate further research and development of such materials. | 6 | Discovery and preclinical development of ⅢM-160, a Bergenia ciliatabased anti-inflammatory and anti-arthritic botanical drug candidate显示文摘Objective: Bergenia ciliata(Haw.) Sternb. is used in the Indian traditional system of medicine to treat various ailments including rheumatism and to heal wounds. The objective of the present study was to perform a preclinical characterization of the B. ciliata-based botanical extract IIIM-160.Methods: ⅢM-160 was chemically standardized and analyzed for heavy metal content, aflatoxins,pesticides and microbial load. The in vitro and in vivo efficacies were determined in suitable models of inflammation, arthritis and nociception. An acute oral toxicity study was performed in Swiss albino mice.A suitable oral formulation was developed and characterized.Results: Bergenin was found to be the major component(9.1% w/w) of ⅢM-160. The botanical lead displayed inhibition of lipopolysaccharide-induced production of proinflammatory cytokines in THP-1 cells, with selectivity toward interleukin-6(IL-6) and had an excellent safety-window. It showed anti-inflammatory, anti-arthritic and antinociceptive activity in animal models and was not toxic at oral doses up to 2 g/kg in Swiss-albino mice. The gastroretentive, sustained-release capsule formulation showed sustained-release of the bergenin over the period of 24 h, resulting in improved plasma-exposure of bergenin in Sprague–Dawley rats.Conclusion: The dual-activity of IL-6 inhibition and antinociception marks the suitability of ⅢM-160 for treating rheumatoid arthritis. | 9 | Validity of subjective assessment as screening tool for dry eye disease and its association with clinical tests显示文摘AIM: To determine the role of subjective assessment using Mc Monnies dry eye questionnaire in diagnosing dry eye disease and its association with clinical tests.METHODS: There were 500 patients screened for dry eye using Mc Monnies dry eye questionnaire between May to October 2013 at the outpatient Department of Ophthalmology of a medical college hospital. All 500 patients were subjected to clinical tests. Dry eye was defined as having one or more symptoms often or all the time. Positive signs were if one or both eyes revealed tear film breakup time(TBUT) of ≤10s, a Schirmer test score of ≤10 mm, a Rose Bengal staining score of ≥1, a Lissamine green staining score of ≥1 or existence of meibomian gland disease(≥grade 1). Statistical analysis was performed to describe the distribution of symptoms and signs, to assess the correlations between Mc Monnies score(MS) and variable clinical signs of dry eye, and to explore the association between dry eye symptoms and variable clinical signs. Analysis was performed using software package Epi info. A Probability(P) value using Chi-square test of <0.005 was taken as significant.· RESULTS: Dry eye prevalence with symptoms(questionnaire), Schirmer test, TBUT, Rose Bengal staining and Lissamine green staining was 25.6%,15.20%, 20.80%, 23.60%, and 22.60% respectively. Among those with severe symptoms(MS>20), 75.86% had a low TBUT(<10s), 58.62% had a low Schirmer's I test(≤10 mm),86.20% had Rose Bengal staining score of ≥1, 79.31%had Lissamine green staining score of ≥1. We found statistically significant associations between positive Schirmer test and arthritis(P <0.002), dryness elsewhere(P <0.001), contact lens use(P < 0.002), systemic medication(P <0.0001), sleeping with eyes partly open(P <0.002), history of dry eyes treatment(P <0.0001),environmental factors(P <0.001), swimming(P <0.001).· CONCLUSION: Subjective assessment plays an important role in diagnosing dry eye disease. | 10 | River Rafting in Mountainous Regions of Uttarakhand:Impacts,Suggested Mitigation Measures and Sustainability显示文摘Uttarakhand state in India is well known for its mountainous ecosystems,traditional communities and a variety of ecotourism destinations.Among various tourism activities,River Rafting along the banks of the Ganges River has increased tremendously for two decades,and has had unprecedented impacts on the traditional communities and the bio-diversity.Therefore,it is meaningful to do a comprehensive study on the various impacts associated with river rafting so as to suggest the pathways to achieve the environmental sustainability in this region.In this study,we collected primary data from randomly selected population units across all stake holders such as local people(n = 100),and camp personnel(n = 22),through a pre-tested questionnaire survey between August 2009 and May 2010.The questionnaire contains issues on