1Charlson comorbidity index helps predict the risk of mortality for patients with type 2 diabetic nephropathy显示文摘研究目的:探讨Charlson合并症指数(CCI)对2型糖尿病肾病死亡率的预测作用。研究方法:建立2型糖尿病肾病研究队列,根据CCI评分将患者合并症的严重程度分为三度:轻度(CCI1–2分)、中度(CCI 3–4分)、重度(CCI≥5分)。将影响死亡率的因素及按CCI分层后的组间差异指标进行Logistic回归分析及方差分析(ANOVA)。Kaplan-Meier生存曲线分析CCI指数对生存时间及死亡率的影响。重要结论:533例2型糖尿病肾病患者纳入研究。所有患者的CCI评分均大于1,44.7%(238/533)的患者死亡。患者的死亡率随CCI评分增加而增加,CCI 1–2分患者的死亡率为21.0%(50/238),CCI 3–4分患者的死亡率为56.7%(135/238),CCI≥5分患者的死亡率为22.3%(53/238)。Logistic回归分析显示CCI评分、血红蛋白和血浆白蛋白水平是患者死亡率的预测因子(P<0.05)。方差分析结果显示,与CCI评分相对较低的患者相比,CCI评分越高的患者其血红蛋白水平较低,而血清肌酐较高,死亡率也更高。Kaplan-Meier生存曲线显示CCI评分越高的患者生存时间越短。CCI评分是一种简单易行且实用的评估疾病合并症的方法,可用于预测2型糖尿病肾病患者的死亡率。关注2型糖尿病肾病患者的合并症将有利于早期、有效治疗及改善预后。You-qun HUANG Rong GOU Yong-shu DIAO Qing-hua YIN Wen-xing FAN Ya-ping LIANG Yi CHEN Min WU Li ZANG Ling LI Jing ZANG Lu CHENG Ping FU Fang LIU 2014Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2014,15,1:23

4Effect of high glucose levels on the calcification of vascular smooth muscle cells by inducing osteoblastic differentiation and intracellular calcium deposition via BMP-2/Cbfα-1 pathway显示文摘In this paper,we investigate the effect and the possible mechanism of high glucose levels on the calcification of human aortic smooth muscle cells (HASMCs).HASMCs were divided into four groups: normal glucose group (NG),osmolality control group (OC),high glucose group (HG,HASMCs culture medium containing 30 mmol/L glucose),and high glucose plus recombinant human Noggin protein (bone morphogenetic protein-2 (BMP-2) antagonist) group (HN).The mRNA levels and the protein expressions of BMP-2 and core binding factor alpha-1 (Cbfα-1) were measured by real-time quantitative polymerase chain reaction (PCR) and Western blot.After induced by 10 mmol/L β-glycerol phosphoric acid,cells were harvested for assessments of alkaline phosphatase (ALP) activities at Days 1,2,and 3,and intracellular calcium contents at Days 7 and 14,respectively.High glucose levels increased the mRNA levels and the protein expressions of BMP-2 and Cbfα-1 (P<0.05).The expression of Cbfα-1 was partially blocked by Noggin protein (P<0.05),while BMP-2 was not (P>0.05).After being induced by β-glycerol phosphoric acid,high glucose levels increased the ALP activity [(48.63±1.03) vs.(41.42±2.28) U/mg protein,Day 3;P<0.05] and the intracellular calcium content [(2.76±0.09) vs.(1.75±0.07) μmol/mg protein,Day 14;P<0.05] in a time-dependent manner when compared with the NG group,while the ALP activity could not be blocked by Noggin protein [(48.63±1.03) vs.(47.37±0.97) U/mg protein,Day 3;P>0.05].These results show that high glucose levels can evoke the calcification of HASMCs by inducing osteoblastic trans-differentiation and intracellular calcium deposition via the BMP-2/Cbfα-1 pathway,which can be partially blocked by Noggin protein.Fang LIU Hui ZHONG Jing-yuan LIANG Ping FU Zhi-juan LUO Li ZHOU Rong GOU Jun HUANG 2010Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,12:12

7Analysis of reconstructed annual precipitation from tree-rings for the past 500 years in the middle Qilian Mountain显示文摘The ring-width chronology of a Juniperus przewalskii tree from the middle of the Qilian Mountain was constructed to estimate the annual precipitation (from previous August to current July) since AD 1480.The reconstruction showed four major alternations of drying and wetting over the past 521 years.The rainy 16th century was followed by persistent drought in the 17th century.Moreover,relatively wet conditions persisted from the 18th to the beginning of 20th century until the recurrence of a drought during the 1920s and 1930s.Based on the Empirical Mode Decomposition method,eight Intrinsic Mode Functions (IMFs) were extracted,each representing unique fluctuations of the reconstructed precipitation in the time-frequency domain.The high amplitudes of IMFs on different timescales were often consistent with the high amount of precipitation,and vice versa.The IMF of the lowest frequency indicated that the precipitation has undergone a slow increasing trend over the past 521 years.The 2-3 year and 5-8 year time-scales reflected the characteristics of inter-annual variability in precipitation relevant to regional atmospheric circulation and the El Ni?o-Southern Oscillation (ENSO),respectively.The 10-13 year scale of IMF may be associated with changing solar activity.Specifically,an amalgamation of previous and present data showed that droughts were likely to be a historically persistent feature of the Earth's climate,whereas the probability of intensified rainfall events seemed to increase during the course of the 19th and 20th centuries.These changing characteristics in precipitation indicate an unprecedented alteration of the hydrological cycle,with unknown future amplitude.Our reconstruction complements existing information on past precipitation changes in the Qilian Mountain,and provides additional low-frequency information not previously available.TIAN QinHua ZHOU XiuJi GOU XiaoHu ZHAO Ping FAN ZeXin Samuli HELAMA 2012Science China Earth Sciences2012,55,5:7