culture,social,economic,institutional and associated perceived impacts on pollution and biodiversity including views for sustainability.Secondary information was also collected from various sources and government records to supplement and strengthen the analysis.The impacts were analyzed qualitatively through a ranking mechanism to facilitate the decision making process.The perception of the interviewee about the various possible impacts of rafting was discussed with mitigating mechanism.The ranking analysis as percollected data reveals that economy and education of local community was improved significantly;however aquatic and terrestrial fauna,social cohesion and pollution(water,air,land) were significantly deteriorated.The results show that the existing practices are not sufficient to address the adverse impacts.Improvement in practices is necessary,mainly in the policy regime.Based on the analysis,some measures are recommended on how to protect community interest and environment with the development of river rafting as an ecotourism opportunity. | Parth Sarathi MAHAPATRA Rajiv PANDEY Sonali PRADHAN | 2012 | Journal of Mountain Science2012,9,4: | 2 |
| 11 | Flood susceptibility modelling using advanced ensemble machine learning models显示文摘Floods are one of nature's most destructive disasters because of the immense damage to land,buildings,and human fatalities.It is difficult to forecast the areas that are vulnerable to flash flooding due to the dynamic and complex nature of the flash floods.Therefore,earlier identification of flash flood susceptible sites can be performed using advanced machine learning models for managing flood disasters.In this study,we applied and assessed two new hybrid ensemble models,namely Dagging and Random Subspace(RS)coupled with Artificial Neural Network(ANN),Random Forest(RF),and Support Vector Machine(SVM)which are the other three state-of-the-art machine learning models for modelling flood susceptibility maps at the Teesta River basin,the northern region of Bangladesh.The application of these models includes twelve flood influencing factors with 413 current and former flooding points,which were transferred in a GIS environment.The information gain ratio,the multicollinearity diagnostics tests were employed to determine the association between the occurrences and flood influential factors.For the validation and the comparison of these models,for the ability to predict the statistical appraisal measures such as Freidman,Wilcoxon signed-rank,and t-paired tests and Receiver Operating Characteristic Curve(ROC)were employed.The value of the Area Under the Curve(AUC)of ROC was above 0.80 for all models.For flood susceptibility modelling,the Dagging model performs superior,followed by RF,the ANN,the SVM,and the RS,then the several benchmark models.The approach and solution-oriented outcomes outlined in this paper will assist state and local authorities as well as policy makers in reducing flood-related threats and will also assist in the implementation of effective mitigation strategies to mitigate future damage. | Abu Reza Md Towfiqul Islam Swapan Talukdar Susanta Mahato Sonali Kundu Kutub Uddin Eibek Quoc Bao Pham Alban Kuriqi Nguyen Thi Thuy Linh | 2021 | Geoscience Frontiers2021,12,3: | 2 |
| 12 | In vivo antianxiety and antidepressant activity of Murraya paniculata leaf extracts显示文摘OBJECTIVE: Different parts of Murraya paniculata have been used traditionally for treating several ailments including mental disorders. The present study was designed to evaluate the antianxiety and antidepressant potential of M. paniculata leaves using elevated plus maze model and forced swim test, respectively.METHODS: Extracts of M. paniculata made with petroleum ether(60–80 °C), chloroform, ethanol and water were evaluated for antianxiety and antidepressant activity. The anxiolytic chloroform extract was subjected to column chromatography, yielding five fractions(F_1–F_5). Fraction F_5(100 mg/kg), which showed notable anxiolytic activity, was further chromatographed to get four subfractions(F_(5.1)–F_(5.4)). Simultaneously, the ethanol extract was partitioned with ethyl acetate to obtain ethyl acetate soluble fraction(EASF) and ethyl acetate insoluble fraction. Phytochemical screening of bioactive extracts/fractions and detection of mahanimbine in M. paniculata leaf extract by thin-layer chromatography was also carried out.RESULTS: Fraction F_(5.3)(25 mg/kg) and EASF(20 mg/kg) showed significant anxiolytic and antidepressant activity, respectively. Thin-layer chromatography and phytochemical screening demonstrated the absence of mahanimbine in M. paniculata leaves.