8Establishment of a transgenic mouse model with liver-specific expression of secretory immunoglobulin D显示文摘Mutation of mevalonate kinase(MVK) is thought to account for most cases of hyperimmunoglobulinemia D syndrome(HIDS) with recurrent fever.However,its mechanism and the relationship between elevated serum immunoglobulin D(IgD) and the clinical features of HIDS are unclear.In this study,we generated by fusion PCR a vector to express high levels of chimeric secretory IgD(csIgD) specifically in the liver.We then generated seven founder lines of transgenic mice by co-microinjection,and verified them using genomic PCR and Southern blotting.We detected the expression of csIgD by reverse transcription PCR,quantitative PCR,western blotting,and enzyme-linked immunosorbent assays.We demonstrated that csIgD could be specifically and stably expressed in the liver.We used flow cytometry to show that overexpression of csIgD in the bone marrow and spleen cells had no effect on B cell development.Morphologic and anatomical observation of the transgenic mice revealed skin damage,hepatosplenomegaly,and nephromegaly in some transgenic mice;in these mice,pathological sections showed high levels of cell necrosis and protein-like sediments in the liver,spleen,and kidney.We demonstrated that the genomic insertion sites of the transgenes did not disrupt the MVK gene on mouse chromosome 5.This transgenic mouse will be useful to explore the pathogenesis of HIDS.WANG Ping WEI ZhiGuo YAN BoWen HUANG Tan GOU KeMian DAI YunPing ZHENG Min WANG MeiLi CHENG XueQian WANG XiFeng XU Chen SUN Yi 2012Science China(Life Sciences)2012,55,3:6
9Formation Mechanism and Binding Energy for Equilateral Triangle Structure of He3^+ Cluster显示文摘The formation mechanism for the equilateral triangle structure of the He3+ cluster is proposed. The curveof the total energy versus the internuclear distance R for this structure has been calculated by the method of a modifiedarrangement channel quantum mechanics. The result shows that the curve has a minimal -7.81373 a.u at 1 = 1.55 a0.The binding energy of He3+ with respect to He+He+ +He was calculated to be 0.1064 a.u. (about 2.89 eV). 10SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19.Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan 2021Cell Research2021,31,12:6
11Generation of electricity from CO_2 mineralization: Principle and realization显示文摘Current CO2 reduction and utilization technologies suffer from high energy consuming. Thus, an energy favourable route is in urgent demanding. CO2 mineralization is theoretically an energy releasing process for CO2 reduction and utilization, but an approach to recovery this energy has so far remained elusive. For the first time, here we proposed the principle of harvesting electrical energy directly from CO2 mineralization, and realized an energy output strategy for CO2 utilization and reduction via a CO2-mineralization fuel cell(CMFC) system. In this system CO2 and industrial alkaline wastes were used as feedstock, and industrial valuable Na HCO3 was produced concomitantly during the electricity generation. The highest power density of this system reached 5.5 W/m2, higher than many microbial fuel cells. The maximum open circuit voltage reached 0.452 V. Moreover, this system was demonstrated viable to low concentration CO2(10%) and other carbonation process. 15Application of regional citrate anticoagulation in patients at high risk of bleeding during intermittent hemodialysis:a prospective multicenter randomized controlled trial显示文摘Objective:Safe and effective anticoagulation is essential for hemodialysis patients who are at high risk of bleeding.The purpose of this trial is to evaluate the effectiveness and safety of two-stage regional citrate anticoagulation(RCA)combined with sequential anticoagulation and standard calcium-containing dialysate in intermittent hemodialysis(IHD)treatment.Methods:Patients at high risk of bleeding who underwent IHD from September 2019 to May 2021 were prospectively enrolled in 13 blood purification centers of nephrology departments,and were randomly divided into RCA group and saline flushing group.In the RCA group,0.04 g/mL sodium citrate was infused from the start of the dialysis line during blood draining and at the venous expansion chamber.The sodium citrate was stopped after 3 h of dialysis,which was changed to sequential dialysis without anticoagulant.The hazard ratios for coagulation were according to baseline.Results:A total of 159 patients and 208 sessions were enrolled,including RCA group(80 patients,110 sessions)and saline flushing group(79 patients,98 sessions).The incidence of severe coagulation events of extracorporeal circulation in the RCA group was significantly lower than that in the saline flushing group(3.64%vs.20.41%,P<0.001).The survival time of the filter pipeline in the RCA group was significantly longer than that in the saline flushing group((238.34±9.33)min vs.(221.73±34.10)min,P<0.001).The urea clearance index(Kt/V)in the RCA group was similar to that in the saline flushing group with no statistically significant difference(1.12±0.34 vs.1.08±0.34,P=0.41).Conclusions:Compared with saline flushing,the two-stage RCA combined with a sequential anticoagulation strategy significantly reduced extracorporeal circulation clotting events and prolonged the dialysis time without serious adverse events.Xiaoyan TANG Dezheng CHEN Ling ZHANG Ping FU Yanxia CHEN Zhou XIAO Xiangcheng XIAO Weisheng PENG Li CHENG Yanmin ZHANG Hongbo LI Kehui LI Bizhen GOU Xin WU Qian YU Lijun JIAN Zaizhi ZHU Yu WEN Cheng LIU Hen XUE Hongyu ZHANG Xin HE Bin YAN Liping ZHONG Bin HUANG Mingying MAO 2022Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,11:4
16Formation Mechanism and Binding Energy for Body-Centred Regular Icosahedral Structure of Li_(13) Cluster显示文摘The formation mechanism for the body-centred regular icosahedral structure of Li_(13)cluster is proposed.The curve of the total energy versus the separation R between the nucleus at the centre and nuclei at the apexes for thisstructure of Li_(13)has been calculated by using the method of Gou's modified arrangement channel quantum mechanics(MACQM).The result shows that the curve has a minimal energy of-96.951 39 a.u.at R=5.46a_0.When R approachesto infinity,the total energy of thirteen lithium atoms has the value of-96.564 38 a.u.So the binding energy of Li_(13)with respect to thirteen lithium atoms is 0.387 01 a.u.Therefore the binding energy per atom for Li_(13)is 0.029 77 a.u.or 0.810 eV,which is greater than the binding energy per atom of 0.453 eV for Li_2,0.494 eV for Li_3,0.7878 eV for Li_4,0.632 eV for Li_5,and 0.674 eV for Li_7 calculated by us previously.This means that the Li_(13)cluster may be formedstably in a body-centred regular icosahedral structure with a greater binding energy.LIU Wei-Na LI Ping GOU Qing-Quan ZHAO Yan-Ping 2008Communications in Theoretical Physics2008,50,11:2
17A digital open-loop Doppler processing prototype for deep-space navigation显示文摘A prototype based on digital radio technology with associated open-loop Doppler signal processing techniques has been developed to measure a spacecraft's line-of-sight velocity. The prototype was tested in China's Chang'E-1 lunar mission relying on S-band telemetry signals transmitted by the sat-ellite,with results showing that the residuals had a RMS value of ~3 mm/s (1σ ) using 1-sec integration,which is consistent with the Chinese conventional USB (Unified S-Band) tracking system. Such preci-sion is mainly limited by the short-term stability of the atomic (e.g. rubidium) clock at the uplink ground station. It can also be improved with proper calibration to remove some effects of the transmission media (such as solar plasma,troposphere and ionosphere),and a longer integration time (e.g. down to 0.56 mm/s at 34 seconds) allowed by the spacecraft dynamics. The tracking accuracy can also be in-creased with differential methods that may effectively remove most of the long-term drifts and some of the short-term uncertainties of the uplink atomic clock,thereby further reducing the residuals to the 1 mm/s level. Our experimental tracking data have been used in orbit determination for Chang'E-1,while other applications (such as the upcoming YH-1 Mars orbiter) based on open-loop Doppler tracking will be initiated in the future.
